Anti-Proliferative Activity of Ethanol Extracts from Taxilli Ramulus (Taxillus chinensis (DC.) Danser) Through Cyclin D1 Proteasomal Degradation in Human Colorectal Cancer Cells

  • In this study, we elucidated anti-cancer activity and potential molecular mechanism of 70% ethanol extracts from Taxilli Ramulus (Taxillus chinensis (DC.) Danser) (TR-E70) against human colorectal cancer cells. Anti-cell proliferative effect of TR-E70 was evaluated by MTT assay. The effect of TR-E70 on the expression of cyclin D1 in the protein and mRNA level was evaluated by Western blot and RT-PCR, respectively. TR-E70 suppressed the proliferation of human colorectal cancer cell lines, HCT116 and SW480. Although TR-E70 decreased cyclin D1 expression in protein and mRNA level, decreased level of cyclin D1 protein by TR-E70 more dramatically occurred than that of cyclin D1 mRNA. Cyclin D1 downregulation by TR-E70 was attenuated in presence of MG132. In addition, TR-E70 phosphorylated threonine-286 (T286) of cyclin D1. TR-E70-mediated cyclin D1 degradation was blocked in presence of LiCl as an inhibitor $GSK3{\beta}$ but not PD98059 as an ERK1/2 inhibitor and SB203580 as a p38 inhibitor. Our results suggest that TR-E70 may downregulate cyclin D1 as one of the potential anti-cancer targets through $GSK3{\beta}$-dependent cyclin D1 degradation. From these findings, TR-E70 has potential to be a candidate for the development of chemoprevention or therapeutic agents for human colorectal cancer.

Conditional PTEN-deficient Mice as a Prostate Cancer Chemoprevention Model

  • Background: We generated a mouse model of prostate cancer based on the adult-prostate-specific inactivation of phosphatase and tensin homolog (PTEN) using the Cre-loxP system. The potential of our mice as a useful animal model was examined by evaluating the chemopreventive efficacy of the anti-androgen, chlormadinone acetate (CMA). Materials and Methods: Six-week-old mice were treated subcutaneously with $50{\mu}g/g$ of CMA three times a week for 9 or 14 weeks and sacrificed at weeks 15 and 20. Macroscopic change of the entire genitourinary tract (GUT) and histologically evident prostate gland tumor development were evaluated. Proliferation and apoptosis status in the prostate were examined by immunohistochemistry. Results: CMA triggered significant shrinkage of not only the GUT but also prostate glands at 15 weeks compared to the control (p=0.017 and p=0.010, respectively), and the trend became more marked after a further five-weeks of treatment. The onset of prostate adenocarcinoma was not prevented but the proliferation of cancer cells was inhibited by CMA, which suggested the androgen axis is critical for cancer growth in these mice. Conclusions: Conditional PTEN-deficient mice are useful as a preclinical model for chemoprevention studies and serve as a valuable tool for the future screening of potential chemopreventive agents.

  • Based on the success of green tea as a cancer preventive, herbal medicines are now also attracting attention as potential sources of cancer preventive agents. Using inhibition of TNF-$\alpha$ release assay, we studied Acer nikoense (Megusurino-ki in Japanese): Inhibitory potential was found in the leaf extract, and the main active constituents were identified as geraniin and corilagin. The $IC_{50}$/ values for TNF-$\alpha$ release inhibition were 43 $\mu$M for geraniin and 76 $\mu$M for corilagin, whereas that for (-)-epigallocatechin gallate (EGCG)was 26 $\mu$M.(omitted)

Activating Transcription Factor 3 is a Molecular Target for Apoptotic Effect of Silymarin in Human Colorectal Cancer Cells

  • Apoptosis has been regarded as a therapeutic target because apoptosis is typically disturbed in human cancer. Silymarin found in the seeds of the milk thistle (Silybum marianum) has been reported to exert anti-cancer properties through apoptosis. This study was performed to investigate the molecular target for silymarin-mediated apoptosis in human colorectal cancer cells. Silymarin reduced the cell viability and induced an apoptosis in human colorectal cancer cells. ATF3 overexpression increased PARP cleavage by silymarin. Increased ATF3 expression in both protein and mRNA was observed in silymarin-treated cells. In addition, silymarin increased the luciferase activity of ATF3 promoter. Inhibition of JNK and IκK-α blocked silymarin-mediated ATF3 expression. The results suggest that silymarin induces apoptosis through JNK and IκKα-dependent ATF3 expression in human colorectal cancer cells.

유황시비처리가 열무의 Quinone Reductase 유도물질 생성에 미치는 영향 (Quinone Reductase Inducer from Radish Leaf Cultivated in the Soil Containing Sulfur)

  • 본 연구는 열무에서 다른 십자화과 식물에서 발견되는 sulforaphane 유사물질의 분획과 quinone reductase inducing activity를 측정하였고 열무생육시 유황을 첨가해 주면 이러한 활성이 현저히 증가되는 것을 보았다. Sulforaphane 유사물질의 분획은 유황을 토양에 1,818 g/㎥ 첨가시 가장 높았고, 열무의 생산성을 고려하여 토양을 석회로 중화시켰을 때 감소되었다. 유황을 처리하여 키운 열무에 생리활성성분을 가진 유황화합물이 증가되므로 이러한 열무는 생체에 섭취시 detoxification enzyme을 유도하여 암발생을 지연시키거나 낮출 수 있는 chemoprevention의 효율이 기대된다.

Recently, considerable attention has been focused on the role of cyclooxygenase-2 (COX-2) in the carcinogenesis as well as inflammation.   • Nutrition influences cancer incidence and offers a variety of preventive dietary factors including non-nutritive plant metabolites. To identify novel potential chemopreventive agents, we have set up cell- and enzyme-based in vitro marker systems relevant for prevention of carcinogenesis in vivo. This experimental approach led to the identification of Xanthohumol (Xn), a prenylated chalcone from hop (Humulus lupulus L.) as a most promising broad-spectrum chemopreventive agent.(omitted)

  • Persistent cellular oxidative stress and enhanced lipid peroxidation (LPO) of PUFAs, leading to macromolecular damage and disruption of signaling pathways, are implicated in the development of human malignancies and other chronic degenerative diseases. LPO generates by oxidation of linoleic acid (LA) or arachidonic acid ($\omega$ -6 PUPAs) reactive aldehydes, such as trans-4-hydroxy-2-nonenal, which form etheno $\varepsilon$ -DNA adducts in a variety of human tissues and thus can contribute to diet-related cancers.(omitted)
