Isolation, Culture and Electroporation of Rice Protoplasts (벼 원형질체의 분리, 배양 및 Electroporation에 관한 연구)

  • Culture of embryogenic callus and suspension were induced from rice seeds in MS2.5 medium. In hormone free N6 medium, whole plantlets were regenerated from embryogenic callus. We observed cell division and reformation of embryogenic callus on culture of protoplast isolated from embryogenic cell suspensions. In addition, we studied the influencing factors on viability of protoplast treated with electroporation. Viability was decreased according to the increase of voltage and capacitance during electroporation. An optimal level of viability was obtained after treatment with $200-300\;V/1180\;\mu\textrm{F}$ in HEM buffer at $4^{\circ}C$..

  • Ultrathin sections of intact mycelia, released protoplast and fused protoplast of Micromonospora rosaria were observed by electron microscopy Intact mycelia showed a typical gram-positive bacterial cell wall structure and mesosomes. Released protoplasts had no cell wall components and fibrous nuclear region was distinguished from cytoplsmic region clearly. Protoplasts which treated with sucrose supplemented buffer were stable. But those treated with buffer without sucrose were extensively damaged, forming mom braneous vesicles. It was surmised that those vesicles originated from the damaged cytoplasmic membrane. High frequency of fusion was achieved by 50%(w/v). polyethylene-glycol 1,000 Fusion bodies in different stage of fusion were observed. Cell membrane barrier was stepwise relieved.

  • The cell of Kluyveromyces fragilis yeast, which is worthy of an algal substitute, was disrupted by a chemical treatment to increase the digestion of filter-feeders that yeasts are fed to. The optimum conditions of the chemical treatment were obtained by incubating yeasts at 3$0^{\circ}C$ for one hour after treated by 1 M of Na$_{2}$-EDTA that was dissolved in 0.2 M of Tris-buffer and by 0.3 m of 2-mercaptoethanol. The percentage of protop[last production was about 30%. The percentage could be doubled by the pretreatment of three times of 30 seconds sonication.

  • B-ISDN has many advantages to provide multimedia, image transfer, etc. Recently, ABR service has been proposed in order to satisfy user's various requirements in it. In this paper, we propose a new ABR flow control algorithm called CAPRO. This algorithm uses the buffer proportional to propagation delay and controls the traffic on a link-by-link basis in order to minimize the effect of propagation delay. In order to use buffer more efficienctly, we define the request cell and the control cell. Then, we analyze our algorithm using mathematical model, simulate it using SLAM system, and compare to the existing EPRCA. As a result, our algorithm has the benifit of the throughput, cell loss probability, and fairness.

Development of Parts Sequencing Rule in a Two-machine Robotic Cell

  • This paper suggests a new heuristic to improve robotic cells efficiency with a finite buffer. An efficient heuristic for parts sequencing with this configuration is developed. Analyzing robot\\`s movements and defining the events for the completion times of robot\\`s segmental activities enable us to develop a mathematical model that can be used to estimate, the completion time in the robotic cell. The robotic cell is consisting of two computer numerical control machines and a finite buffer to manufacture multiple parts. The developed heuristic can be used to estimate an optimum or near optimum parts sequences for this configuration. Numerical examples are followed to show the validity of the heuristic.

Quantitative Analysis of Indomethacin by the Portable Near-Infrared (NIR) System (근적외분광분석법을 이용한 인도메타신의 정량분석)

  • Near-infrared (NIR) system was used to determine rapidly and simply indomethacin in buffer solution for a dissolution test of tablets and capsules. Indomethacin standards were prepared ranging from 10 to 50 ppm using the mixture of phosphate buffer (pH 7.2) and water (1 : 4). The near-infrared (NIR) transmittance spectra of indomethacin standard solutions were collected by using a quartz cell in 1 mm and 2 mm pathlength. Partial least square regression (PLSR) was explored to develop calibration models over the spectral range 1100∼1700 nm. The model using 1 mm quartz cell was better than that using 2 mm quartz cell. The PLSR models developed gave standard error of prediction (SEP) of 0.858 ppm. In order to validate the developed calibration model, routine analysis was performed using another standard solutions. The NIR routine analysis showed good correlation with actual values. Standard error of prediction (SEP) is 1.414 ppm for 7 indomethacin samples in routine analysis and its error was permeable in the regulation of Korean Pharmacopoeia (VII). These results show the potential use of the real time monitoring for indomethacin during a dissolution test.

