A study of the formation of artificial plaque on orthodontic brackets (교정용 브라켓상의 인공치태 형성에 대한 연구)

  • 구강내의 교정 장치는 미생물 전파에 있어서 다양한 장소를 제공한다. 치태는 치아 우식증 발생에 있어 매우 큰 역할을 하며 미생물, 비세포성 물질로 구성되어 있다. 본 연구의 목적은 생체외에서 교정용에서의 인공 치태 생성에 영향을 주는 요인을 평가하는 것이다. Streptococcus mutans type c는 $CO_2$ incubator내의 $37^{\circ}C$에서 brain heart infusion broth에서 배양 되었다. 중절치에 사용하는 9개의 $.018"{\times}.025"$ standard edgewise brackets을 3개씩 비이커의 배양액에 매달았다. 3개 비이커의 배양액 pH는 각 각 pH 5.5, 7.0 그리고 8.5로 조절되었다. 5시간 후에 비이커에서 각각의 bracket을 꺼내서, bracket의 평균 무게를 측정하였다. 배지의 stirring effect를 측정하기 위하여 3개씩 $.018"{\times}.025"$ standard edgewise brackets을 2개의 비이커에 위치시켰다. 12개의 brackets을 $CaCl_2$(0.25, 1.0, 4.0 그리고 16.0mM). KCl(2.5, 10, 40 그리고 160mM) 그리고 $MgCl_2$(0.1, 0.4, 1.6, 그리고 6.4mM) 용액에 각각 매달았다. 6개의 $.018"{\times}.025"$ standard edgewise brackets, 6개의 $.022"{\times}.028"$ Roth brackets과 6개의 $.022"{\times}.028"$ Broussard brackets을 각각의 비이커 내에 매달았다. 배양액 내에서 5시간 동안 배양한 후 각각의 brackets을 근사값의 milligram 단위로 측정하였다. 그룹 사이의 차이는 Mann-Whitney와 Kruskal-Wallis tests를 이용하여 비교하였다. p value<0.05의 조건에서 이들의 차이는 통계학적 유의성을 갖는다. 5시간 동안 pH 5.5에서 배양된 Streptococcus mutans에 의해 형성된 인공 치태는 pH 7.0 이나 pH 8.5에서 배양된 것보다 작았다(p<0.05). 인공 치태는 배양하는 동안 저어졌을 때 더 많이 형성되었다(p<0.05). 결론적으로 bracket에 형성된 인공 치태는 좀 더 높은 알칼리성 배지에서 배양됨으로써, 그리고 배양 동안 배지를 저어줌으로써 유의성있게 증가하였다. 그러나 배지의 $CaCl_2,\;KCl,\;MgCl_2$의 농도와 상업적으로 다른 종류의 이용 가능한 교정용 bracket에 대해서는 유의한 차이가 없었다.

Effects of pH, Water Temperature and Chlorine Dosage on the Formation of Disinfection Byproducts at Water Treatment Plant (pH, 수온, 염소주입량이 정수장 소독부산물 생성에 미치는 영향)

  • This study investigated formation potential of 16 disinfection byproducts (DBPs) (e.g., g trihalomethanes, haloacetic acids, haloacetonitriles, chloral hydrate, etc.) upon chlorination of raw water at various pH, water temperatures, and chlorine doses. We also compared the DBP formation potential (DBPFP) of raw and filtered waters. Most of DBPs were formed higher at neutral pH, but dichloroacetic acid, chloroform, and bromodichloromethane were formed higher over pH 7. As water temperature increased, concentrations of chloral hydrate, haloacetic acids, and haloacetonitriles linearly increased while that of trihalomethanes exponentially increased. Formation of chloral hydrate, trihalomethanes, and trihaloacetonitriles significantly increased up to 2.0 mg/L $Cl_2$ of chlorine addition, then gradually increased at 2.0~5.6 mg/L $Cl_2$. Filtered water formed less DBPs than raw water in most DBPs except for trihalomethanes.

