산화스트레스와 치매 (Oxidative Stress and Alzheimer's Disease)

  • Oxidative stress is a cascade reaction characterized by a significant increase in the amount of oxidized components. Free radicals produced by oxidative stress are one of the common features in several experimental models of disease, and contribute to wide range of neurodegenerative diseases, including Alzheimer's disease. Iron (II) species can participate in the Fenton, and Fenton-like reactions, to react with hydrogen peroxide and generate hydroxyl radical. As iron accumulation and oxidative stress are associated with the pathological progression of neurodegenerative diseases, iron chelation and antioxidant therapies have become strategies to combat these diseases. Due to the complexity of the redox system in vivo, a multifaceted approach may be an attractive therapeutic strategy. Further investigations are highly expected for the prevention and treatment of neurodegenerative diseases in future.

  • Dendrimers are well-defined macromolecules exhibiting a tree-like structure, first derived by the cascade molecule approach. Peculiar features of the dendritic geometry are the large number of end groups as well as the shape persistence in higher generations, approaching spherical geometry. And one of the most peculiar characteristics of dendritic macromolecules is their controlled molecular structure and orientation, which means that they have a practical application in achieving a highly organized molecular arrangement. We attempted to fabricate a dendrimer LB films containing 48 pyridinepropanol functional end group. As the pyridinepropanol functional group could form a complex structure with metal ions. We investigated the surface activity of dendrimer films at air-water interface compared with pure dendrimer and its complex with $Fe^{2+}$ ions into subphase. We though that metal ions are contributed to networking or branching reaction between dendrimers. And we expected that it can result in the differences on the electrical properties. We have studied the electrical properties of the ultra thin dendrimer LB films investigated by the current-voltage characteristics of metal dendrimer LB films/metal (MIM) structure.

Simvastatin이 메추리 근육 세포에 미치는 영향 (Simvastatin Induces Avian Muscle Protein Degradation through Muscle Atrophy Signaling)

  • Many studies on poultry have been conducted in the poultry industry to improve their important economic traits, such as egg production, meat quality, and carcass yield. Environmental changes affect the poultry's economic traits, including muscle growth. The purpose of this study is to investigate the mechanisms by which simvastatin causes muscle injury in quail muscle cells. Following treatment with various doses of simvastatin, LD50 in the quail myoblast cells was determined using a cell viability test; cell death was caused by apoptosis and/or necrosis. Thereafter, the expression patterns of the atrophy marker genes were examined via quantitative reverse-transcription polymerase chain reaction (qRT-PCR). The results showed that the transcriptional levels of the muscle atrophy marker genes (Atrogin-1, TRIM63) and the upstream genes in their signaling cascade were increased by simvastatin treatment. This indicated that simvastatin induced myogenic cell death and muscle injury via protein degradation through muscle atrophy signaling. Further studies should focus on identifying the mechanism by which simvastatin induces the protein degradation signaling pathway in quail muscle..

Bidirectional Interactions between Green Tea (GT) Polyphenols and Human Gut Bacteria

  • Green tea (GT) polyphenols undergo extensive metabolism within gastrointestinal tract (GIT), where their derivatives compounds potentially modulate the gut microbiome. This biotransformation process involves a cascade of exclusive gut microbial enzymes which chemically modify the GT polyphenols influencing both their bioactivity and bioavailability in host. Herein, we examined the in vitro interactions between 37 different human gut microbiota and the GT polyphenols. UHPLC-LTQ-Orbitrap-MS/MS analysis of the culture broth extracts unravel that genera Adlercreutzia, Eggerthella and Lactiplantibacillus plantarum KACC11451 promoted C-ring opening reaction in GT catechins. In addition, L. plantarum also hydrolyzed catechin galloyl esters to produce gallic acid and pyrogallol, and also converted flavonoid glycosides to their aglycone derivatives. Biotransformation of GT polyphenols into derivative compounds enhanced their antioxidant bioactivities in culture broth extracts. Considering the effects of GT polyphenols on specific growth rates of gut bacteria, we noted that GT polyphenols and their derivate compounds inhibited most species in phylum Actinobacteria, Bacteroides, and Firmicutes except genus Lactobacillus. The present study delineates the likely mechanisms involved in the metabolism and bioavailability of GT polyphenols upon exposure to gut microbiota. Further, widening this workflow to understand the metabolism of various other dietary polyphenols can unravel their biotransformation mechanisms and associated functions in human GIT.

