Anti-inflammatory activities of carrot(Daucus carota) leaf Fermented by Weizmannia coagulans KK7 (김치에서 분리한 Weizmannia coagulans KK7으로 발효한 당근 잎 추출물의 함염증 활성 연구)

  • In this study, the extracts of carrot (Daucus carota var. sativa) leaf fermented with Weizmannia coagulans KK7 strain were investigated for the anti-inflammatory activities and component changes. The KK7 strain was isolated from kimchi, a Korean fermented vegetable. The high-performance liquid chromatography was performed to analyze the changes in the components of the carrot leaf extracts before and after fermentation. It was confirmed that the content of luteolin, a kind of flavonoid, was significantly increased after fermentation. The anti-inflammatory activities of the carrot leaf extracts and the fermented carrot leaf extracts were evaluated by the inhibition of NO (nitric oxide) production in LPS-induced RAW 264.7 cells. The NO scavenging ability of the fermented carrot leaf extracts was higher than the other extracts. The protein expression of iNOS, an enzyme responsible for the NO production was significantly reduced in a concentration-dependent manner by treatment with the fermented carrot leaf extracts. In conclusion, we found that the anti-inflammatory effect of carrot leaf was increased by microbial fermentation, suggesting that carrot leaf generally discarded could be used as new food and cosmetic materials through fermentation.

  • Bacterial leaf blight in carrot is one of the most important diseases in the worldwide. In the past decade, its introduction into Korea is causing great concern due to the potential damage to carrot crops domestically. This bacterial disease is caused by Xanthomonas hortorum pv. carotae (Xhc). This study aimed to isolate and identify bacterial strains from the soil of carrot farms in Jeju Island. The bacterial isolates showing characteristics similar with those of Xhc were selected when cultured on artificial media. Through DNA sequencing and analysis based on NCBI data, some of the selected bacterial strains were identified as Xhc. Furthermore, the bacterial strains caused the typical symptom of bacterial leaf blight after inoculation on carrot leaves. The results of this study showed the potential establishment of Xhc in the soil of Jeju Island and it may be valuable data for establish a strategy preventing the domestic spread of carrot bacterial leaf blight in the future.

Selection of Desirable Cultivar for Organic Cultivation of Carrot (당근 유기재배를 위한 병해충 저항성 품종 선발)

  • This research was carried out to selection of desirable cultivar for organic cultivation of carrot in Korea. The assay of disease resistance, insect tolerance and rate of bolting were investigated on 32 cultivars of genetic resources of carrot. This experiment was carried out at experimental field and greenhouse of Gangneung-Wonju National University. 'PI 223360' was resistant against powdery mildew. Ten cultivars including 'Oxheart Carrot Heirloom' were moderately resistant, 22 cultivars including 'Long Impeator #58' were susceptible to powdery mildew. Although there was no resistant cultivar against black rot and leaf blight, our results showed that several cultivars have moderate resistance. Thirteen cultivars including 'Oxheart Carrot Heirloom' were having insect (Erythroplusia pyropia) tolerance and 'SA 102' and 'Scarlet Keeper Carrot Rare' were susceptible. According to the investigation of bolting, 6 cultivars including 'Hongsim Ouchon Carrot' were early bolting and 4 cultivars including 'Japanese Imperial Long Carrot' were moderate bolting. Other cultivars were identified as late bolting. From above results, we confirmed that 'Oxheart Carrot Heirloom' was suitable cultivar for organic cultivation among 32 genetic resources of carrot.

Factors Affecting Spore Formation of Carrot Leaf Blight Caused by Alternaria dauci In Vitro (배지에서 당근검은잎마름병균의 포자 형성에 영향을 미치는 요인)

  • In order to examine the pathogenicity of Alternaria dauci, the causal agent of carrot leaf blight, it is necessary to standardize sporulation conditions to ensure the optimal quantity and quality of spore inoculum. Therefore, in this study, the effects of the growth medium, aeration treatment, and UV treatment with 12-hr photoperiod on the sporulation of A. dauci KACC42997 were investigated. A. dauci KACC42997 was pre-cultured for 7 days in a potato dextrose agar medium at 25℃ in the dark condition. When the pre-culture, after removing aerial mycelia, was re-incubated for 5 days, with simultaneous aeration treatment and 12-hr cycle UV treatment at 20℃, the largest number of spores was produced. One hundred seventy isolates of A. dauci were isolated from major carrot growing regions such as Pyeongchang, Gumi and Jeju and tested for sporulation under the conditions established in this study. Except for 20 strains, all strains produced spores. Statistically significant differences in the extent of sporulation were found among local populations of A. dauci isolates, but no difference was observed in their pathogenicity on carrots.

