Comparative Analysis on Concentration and Uptake Amount of Mineral Nutrients in Different Growth Stages and Temperatures of Panax ginseng C. A. Meyer Grown with Hydroponic Culture (수경재배 인삼의 온도와 생육시기별 무기영양성분 함량과 흡수량 변이 양상 비교)

  • The management and control of mineral nutrients is one of most important techniques to increase the productivity and the quality of Korean ginseng. The mineral nutrients are measured with different plant tissues and different growth stages of 2-year-old ginseng grown under hydroponic culture with two different temperatures. The content of N, P, Ca, and Mg were higher at low temperature in both leaves and roots than those at high temperature. However, the content of K was high in leaves at low temperature compared to that of high temperature, while it was not significantly different in roots. The uptake amounts of N and K was higher throughout the experimental period at low temperature in both leaves and roots than those at high temperature. However, the uptake amount of P was not clearly different between two different temperatures and among six different growth stages. The uptake amount of N, P, K was generally decreased in leaves from June to August, while it was increased in roots. The relationship between dry weight and mineral nutrients in leaves was appeared positive with N, K, Ca, and Mg, but negative P. In roots, N, K, Ca, and Mg were negative, showing that was positive with only P. Comparing the correlation coefficients among mineral nutrients in leaves, N and K were significantly positive correlation each other. P was significantly positive correlation with Na and Zn. In case of roots, N was highly significant positive correlation with K, Mg, and Mn, but P was negatively correlated with Ca, Cu, Na, Fe, and Zn.

Effects of L-trans-pyrrolidine-2,4-dicarboxylate, a Glutamate Uptake Inhibitor, on NMDA Receptor-mediated Calcium Influx and Extracellular Glutamate Accumulation in Cultured Cerebellar Granule Neurons

  • Glutamate uptake inhibitor, L-trans-pyrrolidine-2, 4-dicarboxylate (PDC, $20{\mu}M$) elevated basal and N-methyl-D-aspartate (NMDA, $100{\mu}M$)-induced extracellular glutamate accumulation, while it did not augment kainate $100{\mu}M$-induced glutamate accumulation in cultured cerebellar granule neurons. However, pretreatment with PDC for 1 h significantly reduced NMDA-induced glutamate accumulation, but did not affect kainate-induced response. Pretreatment with glutamate $(5{\mu}M)$ for 1 h also reduced NMDA-induced glutamate accumulation, but did not kainate-induced response. Upon a brief application (3-10 min), PDC did neither induce elevation of intracellular calcium concentration $([Ca^{2+}]_i)$ nor modulate NMDA-indLiced $[Ca^{2+}]_1$ elevation. Pretreatment with PDC for 1 h reduced NMDA-induced $[Ca^{2+}]_1$ elevation, but it did not reduce kainate-induced $[Ca^{2+}]_1$ elevation. These results suggest that glutamate concentration in synaptic clefts of neurana cells is increased by prolonged exposure (1 h) of the cells to PDC, and the accumulated glutamate subsequently induces selective desensitization of NMDA receptor.

Changes in the Expressional Levels of Sarcoplasmic Reticulum $Ca^{2+}-regulatory$ Proteins in the Postnatal Developing Rat Heart

