YY1 and CP2c in Unidirectional Spermatogenesis and Stemness

  • Spermatogonial stem cells (SSCs) have stemness characteristics, including germ cell-specific imprints that allow them to form gametes. Spermatogenesis involves changes in gene expression such as a transition from expression of somatic to germ cell-specific genes, global repression of gene expression, meiotic sex chromosome inactivation, highly condensed packing of the nucleus with protamines, and morphogenesis. These step-by-step processes finally generate spermatozoa that are fertilization competent. Dynamic epigenetic modifications also confer totipotency to germ cells after fertilization. Primordial germ cells (PGCs) in embryos do not enter meiosis, remain in the proliferative stage, and are referred to as gonocytes, before entering quiescence. Gonocytes develop into SSCs at about 6 days after birth in rodents. Although chromatin structural modification by Polycomb is essential for gene silencing in mammals, and epigenetic changes are critical in spermatogenesis, a comprehensive understanding of transcriptional regulation is lacking. Recently, we evaluated the expression profiles of Yin Yang 1 (YY1) and CP2c in the gonads of E14.5 and 12-week-old mice. YY1 localizes at the nucleus and/or cytoplasm at specific stages of spermatogenesis, possibly by interaction with CP2c and YY1-interacting transcription factor. In the present article, we discuss the possible roles of YY1 and CP2c in spermatogenesis and stemness based on our results and a review of the relevant literature.

  • Reaction of [$Cp^*Rh(NO_3)OTf$] (1) with 2,6-diisopropylaniline under argon led to the formation of $[Cp^*Rh({\eta}^6-2,6-diisopropylaniline)](OTF)_2$, (2). Compound 2 was characterized by NMR, IR, elemental analysis, and X-ray diffraction. Crystallrographc data for 2: orthorhombic, space group $Cmc2_1$, $a=19.850(16){\AA}$, $b=9.870(8){\AA}$, $c=15.774(13){\AA}$, Z = 4, $R(wR_2)=0.0362(0.0938)$.

Effect of Different GC Columns on the Quantitative Analysis of Long Chain Alkyl Diols (LCDs) (특성이 다른 GC 컬럼이 long chain alkyl diols (LCDs)의 정량 분석에 미치는 영향)

  • Long chain alkyl diols (LCDs) have been reported in sediments from various marine environments. Rampen et al. (2012) introduced the paleo-sea surface temperature (SST) proxy, Long chain Diol Index (LDI) based on the relative abundance of $C_{30}$ 1,15-diol, $C_{28}$ 1,13-diol, and $C_{30}$ 1,13-diol. In general, CP-Sil5CB and DB-5ms columns have been used for the quantitative and qualitative analysis of LCDs with a GC-MS. In this study, we examined the effect of three different columns (CP-Sil5CB, HP-5ms and DB-5) on the quantitative analysis of LCDs using marine sediments from the East Sea of Korea and the western Arctic Ocean. In general, our study showed that the results of CP-Sil5CB differed significantly from those of HP-5ms and DB-5. However, the differences of the LDI-derived SSTs among three columns were $0.1-0.2^{\circ}C$ for the East Sea and $0.2-0.7^{\circ}C$ for the western Arctic Ocean, which were well within the calibration error range (${\pm}1{\sigma}$). Accordingly, our study showed that the use of different columns resulted in significant differences of LCDs concentrations, but its effect on the LDI was relatively insignificant. Therefore, it appears that the different columns can be used for the paleo-SST reconstruction in the East Sea and the western Arctic Ocean using the LDI proxy.

Cold Plasma Treatment Application to Improve Microbiological Safety of Infant Milk Powder and Onion Powder (콜드 플라즈마 처리를 이용한 분유와 양파분말 살균)

  • The potential of applying cold plasma (CP) treatments to improve microbiological safety of powdered products has been investigated using infant milk powder (IMP) and onion powder (OP). Among the different kinds of CP-forming gases, He-$O_2$ (99.8:0.2) gas mixture and He gas were most effective in reducing the number of Cronobacter sakazakii in IMP and Bacillus cereus spores in OP, respectively. C. sakazakii counts in IMP decreased by $0.9{\pm}0.1{\log}\;CFU/g$ after CP treatment, and the extent of C. sakazakii inhibition increased in a time-dependent manner. CP treatment at 900 W for 20 min reduced the number of B. cereus spores by ~0.4 log spores/g. Treatments that integrated CP with microwave (MW-CP treatment) as well as those that integrated CP with heat and microwave (H-MW-CP treatment) resulted in a 90% reduction in the number of spores in OP. Thus, CP treatments demonstrated potential for decontaminating foodborne pathogens from powdered products, in combination with heat for improved effect.

