Mechanism of Cadmium Accumulation in the Cell of Cadmium-Tolerant Bacterium, Pseudomonas putida (카드뮴내성균(耐性菌)(Pseudomonas putida)에 의한 균체내(菌體內) 카드뮴 축적(蓄積) 기작(機作))

  • A mechanistic study by which Cadmium-tolerant P.Putida C1 accumulates high conc of Cd in its cell body was performed. Approximately 57% Cd accumulated was distributed on the cell wall and the other 43% portion was in cytoplasm. 84% Cd of the Cd in the cell wall fractions present in the polyphosphate-polysaccharide fractions, but most of Cd in the cytoplasm fraction was in protein and nucleic acid. Cadmium affected the protein synthesis in P. Putida. The intracellular protein content was decreased by cadmium addition, but the soluble protein precipitated by ammonium sulfate($30{\sim}75%$ satruation) was increased as compared to that from the cells grwon without cadmium. Furthermore, in the cells grown with of cadmium, high-molecular-weight soluble protein was increased, with of cadmium, high-molecular-weight soluble protein was increased, compared with the cells grown without cadmium, but low-molecular-weight soluble protein was decreased. These results indicate that Cd inhibited the intracellular protein biosynthesis but enhance biosynthesis of the high-molecular-weight soluble protein precipitate by ammonium sulfate($30{\sim}75%$ saturation).

Effects of Cadmium on Glucose Transport in L6 Myocytes (L6 근육세포에서 포도당 수송능에 미치는 $CdCl_2$의 영향)

  • This study was aimed to know the effect of cadmium chloride (CdCl₂) on glucose transport in L6 myotube and its action mechanism. CdCl₂ increased the 2-deoxy- (l-3H)-D-glucose (2-DOG) uptake 1.9 and 2.4 fold at 10 and 25 μM respectively. To investigate the stimulating-mechanism of glucose transport induced by CdCl₂, the wortmannin and PD98059 were used as PI3K (phosphatidylinositol 3-kinase) inhibitor and MAPK inhibitor respectively, which did not affect 2-DOG uptake. This fact suggests that CdCl₂ induced 2-DOG uptake may not be concerned to the insulin signalling pathway. Whereas nifedipine, a calcium channel blocker, and trifluoperazine, a calmodulin inhibitor, were found to inhibit the 2-DOG uptake stimulted by CdCl₂. In addition, we also measured the ROS (reactive oxygen species) production and GSH level in L6 myotube to investigate the correlation between the glucose uptake and ROS. CdCl₂(25 μM) increased ROS generation approximately 1.5 fold and changed the cellular GSH level, but GSSG/GSH ratio remained unchanged. CdCl₂ stimulated 2-DOG uptake and ROS generation were inhibited by N-acetylcystein. And BSO pretreatment, a potent inhibitor of γ-GCS, resulted in the dramatic decrease of 2-DOG uptake and also the increase of the sensitivity to cadmium cytotoxicity. The obtained results suggest that CdCl₂-stimulated glucose uptake might be based on the activation of HMP shunt as an antioxidant defense mechanism of the cells.

A Study on the Electrical and Optical Properties of CdS Thin Films Deposited with Different Conditions for Solar Cell Applications (태양전지용 CdS 박막의 제조 조건에 따른 전기적 광학적 특성에 관한 연구)

  • Cadmium sulphide (CdS) thin film, which is used as a window layer of heterojunction solar cell, on the glass substrate was deposited by vacuum evaporation. Effects of deposition conditions such as the source and substrate temperature on electrical and optical properties of CdS films was investigated. As the source temperature was increased, the deposition rate of CdS films was increased. In addition, the optical transmittance and the electrical resistivity of CdS films were decreased as the source temperature was increased. This results were attributed to the increase of excess Cd amount in the film. The crystal structure of CdS films exhibited the hexagonal phase with preferential orientation of the (002) plane. As the substrate temperature was increased, the crystal structure of CdS films was improved and the resistivity of the films was increased due to the decrease of excess Cd in film.

  • The synthetic conditions of $ ZrSiO_4-Cd(S_xS_{1-x})$ stains from CdS, Se, $SiO_2$ , 4ZrO_2$ and Lif were investigated and the colors were examined. Colors of the stains prepared were yellow orange red pink ruby and violet in relation to both the content of CdS-Se in $ZrSiO_4$ and firing temperature. Colors of these series stains were thermally stabilized probably by the structural stability of zirconium silicate. Furthermore by the result of X-ray diffraction analysis it is assumed that color of the zircon cadmium sulphoselenide $ ZrSiO_4-Cd(S_xS_{1-x})$ stain is developed by neither the coloring ions in $ZrSiO_4$ lattice nor the solid solution of $ZrSiO_4$ and $Cd(S_xS_{1-x})$ but by the small crystals of $Cd(S_xS_{1-x})$ being occluded by the zirconium silicate during sintering process.

