Studies on the Antioxidative Effect of Korean Traditional Medicinal Plants as Skin Care Product (피부보호용품제조를 위한 한국재래약초의 항산화성 연구)

  • The total phenolic contents and antioxidative activities of the ethanol and methanol extracts from 9 Korean medicinal herbs were tested for the application of skin care products such as soaf and lotion. For the evaluation of antioxidative activity of the extract the method of electron donating ability with resolution reagent DPPH(1 ,1-diphenyl-2-picryl hydrazyl) and TBARS (Thiobarbituric acid reactive substances) value were used in comparison with α-tocopherol and BHA. As the results, the total phenolic contents of the extracts calculated as chlorogenic acid were ranged from 0.063-0.124 mg/ml. All the medicinal plant extracts showed some appreciable antioxidative effects. Among them, Zanthoxylum piperitum, Cinnamomum cassia and Caesalpinia sappan were shown to have relatively high antioxidative effects comparable with BHA and α-tocopherol..

  • 천연염색은 독성과 환경오염이 적으며 합성염료로는 얻기 어려운 자연스러운 색감 등을 표현할 수 있는 장점에 비하여 염색과정 및 보관의 어려움, 재현성 부족, 낮은 염착량과 견뢰도, 염색과정에서 노동력이 많이 소비되어 대량생산의 어려움 등의 문제점으로 인해 아직 공업화되지 못하고 있다. 천연염료는 합성염료에 비해 색상이 차분하고 은은하며 변퇴색이 일어나도 안정된 색감을 나타낼 뿐 아니라 매염제에 의해 다양한 명도와 색상변화를 꾀할 수 있어 천연염색물에 대한 소비자의 수요가 최근 증가하고 있다. 본 연구에서는 천연염색 공정의 기계화를 통해 대량생산 및 재현성 확보가 가능한 100yd급 염색시스템을 설계 제작하고 천연염료인 소목을 이용하여, 면 및 견을 포함한 10종의 원단에 염액비(1:20), 매염제($FeSO_4{\cdot}7H_2O$)를 사용하여 염색 온도변화($20^{\circ}C{\sim}80^{\circ}C$)에 따른 원단별 염색특성을 평가하였다. 평가결과 세탁견뢰도는 온도가 증가할수록 대부분의 원단에서 견뢰도가 상승하였으며, 염색된 원단내의 색차(${\Delta}E$) 평가에서 대부분의 원단에서 1.0미만의 균염성을 나타내었다.

  • Chitin is a derived product from the shell of shrimp or crab. Chitosan, a deacetylated product of chitin, has widely been used in the biomedical sector, food industry, and textile industry. Chitosan exhibits fiber-forming property under certain conditions. Nonwoven fabrics made of chitosan fibers may have diverse applications in the industry. Previous studies have revealed that the dye uptake properties of natural dyestuffs improved by the chitosan pretreatment on the fabric specimens. In this case, fabric specimen is coated with acidic salt form of chitosan, which is different from the pure chitosan, since the coating process employes coating with the acidic solution of the chitosan and subesquent drying. In this study, chitosan nonwoven fabric samples were prepared from chitosan sample having deacetylation degree of $100\%$ and molecular weight of 650,000. Chitosan nonwoven fabrics maintain the form of $-NH_2$ end-group. These in turn exhibit higher dye uptake ability than the fabrics coated with chitosan acidic solutions do.

