The composition of essential oil from O. javanica was analyzed by gas chromatography-mass spectrometry. Using the broth dilution method and disk diffusion test, anti-microbial activities of the oil fraction and its main components were evaluated against various antibiotic-susceptible and resistant strains of pathogenic microorganisms. As a result of GC-MS analysis, 57 compounds, including ${\alpha}-terpinolene$ (28.1%), dl-limonene (16.0%), ${\gamma}-terpinene$ (10.3%), ${\beta}-pinene$ (9.7%) and ${\alpha}-pinene$ (6.0%) were identified in the essential oil fraction. The essential oil fraction of O. javanica and its main components exhibited significant inhibitory activities, particularly against Candida albicans (antibiotic-susceptible strains) and Streptococcus pneumoniae (antibiotic- susceptible and resistant strains). The main components of the O. javanica oil fraction displayed different patterns of activity against the three tested Candida species as exemplified by the differential minimum inhibiting concentration (MIC) values. The disk diffusion test showed that the activities were dose dependent.

The effectiveness of three antifungal antibiotics amphotericin B, clotrimazole and 5-fluorocytosine was tested against 120 clinical isolated Candida species. The minimum inhibitory concentrations of amphotericin B was considerably lower than those of clotrimazole and 5-fluorocytosine. High-level resistance to 5-fluorocytosine was present in 50% of the isolates. The combination of clotrimazole and 5-fluorocytosine produced synergistic inhibition against all 20 strains of Candida albicans tested that were relatively resistant to both antifungal agents.

Oak tree death caused by symbiosis of an ambrosia beetle, Platypus koryoensis, and an ophiostomatoid filamentous fungus, Raffaelea quercus-mongolicae, has been a nationwide problem in Korea since 2004. In this study, we surveyed the yeast species associated with P. koryoensis to better understand the diversity of fungal associates of the beetle pest. In 2009, a total of 195 yeast isolates were sampled from larvae and adult beetles (female and male) of P. koryoensis in Cheonan, Goyang, and Paju; 8 species were identified by based on their morphological, biochemical and molecular analyses. Meyerozyma guilliermondii and Candida kashinagacola were found to be the two dominant species. Among the 8 species, Candida homilentoma was a newly recorded yeast species in Korea, and thus, its mycological characteristics were described. The P. koryoensis symbiont R. quercusmongolicae did not show extracelluar CM-cellulase, xylanase and avicelase activity that are responsible for degradation of wood structure; however, C. kashinagacola and M. guilliermondii did show the three extracellular enzymatic activities. Extracelluar CM-cellulase activity was also found in Ambrosiozyma sp., C. homilentoma, C. kashinagacola, and Candida sp. Extracelluar pectinase activity was detected in Ambrosiozyma sp., C. homilentoma, Candida sp., and M. guilliermondii. All the 8 yeast species displayed compatible relationships with R. quercus-mongolicae when they were co-cultivated on yeast extract-malt extract plates. Overall, our results demonstrated that P. koryoensis carries the yeast species as a symbiotic fungal associate. This is first report of yeast diversity associated with P. koryoensis.
Candida is one of the most common causes of bloodstream infections and a leading cause of morbidity and mortality among hospitalized patients. The purpose of this study was to provide important information for formulating empirical treatment plans for candidemia by investigating the antifungal resistance rate of Candida. Among the Candida strains (973 cases) isolated from blood culture tests at the S hospital in 2009~2018, 4.7% (N=44) comprising the Candida spp. (932 strains) showed resistance to fluconazole. The resistant strains included C. albicans, C. parapsilosis, C. tropicalis, and C. glabrata. In addition Candida spp. (947 strains) showed resistance to amphotericin B (N=6, 0.6%), flucytosine (N=23, 2.4%) and voriconazole (N=24, 3.1%). C. albicans was resistant to fluconazole (N=23, 6.9%) and voriconazole (N=21, 6.0%), The statistical analysis showed that C. albicans and non-albicans Candida species were resistant to fluconazole (P=0.039) and voriconazole (P<0.001). A monitoring system to understand the rate of candidiasis infections in a hospital setting is required. It is also important to make the right choice of the antifungal agent based on drug susceptibility patterns. Therefore, an infection surveillance policy that tracks Candida resistance through regular antifungal susceptibility tests is necessary.
5-Arylamino-6-chloro-4.7-dioxoindazoles (DZs) were newly synthesized for the evaluation of antifungal activities. The compounds DZs were prepared by regioselective nucleophilic substitution of 5.6-dichloro-4.7-dioxoindazoles with appropriate arylamines in high yield. DZs were tested for their growth inhibitory activities against Candida species and Aspergillus niger. The MIC values were determined by the two-fold dilution method. In general. DZs showed in vitro antifungal activities. Among the tested compounds, DZ1, 3, 6, 7 and 12 showed potent antifungal activities against Candida species and Aspergillus niger. DZ7 was the most effective in preventing the growth of Candida species.
Objectives : This experimental study was performed to investigate the efficacy of Fel Ursi, Bovis Calculus & Moschus pharmacopuncture solution manufactured with freezing dryness method to use eye drop. Methods : After administering Fel Ursi, Bovis Calculus & Moschus pharmacopuncture solution on bacterial species(Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans) which cause Keratitis, MIC(Minimum Inhibition Concentration) and the size of inhibition zone were measured. Anti-bacterial potency was also measured using the size of inhibition zone. Results : There was no response to MIC on bacterial species(Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans) after Fel Ursi, Bovis Calculus & Moschus pharmacopuncture solution was medicated. Conclusions : The present study suggests that Fel Ursi, Bovis Calculus & Moschus pharmacopuncture solution dosen't have anti-bacterial effects on bacterial species(Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans) which cause Keratitis. These study result recommends that more research on other herbal medicines of eye drop for Keratitis are required.

