The expression and localization of V-ATPase and cytokeratin 5 during postnatal development of the pig epididymis

  • Objective: We examined the localization and expression of H+ pumping vacuolar ATPase (V-ATPase) and cytokeratin 5 (KRT5) in the epididymis of pigs, expressed in clear and basal cells, respectively, during postnatal development. Methods: Epididymides were obtained from pigs at 1, 7, 21, 60, 120, and 180 days of age; we observed the localization and expression patterns of V-ATPase and KRT5 in the different regions of these organs, namely, the caput, corpus, and cauda. The differentiation of epididymal epithelial cells was determined by immunofluorescence labeling using cell-type-specific markers and observed using confocal microscopy. Results: At postnatal day 5 (PND5), the localization of clear cells commenced migration from the cauda toward the caput. Although at PND120, goblet-shaped clear cells were detected along the entire length of the epididymis, those labeled for V-ATPase had disappeared from the corpus to cauda and were maintained only in the caput epididymis in adult pigs. In contrast, whereas basal cells labeled for KRT5 were only present in the vas deferens at birth, they were detected in all regions of the epididymis at PND60. These cells were localized at the base of the epithelium; however, no basal cells characterized by luminally extending cell projections were observed in any of the adult epididymides examined. Conclusion: The differentiation of clear and basal cells progressively initiates in a retrograde manner from the cauda to the caput epididymis. The cell-type-specific distribution and localization of the epithelial cells play important roles in establishing a unique luminal environment for sperm maturation and storage in the pig epididymis.

  • Several types of the epithelial cells were classified by ultrastructural observation through transmission electron microscope in the rat epididymis. Ultrastructural studies showed that the principal cells, basal cells and narrow cells are located in all the regions of the epididymis and the light cells are present only in the corpus and cauda epididymis. It was observed that the columnar epithelial cells like the principal cells, light cells and narrow cells contain several secretory vesicles and there are halo cells migrating in the several regions. The basal cells showed the elliptical forms in the caput and corpus region and the global forms in the cauda region.

  • The spermatozoa become mature in the epididymis which is divided into initial segment and caput, corpus, and cauda epididymis. The water movement across the epididymal epithelium is important for creating luminal microenvironment for sperm maturation. Aquaporins (Aqps) are water channel proteins, and expression of Aqps is regulated by androgens. The current research was focused to examine expressional regulation of Aqp1 and Aqp9 by an androgenic-anabolic steroid, nandrolone decanoate (ND). The ND at the low dose (2 mg/kg body weight/week) or high dose (10 mg) was subcutaneously administrated into male rats for 2 or 12 weeks. Transcript levels of Aqp1 and Aqp9 were determined by quantitative real-time polymerase chain reaction (PCR) analyses. In the initial segment, level of Aqp1 was decreased with 12 week-treatment, while Aqp9 level was decreased by the high dose treatment for 12 weeks. In the caput epididymis, Aqp9 expression was decreased by the low dose treatment. The 2 week-treatment resulted in an increase of Aqp1 level but a decrease of Aqp9 expression in the corpus epididymis. In the corpus epididymis, the 12 week-treatment at the low dose caused the reduction of Aqp1 and Aqp9 levels, but the high dose treatment resulted in an increase of Aqp1 expression and a decrease of Aqp9 level. In the cauda epididymis, Aqp1 expression was decreased by 2 and 12 week-treatments, while increases of Aqp9 levels was detected with the high dose treatment for 2 weeks and with 12 week-treatment. These findings indicate differential regulation of Aqp1 and Aqp9 expression among epididymal segments by ND.

Expression of selenium-independent glutathione peroxidase 5 (GPx5) in the epididymis of Small Tail Han sheep