Effects of Metal-ions on Enzyme Activities from Hansenula anomala B-7 Grown in Medium Containing Cadmium (카드뮴 함유 배지에서 배양된 Hansenula anomala B-7의 Malate Dehydrogenase 활성에 미치는 금속 이온의 영향)

  • This study was carried out to investigate the effect of cadmium ion on activities of cadmium-adapted malate dehydrogenase (adapted-MDH), which is defined to be an enzyme obtained from an extreme cadmium-tolerant yeast Hansenula anomaul B-7 grown in medium containing 1 mM cadmium ion. Cadmium-nonadapted malate dehydrogenase (nonadapted-MDH), which is defined to be enzyme expressed in the cells grown in $Cd^{2+}$ -free medium was also characterized by the same manner. Activities of the adapted-MDH and the nonadapted-MDH were strongly induced to 450% and to 150% in comparision with the control examined with 1 mM $Cd^{2+}$, respectively. The adapted-MDH activity was stimulated to 147%, 150%, and 135% compared with the control analyzed with 1 mM $Zn^{2+}$, 1 mM $Mn^{2+}$, and 1 mM $Ca^{2+}$, respectively and to 925%, and 250% compared with the control analyzed in the presence of 2 mM $Cd^{2+}$, and 2.5 mM $Zn^{2+}$, respectively. Km values of the adapted-MDH and the nonadapted-MDH were calculated to be the same 6.9 mM for L-malate, respectively. The Km value of the nonadapted-MDH was not changed by $Cd^{2+}$ while Vmax of the nonadapted-MDH was increased by $Cd^{2+}$. In contrast, both the Km and the Vmax values of the adapted-MDH were changed by $Cd^{2+}$.

Effect of Mungbean Sprouts Juice on Cadmium-Induced Hepatotoxicity in Rats (녹두나물 생즙이 카드뮴에 의한 흰쥐의 간손상에 미치는 영향)

  • The effects of mungbean sprout juice on cadmium-induced hepatotoxicity in rats were investigated. Sprague-Dawley rats weighing about 90g were divided into 4 groups and raised for 6 weeks. ; control group(CON), mungbean sprouts juice-administered group(MSJ), cadmium-administered group(CAD) fed water containing 40 ppm cadmium chloride and mung bean sprouts juice and cadmium-administered group(MCD). The diet was supplied every day for the measurement of feed efficiency ratio(FER) and net weight gain was measured every 3 days. The activities of serum glutamic oxaloactic transaminase(GOT) and glutamic pyruvic transaminase(GPT), superoxide dimutase(SOD), catalase and glutathione peroxidase(GSH-Px) in the liver and the hepatic contents of glutathione were examined. The contents of Cd in liver and kidney of the rats were determined by using ICP(Inductively Coupled Plasma Emission Spectrophotometer). Growth rate and FER were decreased in CAD group, compared with CON group but the changes were not significant in MCD group. The activities of serum GOT and GPT, SOD, catalase and GSH-Px in the liver were increased by Cd administration, but the alterations were decreased by supplementation with mungbean sprouts juice. The level of glutathione decreased in CAD group(26.8$\pm$9.0mg/g liver), whereas it increased in MCD group(36.4$\pm$15.8mg gliver). The content of Cd in the liver and kidney in MCD group(9.57 ppm, 4.88 ppm) was decreased, compared with CAD group(12.81 ppm, 5.46 ppm). This result suggested that mungbean sprout juice has a lowering effect on the accumulation of Cd in the liver and kidney and it is believed that the juice has some protective effects to Cd-induced hepatotoxicity in rats, but the mechanism of these effects was obscure.

  • Concanavalin A (ConA) interacts with carbohydrates as a lectin, and recent reports proposed its application for detecting a diversity of viruses and pathogens. Structural studies have detailed the interaction between ConA and carbohydrates and the metal coordination environment with manganese and calcium ions (Mn-Ca-ConA). In this study, ConA was crystallized with a cadmium-containing precipitant, and the refined structure indicates that $Mn^{2+}$ was replaced by $Cd^{2+}$ (Cd-Ca-ConA). The structural comparison with ConA demonstrates that the metal-coordinated residues of Cd-Ca-ConA, that is Glu8, Asp10, Asn14, Asp19, and His24, do not have conformational shifts, but residues for sugar binding, including Arg228, Tyr100, and Leu99, reorient their side chains, slightly. Previous studies demonstrated that excess cadmium ions can coordinate with other residues, including Glu87 and Glu183, which were not coordinated with $Cd^{2+}$ in this study. The trimeric ConA in this study coordinated $Cd^{2+}$ with other residues, including Asp80 and Asp82, for complex generation. The monomer does not have specific interaction near interface regions with the other monomer, but secondary cadmium coordinated with two aspartates (Asp80 and Asp82) from monomer 1 and one aspartate (Asp16) from monomer 2. This study demonstrated that complex generation was induced via coordination with secondary $Cd^{2+}$ and showed the application potential regarding the design of complex formation for specific interactions with target saccharides.