  • In the present study, the postnatal developmental changes in the expressional levels of cardiac sarcoplasmic reticulum (SR) $Ca^{2+}$ regulatory proteins, i.e. $Ca^{2+}-ATPase,$ phospholamban, and $Ca^{2+}$ release channel, were investigated. Both SR $Ca^{2+}-ATPase$ and phospholamban mRNA levels were about 35% of adult levels at birth and gradually increased to adult levels. Protein levels of both SR $Ca^{2+}-ATPase$ and phospholamban, which were measured by quantitative immunoblotting, were closely correlated with the mRNA levels. The initial rates of $Ca^{2+}$ uptake at birth were about 40% of adult rates and also increased gradually during the myocardial development. Consequently, the relative phospholamban/$Ca^{2+}-ATPase$ ratio was 1 in developmental hearts. $Ca^{2+}$ release channel (ryanodine receptor) mRNA was about $50{\sim}60%$ at birth and increased gradually to adult level throughout the postnatal rat heart development. $^3[H]ryanodine$ binding increased gradually during postnatal myocardial development, which was closely correlated with ryanodine mRNA expression levels during the development except the ryanodine mRNA level at birth. These findings indicate that cardiac SR $Ca^{2+}-ATPase,$ phospholamban, and $Ca^{2+}$ release channel are expressed coordinately, which may be necessary for intracellular $Ca^{2+}$ regulation during the rat heart development.

  • Cryptosporidium andersoni ATP-binding cassette (CaABC) is an important membrane protein involved in substrate transport across the membrane. In this research, the nucleotide binding domain (NBD) of CaABC gene was amplified by PCR, and the eukaryotic expression vector of pEGFP-C1-CaNBD was reconstructed. Then, the recombinant plasmid of pEGFP-C1-CaNBD was transformed into the mouse intestinal epithelial cells (IECs) to study the iron transportation function of CaABC. The results indicated that NBD region of CaABC gene can significantly elevate the transport efficiency of $Ca^{2+}$, $Mg^{2+}$, $K^+$, and $HCO_3{^-}$ in IECs (P<0.05). The significance of this study is to find the ATPase inhibitors for NBD region of CaABC gene and to inhibit ATP binding and nutrient transport of CaABC transporter. Thus, C. andersoni will be killed by inhibition of nutrient uptake. This will open up a new way for treatment of cryptosporidiosis.

N-acetyl-L-cysteine and cysteine increase intracellular calcium concentration in human neutrophils

  • N-acetyl-L-cysteine (NAC) and cysteine have been implicated in a number of human neutrophils' functional responses. However, though $Ca^{2+}$ signaling is one of the key signalings contributing to the functional responses of human neutrophils, effects of NAC and cysteine on intracellular calcium concentration ($[Ca^{2+}]_i$) in human neutrophils have not been investigated yet. Thus, this study was carried out with an objective to investigate the effects of NAC and cysteine on $[Ca^{2+}]_i$ in human neutrophils. We observed that NAC ($1{\mu}M{\sim}1mM$) and cysteine ($10{\mu}M{\sim}1mM$) increased $[Ca^{2+}]_i$ in human neutrophils in a concentration-dependent manner. In NAC pre-supplmented buffer, an additive effect on N-formyl-methionine-leucine-phenylalanine (fMLP)-induced increase in $[Ca^{2+}]_i$ in human neutrophils was observed. In $Ca^{2+}$-free buffer, NAC- and cysteine-induced $[Ca^{2+}]_i$ increase in human neutrophils completely disappeared, suggesting that NAC- and cysteine-mediated increase in $[Ca^{2+}]_i$ in human neutrophils occur through $Ca^{2+}$ influx. NAC- and cysteine-induced $[Ca^{2+}]_i$ increase was effectively inhibited by calcium channel inhibitors SKF96365 ($10{\mu}m$) and ruthenium red ($20{\mu}m$). In $Na^+$-free HEPES, both NAC and cysteine induced a marked increase in $[Ca^{2+}]_i$ in human neutrophils, arguing against the possibility that $Na^+$-dependent intracellular uptake of NAC and cysteine is necessary for their $[Ca^{2+}]_i$ increasing activity. Our results show that NAC and cysteine induce $[Ca^{2+}]_i$ increase through $Ca^{2+}$ influx in human neutrophils via SKF96365- and ruthenium red-dependent way.

