Identification of Quantitative Traits Loci (QTL) Affecting Growth Traits in Pigs

  • Molecular genetic markers were used to detect chromosomal regions which contain economically important traits such as growth, carcass, and meat quality traits in pigs. A three generation resource population was constructed from a cross between Korean native boars and Landrace sows. A total of 240 F2 animals from intercross of F1 was produced. Phenotypic data on 17 traits, birth weight, body weights at 3, 5, 12, and 30 weeks of age, teat number, carcass weight, backfat thickness, body fat, backbone number, muscle pH, meat color, drip loss, cooking loss, water holding capacity, shear force, and intramuscular fat content were collected for F2 animals. Animals including grandparents (F0), parents (F1), and offspring (F2) were genotyped for 80 microsatellite markers covering from chromosome 1 to 10. Least squares regression interval mapping was used for quantitative trait loci (QTL) identification. Significance thresholds were determined by permutation tests. A total of 10 QTL were detected at 5% chromosome-wide significance levels for growth traits on SSCs 2, 4, 5, 6, and 8.

High-level dietary crude protein decreased backfat thickness and increased carcass yield score in finishing Hanwoo beef cattle (Bos taurus coreanae)

  • Recently, a high level of dietary crude protein (CP) has become of interest as a possible practice to improve the carcass quality of beef cattle, and its level has been increasing in the field. However, there is little scientific evidence that supports this. This study was conducted to test whether a high dietary CP level would improve growth performance, body metabolism, and carcass traits in Hanwoo beef cattle. A total of 32 Hanwoo finishing beef cattle (18 multiparous cows, six heifers, and eight steers) participated in a 12-weeks feeding trial. Two kinds of total mixed rations were prepared to contain two different CP; 156 g/kg for the control (CON) and 173 g/kg of CP for the treatment (HCP), while maintaining a similar level of metabolizable energy. The experiment was ended when more than half of the steers reached the target body weight (730 kg). Blood was collected at the end of the experiment. After harvesting, the carcass trait was evaluated at the slaughterhouse according to Korean standards. The carcass yield score and grade were also calculated based on revised criteria. Overall, dry matter intake, average daily gain, blood metabolites concentration, and the carcass traits, except for backfat thickness and the yield score, did not differ between the treatments. The HCP had lower backfat thickness than those of CON. There was no difference in the carcass yield grade, but the yield score was higher in the HCP treatment. According to the newly revised carcass grading criteria, both yield score and grade were higher in HCP than in CON. Increasing CP supply decreased the carcass's backfat thickness without altering growth performance and body metabolism, resulting in improved yield score and grade. Therefore, feeding a high CP diet may be beneficial in the farm income, although it may also increase feed cost and nitrogen excretion to the environment.

Genetic Analysis of Ultrasound and Carcass Measurement Traits in a Regional Hanwoo Steer Population

  • Ultrasound measurements of backfat thickness (UBF), longissimus muscle area (ULMA) and marbling score (UMS) and carcass measurements of carcass weight (CW), backfat thickness (BF), longissimus muscle area (LMA), and marbling score (MS) on 7,044 Hanwoo steers were analyzed to estimate genetic parameters. Data from Hanwoo steers that were raised, finished in Hoengseong-gun, Gangwon-do (province) and shipped to slaughter houses during the period from October 2010 to April 2013 were evaluated. Ultrasound measurements were taken at approximately three months before slaughter by an experienced operator using a B-mode real-time ultrasound device (HS-2000, FHK Co. Ltd., Tokyo, Japan) with a 3.5 MHz linear probe. Ultrasound scanning was on the left side between 13th rib and the first lumbar vertebrae. All slaughtering processes and carcass evaluations were performed in accordance with the guidelines of beef grading system of Korea. To estimate genetic parameters, multiple trait animal models were applied. Fixed effects included in the models were: the effects of farm, contemporary group effects (year-season at the time of ultrasound scanning in the models for UBF, ULMA, and UMS, and year-season at slaughter in the models for CW, BF, LMA, and MS), the effects of ultrasound technicians as class variables and the effects of the age in days at ultrasound scanning or at slaughtering as linear covariates, respectively for ultrasound and carcass measures. Heritability estimates obtained from our analyses were 0.37 for UBF, 0.13 for ULMA, 0.27 for UMS, 0.44 for CW, 0.33 for BF, 0.36 for LMA and 0.54 MS, respectively. Genetic correlations were strongly positive between corresponding traits of ultrasound and carcass measures. Genetic correlation coefficient between UBF and BF estimate was 0.938, between ULMA and LMA was 0.767 and between UMS and MS was 0.925. These results suggest that ultrasound measurement traits are genetically similar to carcass measurement traits.