Neuroprotective Effects of Medicinal Herbs in the Transient Focal Ischemia

  • It is thought that highly reactive oxygen radicals generated at the ischemia-reperfusion in case of strokes play an important role in damaging the brain. We examined the neuroprotective effects from the several medicinal herbs in the transient ischemic rat model and compared their effects with the free radical scavenging activities. Transient ischemia was induced by intraluminal occusion of the right middle cerebral artety for 120 min and reperfusion was continued for 22 h in rats. The free radical scavenging properties of medicinal herbs were examined in vitro by determination of the interaction with the 1,l-diphenyl-2-picrylhydrazyl (DPPH) stable free radical. Aqueous extracts of 11 medicinal herbs (200 mg/kg) were orally administered, promptly prior to reperfusion and 2 h after reperfusion. Total infarction volume in the ipsilateral hemisphere of ischemia reperfusion rats was significantly lowered by the treatment of 7 medicinal herbs (Sophora flavescens, Lycopus lucidus, Sanguisorba officinalis, Caesalpinia sappan, Albizia julibrissin, Rubia akane, Psoralea corylifolia, Prunella vulgaris). However, all of these medicinal herbs did not show antioxidative activities. These results suggest that neuroprotective effects of several drugs are not always correlated with their antioxidative properties.

  • One hundred and seventy crude drugs were screened for prolyl endopeptidase (PEP) inhibitors. Among them, 80% methanolic extract of 18 medicinal plants such as Polygonum cuspi data, Sanguisorba officinalis, Eugenia caryophyllata, Rubus coreanus, Cinnamomum cassia (Cassiae Cortex and Cinammomi Ramulus), Rheum palmatum, Ulmus pumila, Sorbus commixta, Areca catechu, Uncaria sinensis, Terminalia chebula, Caesalpinia sappan, Nelumbo nucifera, Machilus thunbergii, Paeonia moutan, Elscholtzia patrini and Cynomorium coccineum inhibited more than 70% of PEP activity at a concentration of 40 ppm. The active principles of P. moutan, M. thunbergii, T chebula, A. catechu, S. commixta, R. palmatum, R. coreanus, E. caryophyllata and P. cuspidata were transferred into organic solvents, which showed more than 75% inhibition at 5 ppm.

Screening of Some Plant Extracts for Inhibitory Activities on Hepatitis B Virus Replication (수종 생약재의 간염 B형 바이러스 증식 억제 활성 검색)

  • This study was undertaken to test for anti-hepatitis B virus (HBV) activity of the aqueous extracts prepared from 9 medicinal plants of Korea (Cornus officinalis, Caesalpinia sappan, Rubus coreanus, Lycium chinense, Artemisia capillaris, Isatis tinctoria, Phyllanthus urinaria, Lysimachia christinae, Lonicera japonica). Aqueous extracts were tested for cytotoxicity and assayed for inhibition of HBV replication by measurement of HBV DNA and surface antigen (HBsAg) levels in the extracellular medium f HepG2 2.2.15 cells. The extract from Rubus coreanus, Artemisia capillaris, Phyllanthus urinaria decreased the levels of extracellular HBV virion DNA at concentrations ranging from 128 to $256\;{\mu}g/ml$ and inhibited the production fo HBsAg dose-dependently without showing cytotoxicity. Our findings suggest that these three hebal medicinal plants may have potential to develop as specific anti-HBV drugs in the future.

Effects of Brazilin on Erythrocyte Deformability and Its Related Biochemical Factors in Streptozotocin Induced Diabetic Rats

  • Impaired erythrocyte deformability was considered to paly an important role in microcirculatory disturbances. We recently confirmed that the brazilin, the main active principle of Caesalpinia sappan, enhanced activity of erythrocyte deformability and reduced blood viscosity. In this study, we examined the effects of brazilin on three biochemical parameters (ATP, 2, 3-diphosphoglycerate, and calcium) which influenced erythrocyte deformability. Treatment with barzilin increased erythrocyte deformability and ATP concentrations in streptozotocin-induced diabetic rats. Concentrations of 2, 3-diphosphoglycerate and calcium in diabetic rats following brazilin administration were decreased significantly compared to those of diabetic control rats. The results suggest that brazilin have a potential effect to improve rheological abnormalities in diabetes.

