Mebendazole (MBZ), a microtubule depolymerizing drug commonly used for the treatment of helminthic infections, has been suggested as a repositioning candidate for the treatment of brain tumors. However, the efficacy of MBZ needs further study to improve the beneficial effect on the survival of those patients. In this study, we explored a novel strategy to improve MBZ efficacy using a drug combination. When glioblastoma cells were treated with MBZ, cell proliferation was dose-dependently inhibited with an IC50 of less than 1 µM. MBZ treatment also inhibited glioblastoma cell migration with an IC50 of less than 3 µM in the Boyden chamber migration assay. MBZ induced G2-M cell cycle arrest in U87 and U373 cells within 24 h. Then, at 72 h of treatment, it mainly caused cell death in U87 cells with an increased sub-G1 fraction, whereas polyploidy was seen in U373 cells. However, MBZ treatment did not affect ERK1/2 activation stimulated by growth factors. The marked induction of autophagy by MBZ was observed, without any increased expression of autophagy-related genes ATG5/7 and Beclin 1. Co-treatment with MBZ and the autophagy inhibitor chloroquine (CQ) markedly enhanced the anti-proliferative effects of MBZ in the cells. Triple combination treatment with temozolomide (TMZ) (another autophagy inducer) further enhanced the anti-proliferative effect of MBZ and CQ. The combination of MBZ and CQ also showed an enhanced effect in TMZ-resistant glioblastoma cells. Therefore, we suggest that the modulation of protective autophagy could be an efficient strategy for enhancing the anti-tumor efficacy of MBZ in glioblastoma cells.
In this paper, we deal with a method of efficiently processing continuous queries in a data stream environment. We classify previous query processing methods into two dual categories - data-initiative and query-initiative - depending on whether query processing is initiated by selecting a data element or a query. This classification stems from the fact that data and queries have been treated asymmetrically. For processing continuous queries, only data-initiative methods have traditionally been employed, and thus, the performance gain that could be obtained by query-initiative methods has been overlooked. To solve this problem, we focus on an observation that data and queries can be treated symmetrically. In this paper, we propose the duality model of data and queries and, based on this model, present a new viewpoint of transforming the continuous query processing problem to a multi-dimensional spatial join problem. We also present a continuous query processing algorithm based on spatial join, named Spatial Join CQ. Spatial Join CQ processes continuous queries by finding the pairs of overlapping regions from a set of data elements and a set of queries defined as regions in the multi-dimensional space. The algorithm achieves the effects of both of the two dual methods by using the spatial join, which is a symmetric operation. Experimental results show that the proposed algorithm outperforms earlier methods by up to 36 times for simple selection continuous queries and by up to 7 times for sliding window join continuous queries.

The equation of the step-drawdown test "$s_w=BQ+CQ^p$" written by Rorabaugh (1953) is suitable for drawdown increased non-linearly in the fractured rocks. It was found that value of root mean square error (RMSE) between observed and calculated drawdowns was very low. The calculated $C$ (well head loss coefficient) and $P$ (well head loss exponent) value of well head losses ($CQ^p$) ranged $3.689{\times}10^{-19}{\sim}5.825{\times}10^{-7}$ and 3.459~8.290, respectively. It appeared that the deeper depth in pumping well the larger drawdowns due to pumping rate increase. The well head loss in the fractured rocks, unlike that in porous media, is affected by properties of fractures (fractures of aperture, spacing, and connection) around pumping well. The $C$ and $P$ value in the well head loss is very important to interpret turbulence interval and properties of high or low permeability of fractured rock. As a result, regression analysis of $C$ and $P$ value in the well head losses identified the relationship of turbulence interval and hydraulic properties. The relationship between $C$ and $P$ value turned out very useful to interpret hydraulic properties of the fractured rocks.

We present a method of runtime configuration scheduling in reconfigurable SoC design. As a model of computation, we use a popular formal model of computation, hierarchical FSM (HFSM) with synchronous dataflow (SDF) model, in short, HFSM-SDF model. In reconfigurable SoC design with HFSM-SDF model, the problem of configuration scheduling becomes challenging due to the dynamic behavior of the system such as concurrent execution of state transitions (by AND relation), complex control flow (HFSM), and complex schedules of SDF actor firing. This makes it hard to hide configuration latency efficiently with compile-time static configuration scheduling. To resolve the problem, it is necessary to know the exact order of required configurations during runtime and to perform runtime configuration scheduling. To obtain the exact order of configurations, we exploit the inherent property of HFSM-SDF that the execution order of SDF actors can be determined before executing the state transition of top FSM. After obtaining the order information and storing it in the ready configuration queue (ready CQ), we execute the state transition. During the execution, whenever there is FPGA resource available, a new configuration is selected from the ready CQ and fetched by the runtime configuration scheduler. We applied the method to an MPEG4 decoder and IS95 design and obtained up to 21.8% improvement in system runtime with a negligible overhead of memory usage.
Multicellular tumor spheroids of HeLa cells have been grown in a static culture system. Samples of spheroids were exposed for 2 h to graded concentration of cis-platinum and its analogue, carboplatin, and then response assayed by survival of clonogenic cells. The purpose of present experiment is to clarify the effectiveness of these platinum compounds and to evaluate intrinsic radiosensitivity of cells using spheroids of HeLa cells as an experimental in vitro model. Variations of the drug sensitivity of monolayers as well as spheroids were also evaluated in cell-survival curves. In cis-platinum concentration-survival curve, there was a large shoulder extending as far as $Cq=3.4{\mu}M$, after which there was exponential decrease in survival curve having a Co Value of $1.2{\mu}M$ in spheroids. While the Co for the spheroids was essentially no significant change, but Cq value was larger than that of monolayers. This suggest that the effect of cis-platinum is greater En the monolayer with actively proliferaing cells than hypoxic one. In the carboplatin concentration-survival curves, the Co value of spheroids was $15.0{\mu}M$ and the ratio with the Co from monolayer cell $(32.5{\mu}M)$ was 0.40, thus indicating that the spheroids had a greater sensitivity to carboplatin than monolayers. Therefore, the effect of carboplatin is mainly on the deeper layers of spheroids acting as hypoxic cell sensitizer. The enhanced effect was obtained for monolayer cells using combined X-ray and carboplatin treatment 2 hours before irradiation. The result shown in isobologram analysis for the level of surviving fraction at 0.01 indicated that the effect of two agents was trusty supra-additive. From this experimental data, carboplatin has excited much recent interest as one of the most promising, since it is almost without nephrotoxicity and causes less gastrointestinal toxicity than cis-platinum. Interaction between carboplatin and radiation might play an important role for more effective local tumor control.

