Appropriateness Evaluation of Plug Seedling Cultivation for Replanting of Rehmannia glutinosa (Gaertn.) Libosch. ex Steud (보식용 지황의 플러그 육묘 재배를 위한 적정성 평가)

  • Background: Rehmannia glutinosa is a perennial herb belonging to the family Scrophulariaceae and is used in traditional oriental medicine. This study was conducted to evaluate the appropriateness of plug-seedling cultivation for replanting of R. glutinosa. Methods and Results: The seed rhizomes of Rehmannia glutinosa 'Jihwang 1' were harvested in the middle of March, 2018. Direct seedling cultivation was conducted after air curing for 3 days. The seedlings for transplanting were cultivated for 30 (30D), 45 (45D), and 60 days (60D). The mat formation rates of the 30D, 45D and 60D treatment seedling were 81.3%, 100%, and 100%, respectively. Of the tree treatment groups, flowering occurred only in the 45D and 60D treatment groups, and their flowering rates were $7.3{\pm}3.06%$ and $13.3{\pm}1.15%$, respectively. The agronomic characteristics and active components of the three treatment groups were evaluated in comparison with the DS1 (directly seeded group, control) and DS2 (directly seeded group, replanting) groups. As a result, the leaf length and leaf width of the 30D seedlings were higher than other treatment groups, but the number of leaves per plant was statistically the same. In addition, root diameter and yield of the 30D seedling were statistically the same as DS1. Moreover the catalpol contents of 30D, 45D, DS1 and DS2 group seedlings were 2.23%, 1.86%, 2.50% and 2.08%, respectively. Conclusions: Taken together, these findings suggest it is feasible to use the plug-seedling cultivation method for replanting of R. glutinosa. seedlings. However, further evaluation of the economic aspect is needed.

Characteristic Effects of Dangnyo-hwan for Diabetes Control Studied Using LC-MS/MS and ICP (LC-MS/MS 및 ICP를 이용한 당뇨환의 화학적 특성 분석)

  • Objectives: The primary aim of this study was to identify bioactive compounds in Dangnyo-hwan, a Korean herbal medicine, through instrumental analysis using LC-MS/MS and ICP, and investigate its potential use in diabetes treatment. Methods: The extract of Dangnyo-hwan has 12 medicinal herbs; these were compared with 18 marker substances selected from literature survey. Results: LC-MS/MS analysis could detect 9 of the 18 bioactive compounds: citruline, catalpol, berberine, ginsenoside Rb1, ginsenoside Rg1, oleanolic acid, β-sitosterol, mangiferin, and schizandrin. While harmful heavy metals such as As, Pb, Cd, Hg, Ni, and Cu were not present in high concentrations, Zn concentration was 4.2 mg in 100 g Dangnyo-hwan. Conclusions: Instrumental analysis such as LC-MS/MS and ICP was successfully used to identify bioactive compounds in Dangnyo-hwan. Detection of 9 bioactive substances and Zn from the herb medicine is a valuable finding, and suggests that Dangnyo-hwan is a candidate medicine for diabetes. Further investigations like in vitro assay, percent GPR 119 activity, and percent human DGAT-1 inhibition are underway.

Effects of Iridoid Compounds on RNA and Protein Biosyntheses in Sarcoma 180 cells (Iridoid Compounds가 RNA 및 Protein 생합성에 미치는 영향)

  • To investigate a possible biological activity of iridoid glucosides, six compounds, aucubin, catalpol, gardenoside, geniposide, rehmannioside and swertiamarin, were studied in relation with their potential influences in RNA and protein biosyntheses in murine tumor cell, sarcoma 180, in vitro. Protein biosynthesis was slightly inhibited by aucubin, gardenoside and swertiamarin. Degree of inhibition of RNA biosynthesis by those iridoid appeared to be more sensitive than that of protein biosynthesis. When aucubin was pretreated with ${\beta}-glucosidase$ to produce its genin form and the sarcoma 180 cells were exposed to this aucubigenin, the protein and RNA biosyntheses in the cells were profoundly inhibited. The results indicate that a biologically active from of iridoid compounds is the hydrolytic products of glycoside, i.e. genin form. It is also suggested that sarcoma 180 cells used in the experiments appear to lack of ${\beta}-glucosidase$, since the inhibitory actions of iridoid glucosides were so slight that those glucosides were not hydrolysed by the enzyme to their genin forms.

