Forty-six Holstein heifers were used in a completely randomized design and assigned to 1 of 2 treatments to evaluate the effects of 2 diets varying in ruminal fermentable carbohydrate sources, namely ground corn (GC) and rolled wheat (RW), on metabolism and performance of primiparous cows in the periparturient period. The heifers were fed diets as a total mixed ration (TMR) with similar energy and crude protein content including i) 18.57% GC, or ii) 18.57% RW from -24.13${\pm}$7.73 d relative to expected calving until calving. After calving, all animals received the same lactation diet until 28 d. Animals were group fed from the beginning of the study to -7 d relative to expected calving, fed individually from d -7 to 7 days in milk (DIM), and again group fed to 28 DIM. The pre-partum diets affected (p<0.05) dry matter intake (DMI), energy intake, energy balance (EB) and urinary pH during the last week pre-partum. There was no effect of pre-partum carbohydrate source on overall plasma concentration of glucose, nonesterified fatty acid (NEFA), $\beta$-hydroxybutyrate (BHBA), albumin, triglyceride (TG), cholesterol, aspartate aminotransferase (AST), insulin, and cortisol during the periparturient period. Cows fed the RW diet during the pre-partum period had greater calcium for the first week (p<0.05) and during 28 d (p = 0.08) of lactation compared with heifers fed the GC diet. Primiparous cows fed the RW diet produced greater milk protein content and yield (p<0.05). Primiparous cows fed the RW diet had lower milk urea nitrogen (MUN) and somatic cell count (SCC) than cows fed the GC diet (p<0.05). The results of this study show that feeding pre-partum diets with a rapidly fermentable source of starch but low energy content can improve animal metabolism and performance and smooth the transition of primiparous Holstein cows from gestation to lactation.
In spite of potential benefits of anther culture, low productivity of plant regeneration in some genotypes; e.g. tonsil and indica rice, is one of the major obstacles for practical use of anther culture. This study was conducted to improve cold shock method and carbohydrate source for increasing the efficiency of anther culture in rice. The most common carbon source, sucrose was replaced to maltose, which has two molecules of glucose. Maltose increased callus induction 1.4-to 1.8-fold higher in japonica rice, 3.2-to 11.6-fold in tongil types and 2.7-fold in indica rice IR50. Callus induction was increased from 0.2% to 12.5% in maltose medium compared to the medium supplemented with sucrose plus glucose in indica rice "Tetep". A simple procedure of vacuum packaging of panicles during cold shock treatment prolonged not only anther viability more than 15 days but also increased callus induction more than 2-fold compared to open-air storage (conventional method). Combining of above two methods, callus induction was increased 28 to 56% in japonica, 13 to 33% in tonsil type and 12 to 31% in indica rice. Plant regeneration was increased 14 to 35% in japonica, 10 to 20% in tonsil and 4 to 15% in indica rice, respectively.
Apparent digestibility coefficients (ADCs) of dry matter, crude protein, crude lipid, nitrogen-free extract, and energy in selected carbohydrate sources including wheat flour (WF), ${\alpha}-potato$ starch (PS), ${\alpha}-corn$ starch (CS), Na alginate (AL), dextrin (DEX), and carboxymethyl cellulose (CMC) were determined for olive flounder. The olive flounder averaging $150{\pm}8.0g$ were held in 300-L tanks at a density of 30 fish per tank. Chromic oxide was used as the inert marker. Feces were collected from the flounder by a fecal collector attached to a fish rearing tank. Apparent dry matter and energy digestibilities of flounder fed WF, PS, CS, and DEX diets were significantly higher than those of fish fed AL and CMC diets. Apparent crude protein digestibility coefficients of flounder fed PS and CS diets were significantly higher than those of fish fed AL, DEX, and CMC diets. Apparent crude lipid and nitrogen-free extract digestibility coefficients of flounder fed PS and DEX diets were significantly higher than those of fish fed WF, CS, AL, and CMC diets. The present findings indicate that PS and DEX could be effectively used as dietary carbohydrate energy compared to WF, CS, AL, and CMC for olive flounder.
