Effects of Hwanggeum-tang Water Extract on the Expression of Pro-inflammatory Responses Elicited by Advanced Glycation End Products in THP-1 Cells (황금탕(黃芩湯) 추출물이 THP-1 세포에서 당화종말산물에 의한 염증반응에 미치는 효과)

  • Hwanggeum-tang (HGT) was recorded in Dongeuibogam as being able to treat Sogal whose concept had been applied to Diabetes Mellitus (DM). Advanced glycation end products (AGEs) play important roles in the development of diabetic complications such as atherosclerosis by eliciting inflammatory responses. In this study, we examined the suppressive effects of HGT against inflammation elicited by AGEs. AGEs treatment increased the expression of pro-inflammatory cytokine gene TNF-${\alpha}$; chemokines MCP-1, IP-10; pro-inflammatory cyclooxygenase COX-2 on the THP-1 cells. HGT had suppressed the expression of pro-inflammatory genes and protein levels in AGE-treated THP-1 cells. HGT had also decreased intracellular ROS production stimulated by AGEs. These results suggest that HGT has beneficial effects for the improvement diabetic vascular complication through suppressing inflammatory responses elicited by AGEs.

Inhibitory Effects of Ginsenoside Rb1,Rg3, and Panax ginseng Head Butanol Fraction on Inflammatory Mediators from LPS-Stimulated RAW 264.7 Cells

  • Panax ginseng C.A. Mayer (Araliaceae, P. ginseng) has been used for the enhancement of vascular and immune functions in Korea and Japan for a long time. Ginsenoside $Rb_1$ and $Rg_3$ isolated from P. ginseng head-part butanolic extract (PGHB) were investigated for anti-inflammatory activity. Ginsenosides and PGHB did not affect the cell viability within $0\;-\;100\;{\mu}g/ml$ concentration to RAW 264.7 murine macrophage cells. Ginsenosides and PGHB inhibited partly lipopolysaccharide (LPS)-induced nitrite production in a dose-dependent manner. The ginsenosides and PGHB showed partially chemical nitric oxide (NO) quenching (maximum 40%) in the cell-free system. Also, ginsenoside $Rb_1$ and $Rg_3$ inhibited markedly approximately 74 and 54% of inducible nitric oxide synthase (iNOS) mRNA transcription from LPS-induced RAW 264.7 cells. Taken together, the inhibitory effect of ginsenosides and PGHB on NO production did not occur as a result of cell viability, but was caused by both the chemical NO quenching and the regulation of iNOS. Additionally, the ginsenoside $Rb_1$ and PGHB inhibited prostaglandin $E_2$ ($PGE_2$) synthesis in a concentration-dependent manner, showed approximately 70-98% inhibition at $100\;{\mu}g/ml$ concentration. And the treatment with ginsenosides and PGHB attenuated partially LPS-upregulated cyclooxygenase-2 (COX-2) gene transcription. Ginsenoside $Rg_3$ suppressed LPS-stimulated interleukin-6 (IL-6) level to the basal in RAW 264.7 cells. From these results, ginsenoside $Rb_1,\;Rg_3$, and PGHB may be useful for the relief and retardation of immunological inflammatory responses and its action may occur through the reduction of inflammatory mediators, including NO, $PGE_2$, and IL-6 production.

The effect of hyaluronic acid on anti-inflammatory action in mouse (마우스에서 히알우론산 나트륨이 항염효과에 미치는 영향에 관한 연구)

  • Purpose: The purpose of this study was not only to evaluate the relative mRNA expression of interleukin-$1{\beta}$(IL-$1{\beta}$), cyclooxygenase2 (COX-2) and prostaglandin E2 (PGE2) by RT-PCR analysis but to observe pattern of edema by light microscopic and electron microscope after topical apply of hyaluronic acid in inflammation-guided mouse. Material and methods: Mice of this study were devided into 4 groups: Control group (no inflammation guided), Positive control (inflammation guided + vaselin apply), Protopic group (inflammation guided + protopic apply), Hyaluronic group (inflammation guided + hyaluronic acid apply). Results: Hyaluronic group showed less expressions of IL-$1{\beta}$, COX-2, PGE2 than those of positive control & protopic group. Hyaluronic group revealed a decreased inflammation than positive control & protopic group in Light Microscope. Hyaluronic group appeared decreased edema of ear compare to positive control & protopic group in Elecron Microscope. Conclusion: It was considered that hyaluronic acid has an antiinflammatory effect for intercepting the gene expression of cytokines related to inflammation.

