  • 집속초음파 추출법(Innovative Extraction by Focused Ultrasound, INEFU)은 기존의 추출법에 비교하여 식물세포벽으로부터 기능성 성분을 낮은 온도에서 높은 추출 효율을 얻기 위한 추출공정으로 적용하기 위하여 연구되고 있다. 본 연구에서는 그린커피빈(COFFEA ARABICA SEED)에서 Chlorogenic acid, Caffein, Caffeic acid의 기능성 성분 추출을 위해 INEFU를 적용하여 추출 효율에 미치는 요인을 평가하였다. 추출 변수의 최적화를 위해 추출 온도 및 추출 시간을 요인으로 하여 순차적인 최적화를 진행하였다. 동일한 추출조건에서 저온추출(CEM)과 INEFU를 비교하였을 때 INEFU에서 Chlorogenic acid, Caffein이 각각 2.1 와 2.2배 증가함을 확인하였다. 또한 Caffeic acid는 저온추출(CEM)에서는 추출되지 않았으나, INEFU에서는 추출되는 것으로 확인하였다. 본 연구를 통해 INEFU가 기존의 추출 공정에 비해 기능성 성분 생산에 보다 효과적임을 확인하였다.

Protective Effects of the Phenolic Compounds from the Leaves of Hedera rhombea on Hepatic Injury (송악의 페놀성 물질의 간 보호효과)

  • Hedera rhombea (HR) has been used for treatments of hemorrage, chronic catarrh, jaundice, lithisis and convulsion. This study was done to isolate active compounds that have protective effect on liver damage. BuOH and EtOAc fractions of HR recovered serum glutamic pyruvic transaminase (GPT), glutamic oxaloacetic transaminase (GOT) and ${\gamma}-glutamyltranspeptidase$ (${\gamma}-GTP$) activities in CCl4 treated rats. We isolated 7 phenolic compounds from BuOH and EtOAc fractions, which were identified as 3-caffeoyl quinic acid, 3,4-di-O-caffeoyl quinic acid, 3,5-di-O-caffeoyl quinic acid, 4,5-di-O-caffeoyl quinic acid, caffeic acid, methyl 3,4-di-O-caffeoyl quinic acid and methyl 3,5-di-O-caffeoyl quinic acid by chemical and spectral analysis. These compounds reduced significantly serum GOT and GPT elevated by CCl4 treatment in rats, and 3-caffeoyl quinic acid, 3,5-di-O-caffeoyl quinic acid and caffeic acid also showed mild inhibitory activity against human immunodeficiency virus.

  • The antioxidant and anti-neuroinflammatory activities of water, 30, 70, and 100% ethanol extracts of leaves of three different species of bamboo (Phyllostachys nigra, P. bambusoides, and Sasa borealis) were investigated. The levels of total polyphenol and flavonoid were measured, and antioxidant activity was evaluated using various antioxidant assays (DPPH, ABTS, and FRAP). Lipopolysaccharide (LPS)-induced BV2 microglial cell activation was used to evaluate the anti-neuroinflammatory properties of the bamboo leaf extracts. Treatment with both aqueous and ethanolic extracts showed no cytotoxicity in BV-2 microglial cells. Pre-treatment of BV-2 cells with bamboo leaf extracts significantly inhibited LPS-induced excessive production of nitric oxide in a dose-dependent manner. Moreover, phytochemical analysis based on the extraction solvent showed that caffeic acid, p-coumaric acid, and tricin are the principal constituents of all three bamboo leaf extracts. Therefore, our findings suggest that bamboo leaf extract contains potent antioxidants and anti-neuroinflammatory compounds that can be used as potential therapeutic agents for the treat neuroinflammatory diseases.

Quantification of Caffeic Acid and Rosmarinic Acid and Antioxidant Activities of Hot-water Extracts from Leaves of Perilla frutescens (품종별 들깻잎 열수추출물의 caffeic acid와 rosmarinic acid의 함량 및 항산화력 비교)

