Porous calcium phosphates were successfully fabricated by using a cuttlefish bone. The cuttlefish bone, which is composed of $CaCO_3$, showed a special porous microstructure containing uniform-sized voids. In this study, the pre-forms infiltrated distilled phosphoric acid were sintered at $1200^{\circ}C$ in an air atmosphere. The porous microstructure of the pre-forms was kept their original pattern after sintering with a synthesis of calcium phosphate. The obtained porous calcium phosphate, sintered at $1200^{\circ}C$ for 3 h at 17% concentration of phosphoric acid, showed uniform open pores of 150 ${\mu}m$ in size and $\beta$-TCP phase in the XRD patterns. Above 16% concentration, CaO phase, derived from the decomposition of $CaCO_3$, decreased gradually in the sintered samples, and the measured Ca/P ratios of the samples prepared from 16% and 18% concentration were 1.67 and 1.43, which are close to stoichiometric HA (1.66) and $\beta$-TCP (1.50).

Polycystic kidney disease 2-like-1 (PKD2L1), polycystin-L or transient receptor potential polycystin 3 (TRPP3) is a TRP superfamily member. It is a calcium-permeable non-selective cation channel that regulates intracellular calcium concentration and thereby calcium signaling. Although the calmodulin (CaM) inhibitor, calmidazolium, is an activator of the PKD2L1 channel, the activating mechanism remains unclear. The purpose of this study is to clarify whether CaM takes part in the regulation of the PKD2L1 channel, and if so, how. With patch clamp techniques, we observed the current amplitudes of PKD2L1 significantly reduced when co-expressed with CaM and $CaM{\triangle}N$. This result suggests that the N-lobe of CaM carries a more crucial role in regulating PKD2L1 and guides us into our next question on the different functions of two lobes of CaM. We also identified the predicted CaM binding site, and generated deletion and truncation mutants. The mutants showed significant reduction in currents losing PKD2L1 current-voltage curve, suggesting that the C-terminal region from 590 to 600 is crucial for maintaining the functionality of the PKD2L1 channel. With PKD2L1608Stop mutant showing increased current amplitudes, we further examined the functional importance of EF-hand domain. Along with co-expression of CaM, ${\triangle}EF$-hand mutant also showed significant changes in current amplitudes and potentiation time. Our findings suggest that there is a constitutive inhibition of EF-hand and binding of CaM C-lobe on the channel in low calcium concentration. At higher calcium concentration, calcium ions occupy the N-lobe as well as the EF-hand domain, allowing the two to compete to bind to the channel.
The inward tail current after a short depolarizing pulse has been known as Na-Ca exchange current activated by intracellular calcium which forms late plateau of the action potential in rabbit atrial myocytes. Chloride conductance which is also dependent upon calcium concentration has been reported as a possible tail current in many other excitable tissues. Thus, in order to investigate the exsitance of the calcium activated chloride current and its contribution to tail current, whole cell voltage clamp measurement has been made in single atrial cells of the rabbit. The current was recorded during repolarization following a brief 2 ms depolarizing pulse to +40mV from a holding potential of -70mV. When voltage-sensitive transient outward current was blocked by 2 mM 4-aminopyridine or replacement potassium with cesium, the tail current were abolished by ryanodine$(1{\mu}M)$ or diltiazem$(10{\mu}M)$ and turned out to be calcium dependent. The magnitudes of the tail currents were increased when intracellular chloride concentration was increased to 131 mM from 21 mM. The current was decreased by extracellular sodium reduction when intracellular chloride concentration was low(21 mM), but it was little affected by extracellular sodium reduction when intracellual chloride concentration was high(131 mM). The current-voltage relationship of the difference current before and after extracellular sodium reduction, shows an exponential voltage dependence with the largest magnitude of the current occurring at negative potentials, with is similar to current-voltage relationship at negative potentials, which is similar to current-voltage relationship of Na-Ca exchange current. The current was also decreased by $10{\mu}M$ niflumic acid and 1 mM bumetanide, which is well known anion channel blockers. The reversal potentials shifted according to changes in chloride concentration. The current-voltage relationships of the niflumic acid-sensitive currents in high and low concentration of chloride were well fitted to those predicted as chloride current. From the above results, it is concluded that calcium activated chloride component exists in the tail current with Na-Ca exchange current and it shows the reversal of tail current. Therefore it is thought that in the physiologic condition it leads to rapid end of action potential which inhibits calcium influx and it contributes to maintain the low intracellular calcium concentration with Na-Ca exchange mechanism.
