Identification and Quantification of Glucosinolates in Rapeseed (Brassica napus L.) Sprouts Cultivated under Dark and Light Conditions

  • BACKGROUND: This study was performed for the identification and quantification of glucosinolate (GSL) contents in seven varieties of rapeseed (Brassica napus L.) sprouts cultivated under dark and light conditions. METHODS AND RESULTS: Crude glucosinolates (GSLs) were desulfated by treating with aryl sulfatase and purified using diethylaminoethyl sepharose (DEAE) anion exchange column. Individual GSLs were quantified using high-performance liquid chromatography (HPLC) with electrospray ionization-tandem mass spectrometry (ESI-MS/MS). Eleven GSLs including six aliphatic (progoitrin, sinigrin, glucoalyssin, gluconapoleiferin, gluconapin, and glucobrassicanapin), four indolyl (4-hydroxyglucobrassicin, glucobrassicin, 4-methoxyglucobrassicin, and neoglucobrassicin) and one aromatic (gluconasturtiin) were identified based on the fragmentation patterns of MS spectrum. Aliphatic GSLs were noted as the predominant group with average 85.2% of the total contents. The most abundant GSLs were progoitrin which was ranged at $8.14-118.68{\mu}mol/g$ dry weight (DW). The highest total GSL amounts were documented in 'Hanra' ($146.02{\mu}mol/g$ DW) under light condition and 'Mokpo No. 68' ($86.67{\mu}mol/g$ DW) in dark condition, whereas the lowest was in 'Tamra' (30.13 and $14.50{\mu}mol/g$ DW) in both conditions. The sum of aliphatic GSLs attributed > 80% in all varieties, except 'Tamra' (67.7% and 64.9% in dark and light conditions, respectively) in the total GSL accumulation. Indolyl GSLs were ranged $2.41-15.73{\mu}mol/g$ DW, accounted 2.78-33.6% of the total GSLs in rapeseed varieties. CONCLUSION(S): These results provide valuable information regarding potential beneficial GSL contents individually. This study attempts to contribute to knowledge of the nutritional properties of the different varieties of rapeseed plants. These results may be useful for the evaluation of dietary information.

Evaluation of Commercial Varieties of Carrot in Jeju Island (제주 지방에서 당근 재배품종의 특성평가)

  • The overall objective of this study was to determine which variety of carrot-10 Japan varieties or 6 domestic varieties- produces the optimum quality, when cultivated on Jeju Island. Plant height, leaf number and leaf weight were observed to be more superior in the purebred variety ‘Jungang’than other varieties. further studies, however, skewed the crossing variety ‘Bureoun’was superior to all others. Furthermore, when comparing root diameter, root length and root weight, the ‘Bibary’was superior in both the purebred and crossing variety, When researching Japan varieties, the crossing variety, ‘Jung’showed an increase in underground root. The purebred variety ‘Donghae’also showed an increase in underground root. Marketable yields in domestic varieties were greatest in ‘Hapa’, while in Japan varieties, ‘Kaneko’was the most productive. The percentage of root cracked was 15% greater in ‘Bureon’. In the purebred variety, Jungang, the percentage was only 10%. The percentage of roots branched was highest in ‘Hidomi’which showed a rate of 9%, The roots branched rate was 2% for ‘Jung’and 6% for‘Donghae’. Considering the percentage of roots clacked or branched, the marketability of carrots was higher in Japan varieties in comparison with domestic varieties. Therefore we conclude that, in the cropping patterns of carrots, the domestic variety has an advantage over the Japan variety.