Characteristics of organic electroluminescent devices using conducting polymer materials with buffer layers (전도성 고분자를 Buffer층으로 사용한 유기 발광 소자의 제작과 특성 연구)

  • Electroluminescent(EL) devices based on organic thin films have attracted lots of interests in large-area light-emitting display. One of the problems of such device is a lifetime, where a degradation of the cell is possibly due to an organic layer's thickness, morphology and interface with electrode. In this study, light-emitting organic electroluminescent devices were fabricated using Alq$_3$(8-hydroxyquinolinate aluminum) and TPD(N,N'-diphenyl-N,N'-bis(3-methylphenyl)-[1-1'-biphenyl]-4,4'-diamine).Where Alq$_3$ is an electron-transport and emissive layer, TPD is a hole-transport layer. The cell structure is ITO/TPD/Alq$_3$/Al and the cell is fabricated by vacuum evaporation method. In a measurement of current-voltage characteristics, we obtained a turn-on voltage at about 9 V. And we used other buffer layer of PPy(Polypyrrole) with ITO/PPy/TPD/Alq$_3$/Al structure. We observed a surface morphology by AFM(Atomic Force Microscopy), UV/visible absorption spectrum, and PL(Photoluminescence) spectrum. We obtained the UV/visible absorption peak at 358nm in TPD and at 359nm in Alq$_3$, and at 225nm and the PL peaks at 410nm in TPD and at 510nm in Alq$_3$ and at 350nm. We also studied EL spectrum in the cell structure of ITO/TPD/Alq$_3$/Al and ITO/PPy/TPD/Alq$_3$/Al and we observed the EL spectrum peak at 510nm from our cell

Implementation of a Shared Buffer ATM Switch Embedded Scalable Pipelined Buffer Memory (가변형 파이프라인방식 메모리를 내장한 공유버퍼 ATM 스위치의 구현)

  • This paper illustrates the implementation of a scalable shared buffer asynchronous transfer mode (ATM) switch. The designed shared buffer ATM switch has a shared buffet of a pipelined memory which has the access time of 4 ns. The high-speed buffer access time supports a possibility of the implementation of a shared buffer ATM switch which has a large switching capacity. The designed switch architecture provides flexible switching performance and port size scalability with the independence of queue address control from buffer memory control. The switch size and the buffer size of the designed ATM switch can be reconfigured without serious circuit redesign. The designed prototype chip has a shared buffer of 128-cell and 4 ${\times}$ 4 switch size. It is integrated in 0.6um, double-metal, and single-poly CMOS technology. It has 80MHz operating frequency and supports 640Mbps per port.

  • Asynchronous transfer mode (ATM) provides the means to support with variety of multimedia serivices in broadband network. For the support of multimedia services, various data traffic with priortes should be controlled errectively and congestion function is required in the network to carry out the control operation. To accomplish this in a flexible and effective manner, priorty classes for the different services are commonly used. In this paper, we suggested a buffer managment algorithm in ATM network to improve overall network performance with separated buffer which consists of existing buffer with threshold andauxiliary buffer whose input consists of superposition of voice and data traffic. To evaluate the proposed buffer managment algorithms, simulations are excuted with two proiorities and the results are provied that network throughput is improved better than the existing parital buffer method.

  • 기존의 박막 실리콘 태양전지는 TCO와 p-layer 사이의 Bandgap차이가 p-layer, i-layer, n-layer 사이의 Bandgap 차이보다 커서 TCO를 통과한 태양광이 p-layer에 흡수되기 전에 일정량 손실된다. 이를 해결하기 위하여, p-layer 위에 기존의 p-layer보다 높은 Bandgap을 갖는 p buffer layer가 추가된 박막 실리콘 태양전지 구조를 만들어서 흡수되는 태양광의 손실량을 줄이고, 변환효율을 높이고자 하였다. 실험은 ASA Simulator를 이용하여 진행하였으며, Simulation결과 1.92eV의 Bandgap을 갖는 p buffer layer의 추가로 인하여, 기존 10.64%에서 11.16%로 증가된 변환효율을 얻을 수 있었다. Bandgap뿐만 아니라 다른 요소의 최적화도 이루어진다면, 기존의 박막 실리콘 태양전지보다 훨씬 높은 변환효율을 갖는 박막 실리콘 태양전지를 설계 하는 것이 가능 할 것이다.
