Effects of Carbon Substrates on Exopolysaccharide Production by Enterobacter sp. (Enterobacter sp. 의 다당 생산에 미치는 탄소원 기질의 영향)

  • The effects of carbon sources for exopolysaccharide production during batch cultivation of an Enterobacter sp. isolated from the composter were investigated. The highest amount of exopolysaccharide was obtained when lactose was used as carbon source. Lactose in medium was converted into glucose and galactose. Glucose was metabolized fast and was completely consumed, but about $20\%$ of lactose was accumulated as galactose. On the other hand, enzyme activity was about $350\~450$ unit with the increase of lactose concentration. Thus, it was considered that the exopolysaccharide might be produced in the course of that lactose was hydrolyzed into glucose and galactose by $\beta-galactosidase$ with respect to that enzyme activity on lactose hydrolysis was accorded to the exopolysaccharide production. When glucose and galactose were added to lactose medium, respectively, it could be considered that glucose was as a repressor and galactose was as a inducer for $\beta-galactosidase$ synthesis even though the mechanisms were not elucidated. The increase of lactose concentration was almost ineffective to the specific growth rate $(0.133\~0.151\;hr^[-1})$ but showed the difference in the biomass content. The higher carbon source concentration, the more residual sugar remained. It was assumed that the optimum lactose concentration for exopolysaccharide production was $30\~70g/L.$ On the other hand, it was considered that the nitrogen acted as growth limiting nutrients to the cell growth. In the cases of 30 and 70 g/L of the fixed carbon concentrations, the increase of the nitrogen sources concentration caused a remarkable increase within the range of $0.059\~0.225\;hr^{-1}$ and $0.141\~0.237hr^{-1}$ of the specific growth rate, respectively, while there was no significant difference in biomass.

Adsorption Removal of Eosin Y by Granular Activated Carbon (입자상 활성탄에 의한 Eosin Y의 홉착제거)

  • Eosin Y is used a colorant and dye but eosin Y is harmful toxic substance. In this study, the adsorption characteristics of granular activated carbon have been investigated for the adsorption of eosin dye dissolved in water. The effects of initial dye concentration, contact time, pH and temperature on adsorption of eosin by a fixed amount of activated carbon have been studied in batch adsorber and fixed bed. The adsorptivity of activated carbon for eosin Y were largely improved by pH control. When the pH was 3 in the sample, the eosin Y could be removed 99% of initial concentration (10 mg/L). The adsorption equilibrium data are successfully fitted to the Freundlich isotherm equation in the temperature range from 293 to 333 K. The estimated values of k and ${\beta}$ are 19.56-134.62, 0.442-0.678, respectively. The effects of the operation conditions of the fixed bed on the breakthrough curve were investigated. When the inlet eosin Y concentration is increased from 10 to 30 mg/L, the corresponding adsorption breaktime appears to decrease from 470 to 268 min at bed height of 3 cm and a constant flow rate of 2 g/min. When the initial eosin Y flow rate is increased from 1 to 3 g/min, the corresponding adsorption breaktime appears to decrease from 272 to 140 min at bed height of 3 cm and inlet concentration of 10 mg/L. Also, breaktime increased with increasing bed height at flow rate of 2 g/min and inlet concentration of 10 mg/L. And length of adsorption zone showed similar patterns.

Comparison of Airborne Nanoparticle Concentrations between Carbon Nanotubes Growth Laboratories based on Containment of CVD (탄소나노튜브 성장 실험실에서 CVD 밀폐 여부에 따른 공기 중 나노입자 농도 비교)

  • Although the usage of nanomaterials including carbon nanotubes (CNTs) has increased in various fields, scientific researches on workers' exposures and controls of these materials are very limited. The purpose of this study was to compare the airborne nanoparticles concentrations from two university laboratories conducting experiments of CNTs growth based on containment of thermal chemical vapor deposition (CVD). Airborne nanoparticle concentrations in three metrics (surface area concentration, particle number concentration, and mass concentrations) were measured by task using three direct reading instruments. In a laboratory where CVD was not contained, the surface area concentration, number concentration and mass(PM$_1$) concentration of airborne nanoparticles were 1.5 to 3.5 times higher than those in the other laboratory where CVD was confined. The ratio of PM$_1$ concentration to total suspended particles(TSP) in the laboratory where CVD was not confined was about 4 times higher than that in the other laboratory. This indicates that CVD is a major source of airbone nanoparticles in the CNTs growth laboratories. In conclusion, researchers performing CNTs growth experiments in these laboratories were exposed to airborne nanoparticles levels higher than background levels, and their exposures in a laboratory with the unconfined CVD were higher than those in the other laboratory with the confined CVD. It is recommended that in the CNTs growth laboratories adequate controls including containment of CVD be implemented for minimizing researchers' exposures to airborne nanoparticles.