돼지 MC4R 유전자 892G>A 다형성이 비육돈의 도체형질에 미치는 영향 (Effects of a Porcine MC4R Polymorphism(892G>A) on Carcass Traits in Commercial Pigs)

  • 임의적으로 선정된 비육돈 집단에서 melano- cortin-4 receptor(MC4R) 유전자의 유전적 변이와 도체형질의 연관성을 조사하였다. 돼지 MC4R 유전자형은 Asp298Asn(nt. 892G>A) 돌연변이를 PCR-RFLP 방법으로 결정하였다. 비육돈 집단에서 유전자형 A/A, A/G, G/G가 모두 출현하였고, 빈도는 각각 28.8, 48.4, 22.8%를 나타내었다. 전체 비육돈에서 유전자형 A/-은 G/G에 비해 근내지방도가 유의적으로 높게 나타났다(P<0.05). 동형접합자 A/A와 G/G가 이형접합자 A/G인 도체에 비해 육색도는 더 낮고 수분삼출도는 높은 것으로 확인되었다(P<0.01). 반면, -/G인 거세돈의 도체중이 A/A에 비해 도체중이 유의적으로 무겁게(약 2.5kg) 나타났다(P<0.05). 각 유전자형의 도체형질에 대한 효과는 미경산돈에서는 전체 비육돈 집단에 대한 효과와 유사하나, 거세돈 집단과는 다소 차이가 있었고, 이는 도체형질에 대한 현재까지 알려지지 않은 성-관련 효과로 추정된다. 본 연구에서 비육돈 생산에 있어 MC4R A/- 유전자형이 육질을 향상시킬 수 있을 것으로 기대된다. 하지만, MC4R 유전자형들이 성과 관련되어 차별적으로 도체형질에 영향을 주고 있기 때문에, 비육돈 생산을 위한 marker-assisted selection을 위해서는 성과 유전자형 모두가 고려되어져야 할 것이다.

한우 도체형질 및 도체 일반성분 유전모수 추정 (Estimates of Genetic Parameter for Carcass Traits and Chemical Composition Analysis in Hanwoo)

  • 본 연구의 목적은 한우 도체형질과 등심에 대한 일반성분(수분, 조지방, 조단백질, 조회분) 분석을 통한 등심의 영양학적 가치와 유전모수 추정을 통한 개량형질 적용 가능성을 확인하기 위하여 실시하였다. 본 연구에 이용된 공시 재료는 한우 후대검정우 47차부터 57차까지 실시한 2,033두에 대하여 등심 1kg을 시료를 채취하여 분석한 자료를 이용하였다. 연구결과 한우 도체형질인 도체중, 등심단면적, 등 지방두께, 근내지방도에 대한 유전력은 각각 0.37, 0.51, 0.44, 0.57로 추정되었으며, 일반성분 분석인 수분, 조지방, 조단백질, 조회분에 대한 유전력은 각각 0.62, 0.64, 0.58, 0.12로 추정되었다. 그리고, 도체형질과 일반성분간의 유전상관은 도체중과 조지방에서 0.35, 근내지방도와 수분, 조지방, 조단백질, 조회분에서 각각 -0.966, 0.964, -0.924, -0.664으로 고도의 유전상관을 보였다. 수분과 조지방, 조단백질간의 유전상관은 각각 -0.998, 0.969로 분석되었으며, 조지방과 조단백질에서 -0.979로 고도의 유전상관을 보였다. 일반성분 분석치인 수분, 조지방, 조단백질에서 고도의 유전력을 보였으므로, 개량형질 이용 가능성이 있을 것으로 사료된다.

제주도 한우 집단에서 Galactose mutarotase (GALM) 유전자의 삽입/결실 다형성과 도체형질의 연관성 분석 (Associations between Insertion/deletion Polymorphisms in Galactose Mutarotaseand Carcass Traits in the Hanwoo Population of Jeju Island)

  • 이 연구는 제주도 한우집단에서 galactose mutarotase (GALM) 유전자형과 도체형질의 연관성을 시험하였다. GALM 유전자형은 3'-비해독부위(3'-UTR)의 14-bp (5'-GGTCTAATGACCAG-3') 삽입/결실 다형성을 이용하였다. 한우 비육우 집단에서 GALM 유전자의 세 가지 유전자형(LL, LS, SS)이 모두 관찰되었다. 연관성 분석결과는 근내지방의 함량과 밀접한 상관을 보이는 육질등급과 근내지방도의 수준과, 등지방두께의 수준이 유전자형에 따른 유의적인 차이를 나타내었다(p<0.05). 동형접합인 SS 유전자형을 보유한 도체에서 LL 또는 LS 유전자형인 도체에 비해 근내지방 함량 수준은 더 높고, 등지방두께도 더 얇은 수준을 보였다. 반면, 도체중, 등심단면적, 육색, 지방색 등은 GALM 유전자형에 따른 유의적인 차이는 없었다(p>0.05). 3'-UTR에서 14-bp 절편의 결실은 RNA의 2차 구조의 변형과 RNA-결합 단백질, microRNA와의 결합능력에 대한 방해를 통해 유전자 발현에 영향을 줄 수 있는 것으로 예측되었다. GALM 유전자의 3'-UTR 영역에서 14-bp 삽입/결실 다형성에 대한 이번 연구결과는 소에서 근육과 등지방 조직에서 galactose 대사에 의한 지방 축적을 통해 성장형질, 도체형질에 영향을 주는 것으로 판단된다.