Detection of Fungicidal Activities against Alternaria dauci Causing Alternaria Leaf Spot in Carrot and Monitoring for the Fungicide Resistance (당근검은잎마름병균 Alternaria dauci에 대한 살균제 효과 검정 및 병원균 집단에 대한 저항성 검정)

  • With 32 fungicides, it was examined the inhibitory effects on the mycelial growth of Alternaria dauci KACC42997 causing Alternaria leaf blight of carrot. Showing the results of the agar dilution method, the fungicides belonging to C2, C5, G1, E2, and E3 group were excellent in inhibiting mycelial growth. Protective fungicides belonging to M group, except for iminoctadine tris-albesilate, and pyraclostrobin belonging to C3 group were effective in inhibiting spore germination of pathogens. The fungicides included into C2 group inhibiting succinate dehydrogenase activity and the G1 group inhibiting demethylase activity showed the excellent inhibitory effect on mycelial growth but the inhibitory effect of spore germination was very low. However, fluazinam belonging to C5 group was excellent in inhibiting spore germination as well as mycelial growth. Especially, when 100 ㎍/ml of fluxapyroxad belonging to the C2 group was treated, 47.1% of spore formation was inhibited on the medium. In comparison of the resistance factors of 3 fungicide groups, as G, C, and E group, in populations of A. dauci isolates collected from Gumi, Pyeongchang, and Jeju, resistance factor in the population of Jeju was the lowest. However, two isolates resistant to fludioxonil belonging to E2 group were found in the isolate group of Pyeongchang, and both showed cross-resistance to iprodione and procymidone.

Genomic Analysis of the Carrot Bacterial Blight Pathogen Xanthomonas hortorum pv. carotae in Korea

  • Bacterial leaf blight of carrots caused by Xanthomonas hortorum pv. carotae (Xhc) is an important worldwide seed-borne disease. In 2012 and 2013, symptoms similar to bacterial leaf blight were found in carrot farms in Jeju Island, Korea. The phenotypic characteristics of the Korean isolation strains were similar to the type strain of Xhc. Pathogenicity showed symptoms on the 14th day after inoculation on carrot plants. Identification by genetic method was multi-position sequencing of the isolated strain JJ2001 was performed using four genes (danK, gyrB, fyuA, and rpoD). The isolated strain was confirmed to be most similar to Xhc M081. Furthermore, in order to analyze the genetic characteristics of the isolated strain, whole genome analysis was performed through the next-generation sequencing method. The draft genome size of JJ2001 is 5,443,372 bp, which contains 63.57% of G + C and has 4,547 open reading frames. Specifically, the classification of pathovar can be confirmed to be similar to that of the host lineage. Plant pathogenic factors and determinants of the majority of the secretion system are conserved in strain JJ2001. This genetic information enables detailed comparative analysis in the pathovar stage of pathogenic bacteria. Furthermore, these findings provide basic data for the distribution and diagnosis of Xanthomonas hortorum pv. carotae, a major plant pathogen that infects carrots in Korea.

Effective Control Strategy against Bacterial Blight on Carrot (당근 세균잎마름병에 대한 효과적 방제 수단)

  • Bacterial blight of carrot caused by Xanthomonas hortorum pv. carotae (Xhc) is one of the serious diseases of carrot, of which control measures has not been still established in the domestic farm. In this study, in order to select effective sterilizer for bacterial blight of carrots, three antibiotics such as streptomycin, oxolinic acid, kasugamycin, two copper compounds like copper hydroxide and copper sulfate basic and three rhizobacteria Burkholderia gladioli MRL408-3, Pseudomonas fluorescens TRH415-2 and Bacillus cereus KRY505-3 were selected to investigate their direct antibacterial effects using artificial media, aiming to identify effective pesticides against Xhc. Among them, treated medium with antibiotics such as streptomycin, oxolinic acid, and the antagonistic rhizobacteria MRL408-3 were formed inhibition zone. The agrochemicals and the rhizobacteria MRL408-3, which showed antibacterial effects on carrot leaves, pre-treated on the carrot leaves and then inoculated with Xhc. High control effects were shown on the carrot leaves pre-treated with both streptomycin and oxolinic acid. Scanning electron microscopy images of the carrot leaf surfaces showed that the population of bacteria decreased significantly on leaves pre-treated with streptomycin and oxolinic acid. From these results, it can be inferred that antibiotics like streptomycin and oxolinic acid exhibit superior control effects compared to other agents. This study provides valuable insights towards establishing an effective control system for bacterial blight of carrot.