  • Rice is the world's three major crops that is the staple food of Asian. Japonica type rice is consumed in Korea. Rice cultivation area decreases steadily. That is affected by decrease of the farm income with the inventory of 1.7 million tons, which is 2 to 3 times more than the optimal amount due to the decrease in the consumption of rice per person and cultivate high yield varieties. In recent years, as income level has improved, interest in health has been high and consumption for health food has been steadily increasing. For health food, rice is added by adding grains such as millet, sorghum, oats, beans and colored rice rather than white rice. In 1997, RDA(Rural Development Administration) developed black rice 'Heuknambyeo', and then 20 varieties were bred until 2017. In CBARES(Chungbuk Agricultural Research and Extension Services), we have developed new rice varieties 'Cheongpungheukchal' in 2010, 'Cheongpungheukhyangchal' in 2014, 'Cheongpungheukchal' is high in farming preference because of high yield. Black rice has high content of GABA and water-soluble phenol, DPPH radical scavenging activity, and consumers are interested in the function in the body. Therefore, functional and antioxidant activities(anthocyanin, total polyphenol, ${\alpha}-glucosidase$ inhibition) and antioxidant activities(ABTs, DPPH) were analyzed by comparing white and black rice. Testing varieties are 'Chucheongbyeo', best quality cultivars 'Daebo' and 'Samgwang' as white rice which are cultivated much in Chungbuk area, and black rice are 10 varieties including 'Cheongpungheukchal', 'Cheongpungheukhyangchal', 'Josaengheukchal' and so on. It has transplanted on 25th May, at CBARES research paddy by 100% fertilizer recommendation rate. Harvesting time was 50 days after heading by varieties, and has researched growth properties, yield and yield components, functional and antioxidant activities. Anthocyanin content was not measured because there was no pigmented in white rice, the highest value of anthocyanin content was 'Shintoheukmi' and in the range of 125.6~249.6mg/100g by black rice varieties. Total polyphenol content was high 'Cheongpungheukchal' and 'Shintoheukmi' and in the range of 96.68~244.34 mg/100g in black rice, white rice lower than blackish rice at 19.84~22.51mg/100g. ${\alpha}-glucosidase$ inhibition was high 'Cheongpungheukchal', 'Cheongpungheukhyangchal' and 'Shintoheukmi' in the rage of 75.87~98.85% by black rice varieties, especially 'Samgwang' was 80.75% and the other white rice was higher than 58~68%.

Effect of Rice Black-Streaked Dwarf Virus(RBSDV) Infection Rate on Forage Productivity of Corn Variety in Middle District of Korea (중부지역에서 검은줄오갈병 발생이 사료용 옥수수 품종의 조사료 생산성에 미치는 영향)

  • This experiment was carried out to study the effect of rice black-streaked dwarf virus(RBSDV) infection rate on forage productivity of corn varieties in Cheonan of chungcheongnamdo from 2006 to 2008. Forage corn varieties of 10 were cultivated with first cropping(seeding in the last ten days of April) and second cropping(seeding in the last ten days of May) system in field and tested the infection rates of RBSDV and productivity of forage. The Infection rate of RBSDV was significant difference between corn varieties in middle district of Korea. Resistant corn varieties for RBSDV were 'Kwanganok', 'P3156', 'Kwangpeyongok' and 'P3394' but susceptible varieties were 'Suwon19', 'DK697', 'GW6959' and NC+7117. Dry matter(DM) yield of forage corn according with infection rates of RBSDV in field was significant difference between varieties(p<0.05). DM yield of susceptible varieties, 'Suwon 19', 'DK697' and 'GW6959' was lower about 20% than that of resistant varieties, 'Kwangpeyongok' and 'P3156'. For increasing the productivity of forage corn, recommend of resistant varieties for RBSDV and control of seeding time are very important in middle district of Korea.