  • Clonorchis sinensis is commonly found in East Asian countries. Clonorchiasis is prevalent in these countries and can lead to various clinical symptoms. In this study, we used overlap extension polymerase chain reaction (PCR) and the Xenopus laevis oocyte expression system to isolate a cDNA encoding the choline transporter of C. sinensis (CsChT). We subsequently characterized recombinant CsChT. Expression of CsChT in X. laevis oocytes enabled efficient transport of radiolabeled choline, with no detectable uptake of arginine, α-ketoglutarate, p-aminohippurate, taurocholate, and estrone sulfate. Influx and efflux experiments showed that CsChT-mediated choline uptake was time- and sodium-dependent, with no exchange properties. Concentration-dependent analyses of revealed saturable kinetics consistent with the Michaelis-Menten equation, while nonlinear regression analyses revealed a Km value of 8.3 µM and a Vmax of 61.0 pmol/oocyte/h. These findings contribute to widen our understanding of CsChT transport properties and the cascade of choline metabolisms within C. sinensis.

2D to 3D Conversion에서 Depth-Map 기반 제작 사례연구 - '명장 관우' 제작 중심으로 - (Propriety analysis of Depth-Map production methods For Depth-Map based on 20 to 3D Conversion - the Last Bladesman)

  • 입체 디스플레이 보급의 보편화가 진행됨에 따라, 입체 콘텐츠의 수요가 증가하고 있다. 이러한 증가에 맞추어 2010년도를 기점으로 2D to 3D 변환 콘텐츠가 부족한 수요를 충족시킬 대안으로 제시되었다. 그러나 입체효과만을 강조한 2D to 3D 변환콘텐츠가 생산되면서 시각적 피로도와 입체감에 대한 품질의 저하가 문제로 지적되고 있다. 본 연구에서는 2011년 개봉한 '명장 관우'에서, 13개 Scene을 선별하여 입체 변환 콘텐츠로 제작하고, 변환에 적용된 Depth-Map의 품질이 시각적 피로도와 입체감을 표현하는데 있어서, 적정성을 가지는가의 여부를 전문가 그룹을 대상으로 인터뷰 및 설문조사를 시행하였다. 움직임의 변화가 많은 영상에 적용한 Depth-Map의 구성방식은 입체변환 기술에 많이 사용되는 방법으로 전(前) 후(後)관계의 분석을 통해 계단식 구성방식으로 깊이 단계 지도를 제작하게 된다. 실험을 통하여, 본 연구에서 제시한 Depth-Map의 구성이 입체변환 콘텐츠 제작에 있어 시각적 피로도를 낮추고 입체감 향상에 타당한지에 대한 결과를 도출하였으며, 실험에 응한 전문가 그룹의 과반수이상이 긍정적인 반응을 표시하였다. 본 연구의 결과 빠른 움직임을 가지는 2D영상을 3D영상으로 변환하는데 적용한 계단식 Depth-map의 구성방식으로도 시각적 피로도를 감소시키고, 입체감 인식을 증가시키는데 효율성을 가진다는 결과를 도출하였다.

Helicobacter pylori inhibited cell proliferation in human periodontal ligament fibroblasts through the Cdc25C/CDK1/cyclinB1 signaling cascade

  • Purpose: Several studies have shown that the oral cavity is a secondary location for Helicobacter pylori colonization and that H. pylori is associated with the severity of periodontitis. This study investigated whether H. pylori had an effect on the periodontium. We established an invasion model of a standard strain of H. pylori in human periodontal ligament fibroblasts (hPDLFs), and evaluated the effects of H. pylori on cell proliferation and cell cycle progression. Methods: Different concentrations of H. pylori were used to infect hPDLFs, with 6 hours of co-culture. The multiplicity of infection in the low- and high-concentration groups was 10:1 and 100:1, respectively. The Cell Counting Kit-8 method and Ki-67 immunofluorescence were used to detect cell proliferation. Flow cytometry, quantitative real-time polymerase chain reaction, and western blots were used to detect cell cycle progression. In the high-concentration group, the invasion of H. pylori was observed by transmission electron microscopy. Results: It was found that H. pylori invaded the fibroblasts, with cytoplasmic localization. Analyses of cell proliferation and flow cytometry showed that H. pylori inhibited the proliferation of periodontal fibroblasts by causing G2 phase arrest. The inhibition of proliferation and G2 phase arrest were more obvious in the high-concentration group. In the low-concentration group, the G2 phase regulatory factors cyclin dependent kinase 1 (CDK1) and cell division cycle 25C (Cdc25C) were upregulated, while cyclin B1 was inhibited. However, in the high-concentration group, cyclin B1 was upregulated and CDK1 was inhibited. Furthermore, the deactivated states of tyrosine phosphorylation of CDK1 (CDK1-Y15) and serine phosphorylation of Cdc25C (Cdc25C-S216) were upregulated after H. pylori infection. Conclusions: In our model, H. pylori inhibited the proliferation of hPDLFs and exerted an invasive effect, causing G2 phase arrest via the Cdc25C/CDK1/cyclin B1 signaling cascade. Its inhibitory effect on proliferation was stronger in the high-concentration group.