Effect of Cassoy-urea Pellet as a Protein Source in Concentrate on Ruminal Fementation and Digestibility in Cattle

  • Four male crossbred native beef cattle (average body weight of 427.7 kg) were randomly allocated to four types of cassoy-urea pellet as a source of protein in concentrate according to a $4{\times}4$ Latin square design to determine effect of diets on ruminal fermentation and nutrient digestibility. The four types of cassoy-urea pellets contained cassava hay, soybean meal, urea and binding agent at 79.2:19.8:0:1 (27.9% CP dry matter), 78.4:19.6:1:1 (30.4% CP), 77.6:19.4:2:1 (33.0% CP) and 99:0:0:1 (23.8% CP) for dietary treatments; 1, 2, 3 and 4, respectively. All four concentrate mixtures contained similar crude protein levels (11% CP) and were fed to animals in two equal parts (0.5% of body weight per day) while urea-treated rice straw (5% urea) was given ad libitum. The experiment revealed that dietary concentrate treatments had no effect on dry matter intake while digestibilities of neutral-detergent fiber and crude protein were higher (p<0.05) in cattle fed dietary treatments 1, 2 and 3 than in cattle fed dietary treatment 4. Ruminal ammonia-nitrogen ($NH_3$-N), was higher and acetic acid concentration (C2) and ratio of C2 to propionic acid (C3) were lower (p<0.05) in cattle fed dietary treatments 1, 2 and 3 than in those on treatment 4. It is concluded that use of cassoy-urea pellet as a protein source in concentrates for cattle resulted in improvement of digestibility, ruminal fermentation and rumen ecology. Further research using cassoy-urea pellet in feeding trials with milking cows and fattening beef should be undertaken.

Physicochemical Properties of a Low Calorie Sweetener, Tagatose (저열량 감미료 Tagatose의 이화학적 특성)

  • The physicochemical properties of tagatose, a low calorie sweetener, was investigated. Rheological property of tagatose solution was found to be Bingham fluid. As the concentration of tagatose increased from 10 to 50% at $25^{\circ}C$, the viscosity increased from 1.65 to 5.14 cp. When the temperature of 40% tagatose solution increased from 15 to $55^{\circ}C$, the viscosity decreased from 4.59 to 2.33cp. The melting onset temperature and endothermic enthalpy of tagatose were $130.4^{\circ}C$ and -202.3 J/g, respectively, which were obtained from the analysis of differential scanning calorimetry. Tagatose showed higher water absorption than sucrose under $85{\sim}100%$ of relative humidity. Tagatose was less soluble than sucrose at $20{\sim}70^{\circ}C$. Water activity of tagatose in 60% concentration was 0.892, which was lower than 0.957 of sucrose solution. Tagatose solution adjusted from pH 2 to pH 12 was stable after 3 days. Amount of tagatose was not changed after heat treatment at $154^{\circ}C$ for 4 hours. But a browning reaction was found and absorbance of a tagatose solution increased with heat treatment.

Transmission of Multiple Drag-Resistance in Hospital Pseudomonas aeruginosa by Intraspecies Conjugation System (병원녹농균(病院綠膿菌)의 동종균접합(同種菌接合)에 의한 다제내성(多劑耐性)의 전달(傳達))

  • Two hundred and ninety-five strains of Peudomonas aeruginosa isolated from clinical sources were tested for drug resistance and demonstration of R plasmids by intraspecies conjugation system. Sixty strains were found highly resistant to two or more of drugs. The rate of resistant strains were 38.9% to kanamycin(km), 33.2% to streptomydn(sm), 22.7% to sulfisomidine(Sa), 14.2% to chloramphenicol(Cp), 13.8% to tetracycline(Tc), 3.0% to carbenicillin(Cb), and to gentamicin(Gm), respectively. But no strains was resistant to nalidixic acid and colistine. They were resistant to per milliliter to more than $400{\mu}g$ per ml. of Tc, $800{\mu}g$ per ml of Cp and of Sm, $6,400{\mu}g$ per ml. of Sa, $200{\mu}g$ per ml. of Cb, $100{\mu}g$ per ml. of Gm, and $25{\mu}g$ per ml. of colistine. Forty-three strains of Pseudomonas aeruginosa could be transferred their resistance to Pseudomonas aeruginosa 2-70, 1005 rifampin resistant FP-auxotrophic mutant. Of sixty multiple resistant strains, forty-three(71.6%) demonstrated R plasmids; nineteen carried resistance to(Tc Cp Sm Sa), six to(Tc Cp Sm), three to(Tc Cp Sa), and Cp, five to(Tc Sm Sa), two to(Tc Sa), (Cp Sm) and Tc, and one to(Cp Sm Sa). Degree of resistance of recipients recieving R plasmids from donors were almost the same level of resistance as the donor in regardless of mating temperature at $25^{\circ}C$ and $37^{\circ}C$. Resistance to Tc, Sm, and Sa were transferred to a very few of recipient cells at five minutes after mating with donor and recipient cells but resistance to Cp were transferred to the majority of recipient cells. The transfer frequency of Tc, Cp, Sm, and Sa resistance from donors to recipients were from $1.0^{-1.4}\;to\;1.0^{-3.5}$ at $25^{\circ}C$ for 18 hours of incubation and were from $1.0^{-1.5}\;to\;1.0^{-3.5}$ at $37^{\circ}C$ for 18 hours of incubation.

