Alpha-alkylcysteines as Inhibitors for Carboxypeptidase A. Synthesis, Evaluation, and Implication for Inhibitor Design Strategy

  • (R,S)- and (R)-2-Benzylcysteine (1) and (R,S)-2-phenethylcysteine (2) were synthesized and evaluated as inhibitors for carboxypeptidase A (CPA) with the expectation that these compounds exhibit improved inhibitory activities over 2-benzyl-3-mercaptopropanoic acid (BMPA), a potent CPA competitive inhibitor, possibly having additional interactions of their amino group with the carboxylate of Glu-270 of the enzyme upon binding to CPA. Contrary to the expectation, however, the CPA inhibitory potencies of these compounds were found to be much reduced compared with that of BMPA, suggesting that the amino group in the inhibitors rather exerts steric hindrance in binding of these inhibitors to CPA.

Effect of Surfactant and Anti-foaming Agent on the Properties of Silicone Rubber Impression Material (계면활성제와 소포제가 실리콘 고무인상재의 물성에 미치는 영향)

  • The three types of surfactants such as nonylphenoxy poly(ethylene) ethanol homologues, caster oil poly(ethylene) ethanol homologues, and sodium perfluoroalkyl carboxylates are used to improve the wettability of rubber impression material. Among the surfactants, the usage of sodium perfluoroalkyl carboxylates containing fluoro group resulted in the lowest surface energy of impression material and the result gave the positive effect on the wettability of rubber impression material to teeth. Also, the anti-foaming agents were used to reduce or remove the hydrogen gas generating on the impression material by reaction. In the case of rubber impression material containing sodium perfluoroalkyl carboxylate as a surfactant, it was found that the tear strength of rubber impression material increased over 3 N/mm with the addition of anti-forming agent. Therefore, the anti-foaming agent could contribute to the mechanical property of rubber impression material without the change of surface property.

  • The crystal structure of the potential active Methyl 8-bromo-3-phenyl-5a,9a-dihydro-3H-chromen [4,3-c][1,2] isoxazole-3a(4H)-carboxylate ($C_{18}H_{15}BrNO_4$) has been determined from single crystal X-ray diffraction technique. The title compound crystallizes in the triclinic space group Pī with unit cell dimension a=8.3129 (3) ${\AA}$, b=9.5847 (4) ${\AA}$ and c=11.1463(4) ${\AA}$ [${\alpha}=98.457(3)^{\circ}$, ${\beta}=102.806(2)^{\circ}$ and ${\gamma}=105.033(5)^{\circ}$]. Single crystals suitable for X-ray diffraction were obtained by slow evaporation method, the isoxazole and six membered pyran rings adopts envelope conformation. In the crystal, molecules are linked via pairs of inter molecular $C-H{\ldots}O$ hydrogen bonds to form dimmers.

Synthesis and Characterization of Mononuclear Octahedral Fe(III) Complex Containing a Biomimetic Tripodal Ligand, N-(Benzimidazol-2-ylmethyl)iminodiacetic Acid

  • The mononuclear iron complex 1, $Fe^{III}$(Hbida)Cl($H_2O$), was synthesized using a tripodal tetradentate ligand, N-(benzimidazol-2-ylmethyl)iminodiacetic acid (H3bida), which has two carboxylate groups, one benzimida- zoyl group, and one tertiary amine where it serves as a tetradentate chelating ligand for the octahedral Fe(III) ion. The four equatorial positions of the octahedral complex are occupied by two monodentate carboxylates, a benzimidazole nitrogen, and an oxygen of a water molecule. One of the axial positions is occupied by an apical nitrogen of the Hbida and the other by a chloride anion. The mononuclear octahedral complex 1 mimics the geometry of the key intermediate structure of the catalytic reaction cycle proposed for the FeSODs, which is a distorted octahedral geometry with three histidyl imidazoles, an aspartyl carboxylate, a superoxide anion, and a water molecule. The redox potential of complex 1, $E_{1/2}$ is -0.11V vs. Ag/AgCl (0.12 V vs. NHE), which is slightly lower than those reported for the most FeSODs. The magnetic susceptibility of complex 1 at room temperature is 5.83 $\mu$B which is close to that of the spin only value, 5.92 $\mu$B of high-spin d5 Fe(III).

