REVASCULARIZATION AND REINNERVATION OF CALCITONIN GENE-RELATED PEPTIDE IMMUNOREACTIVE NERVES IN REPLANTED RAT MOLARS (재식한 흰쥐 구치 치수조직의 재혈관화와 CGRP 면역반응 신경섬유의 변화)

  • The purpose of this study was to investigate the revascularization and reinnervation of calcitonin gene-related peptide immunoreactive nerves in immediately replanted rat molars. First maxillary right molars in 56 rats(35days old) were extracted and immediately replanted. The rats were killed 1, 2, 3, 7, 14, 28 and 42 days after replantation and revascularization of pulpal blood vessels were examined microangiogram with korean traditional ink and reinnervation of pulpal nerve were examined immunohistochemical method using calcitonin gene-related peptide(CGRP) antiserum. The results were as follows; 1. Revascularization and reinnervation of CGRP immunoreactive nerve fibers were observed mesial side whole pulp tissue of replanted teeth. Revascularization and reinnervation of CGRP immunoreactive fiber were made at 2days after replantation in entire pulp of replanted teeth and the distribution density of blood vessels were gradually increased according time elapsed, but did not achieve the density of control. 2. Postoperative dentin formation in replanted teeth revealed at 1week after replantaton and gradually increased according to time elapsed. 3. Revascularization and Reinnervation of CGRP immunoreactive nerve fibers were established at the same time and it seems to be closed relatationship between revascularization and reinnervation.

The Role of PET in Thyroid Cancer (갑상선암에서 PET의 역할)

  • The role of PET in the diagnosis and management of thyroid cancer is discussed. The major role of F-18 FDG PET is on patients with discordant negative I-131 scan and a positive serum thyroglobulin values. F-18 FDG PET scan localized metastatic sites in I-131 scan-negative thyroid carcinoma patients with high accuracy. F-18 PET is also valuable in medullary thyroid cancer with high calcitonin level. Focal thyroid uptake in patients with non-thyroidal diseas has high likelihood of thyroid cancer.

고양이 후두상피내 neuroendocrine cell 의 형태학적 연구

  • amine 전구물질을 흡수하며 신경분비과립을 함유하고 있는 것으로 밝혀진 neuroendocrine cell의 후두상피 내에서의 구조와 분포를 알기 위하여 고양이 후두 상피조직을 이용, 면역조직화학적 염색법으로 관찰하였다. neuroendocrine cell은 방추체 모양이었으며 후두강쪽의 첨부돌기는 미세돌기를 갖고 있었으며 기저막에 접하고 있는 세포질돌기에는 많은 수의 농심과립을 함유하고 있었다. neuroendocrine cell은 calcitonin gene-related peptide, substance P, 5-hydroxytryptamine들 함유하고 있었으며 protein gene product 9.5, neuron-specific enolase에 면역양성반응을 보였으며 성문하부에 가장 많이 분포하고 있었다. 이로 미루어 neuroendocrine cell은 고양이 성대에서 어떤 자극을 받으면 다양한 펩티드를 분비하여 내분비 혹은 paracrine 경로를 통하여 후두 기능 조절에 관여한다고 사료된다.

Effects of Fermented Liquid of Males meles with Medicinal Plants on the Menopausal Symptoms, Estradiol Content and Bone Health Indices in the Postmenopausal Women (한약재를 첨가한 오소리 발효액이 폐경기 증후군과 Estradiol 농도 및 골대사에 미치는 영향)

  • The purpose of this study was to investigated the possibility of Meles meles as an edible functional resource. To assess the effects of Meles meles fermented liquid(ML) with medicinal plants in postmenopausal women, the menopause symptoms, hematological and serum chemical variables and bone health indices were examined. Twenty five postmenopausal women participated in the study. As a result, in postmenopausal women, ML supplements were effective in reducing the incidence of menopause symptoms, such as insomnia(44.0%), sensitivity(40.0%), headache(34.0%), hot flush and decreased vaginal lubrication(20.0%) after 12 weeks. Also serum estradiol and calcitonin and calcium levels were 23.27 pg/mL, 14.88 pg/mL and 8.86 mg/dL before the ML intake. Levels were significantly increased of estradiol and calcitonin and calcium after ML intake during 12 weeks to 30.36 pg/mL, 21.61 pg/mL and 10.01 mg/dL. Osteocalcin and bone alkaline phosphatase activity were 14.36 ng/mL, 21.55 U/L before the ML intake, but according to ML intake, the levels were significantly decreased. In conclusion, ML play benefical roles in the prevention of bone loss and menopause symptoms. Howere current data are not sufficient to determine the effective doses for benefical effect as well as harmful effect and support dietary recommendation level for ML. Therefore, our results should be viewed with caution. But Meles meles fermented liquid with medicinal plants can be used as possible food resources and functional food materials, more studies are needed to identify the proper mechanism of ML.