Assessment of Indoor Air Quality in Commercial Office Buildings (업무용 빌딩 내 사무실의 실내공기질 평가)

  • Recently, concerns regarding indoor air quality in offices have continued to increase. Thirty offices in five metropolitan commercial buildings were surveyed from February to April 2004. Sampling was performed during normal business hours. Thermal comfort factors such as temperature and relative humidity, carbon dioxide ($CO_2$), carbon monoxide (CO), formaldehyde (HCHO), respirable dust, $PM_{10}$ were sampled and analyzed to determine the mean, standard deviation, range, and correlation for each of those parameters. The data was then compared to office as standard of Ministry of Labor, and guideline applicable to the indoor environment. The results represented that the temperature was slightly higher than the standard of American Society of Refrigerating and Air-Conditioning Engineers (ASHRAE), the relative humidity was lower that the standard of ASHRAE. The range of the 8-TWA concentration of $CO_2$ was 639 ~ 786 ppm, but 33.3% of the total thirty offices exceeded the 1000 ppm as ceiling concentration. The concentration of CO was less than 3 ppm, which was similar to that of offices in Japanese. The mean concentration of formaldehyde was 0.032 ppm, and only 2 % of total samples (193) exceeded the 0.1 ppm, standard of formaldehyde in office air. The concentration of respirable dust and $PM_{10}$ was not exceeded the standard of those parameters, $150{\mu}g/m^3$. The concentration of those parameters in the office air was statistically correlated.

Physio-chemical impacts on efficient germination in Platycodon grandiflorum for. duplex pollen culture

  • The highly valued ornamental plant, Platycodon grandiflorum for. duplex was generated by petaloidy of a calyx of Platycodon grandiflorum. The present investigation was executed to explore the several factors having effects on the germination of pollens with a view to acquire the underlying data for the artificial crossing to cultivate the species of Platycodon grandiflorum for. duplex. Both low and high temperature impaired the germination of Platycodon grandiflorum for. duplex pollens. The good germination rate was observed at the temperature of $25^{\circ}C$. The types and concentrations of carbon sources induced the differences in germination rate. The germination rate increased with the increasing concentration of sucrose and glucose except for fructose. Sucrose and glucose showed the highest results at the concentration of 20%. While fructose demonstrated the similar tendency to sucrose and glucose, it reduced the germination rate at the concentration of 20%. The highest germination rate was observed at the concentration of 15%. The appropriate carbon course for germination of pollens of Platycodon grandiflorum for. duplex was glucose of which germination rate was twice as high as that of sucrose and fructose. The germination rate was reduced substantially when the pH was close to alkali, and the potential germination rate was obtained at pH 6. Boric acid enhanced the germination rate at a lower concentration than the higher concentration.

Thermal Distribution of Size-resolved Carbonaceous Aerosols and Water Soluble Organic Carbon in Emissions from Biomass Burning

  • The study of carbonaceous aerosols in the atmosphere is critical to understand the role of aerosols in human health and climate. Using standardized thermal optical transmittance methods, organic carbon (OC), elemental carbon (EC), and water soluble organic carbon (WSOC) were determined using a combustion sampling system for four types of agricultural crop residues (rice straw, red pepper stems, soybean stems, and green perilla stems) and eight types of forest trees (pine stems, pine needles, ginkgo stems, ginkgo leaves, maple stems, maple leaves, cherry stems, and cherry leaves). The aerosol particles between 0.056 and $5.6{\mu}m$ in size were analyzed using a Micro-Orifice Uniform Deposit Impactor (MOUDI). In the current study, the Carbonaceous Thermal Distribution (CTD) by carbon analyzer was discussed in order to understand the carbon fractions from the twelve types of biomass burning. Also, the concentration of OC, EC, WSOC, and water insoluble organic carbon (WIOC) detected in the emissions were described.

Preparation and Electrochemical Properties of Carbon Cryogel for Supercapacitor

  • Electrochemical properties of carbon cryogel electrode for the application of composite electrode materials mixed with metal oxide in supercapacitor have been studied. Carbon cryogels were synthesized by sol-gel polycondensation of resorcinol with form aldehyde, followed by a freeze drying, and then pyrolysis in an inert atmosphere. Physical properties of carbon cryogel were characterized by BET, X-ray diffraction (XRD) and scanning electron microscopy (SEM). It is found that carbon cryogel is amorphous material. The electrochemical properties of carbon cryogel were measured by cyclic voltammetry as a function of concentration of liquid electrolyte, galvanostatic charge-discharge with different scan rates and electrochemical impedance measurements. The result of cyclic voltammetry indicated that the specific capacitance value of a carbon cryogel electrode was approximately 150.2 F/g (at 5 mV/s in 6M KOH electrolyte).

