The heavy use of petroleum products in modern livings has brought ubiquitous environmental contaminants of aromatic compounds, which persist in aquatic and geo-environment without the substantial degradation. The persistence and accumulation of the aromatic compounds, which include xylene, phenol, toluene, phthalate, and so on are known to cause serious problems in our environments. Some of soil and aquatic microorganisms facilitate their growth by degrading aromatic compounds and utilizing degrading products as growth substrates, the biodegradation helps the reentry of carbons of aromatic compounds, preventing their accumulation in our environments. The metabolic studies on the degradation of aromatic compounds by microoganlsms were extensively carried out along with their genetic studies. A Rhodococcus sp. isolated in activated sludges has shown the excellent ability to grow on phenol as a sole carbon source. In the present study investigated a gene encoding phenol-degrading enzymes from a Rhodococcus sp.
The trace toxic metal copper was assayed using mercury immobilized on a carbon nanotube electrode (MCW), with a graphite counter and a reference electrode. In this study, a macro-scale convection motor was interfaced with a MCW three-electrode system, in which a handmade MCW was optimized using cyclic- and square-wave stripping voltammetry. An analytical electrolyte for tap water was used instead of an expensive acid or base ionic solution. Under these conditions, optimum parameters were 0.09 V amplitude, 40 Hz frequency, 0.01 V incremental potential, and a 60-s accumulation time. A diagnostic working curve was obtained from 50.0 to 350 ${\mu}g/L$. At a constant Cu(II) concentration of 10.0 ${\mu}g/L$, the statistical relative standard deviation was 1.78% (RSD, n = 15), the analytical accumulation time was only 60 s, and the analytical detection limit approached 4.6 ${\mu}g/L$ (signal/noise = 3). The results were applied to non-treated drinking water. The content of the analyzed copper using 9.0 and 4.0 ${\mu}g/L$ standards were 8.68 ${\mu}g/L$ and 3.96 ${\mu}g/L$; statistical values $R^2$ = 0.9987 and $R^2$ = 0.9534, respectively. This method is applicable to biological diagnostics or food surveys.
Long-term monitoring results of water qualities at major lakes in Korea showed COD (chemical oxygen demand) concentrations have been increasing while BOD (biochemical oxygen demand) concentrations have been decreasing during last decades. This was mainly due to refractory organic matters have been accumulated in the water body. In this study, the possible causes of COD concentration increase were evaluated. From the statistics, it can be understood that potent pollutant sources including fertilizer consumption, population, livestock, and carbon uptake have increased. Leaching tests were carried out with soils and biomasses sampled at agricultural-forestry area. From the leaching experiments, leachate qualities as a ratio of $COD_{Cr}/BOD$ were in the range of 2.5-5.0, implying that NOM (natural organic matters) discharged from the forestry area was mainly responsible for the COD accumulation. It can be understood from this research that diffuse pollutants from forestry areas should be controlled properly to reduce COD accumulation in the lakes.
Spatial variation of heavy metal accumulation was investigated in Manila clam Ruditapes philippinarum collected from several tidal flats. Sediment metal levels varied highly among the sites, which was attributed primarily to differences in Fe and organic carbon contents and in part to gain size. Significant differences in metal concentrations also were found in the clam tissue among the different sampling sites. However, except for a few metals (Mn, Zn, Pb), which showed some elevation, the variations in the clam tissue were not related to the variations in the sediment. This is likely because most metals in filter-feeding herbivores such as R. philippinarum accumulated as a result of feeding on suspended particles such as phytoplankton and organic detritus in the water column, not in bottom sediment. In addition, tissue weight for a specific shell size varied significantly among the sites, and increased tissue mass indicating a good nutritive condition likely caused a subsequent dilution of body metals leading to reduced weight-specific concentrations of some metals (Cd, Zn, Cu, Co).
Alumina microfibers with porous structures were prepared through hydrothermal reaction, and then used to modify the surface of carbon paste electrode (CPE). After modification with alumina microfibers, the electrochemical activity of CPE was found to be greatly improved. On the surface of alumina microfibers-modified CPE, the oxidation peak current of salvianolic acid B, a main bioactive compound in Danshen with anti-oxidative and anti-inflammatory effects, was remarkably increased compared with that on the bare CPE surface. The influences of pH value, amount of alumina microfibers and accumulation time were studied. Based on the strong signal amplification effects of alumina microfibers, a novel electrochemical method was developed for the detection of salvianolic acid B. The linear range was from 5 ${\mu}gL^{-1}$ to 0.3 mg $L^{-1}$, and the detection limit was 2 ${\mu}gL^{-1}$ (2.78 nM) after 1-min accumulation. The new method was successfully used to detect salvianolic acid B in ShuangDan oral liquid samples, and the recovery was over the range from 97.4% to 102.9%.