  • Objective: Selenium-independent glutathione peroxidase (GPx5) is specifically expressed in the mammalian epididymis and plays an important role in protecting sperm from reactive oxygen species and lipid peroxidation damage. This study investigates GPx5 expression in the epididymis of Small Tail Han sheep. Methods: GPx5 expression was studied in three age groups: lamb (2 to 3 months), young (8 to 10 months), and adult (18 to 24 months). The epididymis of each age group divided into caput, corpus and cauda, respectively. Analysis the expression quantity of GPx5 in epididymis and testis by real-time fluorescent quantitative polymerase chain reaction and Western blot. Finally, GPx5 protein locating in the epididymis by immunohistochemical. Results: The results demonstrate that in the lamb group, the GPx5 mRNA, but not protein, can be detected. GPx5 mRNA and expressed protein were detected in both the young and adult groups. Moreover, both the mRNA and protein levels of GPx5 were significantly higher in the young group than in other two groups. When the different segments of epididymis were investigated, GPx5 mRNA was expressed in each segment of epididymis regardless of age. Additionally, the mRNA level in the caput was significantly higher than that in corpus and cauda within same age group. The GPx5 protein was in the epithelial cells' cytoplasm. However, GPx5 mRNA and protein were not detected in the testis. Conclusion: These results suggest that GPx5 is mainly expressed in the epididymis of Small Tail Han sheep, and that the expression level of GPx5 is associated with age. Additionally, GPx5 was primarily expressed in the epithelial cells of the caput. Taken together, these studies indicate that GPx5 is expressed in the epididymis in all age grades.

Effects of MethyIxanthine Derivatives on Induction and Enhancement of Hamster Epididymal Sperm Motility (Hamster 정소상체 정자의 운동성 유도와 증가에 영향을 미치는 Methylxanthine Derivatives 의 효과)

  • This study were performed the effect of methylxanthine derivatives on hamster epididymal sperm motility. To assess the effect of methylxanthine derivatives on motility kinematics of epididymal sperm, pentoxifylline, 2-deoxy-adenosine and hypoxanthine were added to TALP medium. 1 mM of pentoxifylline, significantly increased VCL, VAP, VSL of spermatozoa obtained from corpus and cauda epididymis. With 1 mM of 2-deoxyadenosine, VCL, VAP, VSL of spermatozoa obtained from corpus and cauda epididymis were significantly increased, but 2 mM ADE for cauda spermatozoa was effective than 1 mM. In the case of hypoxanthine, various concentrations were not significantly effective, but 2 mM HX showed higher effect than other concentrations. pentoxifylline 1 mM and 2-deoxyadenosine 1 mM significantly increased VCL, VAP of corpus and cauda epididymal spermatozoa and VSL of cauda epididymal spermatozoa. From these results it could be concluded that the addition of methylxanthine increase motility kinematics of hamster spermatozoa collected from epididymis.

Blood-Testis Barrier and Sperm Delayed in the Cauda Epididymis of the Reproductively Regressed Syrian Hamsters

  • The Syrian (golden) hamsters are seasonal breeders whose reproductive functions are active in summer and inactive in winter. In experimental facility mimicking winter climate, short photoperiod (SP) induces gonadal regression. The blood-testis barrier (BTB) of the sexually involuted animals have been reported to be permeable, allowing developing germ cells to be engulfed or sloughed off the epithelium of the seminiferous tubules. The expressions of genes related to the tight junction composing of BTB were investigated in the reproductive active and inactive testes. Claudin-11, occludin, and junctional adhesion molecule (JAM) were definitely expressed in the active testes but not discernably detected in the inactive testes. And spermatozoa (sperm) were observed in the whole lengths of epididymides in the active testes. They were witnessed in only cauda region of the epididymides but not in caput and corpus regions in animals with the inactive testes. The results imply that the disorganization of BTB is associated with the testicular regression. The developing germ cells are swallowed into the Sertoli cells or travel into the lumen, as supported by the presence of the sperm delayed in the last region of the epididymis. These outcomes suggest that both apoptosis and desquamation are the processes that eliminate the germ cells during the regressing stage in the Syrian hamsters.

Regional Difference in Distribution of Glycoconjugates in Mouse Epididymis (생쥐 부정소 부위별 당쇄 분포의 차이)

  • To characterize the difference in glycoconjugates of mouse epididymis, lectin labeling of the tissue section was conducted using Ulex europaeus agglutinin I(UEA I), succinylated wheat germ agglutinin(sWGA), and Griffonia simplicifolia lectin-I(GSL-I). UEA I which binds to outer $\alpha$-L-fucose residue that is a terminal sugar of the side chain branched from oligosaccharide chain gave the labeling in the proximal caput epithelia exclusively. Lumen was commonly labeled in all of the organ. It suggested that the glycoconjugates bearing outer $\alpha$ -L-fucose residue were largely expressed in the initial segments ot epididymis and subjected to secretion. GSL-I which binds to terminal $\alpha$ -D-galactosyl residue of glycoconjugates gave the labeling in the cytoplasm of clear cells and basal cells, and cilia in corpus and cauda regions but not in the caput region. There was no vast difference in labeling pattern by sWGA which binds to N-acetyl-glucosamine residue among the epididymal regions. Clear cells in corpus and cauda epithelia showed more intense labeling by sWGA compared to principal cells, suggesting the functional specialization of this type of cells. The labeling intensities of luminal content by UEA I and sWGA decreased in cauda region compared to corpus region suggesting the presence of enzymatic activities responsible for processing the $\alpha$-L-fucose and N-acetyl-glucosamine residues from secreted glycoconjugates. In summary, the difference in glycoconjugates bearing the $\alpha$-L-fucose, $\alpha$-D-galactose, and N-acetyl-glucosamine residues according to the type of epithelial cells and epididymal segments suggests functional specialization and different roles of each segment in the processing of sperm surface antigens during the epididymal transit.