Expression of Antioxidant Isoenzyme Genes in Rice under Salt Stress and Effects of Jasmonic Acid and ${\gamma}$-Radiation

  • Analysis of chlorophyll (Chl) fluorescence implicated treatment of 40 mM NaCl decreased maximal photochemical efficiency of photosystem II (PSII) (Fv/Fm), actual quantum yield of PSII (${\Phi}_{PSII}$), and photochemical quenching (qP) in rice, but increased non-photochemical quenching (NPQ). Decreases in Fv/Fm, ${\Phi}_{PSII}$, and qP were significantly alleviated by $30\;{\mu}M$ jasmonic acid (JA), while NPQ increase was enhanced. Transcription levels of antioxidant isoenzyme genes were differentially modulated by NaCl treatment. Expression of cCuZn-SOD2 gene increased, while those of cAPXb, CATb, and CATc genes decreased. JA prevented salt-induced decrease of pCuZn-SOD gene expression, but caused greater decrease in mRNA levels of cAPXa and Chl_tAPX genes. Investigation of vacuolar $Na^+/H^+$ exchanger (NHX2) and 1-pyrroline-5-carboxylate synthetase (P5CS) gene expressions revealed transcription level of NHX2 gene was increased by JA, regardless of NaCl presence, while that of P5CS gene slightly increased only in co-presence of JA and NaCl. Unlike JA, ${\gamma}$-radiation rarely affected expressions of antioxidant isoenzyme, NHX2, and P5CS genes, except for increase in mRNA level of Chl_tAPX and decrease in that of pCuZn-SOD. These results demonstrate enhanced salt-tolerance in JA-treated rice seedlings may be partly due to high transcription levels of pCuZn-SOD, NHX2, and P5CS genes under salt stress.

Effect of Mashed Red Pepper Admixed with Various Freezing Point Depression Agents on the Quality Characteristics of kimchi (빙점 강하제를 첨가한 다진 홍고추가 김치 품질 특성에 미치는 영향)

  • We investigated the use of mashed red pepper in the manufacture of kimchi using various freezing point depression agents (glucose, pectin, ascorbic acid or NaCl). In sensory evaluation results, overall acceptance seemed to be highly related to NaCl content. Kimchi was prepared using mashed red pepper with different levels of NaCl (3, 5, and 10% w/w), and quality characteristics were investigated during storage at $10^{\circ}C$. Salt concentrations of kimchi prepared with addition of 3, 5, or 10% NaCl, and control kimchi, were 2.28, 2.47, 3.10 and 1.92%, respectively. pH values varied significantly among treatments but acidity levels did not differ significantly except during the early stages of production. With increasing NaCl addition, the acidity of kimchi was lower than that of control samples during later fermentation periods. At day 20, the reducing sugar level in control kimchi had decreased by 70% whereas NaCl treatment caused decreases of 45 - 55%. The vitamin C contents of kimchi prepared with addition of NaCl (125.88-145.23 mg/100 g) were higher than that of control (37.22 mg/100 g). In sensory evaluation tests, appearance and texture did not differ significantly with treatment or fermentation period. When taste and overall acceptance were scored, kimchi prepared with the addition of 3% NaCl attained the highest marks throughout the entire fermentation period.

A Study on the Foaming Properties of Small red bean Protein Isolates at Various Conditions (분리 팥 단백질의 기포 특성에 영향을 주는 제 요인에 관한 연구)

  • The purpose of this study was to determine the foaming properties of two small red bean protein isolates at various conditions. Data concerning the effects of pH, temperature, MaCl concentration, sugar concentration and protein concentration on the properties such as solubility, foam expansion, foam stability were presented. The results were summarized as follows : 1. The crude protein contents of two small red beans were 26.14% and 22.71%. The percentage of nonpolar amino acid group was the highest and that of sulfur containing amino acid group was the lowest. 2. Protein solubility showed the minimum at pH 4.5 which is isoelectric point of small red bean protein isolate adn heat treatment lowered solubility(P<0.05). At pH 4.5, solubility increased sighificantly as 0.4M NaCl was added. However, the effect of sugar concentration in the solubility was not significant. 3. Foam expansion of two small red bean protein isolates was high at pH 4.5 and heat treatment at 10$0^{\circ}C$ lowered foam expansion(P<0.05). While addition of NaCl, sugar did not affect the foma expansion, gradual increment of the protein isolates concentration up to 9% decreased the foma expansion slightly. 4. Foam stability was significantly high at pH 4.5 and heat treatment at 10$0^{\circ}C$ lowered foam stability. Addition of sugar caused slight decrease in foam stability. From 1% to 9% suspension, foma stability increased significantly as protein concentration increased(P<0.05)