Combined Effects of Container Volume and Fertilizer Level on Plant Growth, Physiological Characteristics, and Nutrient Uptake of Vinca Plant (Catharanthus roseus)

  • The aim of our study was to investigate the interactive effects of container size and nutrient supply on plant growth, chlorophyll synthesis, transpiration, $CO_2$ assimilation, water use efficiency (WUE), and nutrient uptake of vinca plant (Catharanthus roseus). A complete experiment utilizing four concentrations of fertilizer and three volumes of containers was conducted. As the container size was increased, the plant height, leaf area, and dry weight of vinca significantly increased regardless of fertilizer level. The leaf area and dry weight of vinca were highly sensitive to the container size. However, the chlorophyll contents of vinca 20 days after the transplant significantly increased with decreasing container sizes and increasing fertilizer concentrations. Significant differences in transpiration and $CO_2$ assimilation occurred with the use of differentfertilizer solutions, but the highest values for transpiration and $CO_2$ assimilation were in plants grown in the 15 cm-diameter containers. The highest water use efficiency was observed in the plants grown in 10 cm-containers with 4 dS/m of fertilizer, and there were no significant differences in WUE values among container sizes with fertilizer concentrations of 0, 1, or 2 dS/m. No significant difference in nutrient uptake was observed among the fertilizer levels or among the container sizes. However, at a fertilizer concentration of 4 dS/m, the uptake of several nutrients, including N, P, K, Ca, Mg, B and Fe, was higher in small containers than in larger ones.

Studies on the Marginal leaf chlorosis of Ginseng Plant (1) The Effect of Excess Manganese uptake on the occurrence of marginal leaf chlorosis (인삼의 엽록형 황증에 관한 연구 제1보 Mn의 과잉흡수가 엽록형 황증발생에 미치는 영향)

  • This study was conducted to determine the cause of the occurence of marginal leaf chlorosis in ginseng plants (Panax ginseng C.A. Meyer), and to determine its emersion in fields (practically) and in pots (experimentally). The following results were obtained. In the Present investigation, ginseng plants raised in acidic soil containing a high a moue t of Mn showed marginal leaf chlorosis. Henre it Ivas suggested that the shoot growth and root weights became grad gractually lower. The leaves having marginal leaf chlorosis contained low amounts of N, P,. Ca, Mg, and Na and the Fe/Mn ratios were low. There was a corresponding increase in Mn uptake. It was founrl that in soils where marginal leaf chlorisis occured the pH urar brlolv 4.2 to 4.9 and the Ca, Mg and Na content was decreased thus effectively increasing the available manganese in the soil. The Mn/Fe ratios in the yellow leaf margins of ginseng Plants affected by the Mn toxicity was over 2.0 compared to the general Mn/Fe ratio of 0.50 for healthily leaves, stems and roots. Typically when ginseng plants grow fields having soil with a pH below about 5.0, there tenor to be an uptake of excess Mn. When ginseng plants are grown in a nutrient sand culture solution It with an increased Mn concentration, they accumulate large amounts of Mn in the roots and in the shoots. In both casts marginal leaf chlorosis appeared in the emersions. In the Present investigation, ginseng plants raised in acidic soil and containing a high amount of Mn showed marginal leaf chlorosis.

Characterization of Cation Exchange and Cesium Selectivity of Synthetic Beta-Dicalcium Silicate Hydrate

  • Solid beta-dicalcium silicate hydrate $(\beta-C_2SH)$ synthesized under hydrothermal conditions at $240^{\circ}C$ and Ca/Si=2 molar ratio shows cation exchange properties towards divalent metal cations such as Fe, Cu, Zn, Cd, or Pb. The ability of metal cation uptake by the solid was found to be in the order: $Fe^{2+}$$Cu^{2+}$$Zn^{2+}$$Cd^{2+}$ = $Pb^{2+}$. Cesium selectivity of the solid was demonstrated in the presence of univalent cation such as $Li^+$, $Na^+$ and $K^+$ and divalent cations such as $Ca^{2+}$, $Mg^{2+}$ and $Ba^{2+}$, which are one hundred times more concentrated than the $Cs^+$. The uptake of $Cs^+$ is maximum in the presence of $Mg^{2+}$ whereas it is minimum in the presence of $K^+$. The different affinities of ${\beta}-C_2SH$ towards divalent metal cations can be used for the separation of those ions. Due to its selectivity for cesium it can be used in partitioning of radioactive Cs+ from nuclear wastes containing numerous cations. The mechanism of the metal cation exchange and cesium selectivity reactions by the solid is studied.