Dose dependent effect of benzene extract of Ocimum sanctum leaves on cauda epididymal spermatozoa of albino rats

  • An attempt has been made to assess whether the dose dependent effect of benzene extract of Ocimum sanctum leaves on the morphological changes in the cauda epididymal spermatozoa and sperm parameters in male albino rats. Scanning Electron Microscope observations illustrate the disturbance in plasma membrane as well as acrosomal membrane. Most of the sperms appear morphologically abnormal in the mid region of the tail; there is formation of balloon like cytoplasmic droplet. Sperm parametric study exhibits decrease in the total sperm count, sperm motility, forward velocity and increase in the percentage of abnormal sperms in dose dependent manner on treatment benzene extract of Ocimum sanctum leaves. The results suggest that the effects may have resulted from a general disturbance in the proteins and alteration in cauda epididymal milieu probably due to androgen deficiency consequent upon antiandrogenic property of Ocimum sanctum leaves.

Ultrastructural Changes of Epididymal Epitheliurn during Sexual Maturation in Mouse (성적 성숙에 따른 생쥐 부정소 상피세포의 미세구조 변화)

  • The ultrastructure of epididymal epithelium of 10, 20, 35 and 80 day-old mouse was observed to study the differentiation and function of the epithelial cells in connection with the absorption and secretion during sexual matruation. The differentiation of epididymis was divided into three phases of, 1) undiflerentiated phase until the day 20 after birth, 2) growing and differentiating phase between the day 20 and 35, and 3) maturating phase up to the adult. Each phase was closely related with the lumination of seminiferous tubule and the influx of spermatozoa within testicular fluid from testis. In adult, the ultrastructural features appeared an absorptive function in the principal cells of proximal caput epididymis, and a strong activity of protein synthesis and secretion in distal caput, corpus and cauda epididymis. Clear cells were predominantly located in corpus and cauda epidiymis, and plenty of absorption vesicles including membranous particles assumed to be the cellular residues from spermatozoa were observed at apical region. Therefore, the distribution of various cell types of epithelium and the ultrastructure even in the same type of epithelial cell, were different according to the epididymal regions.

Effects of Triladyl-egg Yolk Diluents on the Viability of Frozen Korean Black-goat Spermatozoa from Cauda Epididymis and Electro-ejaculated Semen (Triladyl-난황 희석제가 한국 재래 흑염소의 정소상체 및 전기자극 유래 정자의 융해 후 생존성에 미치는 영향)

  • To preserve genetic materials, cryopreservation of the semen from live animals is the main technique to establish cryo-banking system which could be used for artificial insemination and embryo transfer. However, the population of Korean black goat (KBG) becomes to dwindle in number and is now faced genetic erosion by crossbreeding with non-native breeds in small KBG farms. In this study, simple freezing method was used to preserve frozen semen from KBG using spermatozoa of cauda epididymis (CE) and electro-stimulated semen (ES). The negative effects of seminal plasma on fresh sperm was confirmed using precipitation test of Triladyl egg yolk diluent and sperm viability after thawing was compared between CE and ES spermatozoa. When seminal plasma of fresh ES semen was washed with semen washing media (SWM), the rates of live sperm shown no significant difference between CE and ES spermatozoa before freezing. However, the survival rate of frozen/thawed CE sperm was higher than ES ($74.6{\pm}10.6%$ vs $53.8{\pm}5.2%$) with significant difference (p < 0.05). The results of longevity test on frozen/thawed sperm from CE showed healthier sperm than ES. Therefore, spermatozoa from CE could be used for cryo-banking system in KBG lines. The more studies are needed to increase survival rate of ES semen.