Composition and EC of Nutrient Solution on Growth and Quality of Carrot (Daucus carrota L.) in Hydroponics (당근 수경재배시 생육 및 품질에 미치는 배양액 조성 및 농도)

  • Carrot leaves have many nutrients as well as roots, which will increase the demand for carrot leaves in the future. This study was carried out by dividing into two stages: high temperature and low temperature periods, in order to investigate the possibility of cultivation of carrot leaves and the composition and EC of the nutrient solution for growth and quality of carrot leaves. Composition of nutrient solution($NO_3-N:16.0$, $NH_4-N:1.0$, P: 1.0, K: 11.0, Ca: 2.0, Mg: 1.0, $SO_4-S:1.0mM{\cdot}L^{-1}$) developed by analysis of plant. In the high temperature range (From June $29^{th}$ to Sep. $8^{th}$, 2016), the concentration of the developed nutrient solution (JNU) were 1.0, 2.0, 3.0, and $4.0dS{\cdot}m^{-1}$ and the concentration of nutrient solution of Japanese Horticultural Station(JHS) $2.0dS{\cdot}m^{-1}$ was used for comparison. In the low temperature range (From Dec. $31^{st}$, 2015 to Feb. $29^{th}$, 2016), the concentration of the developed nutrient solution 1.0, 2.0, and $3.0dS{\cdot}m^{-1}$ were used. Growth was investigated in root fresh and dry weights, shoot fresh and dry weights, leaf number, and leaf area of carrot. In the high temperature range, the leaf area and shoot fresh and dry weights were good at 1.0 and $2.0dS{\cdot}m^{-1}$. The sugar content of the root was the highest at the EC $2.0dS{\cdot}m^{-1}$, and the chlorophyll content was the highest at the EC $4.0dS{\cdot}m^{-1}$. In the low temperature range, The shoot fresh and dry weights were the highest at EC 1.0 and $2.0dS{\cdot}m^{-1}$. There was no significant difference in sugar content and chlorophyll content. As a result, from the viewpoint of growth and quality of carrot, it is good to cultivate EC 1.0 and $2.0dS{\cdot}m^{-1}$ in high temperature period and low temperature period, but EC $1.0dS{\cdot}m^{-1}$ is economical perspective such as fertilizer input.

Effects of Single Cells of Carrot and Radish on the Fecal Excretion Properties, Mineral Absorption Rate and Structure of Small Intestine and Colon in Rats (당근과 무의 단세포물 섭취가 흰쥐의 배변특성, 무기질 흡수율 및 소장과 대장의 구조에 미치는 영향)

  • The effects of single cells of carrot or radish on the fecal excretion, mineral absorption rate and structure of small intestine and colon were investigated in rats fed 5% single cell diet for 4 weeks. Carrot contained 28.76% of total dietary fiber and 21.45% of insoluble dietary fiber, and radish contained 23.14% of total dietary fiber and 16.77% of insoluble dietary fiber on a dry weight basis. Total dietary fiber contents of the single cell were 44.68% for carrot, 48.0% for radish. Absorption rates of magnesium were higher in the carrot and radish single cell groups than the other groups. Cellulose significantly increased fecal weight and weight of small intestine. The consumption of cellulose and single cells decreased digestibility of lipid. The length of colon were longer in the carrot and the radish group than the other groups. Scanning electron microscopy studies showed that small intestine microvilli with leaf-shaped were seen in cellulose and single cell fed groups. These results suggest that the diet containing 5% single cells of carrot or radish increases the digestibility of dietary fiber, weight gain, and fecal output and shorten the gastrointestinal transit time.

Analysis of Flavonoids in Raw and Blanching of Several Green-Yellow Vegetables (시판 녹황색 채소류 중의 플라보노이드 함량과 조리과정에 의한 변화)

  • Flavonoids in commerical green-yellow vegetables (carrot, mugwort, perilla leaf, leek and water dropwort) were analyzed by HPLC. Flavonoids analyzed in samples were myricetin, luteolin , qercetin, apigenin and kaempferol. When 2N HCI and a reaction period of 1 hours was used, quercetin and kaempferol could be detected in leek and water dropwort, luteolin and apigenin could be detected in perilla leaf. Increasing reaction period up to 6 hours with 2N HCI led a degradation of quercetin and kaempferol to approximately 90% in leek and water dropwort, but it led to a increasing of luteolin and apigenin of maximum 300% in perilla leaf. After the blanching process, the amount of the flavonoids increased whereas the components of the flavonoids were not changed.
