Extrahepatic cholangiocarcinoma (ECC), a malignant tumor of biliary origin, has a poor prognosis with limited treatment options. The KRAS oncogene is the most commonly mutated gene in ECC and one of the factors that predicts a poor prognosis and low survival rate. L1 cell adhesion molecule (L1CAM) is expressed in ECC cells and acts as an independent poor prognostic factor in predicting patient survival. In this study we investigate the functional significance of L1CAM in ECC cells with activating KRAS mutation. We selected an ECC cell line, EGI-1, with activating KRAS mutation, and then confirmed its expression of L1CAM by RT-PCR, western blot analysis, and flow cytometry. The suppression of L1CAM expression (using a specific lentivirus-delivered shRNA) significantly decreased the migratory and invasive properties of EGI-1 cells, without altering their proliferation or survival. Analyses of signaling effectors in L1CAM-depleted and control EGI-1 cells indicated that L1CAM suppression decreased the levels of both phosphorylated MKK4 and total MKK4, together with c-Jun N-terminal kinase (JNK) phosphorylation. Further, exposure to a JNK inhibitor (SP600125) decreased migration and invasion of EGI-1 cells. These results suggest that L1CAM promotes cellular migration and invasion via the induction of MKK4 expression, leading to JNK activation. Our study is the first to demonstrate a functional role for L1CAM in ECC carrying the activating KRAS mutation. Given that KRAS is the most commonly mutated oncogene in ECC, L1CAM may serve as an attractive therapeutic target for ECC cells with activating KRAS mutation.
PURPOSE. To evaluate the effect of surface treatments and repair materials on the shear bond strength and to measure the fracture toughness of CAD/CAM provisional restoration materials. MATERIALS AND METHODS. Four CAD/CAM (3D printing: Nextdent C&B and ZMD-1000B Temporary, CAD/CAM resin block: Yamahachi PMMA disk and Huge PMMA block) and four conventional (monometacrylate: Jet and Alike, dimetacrylate: Luxatemp and Protemp 4) materials were selected to fabricate disk-shaped specimens and divided into six groups according to surface treatment (n=10). CAD/CAM materials were repaired with Jet or Luxatemp, while conventional materials were repaired with their own materials. The shear bond strength was measured by using universal testing machine. Ten rectangular column-shaped specimens for each material were fabricated to measure the fracture toughness by single edge v notched beam technique. Statistical analysis was performed by one-way ANOVA. RESULTS. The highest shear bond strength of CAD/CAM materials was achieved by SiC paper + sandblasting. It was also accomplished when repairing 3D printing materials with Luxatemp, and repairing CAD/CAM resin blocks with Jet. Yamahachi PMMA disk showed the highest fracture toughness. Nextdent C&B showed the lowest fracture toughness value but no statistically significant difference from Alike and Luxatemp (P>.05). CONCLUSION. In order to successfully repair the CAD/CAM provisional restoration, mechanical surface treatment and appropriate repair material according to the CAD/CAM material type should be selected. The CAD/CAM provisional materials have proper mechanical properties for clinical use as compared to conventional materials.

Lunar permanently shadowed regions (PSRs) never see direct sunlight and are illuminated only by secondary illumination - light reflected from nearby topography. The ShadowCam imaging experiment onboard the Korea Pathfinder Lunar Orbiter is acquiring images of these PSRs. We characterize and discuss the nature of secondary illumination for the Shackleton PSR from ShadowCam radiance-calibrated images. We also use modeling to understand the magnitude and direction of the secondary illumination. Results from our analysis highlight the non-homogeneous, dynamic, and complex nature of PSR secondary lighting. Knowledge of the direction of the secondary illumination is crucial for reli-able interpretation of contrasts observed in ShadowCam images. This preliminary analysis of the floor of Shackleton crater from images acquired over multiple secondary illumination conditions does not reveal indications of exposed surface ice, even though temperatures are constantly below 110K.
목적: Biomet 3i 시스템과 Astra 시스템의 내측 연결형 임플란트 고정체에 국내에서 제작된 맞춤형 CAD-CAM 지대주를 체결하여 구성 성분 간의 적합성과 나사의 안정성을 평가하고자 하였다. 재료 및 방법: 24개 3i 임플란트 중 12개의 임플란트에 기성 지대주를 연결하고, 나머지 임플란트에는 맞춤형 CAD-CAM 지대주를 연결하여 각각 Group 1과 Group 2로 분류하였다. 동일 개수와 동일한 방법으로 Astra 임플란트를 각각 Group 3와 Group 4로 분류하였다. 각각의 고정체에 지대주를 장착하여 각 계면 사이의 적합성을 micro-CT로 관찰하고 초기 풀림 토크를 측정한 후 평가하였다. 결과: 고정체-지대주의 접촉길이는 Group 1과 Group 2 사이 뿐만 아니라 Group 3와 Group 4 사이에도 유의한 차이가 없었다(Mann-Whitney test, P>.05). 하지만, 지대주와 나사의 계면 및 고정체와 나사의 계면에서 Group 2와 Group 4가 Group 1와 Group 3에 비해 각기 더 큰 접촉양상을 보여주었다(Mann-Whitney test, P<.05). 또한, Group 2와 Group 4가 Group 1과 Group 3에 비해 각기 더 낮은 나사의 초기 풀림 토크를 기록하였다(Student t-test, P<.05). 결론: CAD-CAM지대주는 임상에 적용할 만한 적합성을 지니고 있으나, 광범위한 임상 적용을 위해서는 풀림 토크가 향상되어야 할 것으로 사료된다.