Effect of Storage Conditions on the Color and the Mechanical Properties of Fabrics dyed with Natural Dyes (천연염색 직물의 환경조건에 따른 변.퇴색 및 물성변화에 관한 연구)

  • Cotton and silk fabrics dyed with brazilein(Caesalpinia Sappan), berberine (Phellodendron Amurense), and shikonin(Lithospermum Erythrorhizon) were stored in the air, in the water, and under the ground for about one year. The change of the color, the flexibility, and the breaking strength were measured at the intervals of few weeks. The results are as follows; 1. The color of the fabrics changed most extensively in the fabrics that were under the ground, then in the order of the ones that were in the water, and in the air. The color of the fabrics changed to the achromatic color over the time. 2. The flexibility change among the dyes was similar. All the fabrics became stiff under the ground and in the water over the storage time. 3. In general, cotton and silk fabrics dyed with berberine showed better strength retention than the fabrics dyed with other substances. 4. The strength retention of cotton fabrics was high in the order of the fabrics which were in the air, in the water, and under the ground. Only minuscule change occurred in the strength of the cotton fabrics. The strength of the fabrics that were in the water and under the ground decreased remarkably after 30 weeks and 20 weeks respectively. 5. The strength retention of silk fabrics was high in the order of the fabrics which were in the air, under the ground, and in the water. In the air, the strength on silk fabrics decreased rapidly after 30 weeks. In the water, the strength of silk fabrics decreased more rapidly than that of the cotton fabrics. Under the ground, the strength retention of silk was higher than that of cotton.

Screening for Antioxidative Activity of Oriental Medicinal Plant Materials (시판 한약재에 대한 항산화 활성 검정)

  • Antioxidative effect of 42 medicinal plant extracts was screened to search natural antioxidants. The extracts of Terminalia chebula, Caesalpinia sappan, Sanguisorba officinalis, Rubus coreanus, and Alpinia katsumadai showed strong inhibition effect on DPPH radical, and LDL oxidation. Those of Eugenia caryophyllata, Alpinia officinarum, Zingiber officinale, Xanthium strumarium, Sophora japonica, Aristolochia contorta, and Alpinia katsumadai exhibited potent antioxidative activity on linoleic acid peroxidation. Among 42 medicinal plant materials, the extract of Terminalia chebula showed the highest scavenging activity $(3.08\;{\mu}g/ml)$ on DPPH radical which was higher than that $(13.52\;{\mu}g/ml)$ of ${\alpha}-tocopherol$. The extract of Terminalia chebula showed also stronger inhibition activity on LDL oxidation than that of ${\alpha}-tocopherol$ did.

Antimicrobial activity of extracts from medicinal herbs against Escherichia coli (Escherichia coli에 대한 한약재추출물의 항균활성)

  • The extracts from 40 different traditional medicinal herbs were used to investigate the antimicrobial activities against Escherichia coli. Among them, the extracts from Paeonia suffruticosa (PS), Siegesbeckia orientalis (SO), Schizandra chinensis (SC), Caesalpinia sappan (CS) and Rhus javanica (RJ) exhibited high antimicrobial activities against E. coli, Minimum inhibitory concentrations (MIC) of the RJ extract against E. coli were 0.8 mg/ml. After heating treatment of these extracts, the antimicrobial activities against E. coli were significantly reduced in case of the CS extract. After alkaline or acid treatment of these extracts, the antimicrobial activities against E. coli were significantly increased in the PS extract but reduced in both SO and RJ extract. Since extracts from RJ and CS exhibited the highest antimicrobial activities, bacterial growth-inhibiting activities against E. coli by these two extracts were further examined. Optical density at 620 nm after 24 hours incubation of E. coli in the presence of 100, 300 or 500 ppm of RJ extract ranged from 0.1 to 0.2 compared to 0.35~0.65 in the absence of RJ extract, indicating that growth of E. coli was significantly inhibited within 24 hours by the addition of at least 100 ppm of RJ extract. Optical density at 620 nm after 24 hours incubation of E. coli in the presence of 300 or 500 ppm of CS extract ranged from 0.01 to 0.25 compared to 0.5~0.55 in the absence of CS extract, indicating that growth of E. coli was also significantly inhibited within 24 hours by the addition of at least 300 ppm of CS extract. In conclusion, these findings suggest that extracts from RJ and CS may play important roles for antimicrobial activities against E. coli causing various animal diseases.
