Effects of High Voltage Pulsed Electric Fields on the Extraction of Carotenoid from Phaffia rhodozyma (Phaffia rhodozyma로부터 Carotenoid 추출에 미치는 고전압 펄스 전기장의 영향)

  • High voltage pulsed electric fields (PEF) technology is a non-thermal technique which is applicable to extract useful components froms biological materials. This research suggested the possibility for extracting carotenoid pigments from Phaffia rhodozyma by PEF treatments. The yeast cell suspensions were treated with high voltage pulses in a recycled PEF treatment chamber which consists a pair of thin plates of stainless steel adhering to a small chamber with approximately $1{\sim}4\;mm$ gap. A 2.5 log reduction in survivability and more than 98% of electropermeabilization of the yeast cells could be achieved by PEF treatment for $300\;{\mu}s$ with an electric field of 30 kV/cm and pulse duration of $1\;{\mu}s$. When the yeast cell suspended in 0.01% NaCl solution were treated with PEF under various conditions, carotenoid pigments were not extracted. However, the PEF treatment of the yeast cell suspensions in 0.01% $CaCl_2$ solution, have positive effects on the extraction of carotenoid pigments ($27.3\;{\mu}g/g$ of dried yeast).

Comparison of Carotenoid Pigments in Flat bitterling, Paracheilognathus rhombea and Korean striped bitterling, Acheilognathus yamatsutae Mori, in the Subfamily Cyprinidae (잉어아과에 속하는 납지리와 줄납자루의 Carotenoid 색소성분의 비교)

  • 어류의 carotenoid에 관한 비교 생화학적 연구의 일환으로서 잉어아과에 속하는 납지리와 줄납자루의 표피와 난의 carotenoid 조성을 비교 검토하여 다음과 같은 결론을 얻었다. 납지리와 줄납자루 표피의 총 carotenoid 함량은 납지리 수컷 2.3mg%, 납지리 암컷 1.1mg%, 줄납자루 수컷 1.9mg%, 줄납자루 암컷 0.8mg%로 나타나 수컷 표피의 총 carotenoid 함량이 암컷 표피보다 다소 높았다. 표피의 carotenoid 조성은 lutein, lutein epoxide, tunaxanthin, zeaxanthin, triol 이 주성분을 이루었고 그외, $\beta-carotene,$ $\alpha-cryptoxantinm$ cyn-thiaxantin 이 함유되어 있었다. 납지리 수컷의 표피에는 줄납자루 수컷에 비해 lutein의 함량치가 높았고 줄납자루 수컷의 경우는 납지리 수컷이 비해 lutein epoxide와 tunaxanthin의 함량치가 높았다. 그리고 납지리 암컷을 표피에는 줄납자루 dkat에 비해 lutein epoxide 함량치가 높았도 줄납자루 암컷의 표피에는 납자리 와 줄납자루 난의 총 carotenoid 함량은 납지리 1.8mg%, 와 줄납자루 2.5mg%로 표피와는 달리 줄납자루 난의 함량치가 높았다. 납지리와 줄납자루 난의 carotenoid 조성은 다 같이 lutein, lutein epoxid 가 주성분을 이루었고 그외 triol은 표피에 비해 적게 함유되어 있었다. 동일한 잉어아과의 납지리와 줄납자루의 표피 및 나의 carotenoid 조성은 서로 유사함을 확인 할 수 있었다.

Carotenoid Pigments of Flounder and Yellowtail (넙치와 방어의 Carotenoid 색소성분)

  • Difference of carotenoid pigments in integuments of the wild and cultured flounder, Paralichthys olivaceus and yellowtail, Seriola quinqueradiata were studied. Total carotenoid contents in integuments of the wild and cultured flounder were 1.38mg% and 1.l6mg%, respectively. The main carotenoids in integuments of the wild flonder were zeaxanthin (19.22%), $\beta$-carotene type triol (17.80%), tunaxanthin C (17.77%), lutein (16.44%) and tunaxanthin B (13.70%). In addition, tunaxanthin A (5.42%), $\alpha$-cryptoxanthin (4.80%), astaxanthin (0.69%) and $\beta$-cryptoxanthin (0.24%) were also contained in small amounts. But in the cultured flounder, lutein (38.21%) and zeaxanthin (29.69%) were contained as main carotenoids. In addition, $\beta$-carotene type triol (7.80%), tunaxanthin C (7.05%), $\alpha$-cryptoxanthin (4.34%), tunaxanthin B (4.21%), as-taxanthin (2.40%) and $\beta$-cryptoxanthin (1.30%) were present in small amounts. Consequently, the wild flounder contained higher amounts of tunaxanthin and trios but contained lower amounts of lutein and zeaxanthin than the cultured flonder. The contents of carotenoids from integuments of wild and cultured yellow-tail were 1.08mg% and 0.09mg%. Wild and cultured yellowtail have similar carotenoid patterns, consisting of tunaxanthin C (44.11%, 43.37%), tunaxanthin B (33.56%, 29.23%) and tunaxanthin A (18.22%, 21.68%), respectively.