Analysis of the Transport System of Cadmium and the Change of Proline Content in Spring Radish Young Plant (봄무우 유식물에서 카드뮴 운반계와 Proline 농도 변화의 분석)

  • Cadmium was transported through the special carrier system into the cells of cotyledons, hypocotyls, and roots spring radish young plants. The transport of cadmium was inhibited by metabolic inhibitor, like DNP. The $K_m$ values for cadmium were 0.7 ppm for cotyledons, 1.72 ppm for hypocotyls, and 0.3 ppm for roots, and $V_{max}$ for cadmium was $40ppm{\cdot}h^{-1}{\cdot}g{\cdot}fresh\;weight^{-1}$ for cotyledons, $313ppm{\cdot}h^{-1}{\cdot}g{\cdot}fresh\;weight^{-1}$ for hypocotyls, and $606ppm{\cdot}h^{-1}{\cdot}g{\cdot}fresh\;weight^{-1}$ for roots. Cadmium cannot prove to be inducer for proline accumulation. Therefore, proline accumulation cannot be used as a marker to test the level of heavy metal pollution.

Effect of Dietary Iron Levels on the Cadmium Accumulation in Cadmium Poisoned Rats (카드뮴의 장기 중독시 철분의 섭취 수준이 흰쥐의 체내 카드뮴 축적에 미치는 효과)

  • This study was undertaken to investigate the effect of dietary Fe levels on Cd accumulation in Cd poisoned rats. Forty male weaning Sprague Dawley rats weighing 80-90g were divided into 4 groups(LFe : low Fe, LFeCd : low Fe and Cd, AFe : adequate Fe, AFeCd : adequate Fe and Cd) according to Cd administration(Cd : 0 or 50ppm in drinking water) and Fe levels (Fe : 6 or 40ppm in diet) for 12 weeks. The food intake and weight gain of LFe and LFeCd were significantly lower than those of AFe and AFeCd(p<0.01, p<0.001). The water intake was not affected significantly by Cd and Fe, therefore Cd intake was no significant difference between groups. The Cd accumulation of kidney in LFeCd was significantly higher than those of AFeCd(p<0.001). But the Cd accumulations of brain, liver and spleen were not significantly different between Cd groups and without Cd groups. The serum Cd content and urinary Cd excretion of LFeCd was significantly higher than those of AFeCd(p<0.01, p<0.01). But the fecal Cd excretion of LFeCd was significantly lower than that of AFeCd(p<0.001). The Cd retention, Cd retention rate, and apparent Cd digestibility of LFeCd were significantly higher than those of AFeCd(p<0.001, p<0.001, p<0.001). It was concluded that adequate Fe supplementation have protective effects on the long term Cd poisoning in rats.

Effect of Dietary CHitin, Chitosan and NOCC on Cadmium Toxicity and Lipid Metabolism in Rats (Chitin, CHitosan 및 수용성 Chitosan 유도체가 흰쥐의 Cadimium 중독과 지방대사에 미치는 영향)

  • This study was to investigate the effects of chitin, chitosan , NOCC and cellulose on cadmium toxicity and lipid metabolism in rats. Fifty male rats of Sprague-Dawley strain weighing 155$\pm$17g were divided into 10 groups according to body weight, and were raised for 5 weeks. Levels of 0 and 400 ppm of cadmium chloride were in the diets. And chitin, chitosan , NOCC and cellulose were given at the level of 0 and 4%(w/w) of the diets. Chitosan and NOCC decreased Cd concentration of liver and kidney, and increased the fecal excretion of Cd. Although cellulose was less effective, cellulose also decreased Cd concentration of liver and increased fecal Cd excretion . However chitin had little effect on alleviating Cd toxicity. In addition, chitosan and NOCC lowered total lipid, cholesterol and triglyceride contents in serum and liver by decreasing the lipid absorption. The lowering of cholesterol especially had a remarkable effect. Although it was less effective than chitosan and NOCC, chitin decreased the lipid absorption ratio and lowered cholesterol contents in serum and liver. In conclusion , chitosan and NOCC showed effects of decreasing the absorption of Cd and lipid.