The role of lactic acid bacteria (LAB) in honey as antifungal activity has received little attention and their mechanism of inhibitory of fungi is not fully understood. In this study, LAB were isolated from honey samples from Malaysia, Libya, Saudi Arabia, and Yemen. Twenty-five isolates were confirmed LAB by catalase test and Gram staining, and were screened for antifungal activity. Four LAB showed inhibitory activity against Candida spp. using the dual agar overlay method. And they were identified as Lactobacillus plantarum HS isolated from Al-Seder honey, Lactobacillus curvatus HH isolated from Al-Hanon honey, Pediococcus acidilactici HC isolated from Tualang honey and Pediococcus pentosaceus HM isolated from Al-Maray honey by the 16S rDNA sequence. The growth of Candida glabrata ATCC 2001 was strongly inhibited (>15.0 mm) and (10~15 mm) by the isolates of L. curvatus HH and P. pentosaceus HM, respectively. The antifungal activity of the crude supernatant (cell free supernatant, CFS) was evaluated using well diffusion method. The CFS showed high antifungal activity against Candida spp. especially The CFS of L. curvatus HH was significantly (p < 0.05) inhibited growth of C. glabrata ATCC 2001, C. parapsilosis ATCC 2201, and C. tropicalis ATCC 750 with inhibitory zone 22.0, 15.6, and 14.7 mm, respectively. While CFS of P. pentosaceus HM was significantly (p < 0.05) effective against C. krusei, C. glabrata, and C. albicans with inhibition zone 17.2, 16.0, and 13.3 mm, respectively. The results indicated that LAB isolated from honey produced compounds which can be used to inhibit the growth of the pathogenic Candida species.

The antifungal activities of amphotericin B, 5-fluorocytosine, and ketoconazole in combination of amphotericin B/ketoconazole and 5-fluorocytosine/ketoconazole were determined against 42 strains of Candida spp. isolated from oral cavity. Among 42 strains of Candida species, 36 strains of Candida albicans, 2 strains of Candida parapsilosis and Candida tropicalis 1 strain of Candida krusei and Candida stellatoidea were identified. The minimum inhibitory concentrations(MICs) of amphotericin B, 5-fluorocytosine and ketoconazole for these strains were ranged from 0.05-1.56 mcg/ml, 12.5->100.0 mcg/ml and 0.2-50.0 mcg/ml. In all of the experimental strains, amphotericin B had the greatest antifungal activity on a dilution basis. When a microtiter checkerboard technique was used 5-fluorocytosine acted synergistically with ketoconazole against all strains, whereas amphotericin B has a reduced effect. The killing curve experiments with on strain of Candida albicans WMC-85024 demonstrated that the combination of amphotericin B/ketoconazole and 5-fluorocytosine/ketoconazole produced a decrease in number of colony forming unit of >3 logs in 72 hours.
Objectives : This experimental study was performed to investigate the effectiveness of Fel Ursi Pharmacopunture solution(FUPS) manufactured by using alcohol/water extraction method. To identify the use of it as eyedrops, antibacterial test on Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, and Candida albicans was performed. Methods : After treatment FUPS on bacterial species(Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans) which cause Keratitis, we investigated anti-bacterial effects of FUPS on Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans in terms of measuring MIC and size of inhibition zone respectively. Results : After FUPS was treated, significant changes of MIC on Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum and Candida albicans were not observed at all. Conclusions : The present study suggests that FUPS doesn't have anti-bacterial effects on Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum and Candida albicans which cause Keratitis. Perhap These results recommend that FUPS doesn't have anti-bacterial effects but have other mechanism which suppress inflammation.
The goal of this study was to isolate wild yeasts from reed fields in the Seocheon-gun county in Chungcheongnam-do, South Korea. Molecular analysis of the D1/D2 domain of the 26S rDNA of the yeasts isolated from soil samples was performed using Basic Local Alignment Search Tool (BLAST). In total, 20 yeast strains from 11 species were isolated from 20 different soil samples. Candida species were observed to be the most common yeasts. Of these 20 strains, Candida subhashii (6 strains), Candida tropicalis (3 strains), Candida boleticola MS008, and Lachancea thermotolerans (2 strains) were identified as alcohol-fermenting yeasts. Further, Bullera japonica YJ10-1, Candida subhashii J7-1, Kluyveromyces yarrowii YJ11-1, and Ustilago shanxiensis Y10-1 were newly recorded yeast strains in Korea, and therefore, their microbiological characteristics were investigated further. All of these unrecorded yeast strains had oval cells; only Candida subhashii J7-1 formed ascospores and pseudomycelia. Kluyveromyces yarrowii YJ11-1 grew in vitamin-free medium, and all of these strains, except Candida subhashii J7-1, grew in 5% NaCl-containing YPD broth.