Composite Structures of $SiC_p$/6063 Aluminum Alloy by Rheo-Compocasting. (Rheo-Compocasting에 의한 $SiC_p$/6063 Al합금의 복합조직)

  • Aluminum alloy matrix composites reinforced by SiC particles were prepared by rheocompocasting, a process which consists of the incoporation and distribution of reinforcement by stirring within a semi-solid alloy. When the volume fraction of SiCp and stirring speed were fixed, the dispersion of SiCp in Al-matrix alloy depended on stirring time and solid volume fraction in slurry. The results were as follows : 1) As a dispersed SiCp during stirring at $647^{\circ}C$ in 6063-Al alloy, SiC was better dispersed than that other temperature, where solid volume fraction was 43% in slurry. 2) When increased solid fraction in slurry, rate of dispersing SiC increased during stirring and porosities decreased in matrix alloy after casting. 3) Inspite of stirring with 800rpm, since solid particles of matrix alloy in slurry joined each other and occured joining growth, so that SiC was not dispersed into solid particle.

Protective Effects of Cardiotonic Pills(CP) on Neuroglia Cells Against Oxidative Stress and the Effects on Regional Cerebral Blood Flow in Normal Rats (심적환(心適丸)이 산화적 손상에 따른 신경교세포 보호효과 및 국소 뇌혈류량 변화에 미치는 영향)

  • Objectives: SimJeok-Hwan(CP, Cardiotonic Pills) was made to treat patients with coronary arteriosclerosis, angina pectoris and hyperlipidemia. This study was designed to investigate the effects of CP on Proliferation rates neuroglia cells and protective effect of CP against oxidative stress, and also investigate the effects on regional Cerebral Blood Flow(rCBF) in normal rats. Methods: In this experiment, effects of CP on proliferation rates of neuroglia cells were measured using modified MTT methods. Oxidative stress was induced by treatment with 200 mM of hydrogen peroxide for 2 hr. rCBF and MABP were measured using Laser doppler flowmeter. Results: Treatment with CP elevated proliferation rates in C6 cells. In addition, CP protected cell death of C6 cells induced by oxidative stress. In results, rCBF was elevated by treatment with CP in normal rats. But, Mean Arterial Blood Pressure(MABP) did not affected by CP. In addition, the elevation of rCBF was blacked by pre-treatment with 1 mg/kg of indomethacin effectively. On the other hand, pre-treatment with 0.01 mg/kg of methylene blue did not affect rCBF level in normal rats. Conclusions: In conclusion, these results suggest that CP can act as anti-oxidant to protect neuroglia cells and also suggest that CP can elevate rCBF, which are involved in cyclooxygenase pathway.

Effect of CpG Oligodeoxynucleotides on Airways of Mice with Established Airways Inflammation (기도 염증이 유발된 생쥐에서 CpG Oligodeoxynucleotides가 미치는 효과)

  • Purpose : Airways eosinophilia and increased IgE, characteristic features of asthma, result from a predominant Th2 response. In this study, we investigated the effect of CpG oligodeoxynucleotides (ODNs) on the inhibition of airways eosinophilia in mice with established airway inflammation. We also investigated the immunological mechanisms involved. Methods : Groups of BALB/c mice were sensitized intradermally with ovalbumin(OVA). At week 10, airway inflammation was induced by intranasal challenge of the mice with OVA. At week 14, the mice were challenged intranasally again with OVA in the presence and without the presence of CpG ODNs. Mice with saline administration served as negative controls. Bronchoalveolar lavage fluids(BALF) were obtained and eosinophils were counted. Th1 and Th2 cytokines in the spleen cell cultures were measured by ELISA. Serum OVA-specific IgE and IgG2a antibodies were also measured by ELISA. Results : BALF eosinophils were significantly inhibited in the CpG ODNs-treated mice(P<0.01). IgE and IgG2a levels increased significantly in both CpG ODNs-treated and untreated groups as compared to the negative control group; there was, however, no significant difference between the two groups four days after intranasal administration of CpG ODNs. Cytokine analysis revealed decreased production of IL-4, IL-5, and IL-13 and increased production of IL-12 in the CpG ODNs-treated group as compared to the untreated group. Interestingly, $IFN-{\gamma}$ levels were not upregulated in the CpG ODNs-treated group. Conclusion : CpG ODNs vaccination is a potentially useful approach for reversing airways eosinophilia in mice with established airways inflammation.