  • The principal objective of this investigation was to identify adequate species for the harvest of Perilla frutescens leaves, which provide profound antioxidant activities, and harbor abundant caffeic acid and rosmarinic acid. Namchun, Donggeul-2, Bora, Sae-bora and Neul-bora variants of the plant were assessed herein. In this study, we evaluated the antioxidant effects of these plants, and utilized an HPLC system to verify their caffeic acid and rosmarinic acid contents. Dried Perilla frutescens leaves were boiled at a temperature of $100^{\circ}C$ for three hours, and were lyopholized in a freezedryer. The extracts were then processed in order to confirm their antioxidant activities via 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical-scavenging activity assay, ferric-reducing antioxidant power (FRAP) assay, total flavonoid content and total polyphenol content assays. According to the observed antioxidant results of the five tested species of Perilla frutescens leaves, Bora and Donggeul-2 were shown to have more potent antioxidant activities than the other tested variants. The scavenge 50% of DPPH radical ($SC_50$ of DPPH) values were 241 ${\mu}g$ DM/mL in Donggeul-2 and 261 ${\mu}g$ DM/mL in Bora. Based on the results of the FRAP assay, the Bora variant showed a value of 796 mM $FeSO_4$ $7H_2O/g$ DM, and Donggeul-2 exhibited a value of 748 mM $FeSO_4$ $7H_2O/g$ DM, of which the total polyphenol contents were measured as 69.4 g GAE/kg DM and 61.8 g GAE/kg DM, respectively. Moreover, the Bora variant had the highest level of caffeic acid, and Donggeul-2 showed the highest rosmarinic acid content among the tested samples (0.87 mg/100 g wet base and 121 mg/100 g wet base, respectively). According to the results of this experiment, we selected two species, Bora and Donggeul-2, which were both verified to contain adequate and favorable antioxidant activities.

Autotoxicity of alfalfa flower extract and its allelopathy to Echinochloa crus-galli (알팔파 꽃 추출물의 Autotoxicity와 돌피에 대한 Allelopathy)

  • The aim of this study was to separate or purify some bioactive compounds from flowers of alfalfa(Medicago sativa L.) and to test of the isolated compounds on alfalfa for their autotoxicity and on Echinochloa crus-galli for their allelopathy for seed germination and seedling weight. Using thin layer chromatography(TLC) of $CHCl_3$ extracts, the most inhibitory band to alfalfa seed germination was determined. Germination inhibition of this extract suggested a complex chemical interaction. Separation and purification of compounds with CHCl$_3$ extract of fresh alfalfa flowers were conducted by a silica gel TLC, and microcrystalline cellulose TLC(MCTLC), followed by droplet countercurrent chromatography(DCCC) bioassay. Preliminary identification was done by high perfomance liquid chromatography(HPLC) on the most inhibitory fractions in DCCC. Ferulic acid, caffeic acid, vanillic acid, rutin, narringin were identified in fraction 5 and ferulic acid, caffeic acid, vanillic acid, rutin, coumarin in fraction 6. The phytotoxicity of their individual compound was tested on alfalfa and Echinochloa crus-galli seed germination and seedling weight. Coumarin and ferulic acid showed the most inhibitory effect on alfalfa seed germination and Echinochloa crus-galli seedling fresh and dry weight. These compounds may be, at least in part, involved in autotoxicity and allelopathy.

  • Regardless of the test species, germination was adversely affected by the different concentrations of the acidic and neutral fractions of sunflower root exudates while the basic and aqueous fractions had no effect on germination. In both test species, root lengths were inhibited slightly more than shoot lengths. Significant reduction in fresh weights of the test species was observed when the test species were treated with the acidic and neutral fractions but not with the basic and aqueous fractions. Six compounds, hydroquinone, ${\beta}$-resorcyclic acid, vanillic acid, caffeic acid, salicylic acid, and quercetin, were characterized from the acidic fraction. Seven compounds, hydroquinone, gentisic acid, ${\beta}$-resorcyclic acid, vanillic acid, caffeic acid, ferulic acid, and quercetin, were elucidated from the neutral fraction.

  • Antithiamine activity of raw and cooked brackens(Pteridium aquilinum) was evaluated under various reaction conditions by means of the thiochrome fluorescence method. The effects of caffeic acid and cysteine on the thiamine decomposition were also determined by thiochrome fluorescence and Lactobacillus viridescens bioassay methods. A water extract of raw bracken exhibited a high antithiamine activity which was increased with higher pH, temperature, incubation time and concentration of bracken. The influence of reaction conditions was less apparent in cooked bracken than in raw bracken. Caffeic acid stimulated the thiamine decomposition whereas cysteine showed a suppressive effect. The effect of cysteine was lower in the decomposition of thiamine by bracken extract.

  • The emergence of new infectious diseases, the resurgence of several infections that appeared to have been controlled and the increase in bacterial resistance have created the necessity for studies directed towards the development of new antimicrobials. In the present study, we have synthesized a novel antioxidant ZnO nanoparticle that is newly designed and prepared by simple surface modification process. Antioxidative functionality is provided by the immobilization of antioxidant 3-(3,4-dihydroxyphenyl)-2-propenoic acid (caffeic acid, CA) onto the surface of ZnO nanoparticles. Microstructure and physical properties of the ZnO@CA nanoparticles were investigated by field emission scanning electron microscopy (FE-SEM), transmission electron microscopy (TEM), infrared spectroscopy (IR) and steady state spectroscopic methods. Antimicrobial Activities of ZnO@CA nanoparticles were measured against various bacterial strains using antibacterial testing methods.