  • Food productivity in North Korea is about 50% lower than in South Korea. In order to increase the productivity of major crops, it is necessary to develop early maturing, disease resistance, and high-yielding varieties and apply them early. Since the late 1990s, North Korea has been actively developing potatoes, rice and com as major food crops, and soybeans are considered important as a protein-supplying crop. Domestic cultivated varieties, which are expected to be most adaptable eco-climatologically, are mainly selected from soil with high nutrient soil. It is necessary to test separately for adaptability in low organic soil. So it is very necessary to apply technology to improve soil improvement through rotational crop selection in the middle and long-term. Therefore, this study was conducted to test the adaptability to low organic soils of domestic cultivars and to select varieties. In 2021 there are twenty two (22) varieties of soybeans were grown in low organic soil at the field of Chungbuk National University. This year twenty two (22) varieties of soybeans were also grown in low organic soil at the field of Chungbuk National University. Sowing was done on June 10, the planting distance was 70cm × 15cm, after opening the cotyledons fully, the soybeans were thinned and leaving two plants per hole. In addition, various types of growth characteristics and quantitative components were investigated to evaluate the adaptability to low organic soil of domestic varieties. This study was conducted to investigate the growth characteristics and quantitative components of soybean varieties grown in low organic soil. The flowering period of 22 varieties of soybeans was about 14 days from July 22 to August 4. The flowers of the beans were white, purple, light purple and the pubescence color was gray and brown where most of them were gray. The highest plant height was up to 130.4 cm and lowest was 20.3 cm, highest stem length was up to 119.5 cm and lowest was 15.3 cm. Highest first pod height (FPH) was up to 34.0 cm and lowest was 3.0 cm. Highest stem diameter was up to 15.76mm and lowest was 1.76 mm. Number of main stem nodes was up to 19 and at least 1. Number of branch was up to 10 and at least 0. The number of pod per plant was up to 121. Bacterial pustule has been spread in soybean field.

Varietal Responses of Rice Growth and Yield to Soil Salt Content (벼 생태형별 염농도에 따른 생육 및 수량)

  • In other to find out the basic information on cultivation and breeding for salt tolerance in rice, these studies were conducted at salinity paddy fields containing 0.2, 0.3, 0.4% of salt and normal paddy field using 6 Japonica type, 5 Tonsil type varieties and Annapurna. On these fields a series of investigation was made for the responses of rice growth and yield to salt content of saline sails. The heading dates of the rice cultivated at salinity 0.2, 0.3 and 0.4% were delayed compared by 2 or 3,5 or 8 and 10 or 13 days respectively in Japonica and Tonsil type varieties to those at the normal paddy fields. The culm length at salinity paddy fields decreased by 13-39% in Japonica type and 14-35% in Tongil type varieties. But the number of panicles per hill decreased by 16-40% in Japonica type and by 14-35% in Tongil type varieties. The number of grains per panicle in saline paddy fields decreased by 10 to 40% in all varieties, but the percent ripened grain decreased by 20-48% in Japonica type and by 19-51% in Tongil type varieties. 1000 grains weight was decreased by lower than 23% in all varieties, but yield was reduced to 20-62% in Japonica and 25-67% in Tongil type varieties.

Comparison of Growth Characteristics and Yield of Soybean Varieties for Soiling Crop Improvement (풋베기용 대두 개량을 위한 품종의 생육특성 및 수량 비교)

  • This Study was carried out to compare growth characteristics and yield of 10 cultivated soybean varieties, Jinpum I, Jinpum II, Hwanggum, Gumjung I, Gumjung II, Seukryangboot, Jangyup, Gumjungol, Bokwang, Manri for improvement of soiling crop. Plant height of Gumjung I(118cm), Gumjung II(114cm) and Jinpum I(114cm) were taller than other varieties based on dough stage. In total fresh weight and DM yield of Jinpum I, Jinpum II, Gumjung II were the highest of all varieties as 23,841kg, 23,499kg and 22,815kg per hectare, and 5,531kg, 5,173kg and 5,236kg per hectare, respectively. The highest leaf/stem ratio of ten soybean varieties were Manri(2.1), Jinpum I(1.9) and Hwanggum(1.9). Except early matured varieties, Seukryangboot(25.9%) and Gumjungol(25.4%), dry matter percent at dough stage were in the range of 22.0~23.4%. According to plant height, fresh weight, dry matter yield, and leaf/stem ratio, Jinpum I and Gumjung II were proper varieties to improve soiling crop.