Bioaccumulation of polychlorinated biphenyls (PCBs) and organochlorine pesticides was studied in sediment dwelling bivalves, Manila clams (Ruditapes philippinarum), collected from the midwestern coast of Korea. As witnessed by the dominance of tetra- to penta-chlorinated congeners in sediments and the penta- to hexa-chlorinated congener dominance in clams, the profile of PCBs in the sediments and Manila clams differed. Lipid and organic carbon-normalized biota-sediment accumulation factors (BSAFs) were determined for organochlorine pesticides. BSAFs of $\beta$-hexachlorocyclohexane ($\beta$-HCH) and $\Sigma$DDTs were in the range of 0.06~1.36 and 0.31~1.06. No clear relationships were found between BSAFs of $\Sigma$DDTs in Manila clams and the concentrations of DDTs in the associated sediment. The accumulated PCBs and organochlorine pesticides were compared in Manila clams and oysters (Crassostrea gigas) collected from 3 sites. Highly chlorinated PCBs were more commonly found in oyster tissues than in clam tissues. The reasons for the different accumulation pattern of organic pollutants in the two organisms are discussed.

This study investigated the effect of high concentration of free ammonia on microalgal growth and substrate removal by applying real wastewater nitrogen ratio. To test of this, the conditions of free ammonia 1, 3, 6, 9, 12, 15 mg-N/L are compared. After 3 days of incubation, algal growth of Chlorella vulgaris and carbon removal rate are respectively lower in the reactors of FA 12, 15 mg-N/L compared to the others. This indicates that the high concentration of free ammonia, in this case, above 12 mg-N/L, has negative effect on algal growth and metabolic activity. Also, high concentration of free ammonia causes the proton imbalance, ammonium accumulation in algae and has toxicity for these reasons. So, we have to consider free ammonia in applying the microalgae to wastewater treatment system by the way of diluting wastewater or controlling pH and temperature.
Microalgae are attracting much attention as promising, eco-friendly producers of bioenergy due to their fast growth, absorption of carbon dioxide from the atmosphere, and production capacity in wastewater and salt water. However, microalgae can only accumulate large quantities of lipid in abiotic stress, which reduces productivity by decreasing cell growth. In this study, the strategy was investigated to increase cell viability and lipid production by overexpressing S-adenosylmethionine (SAM) synthetase (SAMS) in the microalga Chlamydomonas reinhardtii. SAM is a substance that plays an important role in various intracellular biochemical reactions, such as cell proliferation and stress response, and the overexpression of SAMS could allow cells to ithstand the abiotic stress and increase productivity. Compared to wild-type C. reinhardtii, recombinant cells overexpressing SAMS grew 1.56-fold faster and produced 1.51-fold more lipids in a nitrogen-depleted medium. Furthermore, under saline-stress conditions, the survival rate and lipid accumulation were 1.56 and 2.04 times higher in the SAMS-overexpressing strain, respectively. These results suggest that the overexpression of SAMS in recombinant C. reinhardtii has high potential in the industrial-scale production of biofuels and various other high-value-added materials.
In this study, we aimed to enhance the accumulation of chorismate (CHR) and anthranilate (ANT), key intermediates in the shikimate pathway, by modifying a shikimate over-producing recombinant strain of Corynebacterium glutamicum [19]. To achieve this, we utilized a CRISPR-driven genome engineering approach to compensate for the deletion of shikimate kinase (AroK) as well as ANT synthases (TrpEG) and ANT phosphoribosyltransferase (TrpD). In addition, we inhibited the CHR metabolic pathway to induce CHR accumulation. Further, to optimize the shikimate pathway, we overexpressed feedback inhibition-resistant Escherichia coli AroG and AroH genes, as well as C. glutamicum AroF and AroB genes. We also overexpressed QsuC and substituted shikimate dehydrogenase (AroE). In parallel, we optimized the carbon metabolism pathway by deleting the gntR family transcriptional regulator (IolR) and overexpressing polyphosphate/ATP-dependent glucokinase (PpgK) and glucose kinase (Glk). Moreover, acetate kinase (Ack) and phosphotransacetylase (Pta) were eliminated. Through our CRISPR-driven genome re-design approach, we successfully generated C. glutamicum cell factories capable of producing up to 0.48 g/l and 0.9 g/l of CHR and ANT in 1.3 ml miniature culture systems, respectively. These findings highlight the efficacy of our rational cell factory design strategy in C. glutamicum, which provides a robust platform technology for developing high-producing strains that synthesize valuable aromatic compounds, particularly those derived from the shikimate pathway metabolites.
There is general agreement that global warming is occurring and that the main contributor to this probably is the buildup of green house gasses, GHG, in the atmosphere. Two main contributors are the utilization of fossil fuels and the deforestation of many regions of the world. The burning of fossil fuels increases atmospheric carbon while the burning of fuel-wood reducing fossil fuel consumption along with its forest source maintain an atmospheric carbon level. The standing timber in the forests is a carbon sink, as are wood buildings and structures, and fossil fuel in the ground. This paper is designed to examine a number of current issues related to mitigating the global warming problem through forestry. For this purpose, we develop a modeling approach by integrating timber market, fossil fuel market and carbon cycling model. We use discrete time optimal control theory to identify optimal time paths, the laws of motion, and stationary stats solutions of endogenous variables in the model. On the basis of these results, we identify the optimal amounts of subsidies to be provided or taxes to be imposed by the regulatory agency to mitigate atmospheric carbon accumulation. We also present a numerical example to help illustrate the characteristics of variables in the model when the social cost for atmospheric carbon incrementally shifts upward. A surprising result is that the social cost function for atmospheric carbon has a very smaller impact on the optimal rotation period than previous literature suggested.
