The effect of orally administered ginseng ethanol extract on potassium deficient and normal rat skeletal muscles was investigated in terms of Ca uptake by sarcoplastic reticulum fragments. The ginseng ethanol extract (100mg/kg/day) was administered orally to Sprague-Dawley rats for 21 days and their changes of body weights, $K^+$ content in skeletal muscles, and calcium uptake capacity of sarcoplamic reticulurn of each groups were measured. The growth rate of rats fed with the potassium deficient diet was significantly decreased compared to that of normal rats. Ginseng components did not show any effect on the decreased growth rate of the postassium deficient rats. Potassium content in skeletal muscle from potassium deficient rats was significantly reduced compared to that of normal rats. Ginseng components showed the tendency to prevent the reduction in potassium content of potassium deficient rats, but differences were not statistically significant. Calcium uptake of SR prepared from skeletal muscles of potassium deficient rats was increased significantly compared to that of normal rats. Ginseng components prevented such increase of calcium uptake by 30%. In summary, it can be concluded that ginseng may prevent the increase in Cauptake of SR obtained from potassium deficient rats.
본 연구는 폐경 후 여성에게 식이 칼슘염 형태, 에스트로겐 및 에스트로겐/칼슘 혼합요법이 골격 구성 성분에 미치는 영향을 알아보고자 난소절제쥐를 이용한 총 9군으로 분류하여 6주간 사육한 후 그 효과를 살펴보았다. 오른쪽 대퇴골의 젖은 무게, 밀도, 길이, 강도, 회분 함량은 난소를 절제한 군과 난소를 절제하지 않은 군간에 차이는 없었다. 그러나 대퇴골의 젖은 무게는 체중 당으로 표현한 값은 난소를 절제한 군에서 낮았다. 이것은 난소를 절제한 쥐의 체중부하로 인해 골격 구성 성분은 변화가 없는 것으로 사료된다. 난소절제 후 estrogen 투여, estrogen/칼슘 혼합군에서 체중당 젖은 무게는 증가함을 보여 주었다. 난소절제 후 estrogen 점진적 감소군에서 강도, 회분 함량, 칼슘 함량, 인 함량이 가장 낮았다. 견갑골의 젖은 무게, 밀도, 회분 함량, 칼슘 함량, 인 함량은 난소를 절제한 군과 난소를 절제하지 않은 군간에 차이는 없었다. 난소절제 후 estrogen 점진적 감소군에서 뼈의 젖은 무게와 회분 함량이 모든군에 비해 낮았고 고칼슘을 투여하거나, 칼슘을 점진적으로 증가시키는 것이 골손실을 다소 감소시킴을 알 수 있었다. 이상의 성적으로 보아 난소절제 여부에 따른 골격구성 성분에는 변화가 없었다. 난소절제 후 estrogen 및 estrogen/칼슘 혼합요법 골격손실을 상당량 방지 할 수 있었고 estrogen 점진적 감소군은 골격손실을 가져옮을 알 수 있었다. Estrogen 점진적 감소에 고칼슘, 칼슘 점진적 증가투여는 골격 구성 성분의 손실을 다소 방지할 수 있음을 알 수 있었다.
This study was designed to investigate the effect of dietary calcium and phosphate levels on calcium and bone metabolism in rats. The rats were divided into six groups and each of the groups was fed diets with different Ca/P ratios. The experimental periods were 5 weeks . There was no significant different difference in dietary intake, body weight gain, and organ weight among the groups with different calcium and phosphate intake levels. Fecal calcium excretion was not significantly different among the groups, but urinary calcium excretion was increased by the increase in Ca/P ratio. Fecal phosphate excretion was not different but urinary phosphate excretion was increased by the increase in dietary phosphate intake. There was no significant difference in serum alkaline phophatase activity and urinary hydroxyproline levels were not significantly different among the groups. The low calcium-high phosphate(0.25Ca-1.2% P) group showed the lowest total calcium content in femur and scapula. This may be due to it having the lowest Ca/P ratio among groups. The low calcium-high phosphate(0.2%Ca-1.2%P) group showed that mandible is almost lost and osteolyzed Harversian canal was expanded in femur. Results suggest that phosphate intake affects calcium and bone metabolism more with inadequate calcium nutrition that with adequate calcium intake. Thus , for normal bone growth and metabolism , adequate calcium intake and/or high Ca/P ratio are important.