Comparison of Forage Productivity and Outbreak Rate Between Corn Varieties in Rice Black-Streaked Dwarf Virus (RBSDV) Prevalent Area (검은줄오갈병 발생이 심한 지역에서 옥수수 품종의 발병률과 사초 생산성 비교)

  • This experiment was carried out to select a resistant corn variety for rice black-streaked dwarf virus (RBSDV) disease in a RBSDV prevalent area (Gochang of Jeollabukdo) from 2006 to 2008. Ten corn varieties for silage preparation were cultivated with first cropping and second cropping system in an RBSDV field, and were tested outbreak rates of RBSDV and dry matter (DM) yield of forage. The outbreak rates of RBSDV were significantly different between corn varieties. Therefore, the resistance degree of corn varieties for RBSDV were divided into 4 groups; very resistant ('Kwangpeyongok' and 'Kwanganok'), resistant ('P3156' and 'P3394'), mildly resistant ('Cheonganok' and 'P32P75') and susceptible ('Suwon19', 'DK697' and 'GW6959') groups respectively. DM yield of forage in RBSDV fields exhibited comparatively significant differences between varieties (p<0.05). DM yield of resistant varieties ('Kwangpeyongok', 'Kwanganok', 'P3156' and 'P3394'), were higher (14~26%) than those of susceptible varieties ('Suwon 19', 'DK697' and 'GW6959'). Therefore, resistant varieties were recommended for increasing forage productivity in field of RBSDV prevalent areas.

Effects of Water-deficit Stress on Yield and Chlorophyll Fluorescence in Rice during the Early Tillering Stage (분얼기 수분 스트레스가 벼 생육 특성 및 엽록소 형광 반응에 미치는 영향)

  • This study aimed to evaluate the growth, yield, and grain quality characteristics of rice varieties that were subjected to water stress during the tillering stage. We also sought to identify whether water stress could be indicated via the plants' response to chlorophyll fluorescence. In this study, we induced water stress by intentionally cutting off water for 30 days, starting 15 days after transplanting the rice varieties to a greenhouse. We analyzed nine rice varieties, including Ilpum, which is the most frequently cultivated variety in Gyeongsangbuk-do, South Korea. The control group was planted in a paddy field where irrigation was entirely dependent on rainfall. Our results revealed that the heading stage of the nine studied varieties occurred approximately ten days earlier in the rain shelter than in the field. Moreover, the rice yield, head rice rate, and palatability score decreased by 18.6%, 17.1%, and 8.3%, respectively, while protein content increased by 20.2% compared with the control group. The Saenuri and Haimi varieties showed the lowest reduction in yield under the water stress conditions, while the Daebo and Samkwang varieties showed the highest reduction in yield. The chlorophyll fluorescence response after re-irrigation was measured between July 30th and August 17th. The ratio of variable fluorescence to maximum chlorophyll fluorescence (FV/FM) values failed to recover to their baseline values, resulting in either no change or a reduction in fluorescent response, even after re-irrigation of Daebo and Samkwang varieties. These results can be utilized as empirical data for drought-affected farms to select resistant varieties that can respond to spring drought in the southern plains of Gyeongsangbuk-do.

Vitamin B6 Content of Vegetables and Fruits Cultivated in Korea (국내 채소류와 과일류의 비타민 B6 함량)

  • The purpose of this study was to determine the difference in vitamin B6 content according to the varieties, regions, and harvest times of vegetables and fruits in Korea using high performance liquid chromatography. We verified the accuracy of the analytical method with standard reference material 1849a and achieved reliability for internal analysis quality control with a mixture of cereal, whole wheat, and flour. As a result of the analysis, vitamin B6 contents (㎍/100 g) were 6.9~86.5 in peppers, 5.1~17.2 in paprika, 4.4~5.0 in strawberries, 4.0~52.9 in tomatoes, 7.7~7.8 in Chinese cabbage, 17.3~23.3 in radishes, 13.4~37.6 in apples, 2.3~12.7 in peaches, and 3.7~12.7 in grapes. In general, the difference in vitamin B6 content showed by varieties, harvest times, and regions. Peppers showed the most difference as 79.6 ㎍/100 g among the varieties, and apples showed a difference as 22.8 ㎍/100 g by regions. According to the harvest times in grape 'Mihwang', there was a difference of 7.5 ㎍/100 g. We will collect an amount of nutritional data on various food materials, and continue to build a reliable and integrated nutritional database. And then the database will be used in the 10th revision of the Korean Food Composition Table.