A lactose-hydrolyzed milk with low sweetness was developed using nanofiltration. Raw milk was treated with 0.03% ${\beta}$-galactosidase at $4^{\circ}C$ for 24 h to hydrolyze lactose partially. The resultant lactose-hydrolyzed milk containing 0.43% lactose was then concentrated using a nanofiltration membrane to reach concentration factor of 2.13. The concentration factors and coefficients of retention of milk components in nanofiltration were determined. The concentration factor of milk fat was 2.20 which was the highest of the milk components. The coefficient of retention of calcium and riboflavin was 0.96 and 0.76, respectively. However, the coefficient of retention of glucose, galactose, and sodium was 0.21, 0.15, and 0.22, respectively. Raw milk was treated with 0.1% ${\beta}$-galactosidase at $4^{\circ}C$ for 40 h to hydrolyze lactose fully and then concentrated to reach a concentration factor of 1.6 by using nanofiltration. The concentrated milk was reconstituted with water. The lactose-hydrolyzed milk had sweetness similar to milk. The compositional ratios of crude protein, calcium, sodium, and riboflavin of lactose-hydrolyzed nanofiltrated milk to those of raw milk were 99%, 97%, 77%, and 80%, respectively. This study showed that nanofiltration of lactose-hydrolyzed milk to remove galactose and glucose did not cause significant loss of calcium. The lactose-hydrolyzed nanofiltrated milk contained 0.06% lactose and had sweetness similar to milk.
The present study was carried out to investigate the effects of dietary protein and magnesium levels on calcium and magnesium metabolism in male Sprague-Dawley rats. Forty-eight male rats(average weight, 210g) divided into six diet groups; protein 8% and Mg-free diet, protein 8% and Mg 400mg/kg diet, protein 8% and Mg 900mg/kg diet, protein 20% and Mg-free diet, protein 20% nd Mg 400mg/kg diet, and protein 20% and Mg 800mg/kg diet group. After the rats were fed with experimental diets concentration were examined. Kidney weight was significantly higher in protein 8% and Mg-free group compared with other groups. Serum calcium concentration of protein8% group tended to decrease with the increase of the level of magnesium. Serum magnesium concentration in protein 8% and 20% group was lower in Mg-free group than that in other groups(p<0.05) and it also increased as the dietary magnesium level was increased. Urinary calcium and magnesium concentration in Mg-free group were significantly lower than those of other groups.
Objectives: The present study aimed to evaluate the preventive effects of exposure to liquid fermented milk containing various concentrations of added calcium on dental erosion, and to investigate the optimal concentration of calcium effective in reducing dental erosion. Methods: The present study consisted of a total of 6 experimental groups: a mineral water group, a fermented milk with no added calcium (0%) group, and four fermented milk with various concentrations of added calcium (0.1%, 0.5%, 1%, and 2%) groups. Twelve specimens were immersed for 1, 3, 5 and 10 minutes in each experimental drink and the change in surface microhardness was measured. Additionally, the surface was observed using a scanning electron microscope. Results: The difference in surface microhardness before and after 10 minutes of immersion in the experimental drink was the highest in the Ca 0% group, followed by the Ca 0.1%, 0.5%, 1%, 2% group and the mineral water group, in that order. The groups with a calcium concentration of more than 0.5% showed statistically significant differences in surface microhardness compared to the Ca 0% group. In addition, when the surface morphology of enamel was observed under a scanning electron microscope, the results showed that the highest level of surface damage was observed in the Ca 0% group, followed by the Ca 0.1%, 0.5%, 1%, 2% group, in that order. Conclusions:The present study confirms that a higher calcium concentration in fermented milk is associated with a higher possibility of preventing dental erosion. The addition of 0.5% calcium, which is a relatively low concentration, did not completely prevent dental erosion, but significantly inhibited dental erosion compared to fermented milk without any added calcium. Therefore, it is suggested that consumers should be educated and provided with guidance to consider the calcium content when choosing fermented milk.
본 연구에서는 식물추출액을 이용하여 모래 입자 사이에 친환경적으로 탄산칼슘을 석출시켜 모래 고결을 유발하고자 하였다. 식물추출액에는 고결유발 미생물(예: $Sporosarcina$$pasteurii$)과 같이 요소를 탄산이온과 암모늄이온으로 분해하는 우레아제 성분이 포함되어 있다. 분해된 탄산이온과 용액 속에 녹아 있는 칼슘이온이 결합하여 탄산칼슘을 석출시키는 원리를 이용하여 모래의 고결을 유도하였다. 깨끗한 낙동강모래에 식물추출액과 요소 그리고 칼슘원(염화칼슘 또는 수산화칼슘)을 혼합한 용액을 섞어서 비빈 다음 다짐방법으로 공시체를 제작하였다. 요소의 농도를 세 종류로 달리하였으며, 제작된 공시체를 실내($18^{\circ}C$)에서 3일 동안 양생시킨 다음 일축압축시험과 SEM 및 XRD분석을 실시하여 사질토의 고결 정도를 연구하였다. 칼슘원에 관계없이 요소의 농도가 높을수록 탄산칼슘이 더 많이 침전되어 일축압축강도는 식물추출액을 사용하지 않은 경우보다 최대 10배까지 증가하는 경향을 보였다. 염화칼슘을 사용한 공시체가 수산화칼슘을 사용한 경우보다 더 높은 강도를 나타내었다.