  • Twenty-four compounds were isolated from the 70% ethanolic extract of Rehmanniae Radix (Scrophulariaceae) cultivated in Ubo-myeon, Gunwi-gun, Gyeongbuk province, Korea and their structures were identified as four iridoids [6-O-(4''-O-${\alpha}$-L-rhamnopyranosyl) vanilloyl ajugol (17), ajugol (18), aucubin (19), and catalpol (20)], three phenethyl alcohol glycosides [decaffeoyl acteoside (15), isoacteoside (16), and acteoside (21)], five sugar derivatives [ethyl ${\beta}$-D-fructofuranoside (7), eleutheroside C (14), mannitol (22), raffinose (23), and stachyose (24)], two terpenoids [remophilanetriol (4) and glutinolic acid (11)], a lignan, paulownin (2), and eight others [${\beta}$-sitosterol (1), daucosterol (6), monopalmitin (3), pinellic acid (9), uracil (5), adenosine (12), jio-cerebroside (10), aeginetic acid 5-O-${\beta}$-D-quinovoside (8), aeginetoyl ajugol 5''-O-${\beta}$-D-quinovoside (13)]. The chemical structures of these compounds were identified on the basis of spectroscopic methods and comparison with literature values. Among these compounds, paulownin (2), monopalmitin (3), uracil (5), daucosterol (6), ethyl ${\beta}$-D-fructo-furanoside (7), and eleutheroside C (14) were isolated from this plant for the first time.

The chemical structures of these compounds were identified on the basis of spectroscopic methods and comparison with literature values. This is the first report of the triterpenoids oleanolic acid (1), pomonic acid (2), and ursolic acid (5) from the genus Rehmannia, as well as the first report of compounds 5-hydroxymethyl-2-furaldehyde acetate (3), 2,5-dihydroxyacetophenone (4), daucosterol (9), darendoside B (14), 5-(${\alpha}$-D-galactopyranosyloxymethyl)-2-furancarboxaldehyde (15), adenosine (16), phenethyl alcohol 2-O-${\beta}$-D-xylopyranosyl(1${\rightarrow}$6)-${\beta}$-D-glucopyranoside (17), and salidroside (19) from the Rehmanniae Radix Preparata.

Comparison of Physicochemical Properties on Dried Rehmannia glutinosa with Different Cultivars (품종에 따른 건지황의 이화학적 특성 연구)

  • Background: This study was conducted to compare the physicochemical properties of 12 different cultivars of Rehmannia glutinosa, a plant used in Korean traditional herbal medicine. Method and Results: We investigated the following 12 cultivars of R. glutinosa; Jihwang1, Goryeo, Daegyung, Gogang, Togang, Dagang, Wongang, Yeongang, Hwanggang, Dahwang, Segang and Chunggang. We measured water content, chromaticity, hardness, active components levels, and antioxidant activity. Our results showed that Togang has the lowest water content and the highest yield. The catalpol content was the highest in Togang (40.50 mg/g) and the lowest in Chunggang (22.17 mg/g). The aucubin content was the highest in Hwanggang (0.28 mg/g). Chunggang had the lowest aucubin content, but the highest level of verbascoside. Total flavonoid and total polyphenol were the highest in Daegyung and Chunggang, respectively. There was a strong correlation between the total flavonoid and total polyphenol (r = 0.89) content. Conclusions: There were significant differences in physicochemical properties among 12 cultivars. These properties will affect suitability for processing and final product quality. We therefore suggest the selection of appropriate cultivars for different uses.

Changes of Chemical Constituents of Rehmannia Radix during 'Steaming and Drying' Process (증폭(蒸曝)에 의한 지황(地黃)의 성분 변화에 대한 고찰)

  • Objectives : 'Steaming and drying' is a traditional processing method that has been used to produce Suk-ji-hwang (熟地黃; Rehmanniae Radix Preparata) from Ji-hwang (地黃, the fresh root of Rehmannia glutinosa Liboschitz ex Steudel; Rehmanniae Radix). The steaming and drying process, which is proceeded in heating and moisturizing conditions, plays a crucial role in the change of therapeutic effect of Ji-hwang, presumably due to the modification of its chemical constituents. In this article, the chemical influence of the 'Steaming and drying' process was investigated for understanding the underlying mechanism of chemical modification of Ji-hwang. Methods : The articles regarding the modifications of chemical constituents of Ji-hwang during the 'Steaming and drying' process were collected and analyzed to investigate the influence of the processing to Ji-hwang. Results : The results indicated that iridoid glycosides were degraded to their aglycones and sugars, and such degradations occurred faster at a high pressure than at an atmospheric pressure during the process. The contents of catalpol, ajugol, and acteoside were decreased, while those of rehmannioside A and D were slightly increased during the repeated processing. The contents of oligosaccharides, namely sucrose, maltose, raffinose, and stachyose (except for manninotriose), were decreased, while those of monosaccharides, glucose and fructose, were increased by the repeated processing. Conclusions : These results demonstrate that the 'Steaming and drying' process influenced the chemical constituents of Ji-hwang and provide probable basis for the therapeutic modification of Suk-ji-hwang after the processing of Ji-hwang.