We examined the effects of calcium intake levels on mineral metabolism during pregnancy using calcium-deficient young adult rats. Five week-old female Sprague-Dawley rats were fed normal Ca (NCa, 0.5%) and low Ca (LCa, 0.15%) diets for five weeks (pre-pregnancy). The low Ca intake group was then divided into three groups and fed low Ca (0.15%), normal Ca (0.5%) and high Ca (1.5%) diets for 3 weeks (pregnancy). All of the rats were mated with normal male rats. The control group was fed a consistently normal Ca (0.5%) diet during pre-pregnancy and pregnancy. On the day after delivery, dams and their pups were sacrificed. We measured total protein, albumin, alkaline phosphatase and mineral content in serum and weight, length, breaking force, ash and mineral content in the femur and lumbar (L2-L4) of the dams. Whole body mineral content was measured in the pups. There was no difference in weight gain and food intake among the groups. Serum total protein and albumin were in the normal range but a little lower during pregnancy. High Ca supplementation decreased serum Mg and Fe during pregnancy. Weight, ash and Ca of the femur and lumbar significantly decreased in rats fed a chronically low Ca diet during pre-pregnancy and pregnancy. Calcium supplementation levels were above normal during pregnancy and increased the bone weight and breaking force of rats fed the low Ca diet during pre-pregnancy. However, Ca supplementation did not increase the ash and Ca contents in the bones. High Ca supplementation during pregnancy significantly decreased Mg in the bones and increased Ca and P in the kidneys. Ash content of pups from dams fed the chronically low Ca diet decreased but there was no difference in whole body Ca among the groups. Mg and Fe in the whole body decreased in pups from dams fed the high Ca diet. Pregnancy performance was reduced in dams fed the low Ca diet. These results suggest that above normal Ca supplementation levels during pregnancy restored maternal bone status to some extent in rats fed the chronically low Ca diet. The same could not be said for mineral content. Also, high Ca supplementation during pregnancy may deteriorate mineral composition in bones and other tissues. Therefore, more detailed research is needed to facilitate sound recommendations on appropriate calcium intake during pregnancy. (Korean J Nutrition 36(5): 459∼469, 2003)
This study was conducted to investigate the effect of nondigestable substances and calcium such as oligosaccharide, agar, saponin, tannin and calcium on the reduction of lipid status in rats fed high fat diet. In order to make the observation, the lipid content in plasma, liver and the feces, and bile acid excretion were measured of r 4 weeks. the results obtained from this research are as follows. Concentration of total lipid in plasma seemed highest in the control group and were significantly lower in groups oil-gosaccharide, agar and calcium-tannin, compared to the control group. Concentration of total cholesterol in plasma was significantly lower in groups oligosaccharide, agar, calcium and calcium-saponin, compared to the control group. The improvement in lipid status seems to be insignificant with oligosaccharide, showed increase in total bile acids excretion in feces and decrease in total cholesterol in plasma, compared to the control group. These evidence seems to indicate improvement of the plasma lipid status by calcium and agar supplementation.
Chinese cabbage plant was grown hydroponically for 4 weeks in order to examine the temporal relationship of calcium concentration of the nutrient solution with calcium content in the leaf tissue and susceptibility of the tissue to soft rot disease by $Pectobacterium$$carotovorum$ ssp. $carotovorum$ (Pcc). Calcium concentration from 0.5 to 32.0 mM was maintained for 1 week using Hoagland & Arnon solution. The calcium content of the leaf was proportionally increased to the concentration of the nutrient in the solution (r = 0.912). In contrast, the severity of soft rot symptom in the young leaves was inversely related with the amount of calcium supplied to the nutrient solution (r = 0.899). Water-soluble chitosan, prepared by hollow fiber filtration (> 100 kDa) was applied into the nutrient solution from 0.0 to 5,000 ppm. The chitosan of 10 ppm was the most effective to promote calcium uptake of the leaf, showing 155% of the control. The same chitosan solution prohibited most soft rot development of the leaf by Pcc, exhibiting only 53% of the control. Among different molecular weight fractions, chitosan fraction obtained from 30-100 kDa molecular weight cut-off promoted calcium uptake the most up to 163% of the control, and reduced the development of soft rot disease recording merely 36% of the control of the leaf tissue. The results obtained in the present study suggest that large scale production of water-soluble chitosan with an optimum molecular weight and its commercial application to Chinese cabbage production will be important to improve yield and quality of the crop.