With the contemporary CAD/CAM system, where the tool path is generated and verified purely based on the geometric operation, geometric accuracy of the machined surface cannot be guaranteed dut to the cutting mechanics, meaning that the cutting mechanics should be incorporated in some fashion. In this paper, we incorporate the instantaneous cutting force and the tool deflection phenomena in predicting the machined surface for the finish-cut and milling operation. For the given NC dat including cutting conditions, the developed algorithm computes cutting force and deflection amount along the tool trajectory, and outputs the 3D graphic model of the machined surface together with error analysis. The validity and accuracy of the presented method has been tested by the actual cutting experiments. Experimental results and accuracy enhancement method together with implementing architecture of the VMCS (Virtual Machining CAM System) are discussed in the paper.
The latch needle cam system of circular knitting machines is analysed using multibody dynamics. A formulation is made to obtain the vertical stiffness between the needle and the cam. By implementing this formulation into the data of the multibody dynamics program, the motion of the needle is described and the forces and impulses between the needle and the cam are obtained.
In this paper, performance of a dry CVT vehicle with torque cam is investigated. Based on belt transmission mechanism and torque cam actuator characteristics, an algorithm which is able to calculate the speed ratio from given thrust and load torque is suggested and a 3-D map is constructed for the speed ratio-thrust-torque. Using the 3-D map, a steady state characteristic of torque cam and a transient response of a dry CVT vehicle are investigated by simulation. It is found that initial spring compression plays a key role in determining the sliding criteria and the speed ratio velocity.
The cam is used to a main component in a variety of automatic machines and instruments. To meet the demand of a complicated operation and of reducing weight for automatic machine, Curve Blending Technology, in which each of the basic curves suitable for individual interval is connected, is used for the cam design. In the curve blending, it is necessary to select appropriate elementary curve for each interval and to confirm the dynamic continuity at connecting points between adjoining elementary curves. This paper represented the elementary curve selection method to select an appropriate curve for each interval, and executed computation for the follower displacement and angular displacement of each interval. The paper made an analysis and examine closely for elementary curves to synthesizing curve blending, and it performed dynamic conditions clearly at every points on the cam motions. Therefore the curve blending technology presented by the paper turned into easier work.
When restoring a tooth, the dentist tries to choose the ideal material for existing situation. One criterion that is considered is its suitability for restoring coronal strength. As more tooth structure is removed, the cusps are weakened and susceptible to fracture. Further, this increased deformation may cause the formation of intermittent gaps at the margin between the hard tissue and the restoration, facilitating marginal leakage. The improvements in ceramic materials now make it possible for alternatives to amalgams, composites, and cast metal to be of offered for posterior teeth. Of the materials used, ceramics most closely approximates the properties of enamel. The introduction of computer-aided design/computer-aided manufacture(CAD/CAM) systems to restorative dentistry represents a major technological breakthrough. It is possible to design and fabricate ceramic restorations at a single appointment. Additionally, CAD/CAM systems eliminate certain errors and inaccuracies that are inherent to the indirect method and provide an esthetic restoration. The aim of this investigation was to study the loading characteristics of CAD/CAM ceramic inlay and to compare the stress distribution and displacement associated with different designs of cavity(the isthmus width and cavity depth). A human maxillary left first premolar was prepared with standard mesio-occlusal cavity preparation, as recommended by the manufacturer Ceramic inlay was fabricated with CEREC 2 CAD/CIM equipment and cemented into the prepared cavity. Three dimensional model was made by the serial photographic method. The cavity width was varied $\frac{1}{3}$, $\frac{1}{2}$ and $\frac{2}{3}$ of intercuspal distance between buccal and lingual cusp tip. The cavity depth was varied 1.5mm and 2.3mm. So six models were constructed to simulate six conditions. A point load of 500N was applied vertically onto the first node of the lingual slope from the buccal cusp tip. The stress distribution and displacement were solved using ANSYS finite element program(Swanson Analysis System). (omitted)
Objectives: As effective treatments for dementia are lacking in Western medicine, complementary and alternative medicine (CAM) is considered a useful option. While the quality of life (QoL) is a vital outcome for patients with dementia, the QoL of patients receiving CAM for dementia remains ambiguous. This study aimed to determine the effect of CAM on QoL outcomes in dementia patients. Methods: A search was performed using the keywords "dementia," "Alzheimer's," "cognitive impairment," "Chinese," "Korean," "oriental," "herbal," "acupuncture," and "quality of life". All quantitative data were synthesized using R version 4.1.1. Results: Twenty-five randomized controlled trials (RCTs), 16 pre-post trials, and two cohort studies were selected for the systematic review. QoL in Alzheimer's disease (QOL-AD) (n=11, 25.6%) and geriatric QoL in dementia (GQOL-D, n=9, 20.9%) were the most utilized QoL instruments. Significant benefits in QoL were observed after receiving mind, body, combined mind and body, nursing, oriental medicine, and acupuncture therapies. In the meta-analysis, the combined effect was shown to significantly increase QOL-AD compared to before CAM interventions (standardized mean difference, SMD: 0.507; 95% confidence interval (CI), 0.191~0.824; p<0.01). The overall synthesized estimates in the GQOL-D showed a significantly improved QoL (SMD: 0.537, 95% CI: 0.238~0.837 p<0.01; one group; SMD: 1.465, 95% CI: 0.934~1.996, p<0.01). The seven studies assessing the cost-effectiveness of CAM reported uncertain outcomes. Conclusions: This study showed that CAM interventions benefited patients with dementia by improving their QoL. While additional standardized research is required, CAMs are suggested as effective clinical management for patients with dementia. They are also suggested as complementing therapies for these patients.