Purpose: This study aimed to investigate the clinical and metabolic determinants of circulating soluble leptin receptor (CSLR) and free leptin index (FLI) in pre-pubertal obese male children. Methods: We conducted a preliminary cross-sectional study at three tertiary hospitals and one public primary school. Eighty obese male children without growth and developmental abnormalities aged 5-9 years were recruited. In these children, obesity was solely caused by excessive food intake, and not by acute illness, medications, endocrine abnormalities, or any syndrome. Body mass index (BMI), body fat mass, carbohydrate intake, fat intake, high density lipoprotein cholesterol level, low density lipoprotein cholesterol level, triglyceride level, and Homeostatic Model Assessment for Insulin Resistance are the potential determinants for leptin regulation, which is represented by CSLR level and FLI. Results: Carbohydrate was the main source of energy. BMI and body fat mass had negative weak correlation with CSLR and positive weak correlation with FLI. Furthermore, carbohydrate intake was found to be independently associated with CSLR based on the results of the multiple linear regression analysis. Following an increase in carbohydrate intake, CSLR level decreased progressively without any negative peak. Conclusion: Leptin regulation in prepubertal obese male children is associated with body composition and dietary intake. Carbohydrate intake is useful for predicting CSLR. Lipid profiles and insulin resistance are not related to both CSLR and FLI. Treatment and prevention of leptin resistance in obese children should focus on reducing BMI, fat mass, and carbohydrate intake.
This study was designed to evaluate the effects of fructose(F) or sucrose(S) and guar gum intake on carbohydrate and lipid metabolism in 15-week-old male Goto-Kakizaki(GK) rats. Fifty rats were randomly assigned to 5 groups which were different in carbohydrate(25% of carbohydrate) and fiber(5% w/w) sources. The carbohydrate(CHO) sources of each group were comstarch(control group, 100% of CHO), fructose with cellulose(F), fructose with guar gum(FG), sucrose with cellulose(S), and sucrose with guar gum(SG). Each group was fed exterimental diet for 4 weeks. We measured food intake, body weight gain, adipose tissues weight and organs weight. We conducted oral glucose tolerance test(OGTT) and measured plasma insulin concentration to examine carbohydrate metabolism. To evaluate lipid metabolism, we measured the lipid profile of plasma, liver and feces. Food intake and weight gain of FG or SG groups tended to be less than those of F or S groups. Perirenal and epididymal fat pad weights of SG group were significantly lower than those of S group and those of FG group tended to be lower than those of F group. In OGTT, blood glucose values of F or S groups were significantly higher than those of C group, and FG or SG groups tended to be lower than those of F or S groups during the experimental time. The area under the curve(AUC) of C group was significantly highest among the groups, AUC and plasma insulin concentration of FG or SG groups tended to be lower than those of F or S groups. Plasma and hepatic triglyceride (TG) of FG and SG groups were significantly lower than those of F and S groups, plasma and hepatic total lipid(TL) and total cholesterol(TC) of FG and SG groups tended to be lower than those of F and S groups. Fecal TL, TG and TC of FG or SG groups tended to be higher than those of F and S groups. In conclusion, intake of guar gum should improve carbohydrate and lipid metabolism in partial substitution of fructose or sucrose for cornstarch in GK rats.

A series of in vitro studies were carried out to determine i) the effects of enzyme and formaldehyde treatment on the degradation characteristics of carbohydrate and protein sources and on the synchronicity of these processes, and ii) the effects of synchronizing carbohydrate and protein supply on rumen fermentation and microbial protein synthesis (MPS) in in vitro experiments. Untreated corn (C) and enzyme-treated corn (EC) were combined with soy bean meal with (ES) and without (S) enzyme treatment or formaldehyde treatment (FS). Six experimental feeds (CS, CES, CFS, ECS, ECES and ECFS) with different synchrony indices were prepared. Highly synchronous diets had the greatest dry matter (DM) digestibility when untreated corn was used. However, the degree of synchronicity did not influence DM digestibility when EC was mixed with various soybean meals. At time points of 12 h and 24 h of incubation, EC-containing diets showed lower ammonia-N concentrations than those of C-containing diets, irrespective of the degree of synchronicity, indicating that more efficient utilization of ammonia-N for MPS was achieved by ruminal microorganisms when EC was offered as a carbohydrate source. Within C-containing treatments, the purine base concentration increased as the diets were more synchronized. This effect was not observed when EC was offered. There were significant effects on VFA concentration of both C and S treatments and their interactions. Similar to purine concentrations, total VFA production and individual VFA concentration in the groups containing EC as an energy source was higher than those of other groups (CS, CES and CFS). The results of the present study suggested that the availability of energy or the protein source are the most limiting factors for rumen fermentation and MPS, rather than the degree of synchronicity.