Thyroid Hormone-Induced Alterations of $Ca^{2+}-ATPase$ and Phospholamban Protein Expression in Cardiac Sarcoplasmic Reticulum

  • Alterations of cardiovascular function associated with various thyroid states have been studied. In hyperthyroidism left ventricular contractility and relaxation velocity were increased, whereas these parameters were decreased in hypothyroidism. The mechanisms for these changes have been suggested to include alterations in the expression and/or activity levels of various proteins; ${\alpha}-myosin$ heavy chain, ${\beta}-myosin$ heavy chain, ${\beta}-receptors,$ the guanine nucleotide-binding regulatory protein, and the sarcolemmal $Ca^{2+}-ATPase.$ All these cellular alterations may be associated with changes in the intracellular $Ca^{2+}$ concentration. The most important regulator of intracellular $Ca^{2+}$ concentration is the sarcoplasmic reticulum (SR), which serves as a $Ca^{2+}$ sink during relaxation and as a $Ca^{2+}$ source during contraction. The $Ca^{2+}-ATPase$ and phospholamban are the most important proteins in the SR membrane for muscle relaxation. The dephosphorylated phospholamban inhibits the SR $Ca^{2+}-ATPase$ through a direct interaction, and phosphorylation of phospholamban relieves the inhibition. In the present study, quantitative changes of $Ca^{2+}-ATPase$ and phospholamban expression and the functional consequences of these changes in various thyroid states were investigated. The effects of thyroid hormones on (1) SR $Ca^{2+}$ uptake, (2) phosphorylation levels of phospholamban, (3) SR $Ca^{2+}-ATPase$ and phospholamban protein levels, (4) phospholamban mRNA levels were examined. Our findings indicate that hyperthyroidism is associated with increases in $Ca^{2+}-ATPase$ and decreases in phospholamban levels whereas opposite changes in these proteins occur in hypothyroidism.

Phorbol Ester-Induced Periodic Contraction in Isolated Rabbit Jugular Vein

  • The present study was conducted to evaluate the effect of phorbol 12,13-dibutyrate (PDBu) on the contraction of rabbit jugular vein in vitro. PDBu concentrations of greater than 10 nM induced a periodic contraction which was composed of rapid contraction, plateau and slow relaxation. The frequency of periodic contraction increased as PDBu concentration increased. The PDBu-induced contraction was inhibited by staurosporine (100 nM), it was not changed by tetrodotoxin $(1\;{\mu}M).$ In $Ca^{2+}$-free medium, PDBu induced a sustaining contraction, but not periodic contraction. Addition of $Ca^{2+}$ to medium evoked periodic contraction which was inhibited by nifedipine, PDBu concentrations of greater than $0.1\;{\mu}M$ increased ^{45}Ca^{2+}$ uptake without changing $^{45}Ca^{2+}$ efflux. Charybdotoxin and apamin, $Ca^{2+}$-activated K^{+}$ channel blockers, did not affect the PDBu-induced periodic contraction, whereas tetraethylammonium (TEA) abolished the periodicity. Pinacidil $(10\;{\mu}M).$, a potassium channel activator, blocked PDBu induced periodic contraction, which was recovered by glybenclamide $(10\;{\mu}M).$. In high potassium solution, PDBu did not produce the periodic contraction. These results suggest that the PDBu-induced periodicity of contraction is modulated by voltage dependent $Ca^{2+}$ channel and ATP-sensitive $K^{+}$ channel.