Anti-inflammatory Effects of Haedoksamultang in RAW 264.7 cells (해독사물탕(解毒四物湯)의 항염효과(抗炎效果) 및 면역반응(免疫反應)에 관(關)한 연구(硏究))

  • Purpose: This study was performed to determine anti-imflammatory effects of Haedoksamultang. Methods: In this study, I examined the effects of Haedoksamultang on the production of nitric oxide(NO), tumor necrosis factor-${\alpha}$(TNF-${\alpha}$), and interlukin-1${\beta}$(IL-1${\beta}$) as well as the expression of inducible NO synthase(iNOS), cyclooxygenase-2(COX-2), TNF-${\alpha}$, and IL-1${\beta}$ in RAW 264.7 cells. Haedoksamultang inhibited LPS-stimulated NO production. Western blotting and RT-PCR analysis showed that Haedoksamultang suppressed LPS-induced iNOS and COX-2 protein and mRNA expression in RAW 264.7 cells. Haedoksamultang also suppressed the expression and production of LPS-stimulated TNF-${\alpha}$ and IL-1${\beta}$ in RAW 264.7 cells. Haedoksamultang inhibited NF-${\kappa}B$ activation in LPS-treated RAW 264.7 cells. Moreover, this compound blocked $I{\kappa}B-{\alpha}$ phosphorylation and nuclear translocation of the cytosolic NF-${\kappa}B$ p65 subunit, which highly correlated with the production and expression of inflammatory mediators. Results: Haedoksamultang suppresses that inflammation-associated gene expression by blocking NF-${\kappa}B$ activation. Conclusion: These results suggest that Haedoksamultang may be beneficial for treating inflammatory disease.

The Evaluation of the Effect of Herbal Extract on Osteoarthritis: In Vitro and In Vivo Study

  • In this study, the anti-osteoarthritis effects of Cynanchum wilfordii, Phlomis umbrosa, and Angelica gigas extract (CPAE), observed and confirmed in previously clinical studies were further investigated by in vitro and in vivo studies. Anabolic biomarkers related to healthy cartilage maintenance, such as aggrecan, type II collagen ${\alpha}$-1 (Col2a1), sex determining region Y-box-9 (Sox-9), and catabolic biomarkers related to osteoarthritis, such as cyclooxygenase-2 (Cox-2), matrix metalloproteinase-13 (Mmp13), and nuclear factor kappa-light-chain-enhancer of activated B cells ($Nf{\kappa}b$), were evaluated by quantitative reverse transcriptase polymerase chain reaction and reporter gene assay. In vitro study results showed significant changes in both anabolic and catabolic biomarkers. For anabolic factors, significant changes in the level of aggrecan (P<0.05), Col2a1 (P<0.05), and Sox-9 (P<0.01) activation were shown after treatment of cartilage cells with CPAE (50 ng/mL) with similar efficacy compared to insulin growth factor, the positive control (100 ng/mL). For catabolic factors, significant changes in the inhibition activity of Cox-2 (P<0.05), Mmp13 (P<0.01), and $Nf{\kappa}b$ (P<0.05) were shown for CPAE (50 ng/mL) with similar efficacy compared to Celecoxib, the positive control ($10{\mu}M$). In the in vivo carrageenan-induced paw edema model study results showed that CPAE-treated groups (100 mg/kg) and Celecoxib-treated groups (60 mg/kg) showed comparably significant efficacy of inhibition by 37.1% and 52.1%, respectively. Furthermore, CPAE (200 mg/kg) showed similar effect to Celecoxib (60 mg/kg) with an inhibition rate of 54.3%. This result confirms that CPAE effectively inhibited the inflammation-induced osteoarthritis symptoms.

Study of Anti-microbial Activities and Anti-inflammatory Effects of Chamomile (Matricaria chamomilla) Extracts in HaCaT cells (HaCaT 세포주에서 캐모마일 (Matricaria chamomilla) 추출물의 항병원 성 및 항염 효과에 관한 연구)

  • Chamomile (Matricaria chamomilla), a member of the Asteraceae family, is a well-known for medicinal plant and can be found in India and Europe. Chamomile is an effective sedative and various medical effects. But, the effects of acne treatment by chamomile were not investigated. Therefore, we assessed the anti-oxidant effects, anti-microbial activity and anti-inflammatory effects by chamomile extracts in HaCaT keratinocyte cells. Anti-oxidant effects of chamomile extracts were investigated by DPPH assay. Also, results of MTT assay was demonstrated that chamomile extracts did not have a cytotoxic effect in HaCaT cells. To assess the antimicrobial activity, we determined formation of inhibition zone of Propionibacterium acnes by extracts from chamomile. Tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) induces production of inflammatory cytokines such as interleukin-$1{\beta}$ (IL-$1{\beta}$), IL-6 and IL-8 and expression of COX-2. Chamomile extracts could inhibit TNF-${\alpha}$-induced mRNA expression levels of IL-$1{\beta}$, IL-6, IL-8 and COX-2 gene. These results demonstrated the possibility of chamomile for prevention and treatment of skin inflammatory diseases such as acne.

