가축질병 균주에 대한 소목의 항균활성 (Antimicrobial Activity of Caesalpinia sappan against Animal husbandry disease.)

  • 소목추출물을 제조하여 가축질병에 관련이 있는 균에 대한 항균활성을 조사하였다. 소목추출물은 그람양성균에 대해 높은 항균활성을 보였으며 그 중 S. epidermidis에서 가장 높은 항균활성을 보였다. 소목추출물의 최소저해농도는 MeOH추출물은 0.8∼16 mg/ml 범위였으며 EtOH츤출물은 0.8∼10 mg/ml농도에서 항균활성을 보였으며 소목 분회물의 항균활성은 EtOAc층이 가장 높은 활성을 나타냈다. 열에 의한 항균활성 변화에 있어서는 대부분의 균주에서 항균력은 안정적이며 pH에 의한 항균활성의 변화에서 S.aureus와 Sal. typhimurum에서 산 처리 시 감소하는 경향을 보였으나 대부분의 균주에서 안정적이었다. 소목 추출물의 미생물증식억제 효과를 조사하기 위해 증식배지에 0, 100, 300및 500 ppm의 추출물을 첨가하여 균주의 증식 억제효과를 조사하였다. 그람양성균인 S. aureus 와 S. epidermidis는 첨가농도에 관계없이 배양 후 72시간까지 증식이 정지된 상태를 보였으며 그람 음성균에서는 100ppm이상 첨가 시 증식이 저지되거나 억제효과가 있었다. 이런 결과를 종합하면 소목 추출물은 그람음성균에 비해 그람양성균에 더욱 효과적인 항균활성이 있다고 보여 진다.

견직물의 소목염색에서 단백질 전처리 효과 (The Effects of the Pre-treatments with Proteins on Dyeing of Silk Fabric with Caesalpinia sappan)

  • This study investigated the effects of the working mechanisms of proteins, mordants, and dyes, as well as the mordanting order, on dye uptake by silk fabric pre-treated with proteins and dyed with freeze-dried sappan wood water extract. Soybean protein and sodium caseinate were used as the proteins. 1. When Al mordants were not used, the dyeability of the fabrics increased upon protein pre-treatment as compared to the case without treatment. 2. Dyeing with protein pre-treatment, followed by mordanting, led to the highest dye uptake, and the optimal protein concentration was 5%. 3. The K/S values slightly decreased with an increase in the dyeing temperature, and the fabric turned dark red in color when dyeing was carried out at increasing temperature. Fabrics showed the highest dye uptake at $40^{\circ}C$. 4. Regarding the effect of time, the K/S values of the fabrics with and without protein treatment showed almost no increase after the initial dyeing time of 10min; further, there was hardly any difference in the cases with and without protein pre-treatment. 5. In case of protein pre-treatment fabrics, the washing fastness was level 2. The dry cleaning fastness showed very excellent result with level 4-5. The rubbing fastness was better in dry rubbing than in wet rubbing of the fabrics. For the light fastness, all dyed fabrics showed low fastness.

TPA로 유도된 마우스 귀 부종 동물모델에서 소목추출물의 항염증 효과 (Anti-Inflammatory Effects of Extracts from Caesalpinia sappan L. on Skin Inflammation)

  • 본 연구를 통하여 TPA로 유도한 마우스 귀 부종 염증반응에 대한 소목추출물의 항염증 효능과 기전을 확인하였다. 소목추출물은 TPA로 유도한 마우스 귀 부종을 억제하였으며, TPA에 의한 염증관련 단백질인 COX-2 발현 및 cytokine(IL-6, TNF-${\alpha}$ 그리고 IL-$1{\beta}$)의 mRNA 발현을 현저히 감소시켰다. 또한 TPA에 의한 NF-${\kappa}B$ 및 MAPK의 활성을 억제하였다. 본 연구 결과, 소목추출물은 NF-${\kappa}B$ 및 MAPK의 신호전달을 억제함으로서 항염증 효능을 나타내었다.

Brazilin Inhibits of TPA-induced MMP-9 Expression Via the Suppression of NF-${\kappa}B$ Activation in MCF-7 Human Breast Carcinoma Cells

  • Metastasis is the primary cause of from breast cancer mortality. Cell migration and invasion play important roles in neoplastic metastasis. Matrix metalloproteinase-9 (MMP-9), which degrades the extracellular matrix (ECM), plays an important role in cancer cell invasion. NF-${\kappa}B$ is transcription factor important in the regulation of MMP-9, as the promoter of MMP-9 gene contains binding sites for NF-${\kappa}B$. Brazilin, an active component of sappan wood (Caesalpinia sappan), decreases TPA-induced MMP-9 expression and invasion in MCF-7 cells. Also, brazilin suppressed NF-${\kappa}B$ activation in TPA-treated MCF-7 cells. Taken together, we demonstrated that the inhibition of TPA-induced MMP-9 expression and cell invasion by brazilin is mediated by the suppression of the NF-${\kappa}B$ pathway in MCF-7 cells. This result suggest brazilin provide a potential therapeutic app roach for the treatment of breast cancer.

  • Ethanol ($70\%$) extract of Caesalpinia sappan L. and Forsythiae suspensa Vahl. showed $84\%$ and $72\%$ of anti-urease activity against Helicobacter pylori, respectively. Among the fraction of Artemisia asiatica Nakai extract using methanol ($80\%$), water, ethyl acetate and butanol ethyl acetate fraction showed $90\%$ of the anti-urease activity.