A Case of Severe Hypercalcemia Causing Acute Kidney Injury: An Unusual Presentation of Acute Lymphoblastic Leukemia

  • Severe hypercalcemia is rarely encountered in children, even though serum calcium concentrations above 15-16 mg/dL could be life-threatening. We present a patient having severe hypercalcemia and azotemia. A 14-year-old boy with no significant past medical history was referred to our hospital with hypercalcemia and azotemia. Laboratory and imaging studies excluded hyperparathyroidism and solid tumor. Other laboratory findings including a peripheral blood profile were unremarkable. His hypercalcemia was not improved with massive hydration, diuretics, or even hemodialysis, but noticeably reversed with administration of calcitonin. A bone marrow biopsy performed to rule out the possibility of hematological malignancy revealed acute lymphoblastic leukemia. His hypercalcemia and azotemia resolved shortly after initiation of induction chemotherapy. Results in this patient indicate that a hematological malignancy could present with severe hypercalcemia even though blast cells have not appeared in the peripheral blood. Therefore, extensive evaluation to determine the cause of hypercalcemia is necessary. Additionally, appropriate treatment, viz., hydration or administration of calcitonin is important to prevent complications of severe hypercalcemia, including renal failure and nephrocalcinosis.

Effects of KV Pharmacopuncture on MIA-induced Osteoarthritis Rat (KV 약침이 MIA로 관절염을 유도한 랫드에 미치는 효과)

  • The aim of this study is to investigate anti-arthritis activity using a KV pharmacopuncture. KV pharmacopuncture was inserted at BL40 for 4 weeks to SD-Rat, where arthritis was induced by monosodium iodoacetate (MIA) at 60 mg/ml. MMP-9, CTX II, LTB4, calcitonin and glycosaminoglycan level in serum were measured by ELISA. The cartilage of patella volume was examined and 3-D high-resolution reconstructions of the cartilage of patella were obtained using a Micro-CT system. Also, The histopathological change of knee was observed by H&E and safranin-O staining. Production of MMP-9, CTX II and LTB4 level in serum was decreased, respectively, in comparison with control. The other way, production of calcitonin and glycosaminoglycan level in serum was increased, respectively, in comparison with control. The cartilage of patella volume increased significantly. In addition, the KV group showed a increase in the cartilage volume and proteoglycan. These results may be used a remedy for new korea medicine to ease the symptoms mentioned above.

EFFECT OF EUGENOL ON REGULATION OF iCGRP RELEASE FROM THE BOVINE DENTAL PULP (치수에서 Eugenol이 iCGRP(immunoreactive calcitonin gene-related peptide)의 분비 조절에 미치는 영향)

  • Eugenol has been reported to reduce odontogenic pain and is known to have a structure similar to capsaicin, a potent stimulant of certain nociceptors. We have hypothesized that the analgesic effect of eugenol may be due, in part, to inhibition of capsaicin-sensitive nociceptors. To test this hypothesis, we evaluate whether eugenol inhibits capsaicin-sensitive release of immunoreactive calcitonin generated peptide(iCGRP) from bovine dental pulp. Freshly extracted bovine incisors were transported to the lab. on ice, Spilitted and pulp tissue was removed. The tissue was chopped into 200${\mu}m$ slices. Dental pulp was superfused(340 ${\mu}l/min$) in vitro with oxygenated Kreb's buffer. Eugenol and vehicle(0.02% 2-hydroxyl-${\beta}$-cyclodextrin) were administered prior to stimulation of pulp with capsaicin and iCGRP was measured by RIA. The results were as follows: 1. Administration of eugenol has no effect on basal release of iCGRP. 2. In the vehicle treated group, capsaicin evoked a 2.5-fold increase over basal iCGRP levels. 3. Administration of eugenol(600 ${\mu}M$) reduced capsaicin evoked release of iCGRP by more than 40%(153.4${\pm}$41.1% vs 258.9${\pm}$21.7%). 4. 2-hydroxylpropyl-${\beta}$-cyclodextrin of less than 0.02% is found to be an effective vehicle to dissolve eugenol without evoking iCGRP release from dental bovine pulp. These data indicate that eugenol inhibits pulpal capsaicin-sensitive fibers and suggest that intracanal medicament of eugenol may relieve pain, in part, by this mechanism.

Pharmacological Evidence that Calcitonin Gene-Related Peptide is Implicated in Cerebral Autoregulation

  • In the present study, it was aimed to asses the possibility that calcitonin gene-related peptide (CGRP) released in response to transient hypotension may contribute to the reflex autoregulation of cerebral blood flow as a putative modulator. Changes in pial arterial diameter (mean, 33.0 ${\pm}$ 1.1 $\mu\textrm{m}$) with changes in systemic arterial blood pressure (mean, 101.9 ${\pm}$ 2.7 mmHg) were observed directly through a closed cranial window in anesthetized normotensive rats. Image of the pial vessels was captured with a stereoscope connected to a CCD video camera and the diameter was measured with a microscaler. In the capsaicin-treated rats (one day prior to experiment, 50 nmol capsaicin injected intracisternally), both vasodilater and vasoconstrictor responses evoked by a transient hypotension and the reverse of blood pressure were markedly attenuated or almost abolished. When changes in pial arterial diameter were plotted as a function of changes in blood pressure, the slopes of both regression lines (for vasodilators and vasoconstrictors ) were markedly reduced. Similar reductions were evidenced under treatment wi th the CGRP antibody serum (1:1,000) and following CGRP receptor desensitization. However, the autoregulatory mechanics were neither affected by treatment wi th spantide (1 ${\mu}$M), substance P antagonist, nor by substance P receptor desensitization. Suffusion wi th mock cerebrospinal fluid containing CGRP and cromakalim caused a vasodilatation in a concentration-dependent manner, respectively and their effects were antagonized by glibenclamide. Substance P produced a vasodilatation, which was, however, little affected by glibenclamide. These observations indicate that the CGRP released from the perivascular sensory fibers in response to a hypotension is implicated in the modulation of the autoregulation of cerebral blood flow.