Crystal Structures and Thermal Properties of Two Binuclear Cd(II) Supramolecular Complexes Based on Quinolinecarboxylate Ligand

  • Two novel binuclear metal-organic coordination complexes $[Cd_2(L)_2(bpy)_2(H_2O)_2]{\cdot}6H_2O$ (1), $[Cd_2(L)_2(phen)_2-(H_2O)_2]{\cdot}2H_2O$ (2) (where L = 2-methylquinoline-3,4-dicarboxylate dianion, bpy = 2,2'-bipyridine, phen = 1,10-phenanthroline) have been synthesized under hydrothermal conditions and characterized by single crystal Xray diffraction, spectral method (IR), elemental analysis and thermal gravimetric analysis (TGA). Both 1 and 2 consist of two Cd(II) atoms bridged by two monoatomic bridging carboxylate groups from two L ligands, and the second carboxylate group of each L is monodentately coordinated to Cd(II), creating a sevenmembered chelating ring. The coordination at each metal nucleus is completed by a water molecule and a chelating bidentate molecule. The 3D structures of the complexes are stabilized by ${\pi}-{\pi}$ stacking interactions and hydrogen-bonds.

  • The synthesis of a series of 7-[(Z)-2-(2-aminothiazol-4-yl)-2-oxyiminoacetamido]-3-(4,6-diamino-1-alkyl-1,3,5-triazinium-2-yl)thiomethyl-3-cephem-4-carboxylates is described. Variations of an oxyimino moiety in the 7-side chain and quaternary ammonium moiety in the 3-side chain were examined and structure-activity relationship was studied. The 4,6-diamino-1-methyl-1,3,5-triazinium-2-yl derivative of the 7-$\alpha$-methoxyimino series of cephalospoins, particularly 7-[(Z)-2-(2-aminothiazol-4-yl)-2-(methoxyimino)acetamido]-3-(4,6-diamino-1-methyl-1,3,5-triazinium-2-yl)thiomethyl-3-cephem-4-carboxylate(C-1), exhibited broad antibacterial activity against Gram-positive and Gram-negative bacteria including Pseudomonas aeruginosa.

  • A series of N-{2,4-dichloro-5-(3-pyrazolyl)Phenyl}imides 7 and 8 was prepared and evaluated their herbicidal activities. Those compounds in which either carboxylate or carboxamide moiety is on pyrazole moiety 7c-71 showed no herbicidal activity whereas, the compounds in which trifluoromethyl group is substituted in pyrazolyl moiety showed moderate herbicidal activities against barnyardgrass, monochria and flat-sedge under paddy conditions with good rice tolerance at rate 63-250 g/ha.

  • The author has studied comparatively the sealing quality of polycarboxylate cement, one of the newest dental cement systems, and zinc oxide eugenol cement by means of penetration of 2% methylene blue solution through the root apex of human teeth in 72 cases as time elapsed. Followings are the results obtained from this study. 1) In all groups, there was no increase in penetration related to increased time of immersion in the dye. 2) In polycarboxylate cement groups, there was a significant difference in depth of dye penetration of each tooth, but in zinc oxide eugenol cement group, there was a slight difference. 3) The depth of dye penetration of zinc oxide eugenol cement group is slightly lower than that of poly carboxylate cement groups. 4) In polycarboxylate cement groups, Carbolit cement group showed comparatively lower grade of dye penetration than Carbo cement group.

  • Rhodium (II) (isobutyrate)$_{4}L_2$, an antinumor drug, was shown to reactr with nucleic acid base derivatives, A, G, U and C in chloroform solution. When these derivatives were treated with one of novel metal compounds, rhodium carboxylate in chloroform solution, a fairly strong complex formation was observed by spectroscopic techniques. The characteristic of these complex was that binding occured at the two axial positions of rhodium (II) (isobutyrate)$_{4}L_2$ to the NH or )$NH_2$ group of the base in the ligands.

Molecular Characterization of a Transient Expression Gene Encoding for 1-Aminocyclopropane-1-carboxylate Synthase in Cotton (Gossypium hirsutum L.)

  • Ethylene performs an important function in plant growth and development. 1-aminocyclopropane-1-carboxylate (ACC) synthase (ACS), the key enzyme involved in ethylene biosynthesis, has been the focus of most ethylene studies. Here, a cotton ACS gene referred to as Gossypium hirsutum ACS1 (GhACS1), was isolated. The full-length cDNA of GhACS1 encodes for a 476-amino acid protein which harbors seven conserved regions, 11 invariant amino acid residues, and the PLP binding active site, all of which characterize ACC synthases. Alignment analysis showed that GhACS1 shared a high degree of identity with other known ACC synthases from different species. Two introns were detected in the genomic DNA sequence, and the results of Southern blot analysis suggested that there might be a multi-gene family encoding for ACC synthase in cotton. From the phylogenetic tree constructed with 24 different kinds of ACC synthases, we determined that GhACS1 falls into group II, and was closely associated with the wound-inducible ACS of citrus. The analysis of the 5' flanking region of GhACS1 revealed a group of putative cis-acting elements. The results of expression analysis showed that GhACS1 displayed its transient expression nature after wounding, abscisic acid (ABA), and $CuCl_2$ treatments. These results indicate that GhACS1, which was transiently expressed in response to certain stimuli, may be involved in the production of ethylene for the transmission of stress signals.