본 연구에서는 시판되는 3종의 칼슘 용해에 미치는 식초의 영향을 조사하였다. 식초의 초기총산에 따른 영향을 조사한 결과 초기총산과 칼슘첨가량이 높을수록 용해 후 pH와 잔사량은 낮게, 총산과 칼슘함량은 높게 나타났다. 식초 함량에 따른 영향을 조사한 결과 식초함량이 높을수록 pH는 낮게 총산은 높게 나타났으며, 칼슘함량은 해조칼슘과 탄산칼슘에서 높게 나타났다. 식초를 이용하여 해조칼슘과 탄산칼슘의 포화농도를 조사한 결과 해조칼슘은 $7.0\%$ (w/v), 탄산칼슘은 $6.0\%$(w/v) 농도이며 이때 칼슘함량은 $2,234\;mg\%$와 $2,490\;mg\%$로 나타났다.
Objective : The purpose of this study was to investigate the effect of Bee Venom on the lipopolysaccharide, sodium nitroprusside and hydrogen peroxide induced expression phospholipase $A_2$ and calcium concentration in RAW 264.7 cells, a murine macrophage cell line. Method : The expression of phospholipase $A_2$ was determined by western blotting with corresponding antibodies, and the generation of intracellular calcium concentration was investigated by delta scan system in RAW 264.7 cells. Results : 1. Compared with control, expressions of lipopolysaccharide-induced phospholipase $A_2$ were decreased significantly by $1\;{\mu}g/{\mu}l$ of bee venom and decreased by 0.5, $5\;{\mu}g/{\mu}l$ of bee venom. 2. Compared with control, expressions of sodium nitroprusside-induced phospholipase $A_2$ were decreased significantly by $5\;{\mu}g/{\mu}l$ of bee venom but increased by 0.5, $5\;{\mu}g/{\mu}l$ of bee venom. 3. Compared with control, expressions of hydrogen peroxide-induced phospholipase $A_2$ were decreased significaltly by $1{\mu}g/{\mu}l$ of bee venom and decreased by $0.5\;{\mu}g/{\mu}l$ of bee venom but increased by $5\;{\mu}g/{\mu}l$ of bee venom. 4. An experiment was carried out to determine the effect of age and caponization on the development blood and bone characteristics development in male country chickens in Taiwan. A total of two hundred 8-wk-old LRI native chicken cockerels, Taishi meat No.13 from LRI-COA, were used as experimental animals. Cockerels were surgically caponized at 8 wks of age. Twelve birds in each group were bled and dressed from 8 wks to 35 wks of age at 1 to 5 wk intervals. The results indicated that the plasma testosterone concentration was significantly (p<0.05) lower in capons after 12 wks of age (caponized treatment after 4 wks) than that of the intact males. The relative tibia weight, bone breaking strength, cortical thickness, bone ash, bone calcium, bone phosphorus and bone magnesium contents were significantly (p<0.05) higher in intact males, while capons had higher (p<0.05) plasma ionized calcium, inorganic phosphorus and alkaline phosphatase concentration. The plasma testosterone concentration, relative tibia weight, tibia length, breaking strength, cortical thickness, bone ash, calcium, and phosphorus contents of intact males chickens increased significantly (p<0.05) with the advance of age. In addition, the relative tibia weight of capons peaked at 18 wks of age, and declined at 35 wks of age. The bone ash, calcium and phosphorus content increased most after 14 wks of age in male native chickens in Taiwan. Also, tibia length and cortical thickness peaked at 22 wks of age. However, the peak of bone strength was found at 26 wks of age. These findings support the assertion that androgens can directly influence bone composition fluxes in male chickens. Caponization caused a significant increase in bone loss at 4 wks post treatment, which reflected bone cell damage, and demonstrated reductions in the relative tibia weight, breaking strength, cortical thickness, bone ash, calcium, phosphorus and magnesium contents, and increases in plasma ionized calcium, inorganic phosphorus and alkaline phosphatase concentration.