Clamshells, which comprise more than 50% of a clam’s weight, are a major byproduct of the clam industry and are mainly composed of insoluble calcium carbonate. This study investigates the use of clamshells as a natural calcium resource. Highly soluble powdered calcium lactate (LCCL) was prepared from the calcined powdered shells of littleneck clams (LCCP) using response surface methodology (RSM) to predict optimum conditions. These conditions, as derived from pH, solubility, and yield of 11 LCCLs manufactured according to the RSM model, were 1.80 M lactic acid and 1.13 M LCCP. The actual values of pH (6.98), solubility (93.99%), and yield (351.23%) under the optimized conditions were as predicted. The derived LCCL exhibited a strong buffering capacity in the range of pH 2.78-3.90 when combined with less than 2 mL of 1 N HCl. The ranges of calcium content and solubility of LCCL were 7.7-17.5 g/100 g and 96.6-98.9%, respectively. Fourier transform infrared spectroscopy (FT-IR) of the LCCL identified it as calcium lactate pentahydrate, and field emission scanning electron microscopy (FESEM) revealed an irregular and rod-like microstructure. These results confirm the potential use of clamshells, converted to highly soluble organic acid calcium, as an additive to enhance calcium content in food ingredients.
Calcium is an essential nutrient for living organisms, with key structural and signaling roles. Its deficiency in plants can result in poor biotic and abiotic stress tolerance as well as reduced crop quality and yield. Calcium deficiency in humans causes various diseases such as osteoporosis and rickets. Biofortification of calcium in various food crops has been suggested as an economic and environmentally advantageous method to enhance human intake of calcium. Recent efforts to increase the levels of calcium in food crops have used calcium/proton antiporters ($CAXs$) and modified one to increase calcium transport into vacuoles through genetic engineering. It has been reported that overall calcium content of transgenic plants has been increased in their edible portions with some adverse effects. In conclusion, biofortification of calcium will add more value in crops as well as will be beneficial for animal and human. Therefore, more fundamental studies on the mechanisms of calcium ion storage and transporting are essential for more effective calcium biofortification.
This study was to compare the bioavailability of calcium from anchovy and tofu to those of calcium from calcium carbonate(CaCO3)as the control diet and non-fat dry milk(NFDM). Rats weighing 50-60g were placed on experimental diets and deionized water at free access for 4 weeks. Diets contained 0.2% calcium from calcium-carbonate, NFDM, anchovy, tofu or 0.5% calcium as obtained were as follows : 1) No significant differences in the apparent absorption of calcium(62.5%-71.0%) were observed in the rats fed four different calcium sources at the level of 0.2% while 0.5% calcium diet group apparently absorbed calcium less efficiently(52.2%). 2) Bone length of tibia and femur was not significantly different among the groups, though 0.5% calcium of control group showed slightly longer length. 3) Tibia fat-free dry weights of 0.2% calcium of NFDM and anchovy diet groups were not significantly different from that of 0.5% calcium of control group. For femurs NFDM, anchovy and tofu groups were similar in their fat-free dry weight to that of 0.5% calcium group. 4) For calcium contents tibia contents tibia from anchovy treated group showed higher value than calcium-carbonate and tofu groups and the value was not significantly different from that of 0.5% calcium group. In femur NFDM, tofu and calcium-carbonate groups were not significantly different in their calcium content but 0.5% calcium group had higher level of calcium than 0.2% calcium groups. 5) The normalized values(NV) show that there was no significant differences in NV among 0.2% CaCO3 anchovy and tofu groups, while NV of NFDM group was significantly lower than that of calcium-carbonate group. NV of 0.5% calcium group was a little more than 50% of those in 0.2% calcium groups. Though the values obtained for the calcium bioavailability were somewhat variable among experimental products, it was demonstrated that anchovy and tofu are as good as NFDM for the dietary calcium provider when calcium intake is at marginal level.