  • 어류의 carotenoid에 관한 비교 생화학적 연구의 일환으로서, 우리나라 특산의 통어과이 쏘가리와 꺽지의 표피 및 난의 산란 중과 산란 후의 carotenoid 조성이 서로 어떻게 다른가 비교, 검토하여 다음과 같은 결론을 얻었다. 쏘가리와 꺽지 표피의 총 carotenoid 함량은 쏘가리는 산란 중에 2.8mg%, 산란후에 2.1mg%로 나타나 산란후보다 산란중에 높은 함량치를 보였다. 표피의 carotenoid 조성은 쏘가리는 산란중에 tunaxanthin 42.2%, lutein 22.0%이며, 산란 후에는 tunaxanthin 32.7%, lutein 24.5%로 나타났으며, 꺽지는 산란중에는 tunaxanthin 69.4%, lutein 17.0%m 산란후에는 tunaxanthin 37.5%, lutein 24.8%로 나타나 표피의 tunaxanthin 의 함량이 산란 중이 산란 후가 높았다. 쏘가리와 꺽지 난의 총 carotenoid 함량은 쏘가리의 난은 0.3mg%, 꺽지의 난은 1.3mg%로 나타나 쏘가리 보다 꺽지 난의 함량이 높았다. 난의 carotenoid 조성은 쏘가리 $\beta$-carotene 27.4%, zeaxanthin 25.7%, diatoxanthin 23.8%였고, 꺽지는 $\beta$-carotene 27.4%, zeaxanthin 25.3%, diatoxanthin 22.4%로 나타나 모두 $\beta$-carotene, zeaxanthin, diatoxanthin 이 주성분으로 함유 되어 있었으며 그외 cynthiaxanthin, lutein 및 cryptoxanthin 등이 함유되어 있었다 동일한 농어과의 쏘가리와 꺽지 표피 및 난의 carotenoid 조성은 서로 유사함을 확인할 수 있었다.

  • Carotenoid pigments of echiurid, Urechis unicinctus were investigated during March, April and May as a part of comparative biochemical studies of carotenoid pigments for the marine organisms other than pisces. Total carotenoid contents were found to be 1.19 mg/100 g in March, 0.98 mg/100 g in April and 0.84 mg/100 g in May, indicating that total carotenoid content was negatively affected by the temperature of sea water that echiurid resided. The carotenoid isolated in March composed of $16.3\%$ diatoxanthin monoester, $14.8\%$ $\beta$-carotene and $12.6\%$ cynthiaxanthin monoester, $8.4\%$ cynthiaxanthin diester, $8.2\%$ zeaxanthin monoester, $7.3\%$ diatoxanthin diester $4.2\%$ astaxanthin $2.9\%$ diatoxanthin, $2.4\%$ triol, $2.3\%$ cynthiaxanthin, $1.7\%$ isocrytoxanthin, $1.5\%$ zeaxanthin diester, $0.8\%$ zeaxanthin and $0.5\%$ lutein. The carotenoid isolated in April composed of $21.9\%$ diatoxanthin monoester, $17.2\%$ cynthiaxanthin monoester and $16.6\%$ $\beta$-carotene $10.9\%$ zeaxanthin monoester, $5.6\%$ cynthiaxanthin diester, $4.9\%$ diatoxanthin diester, $3.1\%$ astaxanthin, $2.4\%$ triol, $2.3\%$ diatoxanthin, $1.7\%$ isocrytoxanthin, $1.5\%$ lutein, $1.1\%$ zeaxanthin, $1.0\%$ cynthiaxanthin and $1.0\%$ zeaxanthin diester. Similarly, the carotenoid isolated in May composed of $25.3\%$ diatoxanthin monoester, $19.7\%$ cynthiaxanthin monoester, $13.0\%$ $\beta$-carotene, and $12.6\%$ zeaxanthin monoester, $5.8\%$ cynthiaxanthin diester, $5.1\%$ diatoxanthin, $3.0\%$ astaxanthin, $2.4\%$ triol, $2.2\%$ diatoxanthin, $1.3\%$ isocrytoxanthin, $1.2\%$ zeaxanthin, $1.1\%$ zeaxanthin diester, $1.0\%$ lutein and $0.9\%$ cynthiaxanthin. Based on these data, monoester-type carotenoids ($37.1\~57.6\%$) and diester-type carotenoids ($11.5\~17.2\%$) of total carotenoids in echiurid were the major carotenoids. Meanwhile, when the sea water temperature was elevated and the contents of total carotenoid in echiurids were decreased, the contents of zeaxanthin monoester, diatoxanthin monoester and cynthiaxanthin monoester were increased, but the contents of zeaxanthin diester, diatoxanthin diester and cynthiaxanthin diseter were decreased, indicating that changes in ester-type caroteoids were differently affected by the sea water temperature.