Leptin Polymorphisms Associated with Carcass Traits of Meat in Korean Cattle

  • Leptin has been investigated as a candidate gene for fat characteristics in beef cattle. Previously, we have reported 57 sequence variants discovered in Korean cattle (Bos Taurus coreanae). In this study, we examined the association between polymorphisms of leptin and carcass traits (cold carcass weight (CWT) and marbling score (Marb)) in Korean cattle. Among 57 polymorphisms, 11 common polymorphic sites were genotyped in our beef cattle (n = 437). Statistical analysis revealed that one single nucleotide polymorphism in coding exon (c.+411T>C (A137A)) showed a significant association with the yield trait, CWT. The C-bearing genotypes (CC or CT) of c.+411T>C (A137A) showed the higher CWT (p = 0.006). c.+150C>G (S50S) also showed a significant association with the quality trait, Marb (p = 0.01). Our findings suggest that polymorphisms in leptin might be one of the important genetic factors that influence carcass yield and quality in beef cattle, especially in CWT and Marb.

The effect of castration time on growth and carcass production of elk bulls

  • The effects of castration time on growth and carcass traits of elk bulls were investigated. Twelve bulls at 5 years old were raised and fed on concentrate with ad libitum hay. All animals were allocated randomly to each of four treatment groups (3 heads/group). Groups of each treatment were castrated surgically in March, April or June and managed together with non-castration (entire) treatment. All elk bulls in the trial were slaughtered at same time. Growth parameters, carcass yield and composition were recorded. The total gain and average daily gain was higher when castrated in April (p < 0.05). The entire elk produced heaviest and highest in saddle and brisket portions (p < 0.05). It is apparent that the castrate animals carried more total fat weight and percentages than the entire males (p < 0.05). It was found that loin muscles from non-castrated elk, in comparison with those from castrated one, had higher content of moisture and lower content of fat (p < 0.05). In this study, growth parameters, carcass yields and chemical composition were greatly affected by castration time.

Application of Linkage Disequilibrium Mapping Methods to Detect QTL for Carcass Quality on Chromosome 6 Using a High Density SNP Map in Hanwoo

  • The purpose of this study was to detect QTL for carcass quality on bovine chromosome (BTA) 6 using a high density SNP map in a Hanwoo population. The data set comprised 45 sires and their 427 Hanwoo steers that were born between spring of 2005 and fall of 2007. The steers that were used for progeny testing in the Hanwoo Improvement Center in Seosan, Korea, were genotyped with the 2,535SNPs on BTA6 that were embedded in the Illumina bovine SNP 50K chip. Four different linkage disequilibrium (LD) mapping models were applied to detect significant SNPs for carcass quality traits; the fixed model with a single marker, the random model with a single marker, the random model with haplotype effects using two adjacent markers, and the random model at hidden state. A total of twelve QTL were detected, for which four, one, three and four SNPs were detected on BTA6 under the respective models (p<0.001). Among the detected QTL, four, two, five and one QTL were associated with carcass weight, backfat thickness, longissimus dorsi muscle area, and marbling score, respectively (p<0.001). Our results suggest that the use of multiple LD mapping approaches may be beneficial in increasing power to detect QTL given a limited sample size and magnitude of QTL effect.

Effect of Inclusion of Hard Versus Soft Wheat Bran with Different Particle Size on Diet Digestibility, Growth Performance and Carcass Traits of Fattening Rabbits

  • Effect of inclusion of hard vs. soft wheat bran with different particle size on diet digestibility, growth performance and some slaughter traits was evaluated in fattening rabbits. Four isonitrogenous and isocaloric diets were used according to the origin of wheat bran (hard (HWB) - Triticum durum - and soft (SWB) - Triticum aestivum) combined with wheat bran particle size sieved by 2 mm (fine: 2) or by 8 mm (coarse: 8) in a bifactorial (2${\times}$2) study. A growth trial was conducted to measure the effect of treatments on performance in one hundred and twenty New Zealand White${\times}$Californian rabbits fed experimental diets from 50 to 87 days of age. Faecal apparent digestibility was determined within the last week in twenty animals per diet. Digestibility of nutrients was higher (p<0.05) in the diet containing HWB2, except for crude protein, ether extract and ash, than fine and coarse soft wheat bran diets. Final live weight, feed intake and feed consumption of rabbits on the diet with fine hard wheat bran were higher and resulted in greater daily weight gains (p<0.01) than for animals on the other diets. The slaughter yield and percentage value of organs were not significantly (p>0.05) affected by the diets fed; however, the diet containing fine hard wheat bran led to lower (p<0.05) percentages of skin, abdominal fat and carcass drip loss than the other dietary treatments. It is concluded that fine hard wheat bran can be better included in the diet than soft wheat bran to maximize growth performance without affecting carcass traits of fattening rabbits.
