Chicken Insulin-Like Growth Factor-I Stimulates Protein Synthesis of Chicken Embryo Myoblasts Cultured in Serum-Free Medium

  • The effect of chicken IGF-I on protein synthesis of chicken embryo myoblasts cultured in serum-free medium was examined. When myoblasts were expanded to approximate 20-30% of well, the medium was changed to the serum-free medium including 0, 2, 20, 200 or 2000 ng/ml of recombinant chicken IGF-I. The culture medium including 10% fetal calf serum (FCS) was used as positive control. After 1 day of incubation, protein synthesis was measured by the incorporation of [$^3H$]-L-leucine. Thereafter cells were continued to incubate for further 18 hours, and the radioactivity in the protein was measured as an index of protein synthesis. The values for protein synthesis cultured in the serum-free medium without chicken IGF-I or with 2000 ng/ml of chicken IGF-I were the lowest. Protein synthesis was elevated with increasing chicken IGF-I concentration from 0 to 20 ng/ml. The values for protein synthesis in the 20 ng/ml and 200 ng/ml IGF-I groups were about half of that of the FCS group. The present study revealed that the potency of chicken IGF-I at the levels of 20 to 200 ng/ml to stimulate myoblast protein synthesis was about half of that of 10% FCS.

A quantitative study of the transfer of colostral immunoglobulins to the newborn Korean native calf (한우 송아지의 초유섭취에 의한 수동면역 획득 상태)

  • The levels of the serum and colostral total protein and immunoglobulins of the Korean native cows immediately after parturition and colostrum-conferred passive immune status of the Korean native calves were studied, and the results obtained were summerized as follows: The mean total protein, total immunoglobulin, IgG, IgM, and IgA concentrations in sera of 10 Korean native cows immediately after parturition were $6.8{\pm}0.4g/dl$, $24.39{\pm}3.41mg/ml$, $19.49{\pm}2.70mg/ml$, $4.56{\pm}1.14mh/ml$, and $0.35{\pm}0.08mg/ml$ respectively. And the mean total protein, total immunoglobulin, IgG, IgM, and IgA concentrations in colostrum were $15.0{\pm}1.1g/dl$, $116.13{\pm}23.07mg/ml$, $101.51{\pm}22.59mg/ml$, $9.46{\pm}1.99mg/ml$, and $5.17{\pm}1.59mg/ml$ respectively. The mean concentrations of total protein and immunoglobulins in colostrum were 2.1 times to 14.8 times higher than those in serum. The mean concentrations of total protein, total immunoglobuln, IgG, IgM, and IgA in sera from 211 Korean native calves at 2 days old were $6.7{\pm}0.9g/dl$, $35.74{\pm}9.56mg/ml$, $29.06{\pm}8.07mg/ml$, $4.91{\pm}2.99mg/ml$, and $1,9{\pm}0.99mg/ml$ respectively. Although the calves had been suckled the dam's colostrum, each of calves was considerably various in levels of serum total protein and immunoglobulins and 16.1 percent of the calves were in a state of hypoglobulinemia.

Gene Gun-Mediated Human Erythropoietin Gene Expression in Primary Cultured Oviduct Cells from Laying Hens

  • Factors affecting gene gun-mediated expression of the human erythropoietin gene were investigated in primary cultured oviduct cells from laying hens. The human erythropoietin gene was transfected by a gene gun method at $1.25{\mu}g$ per dish, and cultured in a synthetic serum-free medium for 72 hrs. The concentration of human erythropoietin mRNA was determined by RNA : RNA solution hybridization. In experiment 1, the effect of changing the shooting pressure of DNA-coated microparticles with nitrogen gas was tested at 20 and $60kgf/cm^2$. The results showed that the erythropoietin mRNA concentration was significantly higher at 60 than $20kgf/cm^2$. In experiment 2, the effects of supplementing the medium with fetal calf serum at 10%, and raising the shooting pressure from 60 to $80kgf/cm^2$ on the cell number and erythropoietin gene expression were examined. Although supplementation with fetal calf serum significantly increased the cell numbes compared with no supplemented controls (p < 0.05), erythropoietin mRNA concentration per $10^3$ cells was not affected. Raising the shooting pressure from 60 to $80kgf/cm^2$ did not affect either of the parameters, In experiment 3, the effect of supplementing ascorbate 2-phosphate at 0.5 mM was tested. The results indicated that the ascorbate supplementation significantly increased the cell number (p < 0.05), and tended to increase erythropoietin mRNA concentration (p < 0.1). Thus, for human erythropoietin gene expression by using the gene gun method, shooting pressure with nitrogen gas should be sufficient at $60kgf/cm^2$ and supplementation with ascorbate phosphate would be useful to enhance not only the cell proliferation but also erythropoietin gene expression.