  • This paper presents the thermoelastic analysis and 3-D failure analysis of the carbon/carbon brake disk. The mechanical properties of the carbon/carbon brake disk were measured for both in-plane and out of plane directions. The mechanical properties were used as the input of the thermoelastic analysis and 3-D stress analysis for the brake disk. The gap between rotor clip and clip retainer was an important parameter in the loading transfer mechanism of the rotor. The change of gap was considered separating the mechanical deformation and thermal deformation. Because the rotor clip and clip retainers were not contacted, the clip retainers and rivets were excluded from the rotor analysis model. The disk was modeled by using the cyclic symmetry condition and the contact problem between the rotor disk and rotor clip was considered. From the results of the 3-D stress analysis, the stress concentration at the key hole of the brake disk was confirmed.

$H_2S$ Adsorption Characteristics and Property Analyses of Activated Carbon Adsorbent Impregnated with Basic Solutions (염기성용액으로 첨착시킨 활성탄의 물성분석 및 $H_2S$ 흡착특성)

  • The $H_2S$ adsorption characteristics and property analyses of granular activated carbon adsorbent impregnated with basic solution such as NaOH, KOH, and $(CH_2CH_2OH)_2NH$ were investigated. The concentrations of NaOH and KOH reagent ranged over 1 to 5 M, The concentration of $(CH_2CH_2OH)_2NH$ was in the range of 0.1 to 1 M. Adsorption temperature($25{\sim}45^{\circ}C$) and adsorbate ($H_2S$) concentration (18.23 mg/L) were applied. The experimental results showed that the BET surface area of activated carbon impregnated with KOH decreases from $1,050\;m^2/g$ to $750\;m^2/g$, and the acidity of activated carbon impregnated with NaOH decreases from 0.541 meq/g-AC to 0 meq/g-AC, as the concentration of basic solution increases, while the pH of impregnated activated carbon increased from 9.54 to 10.94 for three basic solutions. It was also found that the $H_2S$ adsorption equilibrium capacity of activated carbon impregnated with NaOH, KOH, $(CH_2CH_2OH)_2NH$ increased with increasing temperature and $H_2S$ adsorption equilibrium capacity of the activated carbon impregnated with diethanolamine was much higher than other cases. At adsorption temperature of $45^{\circ}C$, the $H_2S$ adsorption equilibrium capacity of impregnated activated carbon was 2.0~3.3 times lager than that of pure activated carbon.

Monitoring of Particulate Matter and Analysis of Black Carbon and Some Particle Containing Toxic Trace in the City of Yaoundé, Cameroon

  • The concentration and composition of particulate matter (PM) in the atmosphere can directly reflect the environmental pollution. The atmospheric pollution in some Cameroonian cities is increasing with the industrial development and urbanization. Air pollution is inherently complex, containing PM of varied size and composition. This PM exists as a dynamic cloud interacting with sunlight and is modified by the meteorology. The reflectometer and the EDXRF spectrometry are applied to determine the concentration of some specific elements at four sites in the city of Yaound$\acute{e}$. The particular aim of the present work is to put in place data base on air pollution in urban area and elaborate regulations on the emissions issued to industrial and vehicle activities. This study provides an overview of the concentration of black carbon and some specific elements in the air, which have impacts on human health. The measurement was done by distinguishing the size of particle. So that, the particle with aerodynamic diameter between $2.5-10{\mu}m$ (so-called coarse particle) and aerodynamic diameter < $2.5{\mu}m$ (so-called fine particle) were considered to obtain more information about levels of the inhalable fraction of the location. The results obtained in four locations of the city of Yaound$\acute{e}$ show that the black carbon concentration is very considerable, the element sulfur is a major pollutant and the concentration of fine particle is very greater. The results obtained of fine and coarse filters range from $5-17{\mu}g/m^3$ and $10-18{\mu}g/m^3$ for the black carbon. S, Cu, Zn, Pb, Cd, As, Se and Hg are the specific findings of this work. The pollutants with a greater concentration are S, Pb, and Zn. These later seem to be non-uniformly, non-regular in some location and high compared to other countries. This work allows us to make a potential relation between pollutants and emission sources. In this framework, some suggestions have been proposed to reduce emissions for an improvement of the air quality in the environment and thus, the one of the city of Yaound$\acute{e}$.