Quality Properties and Carotenoid Pigments of Yellow Sweet Potato Puree (황색고구마 퓨레의 품질과 Carotenoid색소)

  • Two kinds of sweet potato puree were prepared with Benihayato cultivar of yellow sweet potato with or without $\alpha$-amylase enzyme treatment. Chemical and rheological properties of enzyme-treated puree were different from those of control puree. Reducing sugar content and iodine value increased by $\alpha$-amylase enzyme treatment, while alcohol insoluble solids and viscosity decreased by enzyme treatment. However, the changes of carotenoid content were not significantly different. Hunter-b-values(yellowness) were 27.19 and 23.54 for no enzyme-treated puree(NTP) and enzyme-treated puree(ETP), respectively. Hunter-a values(redness) were 2.24 and 6.05 for NTP abd ETP, respectively. Content of total carotenoid of canned puree heated at 13$0^{\circ}C$ for 30 min decreased by 59 percents.

  • Pigments were extracted by aqueous acetone from the boiled canned oyster colored during storage and then the components were separated by thin layer chromatography. Totally eleven pigments could be isolated including one yellow, one red, five orange or reddish orange and four green components, and their UV-visible spectra were measured. It can be envisioned from the electronic spectral study and color reaction on the indivisual pigments isolated from the brown acetone extracts that the green pigments as well as most of the yellow orange ones may be porphyrin derivatives originated probably from the chlorophyll and some of the orange pigments contains ketocarotenoids. In particular, the pigment of band 8 which is expected to be pheophytin a or its derivatives and the carotenoid band 7 seem to be the major pigment. The close resemblance of the chromatogram of the colored muscle extract to that of the viscera suggests that the brown coloring material is probably originated from the viscera pigments.

Carotenoid Pigments from Suspended and Sinking Particulate Matter in Prydz Bay, Antarctica

  • Suspended and sinking particles were collected during ODP Leg 119 to the Indian Ocean sector of the Antarctic Ocean. Field work was carried out at four sampling sites in Prydz Bay. Two of these sites were located in the Outer Bay, and two in the Inner Bay. At the four locations, a total of ten deployments of a sediment trap array were made. The concentrations of carotenoids both in suspended and sinking particulate matter in Prydz Bay were analyzed using HPLC. Fucoxanthin was the dominant carotenoid pigments both in suspended and sinking particles. The present study also indicates that 19'-hexanoyoxyfucoxanthin-containing prymesiophytes (Phaeocystis spp.) was abundant in the study area. The flux rates of carotenoids were generally highest at 50 m, and approximately double the flux rates at deeper horizons, however, at Inner Bay sites, the mean flux rates of carotenoids were greatest at 200 m, and 3 times greater than that of 50 m. Such anomalous high fluxes at 200 m imply that grazers were locally abundant between 100 m and 200 m at these sites close to land, and this hypothesis is supported by visual evidence of lots of fecal pellets in the 200 m trap. Integrates standing stocks versus sinking pigments data support that particulate material in Prydz Bay was not recycled rapidly.

  • Although the antioxidant capacity of carotenoids and their role in regulating membrane fluidity have been well studied, their ability to confer resistance to hypoosmotic shock is poorly understood. In this work, we analyzed the effect of a mixture of carotenoid pigments obtained from an Antarctic microorganism belonging to the genus Pedobacter on liposomal resistance to hypoosmotic conditions. Intercalation of pigments into liposomal structures resulted in an improvement of membrane resistance by decreasing the percentage of calcein released in comparison to that by liposomes without pigments. Due to these properties, such pigments could be useful for biotechnological applications.