Transfer, Cryopreservation and Production of Bovine Embryos Cultured in Serum-Free System (Serum-Free Medium에서 배양한 한우 배의 내동성과 이식)

  • Serum-contain is commoly used for the production of in vitro-derived bovine embryos. However, were biological activity of serum varies from lot to lot, time consuming to choose better serum with good quality and risks of virus, bacteria and mycoplasma infection. This study established serum-free culture systems of in vitro embryo development to efficiently obtain a large number of blastocysts from ovaries of Hanwoo and oocytes maturation, cell number, tlerance of cryopreservation. Secondly, serum-contain medium is suspected of contributing to the large calf size, dystocia, cersarean sections, calf mortality and confirmed these blastocysts are high quality in terms of cyotolerance, high rates of pregancy and normal birth. For these reasons, Culture media (IVMD101 and IVD101) designed specifically for the preimplantation bovine embryo are rather simplistic, being based on salt solutions with additional energy substrates and growth factors. An improved serum-free medium (IVMD101) was developed for bovine oocytes maturation in vitro. Proportions of embryos developing to the blastocyst stage cultured in both IVD101(32.4%) and IVD101(34.5%) serum-free media were higher than in TCM199+10% FBS(12.4%) serumcontaining medium. Futhermore, the cell numbers per blastosyst obtained in the serum-free media were superior to those of blastocysts developed in serum-supplemented medium. Also, cell numbers of blastocysts obtained in the serum-free media were similar with blastocysts derived in vivo. Survival rate blastocysts after 24 hr incubation after thawing, the blastocysts cultured in both IVD101(94.5%) and IVD101(95.8%) serum-free media were higher than in TCM199+10% FBS (52.5%) serum-containing medium. After 72 hr incubation after thawing, hatching rates of blastocysts developed in IVD101(78.4%) and IVMD101(83.7%) were sighnificantly higher than that developed in the serum-supplemented medium(32.0%). The pregnancy rates almost not different between fresh blastocysts(38.2%) and frozen blastocysts(34.9%). The results suggested that the improved serum-free media(IVMD101 and IVD101) offer several advantages over culture in serum-cotaining medium, including increased rates of blastocyst formation and high cel numbers. Additionally, the survival and hatching rates of embryos product in serum-free media after post-thaw culture were superior to those of embryos produced in the serum-containg medium and useful for the production of high quality bovine embryos for cryo-preservation. These improved serum-free media are beneficial not only for the study of the mechanisms of early embryogenesis but also for mass production of good quality embryos for embryo transfer, cloning and transgenesis.

  • 목적 :크롬친화세포는 카테콜아민 (CA)을 분비하는 새포로서 무신 수질내에 주로 존재한다. 따라서 부신피질의 영향을 배제하여 여러가지 자율신경계 작용 약물의 약리작용을 연구하는데 중요한 조직이다. 그러므로, 부신으로부터 크롬친화세포를 분리하여 배양하는 방법을 습득하여 자율신경계 작용약물연구에 이용코자 히스타민을 이용하여 CA 분비작용을 연구하였다. 방법: 도살장에서 소를 즉사시킨 후 좌우양측 부신을 분리하여 collagenase digestion으로 부신수질로부터 분리하고 Percoll gradient에 의해서 chromaffine cell을 정제하였다. 이렇게 정제한 크롬친화세포는 Dulbecco's modified Eagle medium에 10% fetal calf serum과 함께 culture dish에다 넣어 5% $CO_2$, incubator에서 유지시켜 주면서 실험을 시행하였다. 배양세포는 분리후 일주일 이내에 사용하였다. Catecholamine측정은 electrochemical detector를 연결하여 HPLC로 측정하였다.

Low Serum Medium for CH07E02 Hybridoma (하이브리도마 배양을 위한 저혈청배지의 조성 결정)

  • Up to now, 10% Fetal Bovine Serum(FBS(V/V)) was added to basal medium for the cultivation of hybridoma. For the cultivation of hybridoma cell line, CH07E02, against colon cancer, serum concentration was reduced to 3% FBS without influence on cell growth and maximum cell concentration. By the addition of cell growth promoting substances-insulin (I), pyruvate (P), oxaloacetate(O), Pluronic F-68(P) and 2-mercaptoethanol(2-ME)-to 1% FBS medium, a cell density higher than that with 1% FBS medium alone was achieved. FBS 3% medium was replaced by very cheap 2% Calf Serum (CS) medium without influence on cell growth rate and concentration. Cells grew vigorously in 0.5% CS+IPOP medium. This composition was used during suspension culture and exhibited good viability and high specific growth rate.