  • Brazilin, the main constitutent of Caesalpinia sappan, was examined for its immunomodulating activities in normal CBA mice. Mitogen induced proliferation and production of ConA induced T-cell growth factors (TCGF) of splenocytes were significantly reduced in brazilin treated group, cmpared to control group. It was also found that suppressor activities of splenocytes in brazilin treated group was significantly increased compared to those in normal control group.

  • Brazilin is the phenolic compound isolated from the Caesalpinia sappan. This compound has shown a wide range of physiological properties, such as hypoglycemic, anticonvulsant, vasorelaxing, and immunomodulating effects. In this study, we have found that brazilin induced DNA strand scissions in the presence of Cu(II) and this DNA cleavages were mediated by reactive oxygen species. (omitted)

천연염료의 매염에 따른 염색성 및 물성에 관한 연구;소목과 꼭두서니를 중심으로 (A Study on the Dyeability and Physical Properties of Mordanted and natural-dyed Fabrics)

  • This study was performed to investigate the effect of mordants on the dyeability colorfastness to light and to laundering and biological properties of mordanted and natural-dyed cotton and silk substrates. Natural red dyes were extracted from Caesalpinia Sappan L. (Sappan wood) and Rubia Akane Nakai (Madder) by boil water Five different compounds of Al, Cr, Cu, Fe and Sn were used as mordanting agents. The result of this study are sumarized as follows ; 1. In the of the fabrics mordanted and dyed with sappan wood K/S value increases and λmax shifts to the longer wavelength as mordant concentration increases. Color of the fabrics dyed with sappan wood and madder change to redder bluer and lighter after mordanting. 2. As the number of repetition of dyeing-mordnating process increase the color of the dyed fabrics tend to be redder bluer and darker. Mordanted dyes with Al and Sn make chroma increase. The quantity of mordant absorbed in the silk fabric increase as mordant concentration increase. More Fe is absorbed in silk than Cr is. 3. After being exposed to light the fabrics dyed with sappan wood and madder change to less red Fe and Cu mordanting yeild better colorfastness to light than others does. Natural-dyed fabrics exposed to light tend to be faded and whitened in CIELAB chromaticity co-ordinates. So we might trace back the original color of ancient faded fabrics by changes in color of natural dyes. 4. After washing the colors of natural dyes change to more blue, Fe and Cu mordanting give better colorfastness to wash than others do. 5. The soil-burial cotton is more susceptible to mildew and rot than the silk is. Antimicrobial activity of the natural-dyed cotton can be enhanced by Cu and silk can be enhanced by Cu and Cr mordanting respectively.

Comparing the Dyeing Properties of Synthetic Mordants and Hwangsu Spring

  • To evaluate the Hwangsu spring as natural mordants effect on dyeing. The natural mordant effect of Hwangsu spring was analyzed by pH, temperature, trace elements and organic matter. Hwangsu spring having pH 2.42 and $14.4^{\circ}C$ was harvested at the end of September, 2011, in Yeongcheon(Gyeongbuk Province, Korea) and left on a $0^{\circ}C$ and $25^{\circ}C$ before use. The Result of ICP analysis, it contain bned Fe(414.9 ppm), Al(88.9 ppm), Mn(4.9 ppm). Dyeing and post-mordant procedure; Fabrics(cotton, silk, rayon) were dyed with natural colorants(Sopbora Japonica L., Caesalpinia sappan L., Allium cepa L.) for 20 min at $80^{\circ}C$. Hwangsu spring, aluminium sulfate $14-18H_2O(Al_2(SO_4)_3{\cdot}14-18H_2O)$, iron sulfate peptahydrate($FeSO_4{\cdot}7H_2O$), copper sulfate pentahydrate($CuSO_4{\cdot}5H_2O$) were used by post mordants. But in the case of Caesalpinia sappan L. was dyed after pre-mordanting with Al(II). The fabrics were dyed with each mordant solutions at $25^{\circ}C$ for 10min. The colorless also was measured by color-differnce meter. Comparison with a synthetic mordants, the K/S values of cotton fabrics dyed with Hwangsu sping were increased.

생약제가 면역세포 활성화에 미치는 영향 (Effect of Edible and Medicinal Plants on the Activation of Immune Cells)

  • In order to evaluate the effect of the extracts of eidble and medicinalplant son the activation of immune cells, measurements were made by ELISA and radioimmunoassay on the degree of release for the tumor necrosis factor (TNF) and neopterin by the edible and medicinal plants in peripheral blood cells. The results of measurements of TNF in the supernatant cultured liquid showed nothing in t도 control which does not have any edible and medicinal plants. However, measurements of TNF 9pg/ml) in the samples are given as follows : 716.7 in lipopolysaccharide (LPS 1 g/ml), 465.2 Rheum plamatum L.m302.7 Sanguisorba offciinalis L. 818.2 Rubus coreanus M, 328.3 Terminalia chebula R., 426.6 AReca catechu L. 227.0 Eugenia caryophiliata T., 272.9 Ephedra sinica S., 30.1 Caesalpinia sappan L., 474.0 Chaenomeles japonica L., 396.0 Cornus officinalis S.in edible and medicinalplants. ENopterin (n mole/L) value showed below the check point in the control group, however, the values are 11.0 in LPS, and edible and medicinal plants, 5.3 Rheum palmatum L., 11.6 Eugenia caryophiliata T., 5.5 Ephedra sinica S., 4.5 Caesalpinia sappan L., 4.3 Chaenomelees japonica L.3.7 Cornus officinalis S. In order to find m RNA levels of Cytokines increased by edible and medicinal plants, total RNA was separated from mononuclear cells treated 5 hrs with Rubus coreanus M. and then administrated for RT-PCR. The considerable increases of the m RNA of TNF, IL-1 $\alpha$ and IL-6 were observed.