불소도포시 주로 형성되는 calcium fluoride는 구강내 환경에서 pH에 의해 조절되는 불소의 저장고 역할을 하여 치아우식 예방에 있어서 중요한 역할을 하는 것으로 알려져 있다. 따라서 불소도포의 우식예방 효과는 calcium fluoride를 얼마나 많이 형성시켜 오래 지속시키는 가에 달려 있다. 이에 치아 내산성 증가에 효과 있다고 알려진 Nd:YAG 레이저 조사가 calcium fluoride형성 및 치아 내산성에 미치는 영향에 대해 알아보기 위해서 실험을 시행하였다. 소의 영구전치에서 276개의 시편을 제작하여 아무런 처치를 하지 않은 군을 대조군으로 하고 1.23% acidulated phosphate fluoride (APF) 5분, 30분 도포 및 Nd:YAG 레이저 조사에너지 밀도 $20J/cm^2,\;40J/cm^2$의 조건에 따라 불소 도포군, 레이저 조사군, 불소 도포후 레이저 조사군, 레이저 조사후 불소 도포군으로 분류하였다. 불소도포를 시행한 경우 이를 다시 KOH 처리 여부에 따라 나누어 23개의 실험조건을 만들었다. 각 실험조건에 12개의 시편을 배정하고 이중 10개의 시편은 불소 및 무기인 측정에, 나머지 2개의 시편은 표면 관찰에 사용하였다. KOH 비처치군에서 0.1 N HClO4로 enamel biopsy를 시행하여 탈회 법랑질의 불소농도 및 탈회깊이를 측정하였으며 KOH 처치군에서는 1 M KOH로 24시간 처리하여 calcium fluoride를 정량 후 enamel biopsy를 시행하였다. 실험결과 얻어진 자료 및 주사 전자현미경 관찰 소견을 비교하여 다음의 결과를 얻었다. 1. KOH 비처치군에서 탈회된 법랑질의 불소농도는 레이저조사 후 불소도포시 레이저 에너지 밀도 증가에 따라 증가되는 경향을 보였다. 2. KOH 비처치군에서 법랑질 탈회깊이는 불소를 5분 도포후 레이저 $20J/cm^2$를 조사한 경우를 제외하고는 레이저와 불소를 병행하여 처치시 각각을 단독으로 처치시보다 탈회깊이가 작았다 (p<0.05). 3. Calcium fluoride의 양은 레이저의 조사에 의해 유의 있게 증가하지 않았다 (p>0.05). 4. Calcium fluoride 입자는 불소만 도포한 경우에 비해 레이저 조사후 불소도포시 입자크기가 증가되었으며, 불소도포후 레이저 조사시 입자크기의 증가와 함께 일부 융합된 양상을 보였다. 5. KOH 처치군에서 법랑질의 불소농도는 불소를 30분 도포후 레이저를 조사한 경우를 제외하고는 대조군의 불소농도와 유의한 차이가 없었다(p>0.05). 6. KOH 처치군에서 불소와 레이저를 병행한 경우, 불소만 도포한 경우보다 탈회된 법랑질의 깊이가 작았다 (p<0.05). 7. KOH 비처치군에서 탈회된 법랑질의 불소농도와 탈회깊이의 상관관계(Spearman correlation coefficient: -0.6281)는 KOH 처치군(Spearman correlation coefficient: -0.3792)에 비해 높은 음의 상관 관계를 보였고, 동일한 조건으로 불소도포 및 레이저 조사를 시행한 경우에 있어서 KOH 처리 여부에 따른 탈회법랑질 깊이의 유의차가 있는 경우 calcium fluoride의 형성량이 많았다. 이상의 실험결과로 미루어 보아 레이저 조사가 calcium fluoride의 형성량에 영향을 미치지 않았으나 레이저 조사의 영향으로 calcium fluoride의 용해가 감소하는 것으로 판단된다. 또한 불소도포 및 레이저 조사의 병행은 각각을 단독으로 처치한 경우에 비하여 치아 내산성 증가면에서 유리하므로 임상에서 불소도포주기 연장과 같은 유용한 결과를 얻을 수 있을 것으로 생각된다.