The phosphoinositide 3-kinase/protein kinase B/mammalian target of rapamycin (PI3K/AKT/mTOR) signaling pathway is a promising target for gastric cancer (GC) treatment; however the efficacy of PI3K/mTOR dual inhibitors in GC has not yet been maximized. Additionally, the effect of autophagy regulation by PI3K/mTOR dual inhibitors has not been clearly elucidated in GC treatment. We aimed to show that our newly developed PI3K/mTOR dual inhibitor, CMG002, when combined with an autophagy inhibitor, chloroquine (CQ), potently induces effective cancer cell death in Epstein-Barr virus (EBV)-associated gastric cancer (EBVaGC) cells, where both the PI3K/AKT/mTOR and autophagy pathways play important roles in disease pathogenesis. EBV- and mock-infected AGS and NUGC3 GC cell lines were treated with CMG002 +/- CQ. PI3K/AKT/mTOR signaling pathway mediators, cellular apoptosis and autophagy markers were confirmed by Western blot assay. Cell viability was assessed using the Cell Counting Kit-8 (CCK-8) assay. CMG002 effectively blocked the PI3K/AKT/mTOR pathway by markedly decreasing phosphorylation of AKT and its downstream mediator S6. CMG002 induced G0/G1 cell cycle arrest and enhanced apoptotic cell death in AGS and NUGC3 cells, particularly EBV-infected cells compared with mock-infected cells, as confirmed by flow cytometric analyses and TUNEL (terminal deoxynucleotidyl transferase dUTP nick end labeling) assays. The combination of CMG002 plus CQ synergistically increased apoptotic cell death in EBV-infected GC cell lines when compared with CMG002 alone (P < 0.05). Our results suggest that the new PI3K/mTOR dual inhibitor, CMG002, when used in combination with the autophagy inhibitor, CQ, provides enhanced therapeutic efficacy against EBVaGC.

Contact angle measuring device was developed in this laboratory using laser beam projec-tion. The new method allows for rapid and direct determination of stationary, advancing, and receding contact angles on both planar and nonplanar solid surfaces, including fibers with very small diameters. A narrow laser beam impinges on an edge of an interface of liquid and solid. This makes two projected laser beam lines upon and radiating from the center of a protractor scale on a tangent screen. Contact angle is measured by determining the difference in angle on the protractor scale between the two projected laser beam lines. Contact angles measured on Perspex-CQ using this instrument were in agreement with the literature. it was shown that this instrument provides a novel method for the facile and accurate measurement of contact angles.
In this paper we study the cardinality of the dot product set generated by two subsets of vector spaces over finite fields. We notice that the results on the dot product problems for one set can be simply extended to two sets. Let E and F be subsets of the d-dimensional vector space $\mathbb{F}^d_q$ over a finite field $\mathbb{F}_q$ with q elements. As a new result, we prove that if E and F are subsets of the paraboloid and ${\mid}E{\parallel}F{\mid}{\geq}Cq^d$ for some large C > 1, then ${\mid}{\Pi}(E,F){\mid}{\geq}cq$ for some 0 < c < 1. In particular, we find a connection between the size of the dot product set and the number of lines through both the origin and a nonzero point in the given set E. As an application of this observation, we obtain more sharpened results on the generalized dot product set problems. The discrete Fourier analysis and geometrical observation play a crucial role in proving our results.
A comparative phonetic analysis of Yodel singing and Belcanto singing by the electroglottographic(EGG) measurement was done in three singers. One professional tenor singer(SDI) who is also well trained in Yodel singing, another yodler(KWS) who is not so trained in Belcanto singing, and the other training tenor singer(CSK) who is not well trained both yodel and Belcanto singing. Closed quotient(CQ), speed quotient(SQ) and fundamental frequency (F0) at the initial modal part(I) , middle falsetto part(M), and final modal part(F) of the same phrase were measured by EGG machine and program(Kay model 4338). In the middle part, not only CQ but also SQ of the Yodel singing were much smaller than that of Belcanto singing in all three singers. However, accuracy of parameters in Belcanto singing of the yodler(KWS) and both Yodel singing and Belcanto singing of the training singer(CSK) were inferior to that of trained tenor singer(SDI). Possible advantages of utilizing Yodel singing training under the guidance of feedback control by the EGG for hyperfunctional voice disorders such as vocal nodules were discussed.