Characteristics and Antioxidant Activities of Rehmanniae radix Powder (지황 및 숙지황 분말의 품질 및 항산화 특성)

  • This study was carried out to investigate the quality characteristics and antioxidant activities of Rehmanniae radix (Rehmannia glutinosa Libosch. var. purpurea Makino) freeze-dried powder (RRP) and Rehmanniae radix preparata powder (RRPP). Under the Hunter color system, redness was higher and lightness/yellowness lower in RRPP, compared to RRP. The reducing sugar contents of RRP and RRPP were 0.8% and 6.0%, respectively (p<0.05). The pH was lower in the RRPP (RRP: 6.71, RRPP: 4.23). The amount of catalpol amount in RRPP (47.20 mg/mL) was lower than RRP (144.90 mg/mL). RRPP contained high amounts of 5-HMF (5-hydroxymethyl-2-furaldehyde, 47,231 mg/mL), but 5-HMF was not detected in RRP. Total phenol contents of RRP and RRPP were 2.10 mg/mL and 3.66 mg/mL, respectively. FRAP values of RRP and RRPP were 0.51 mg/mL and 1.99 mg/mL, respectively. The antioxidant activities by DPPH (1,1-diphenyl-2-picryl hydrazyl) and hydroxyl radical scavenging activity of RRPP were much higher than RRP. Based on these results, RRPP is a good candidate for food processing in terms for its physicochemical and antioxidative activities.

  • This study was conducted to evaluate the appropriateness cultivation of R. glutinosa by plug seedling. Cultivation by direct seeding (DS) and plug seedling (PS) of R. glutinosa was conducted in each of the Pyeongchang and Eumseong areas. As a result, locally, for the Eumseong area, aerial parts in leaf length, leaf width and number of leaves per plant were better than for the Pyeongchang area. In the cultivation method, leaf width and number of leaves per plant were better with PS cultivation than with DS cultivation. Locally, rhizome yield of underground parts in the Eumseong area was better than that in the Pyeongchang area, but the rhizome yield with DS cultivation in the Eumseong area and PS cultivation in the Pyeongchang area were statistically the same. The fresh rhizome yield for DS in Eumseong, PS in Eumseong, DS in Pyeongchang and PS in Pyeongchang was 1,253 kg/10a, 1,376 kg/10a, 923 kg/10a and 1,256 kg/10a, respectively. Finally, the catalpol content for DS in Eumseong, PS in Eumseong, DS in Pyeongchang and PS in Pyeongchang was 3.67%, 2.03%, 2.96% and 1.60%, respectively. From these results, it can be seen that the R. glutinosa PS cultivation method can increase the rhizome yield.

Quality Characteristics of Prepared Rehmannia Root with Four Domestic Cultivars (국내 육성 품종별 숙지황의 품질 특성)

  • Rehmannia glutinosa, one of the major medicinal crops in Korea, can be classified into three types: fresh, dried and prepared Rehmannia root. In this study, the quality characteristics of prepared rehmannia root were evaluated using four different cultivars that are commonly used in the market. In making prepared rehmannia root, roots of Jihwang 1, Kokang, Togang, and Dagang were dried, soaked in rice wine, and steamed nine times. At each stage, physiochemical properties were analyzed, including yield, which is one of the most important industrial factors to consider. The yield was the highest in Togang at 23.61% and the lowest in Dagang at 21.16%. These yield values showed a highly negative correlation with the moisture content of roots. The fructose and glucose contents were increased during the 3rd, 4th and 5th steaming but then decreased. The sucrose, raffinose, and stachyose content gradually decreased during the first three steaming and were not detected during the 4th steaming. Additionally, the catalpol content was not detected after the 4th steaming. On the contrary, the 5-hydroxymethylfurfural content was not detected in the raw root but increased during the steaming. Jihwang1 and Togang exceeded the 0.1% Korean Pharmacopoeia standard after the 5th steaming, reaching it faster than did the other cultivars. Overall, Togang was the optimal cultivar considering the overall characteristics of its high yield and short steaming time. These results could provide useful information for the industrial use of prepared Rehmannia root based on the requirements and characteristics of each cultivar.