Whey protein hydrolytic properties and its immunomodulation activity by produced enzyme from Serratia marcescens S3-R1 (Serratia marcescens S3-R1이 생산한 효소에 의한 유청단백질 가수분해물의 특성과 면역조절 활성)

  • Degrees of hydrolysis by alkaline protease produced from Serratia marcescens S3-R1 is 3.95-6.30% of whey proteins during 5, 15, 30, 60, 90, 120,180, 240 min incubation at $40^{\circ}C$. Proteolytic pattern of the whey proteins showed that various low molecular weight peptides were generated during the incubation periods. The biological function of in Raw 264.7 cells treated with whey protein hydrolytic peptides, anti-inflammatory effect showed exhibit in the expression of pro-inflammatory cytokines such as TNF-${\alpha}$, IL-6, COX-2 and iNOS by PCR analysis. COX-2 and iNOS gene expression inhibited in Raw 264.7 cells on whey protein hydrolysates below 3,000 dalton. The protease from Serratia marcescens S3-R1 showed a potential in production of low molecular weight whey protein hydrolysates which could be used for industrial application.

The Effect of Haptoglobin on Expression of Inflammatory Cytokines in 3T3-L1 Preadipocytes. (3T3-L1 지방전구세포에서 합토글로빈에 의한 염증성 cytokine 발현 조절)

  • White adipose tissue is now recognized as an important endocrine organ which secretes various signal factors and proteins termed 'adipokine'. Haptoglobin (Hp), which has been known as an acute phase protein, belongs to the adipokine. However, the function of Hp in adipose tissue remains unclear. To verify the role of Hp in preadipocytes, in this study, 3T3-L1 preadipocyte cells were stably transfected with human Hp gene and Hp-overexpressing cells were made. The Hp had no effect on cell growth of preadipocytes. By RT-PCR and Western blot analysis, the Hp inhibited gene expression of IL-6 and COX-2 and enhanced HO-1 synthesis in preadipocytes. Moreover, invasion assay showed the Hp suppressed migration of monocytes to preadipocytes. These findings suggest that the Hp may inhibit an inflammatory reaction in adipose tissue by regulating the expressions of pro-inflammatory and anti-inflammatory mediators, and by repressing monocytes/macrophages infiltration.

New records of three monogonont and seven bdelloid rotifers from Korea

  • Rotifers collected from mosses, leaf litter, soil, and wetland at six different locations in Korea were investigated. Ten species belonging to six genera in four families of monogonont and bdelloid rotifers were new to Korea: Habrotrocha scepanotrochoides De Koning, 1947, H. tripus (Murray, 1907), Lecane opias (Harring and Myers, 1926), L. perpusilla (Hauer, 1929), Macrotrachela decora (Bryce, 1912), Otostephanos auriculatus auriculatus(Murray, 1911), O. torquatus amoenus Milne, 1916, Scepanotrocha corniculata Bryce, 1910, S. simplex De Koning, 1947, and Testudinella brevicaudata Yamamoto, 1951. Among these 10 Korean new records, Habrotrocha scepanotrochoides, H. tripus, Macrotrachela decora, Otostephanos auriculatus auriculatus, O. torquatus amoenus, Scepanotrocha corniculata, and S. simplex were new to Asia as well. The present study is the first report of the genus Scepanotrocha Bryce, 1910 from Korea. Here, diagnostic characteristics and photomicrographs are provided for each Korean new record. In addition, a partial sequence of mitochondrial cytochrome c oxidase subunit 1 gene (mtCOX1) for S. simplex is presented.

Four bdelloid rotifers new to Korea

  • We conducted a taxonomic study of bdelloid rotifers found from terrestrial habitats such as mosses, leaf litter and soil at four different locations in Korea. The study resulted in four new Korean records belonging to two genera in two families, Habrotrochidae and Philodinidae, of bdelloid rotifers: Habrotrocha soror Donner, 1950; H. thienemanni rubella Donner, 1951; H. valida Milne, 1916 and Philodina nitida nitida Milne, 1916. These new Korean records are rare and have very limited distributions. Habrotrocha thienemanni rubella and H. valida were not found again after their original descriptions until the present study. The present study is the third record for both H. soror and P. nitida nitida. All these four new Korean records are new to Asian fauna as well. Here, diagnostic characteristics and photomicrographs are provided for each new Korean record. In addition, a partial sequence of mitochondrial cytochrome c oxidase subunit 1 gene (mtCOX1) for P. nitida nitida is presented.