제 I 형 알레르기 반응이 유도된 생쥐에서 식물추출 복합물(PEM381)의 효과 (Effects of Phyto-Extract Mixture (PEM381) in Type I Allergic Reaction-Induced Mice)

  • 본 연구에서는 녹차(Camellia sinensis)잎 추출물, 구아바(Psidium guajava)잎 추출물 그리고 장미 (Rosa hybrida)꽃잎 추출물로 이루어진 식물추출 복합물(PEM381)이 실험동물상에서 제 I 형 알레르기 반응에 미치는 영향을 알아보고자 제 I 형 알레르기 반응을 유발시킨 생쥐를 모델로 류코트리엔류, 프로스타글란딘류 그리고 히스타민 농도 및 호산구의 비강내 침윤 정도를 측정하였다. 그 결과 PEM381은 류코트리엔류의 생합성을 억제할 뿐만 아니라 호산구의 비강내침윤도 억제하는 것으로 나타났다. 그리고 비록 통계학적으로 유의적인 차이는 없었으나 PEM381은 히스타민 방출을 억제하는 것으로 나타났다. 그러나 PEM381에 의한 프로스타글란딘류의 생합성에 미치는 영향은 신뢰할만한 결과를 얻어내지 못했다. 이상의 결과로 미루어 PEM381은 알레르기성 비염, 아토피 피부염 및 기관지 천식 같은 제 I 형 알레르기 질환에 대하여 예방 및 개선에 유용하게 사용될 수 있을 것으로 생각된다.

식물추출 복합물(PEM381)의 제I형 알레르기 반응 억제 효과 (Inhibitory Effects of Phyto Extract Mixture (PEM381) on Type I Allergic Reaction)

  • 본 연구에서는 녹차(Camellia sinensis)잎 추출물, 구아바(Psidium guajava)잎 추출물 그리고 장미(Rosa hybrida)꽃잎 추출물로 이루어진 식물추출 복합물(PEM381)에 대하여 제I형 알레르기 반응 억제 효능 및 생화학적 기전을 밝히기 위하여 COX 억제 활성, LO 억제 활성, compound 48/80에 의한 랫드 복강 비만세포의 탈과립과 히스타민 유리에 미치는 영향 그리고 수동피부아나필락시스 반응 억제효능을 측정하였다. 그 결과 PEM381은 5-LO $IC_{50}$ 값이 $14.11{\pm}0.51ppm$으로 나타났고, COX-1에 비해 COX-2를 선택적으로 억제하는 것으로 나타났다. 또한 PEM381은 compound 48/80에 의한 비만세포의 탈과립과 히스타민 유리를 농도 의존적으로 억제할 뿐만 아니라 수동피부아나필락시스 반응도 억제하였다. 이상의 결과로 PEM381은 알레르기성 비염, 아토피 피부염 및 기관지천식 같은 제I형 알레르기 질환에 대하여 예방 및 치료에 유용하게 사용될 수 있을 것으로 생각된다.

Characterization of the MicroRNA Expression Profile of Cervical Squamous Cell Carcinoma Metastases

  • Objectives: MicroRNAs (miRNAs) are important regulators of many physiological and pathological processes, including tumorigenesis and metastasis. In this study, we sought to determine the underlying molecular mechanisms of metastatic cervical carcinoma by performing miRNA profiling. Methods: Tissue samples were collected from ten cervical squamous cancer patients who underwent hysterectomy and pelvic lymph node (PLN) dissection in our hospital, including four PLN-positive (metastatic) cases and six PLN-negative (non-metastatic) cases. A miRNA microarray platform with 1223 probes was used to determine the miRNA expression profiles of these two tissue types and case groups. MiRNAs having at least 4-fold differential expression between PLN-positive and PLN-negative cervical cancer tissues were bioinformatically analyzed for target gene prediction. MiRNAs with tumor-associated target genes were validated by quantitative reverse transcription-polymerase chain reaction (RT-PCR). Results: Thirty-nine miRNAs were differentially expressed (>4-fold) between the PLN-positive and PLN-negative groups, of which, 22 were up-regulated and 17 were down-regulated. Sixty-nine percent of the miRNAs (27/39) had tumor-associated target genes, and the expression levels of six of those (miR-126, miR-96, miR-144, miR-657, miR-490-5p, and miR-323-3p) were confirmed by quantitative (q)RT-PCR. Conclusions: Six MiRNAs with predicted tumor-associated target genes encoding proteins that are known to be involved in cell adhesion, cytoskeletal remodeling, cell proliferation, cell migration, and apoptosis were identified. These findings suggest that a panel of miRNAs may regulate multiple and various steps of the metastasis cascade by targeting metastasis-associated genes. Since these six miRNAs are predicted to target tumor-associated genes, it is likely that they contribute to the metastatic potential of cervical cancer and may aid in prognosis or molecular therapy.