An excess application of N fertilizer causes physiological and morphological disorder known as ammonium ($NH_4{^+}$) toxicity in Chinese cabbage and it has been to be an issue for appropriate N fertilizer management. Hence, the pot experiment was conducted in order to evaluate the alleviating effect of the application of the easily decomposable carbohydrate on $NH_4{^+}$ toxicity in Chinese cabbage. Four levels of urea at 0, 160, 320, and $640kg\;N\;ha^{-1}$, represented as T1, T2, T3, and T4, respectively, were applied. In order to evaluate the alleviating effect of the application of the easily decomposable carbohydrate (sucrose) at T3 and T4 where $NH_4{^+}$ toxicity had occurred, five levels of sucrose were applied to meet C/N ratios of 0, 2, 4, 6, and 10, respectively. Our results showed that the $NH_4{^+}$ toxicity was observed at T3 and T4 at 5 days after treatment (DAT). $NH_4{^+}$ toxicity contributed to decrease fresh weight, length of leaves, length of root, and number of leaves significantly (p<0.05). The application of sucrose as a source of mitigating $NH_4{^+}$ toxicity had a good performance at T3 with the alleviating effect as 73 % and reduced in $NH_4{^+}-N$ content in soil at 29 DAT. In the maximum N rate of T4, however, sucrose application recovered it as 32 % only compared to T2 even though the same C/N ratio was treated. Consequently, sucrose as the easily decomposable carbohydrate played crucial role to reduce $NH_4{^+}$ concentration in soil and finally alleviated $NH_4{^+}$ toxicity in plant.

An efficient protocol for plant regeneration from protoplasts of the indica rice variety IR43 has been developed. The procedure involved plating of embryogenic suspension-derived protoplasts on the surface of a filter membrane overlaying agarose-embedded feeder cells. Lolium multiflorum cell suspensions were preferable to these of Oryza ridleyi as feeder cells and Lolium suspensions supported colony formation from up to 0.68% of the protoplasts, depending on the age of cell suspensions. Plant regeneration frequency was significantly improved by using maltose alone or in a 1:1(w/w) combination with sucrose as carbohydrate source and a simple dehydration treatment using a high concentration of agarose in the regeneration medium. Medium containing maltose or maltose mixed with sucrose increased the plant regeneration frequency compared with medium containing sucrose alone. The plant regeneration frequency was increased to 30.7 to 70.7% following dehydration treatment, while the non-treated controls showed a regeneration frequency of 3.1 to 30.6%. Protoplast-derived plants were transferred to the glasshouse, flowered with morphologically normal.
This study was conducted to evaluate the influence of early defoliation on photosynthesis and carbohydrate reserves when the source leaves of 'Fuji'/M9 apple (Malus domestica Borkh.) trees were removed during the growing period. Bud regrowth rates of 80%, 50% defoliation and non-defoliation treatments were significantly different 82.7%, 45.9% and 2.1% respectively at 30 days after treatment. In all treatments, sucrose and starch concentrations in remaining leaves decreased non-significantly during the 14-day period. No significant changes were observed for total soluble carbohydrates in non-defoliation and 50% defoliation. However, in 80% defoliated treatments, concentrations of sorbitol and total soluble carbohydrates in remaining leaves declined steadily during the 14-day period. It is thought that high sink strength increases the requirements of carbohydrate from remaining leaves more than non-defoliated. The concentrations of starch in the roots tend to decrease non-significantly as percentage of defoliation increased. Photosynthesis of remaining leaves was monitored during the 14-day period after partial defoliation treatments. Net photosynthetic rates (Pn) and stomatal conductance were significantly enhanced in the 80% defoliation. The observed photosynthetic enhancement following partial defoliation may have been due to the enhancement of osmotic potential in leaves. These results were estimated that increasing of photosynthetic rate in the partial defoliation is due to the sink carbohydrate requirements for the current year's secondary growth of buds.