Serum albumin levels and their correlates among individuals with motor disorders at five institutions in Japan

  • BACKGROUND/OBJECTIVES: The level of serum albumin is an index of nourishment care and management. However, the distribution and correlates of serum albumin levels among individuals with motor disorders have not been reported until now. Therefore, we examined the distribution and correlates of serum albumin levels among individuals with motor disorders. SUBJECTS/METHODS: A cross-sectional study on 249 individuals with motor disabilities (144 men, mean age: 51.4 years; 105 women, mean age: 51.4 years) was conducted at five institutions in Ibaraki Prefecture, Japan in 2008. The results were compared with data from the National Health and Nutrition Survey. RESULTS: The mean serum albumin levels were $4.0{\pm}0.4g/dL$ for men and $3.8{\pm}0.5g/dL$ for women. Overall, 17 (11.8%) men and 25 (23.8%) women had hypoalbuminemia (serum albumin level ${\leq}3.5g/dL$); these proportions were greater than those among healthy Japanese adults (${\leq}1%$). Low serum albumin level was related with female sex, older age, low calf circumference, low relative daily energy intake, low hemoglobin (Hb), low blood platelet count, low high-density lipoprotein cholesterol (HDL-C), low $HbA_{1c}$, and high C-reactive protein (CRP) levels. The strongest correlates, based on standardized betas, were Hb (0.321), CRP (-0.279), and HDL-C (0.279) levels. CONCLUSIONS: These results indicate that the prevalence of hypoalbuminemia is higher in individuals with motor disabilities than in healthy individuals and that inflammation is a strong negative correlate of serum albumin levels. Therefore, inflammation should be examined for the assessment of hypoalbuminemia among institutionalized individuals with motor disabilities.

Effect of Placental Serum to Cultures of the Healthy Human Peripheral Blood Lymphocytes (태반혈청이 사람의 임파구 배양에 미치는 영향)

  • To investigate the effects of placental serum to in vitro culture of the normal human lymphocytes the peripheral blood lymphocytes were isolated by ficoll-hypaque separation method in 20 healthy human adults. The blast transformation respone of lymphocytes to mitogens were observed by stimulation with PHA($25{\mu}g/ml$) and Con A($25{\mu}g/ml$) using RPMI 1640 media containing 20% placental serum(PS), tetal calf serum(FCS), or humans AB serum(AB). And one-way mixed lymphocyte culture test was performed between these unrelated person compounded into stimulators and responders to investigate the effect of placental serum. The following results were obtained. 1. In 20 experimental cases, these were no significant diffenence between FCS, AB, and PS in untreated control cultures. 2. In PHA-treated cultures, whereas the blast transformation rate of the FCS groups and AB groups were $40.8{\pm}4.3%$ and $44.6{\pm}4.3%$ respectively, that of PS groups were $21.7{\pm}3.4%$, Similar results were obtained in Con A-meated cultures. Therefore, placental serum inhibited the mitogenic response of lymphocytes significantly. 3. In MLC tests, stimulation index of the FCS groups and AB groups were $18.5{\pm}3.5$ and $20.1{\pm}3.3$ respectively. But placental serum inhibited MLC response of lymphocytes significantly($7.4{\pm}1.9$).

Viability of In Vitro Fertilized Bovine Embryos Following In Vitro Culture and Embryo Transfer (소 체외수정란의 체외배양 및 이식후 생존성)

  • This study was conducted to examine the condition of in vitro culture system and the viability after embryo transfer of in vitro matured-in vitro fertilized (IVM-IVF) bovine embryos. The in vitro development to the blastocyst stage was enhanced by supplying bovine serum albumin(BSA) to co-culture medium with bovine oviduct epithelial tissue(BOET) compared with that in medium supplemented with fetal bovine serum(FBS) (41.2% vs. 26. 3%, P<0.05). After transfer of IVM-IVF blastocysts into the uterine horn of recipient females (Aberdeen Angus), one was pregnant to term and produced a head of male Korean native calf. These results confirm that the in vitro development of IVM-IVF bovine embryos is affected with different protein source in co-culture with BOET, and IVM-IVF embryos can develop to term after in vitro culture and embryo transfer.

The Role of Protein Kinase C for Prolactin Secretion in Chicken Primary Pituitary Cell Culture (산란계의 뇌하수체 세포배양에서 Prolactin의 생성에 관계하는 Protein Kinase C의 역할)

  • A series of experiments were conducted to investigate the role of protein kinase C (PKC) as a second messenger in vasoactive intestinal peptide (VIP) mediated prolactin secretion. Primary pituitary cells (106 cells/treatment) were separated from laying hens and incubated in M-199 with 5% chicken serum and 5% fetal calf serum. The VIP(0.1 $\pi$M) treatment enhanced prolactin Secretion into media upto 9-fold during 48-h incubation. The phorbol 12-myristate 13-acetate (PMA), a PKG agonist, increased prolactin secretion upto 2-fold at 0.1 nM PMA (P<0.01), and the prolactin secretion was not significantly higher than this concentration. Staurosporine (ST; 1.0$\pi$M) a PKC antagonist, decreased by 70% of 0.1 $\pi$M VIP-stimulated prolactin secretion and by 48% of 10 ${\mu}$M PMA-stimulated prolactin secretion (P<0.01). However, pituitary cell prolactin content did not differ in any treatment (P>0.05). In conclusion, these results indicate that the PKC second messenger system is involved in VIP-stimulated prolactin release in chicken primary pituitary cell culture.