  • Minimal s-Urysohn and minimal s-regular spaces are studied. An s-Urysohn (respectively, s-regular) space (X, $\mathfrak{T}$) is said to be minimal s-Urysohn (respectively, minimal s-regular) if for no topology $\mathfrak{T}^{\prime}$ on X which is strictly weaker than $\mathfrak{T}$, (X, $\mathfrak{T}^{\prime}$) is s-Urysohn (respectively s-regular). Several characterizations and other related properties of these classes of spaces have been obtained. The present paper is a study of minimal P-spaces where P refers to the property of being an s-Urysohn space or an s-regular space. A P-space (X, $\mathfrak{T}$) is said to be minimal P if for no topology $\mathfrak{T}^{\prime}$ on X such that $\mathfrak{T}^{\prime}$ is strictly weaker than $\mathfrak{T}$, (X, $\mathfrak{T}^{\prime}$) has the property P. A space X is said to be s-Urysohn [2] if for any two distinct points x and y of X there exist semi-open set U and V containing x and y respectively such that $clU{\bigcap}clV={\phi}$, where clU denotes the closure of U. A space X is said to be s-regular [6] if for any point x and a closed set F not containing x there exist disjoint semi-open sets U and V such that $x{\in}U$ and $F{\subseteq}V$. Throughout the paper the spaces are assumed to be Hausdorff.

The Effect of Rapidly Rotating Shift work on the Fatigue Level, Urinary 17-KS, $Na^+$ and $Cl^-$ Excretion (빠른 교대근무가 피로도, 요중 17-KS, $Na^+,\;Cl^-$ 배설에 미치는 영향)

  • This study was done to investigate the effects of rapidly-rotating shift work of two-day interval on fatigue level and the concentration of urinary 17-KS, $Na^+,\;Cl^-$. The subjects were 20 nursing college students(control group) and 15 nurses in a university hospital and the study was done from Apr. 21 to May 4th, 1999. In the test group, each 5 nurses were allocated to day shift(8 AM-4 PM), evening shift(4 PM-12 MN) and night shift(12 MN-8 AM) respectively. The fatigue level were measured 30 minutes after work start on the 2nd day of work shift. Urine specimens were collected at 8 AM, 4 PM and 12 MN on the 2nd day of work shift in the control group and 30 minutes before and after work on the 2nd day of work shift in the test group. The data were analyzed with SPSS(for Window, ver 7.5). Statistical analysis was performed by using t-test, paired t-test and ANOVA. The results were as follows. 1. The perceived fatigue level in shift work 1) The physical and mental fatigue level were significantly higher in night shift than that in day or evening shift(p<0.05). In the neuro-sensory fatigue level, night shift showed higher tendency than that in day or evening shift, but there were no significant differences between each shifts. 2) Comparison between the control group and the test group: Physical fatigue level was significantly higher in night shift than that in day or evening shift of the control group(P<.001). Mental fatigue level was significantly higher in day or night shift than that in evening shift of the control group(P<.05). In the neuro-sensory fatigue level, test group showed higher tendency than that in the control group, but there were no significant differences between two groups. 3) The total fatigue level was higher in night shift than that in day shift or evening shift(P<.05). In comparing with the control group, night shift and day shift showed higher total fatigue level than that in the control group(p<0.05). 2. The concentration of urinary 17-KS, $Na^+$ and $Cl^-$ In the control group, urinary 17-KS, $Na^+$ and $Cl^-$ showed higher level in afternoon that in morning and night. In the test group, cr in day and evening shift and $Na^+$ in evening shift showed higher level at the end of work. The 17-KS concentration at the begining and the end of work in three shift groups were lower than those in control group(p<0.05), however, $Cl^-$ concentration at the begining of work in day shift, and the end of work in day and evening shift were higher than those in control group(p<0.05). $Cl^-$ concentration at the begining and end of work in night shift were considerably higher than those in control group repectively(p<0.1, p<001). $Na^+$ concentration showed a higher tendency in three shift groups except at the begining of work in night shift, but there were no statistical difference. In comparing concentration of the 17-KS, $Na^+$ and $Cl^-$ among the shift groups, 17-KS concentration showed a lower tendency and $Na^+,\;Cl^-$ showed a higher tendency in night shift: The result of this study showes that biorhythm of shift work nurse was irregular. Fatigue level as the subjective index for evaluating the health problem concerning shift work was higher in night shift and proved to be in accordance with the concentration of urinary 17-KS, $Na^+$ and $Cl^-$ used as objective indices. Disturbation of biorhythm and work stress due to night shift seems to cause the health problem of nurses and decrease of work efficiency. It is considered that work regualtion is necessary for the rational management of the nursing administration.

Characterization and N Terminal Amino Acid Sequence Analysis of Catechol 1,2-Dioxy-genase from Benzoate Degrading Acinetobacter sp. KS-1 (Benzoate 분해세균 Acinetobacter sp. kS-1에서 분리된 catechol 1,2-dioxygenase의 특성 및 N 말단 아미노산 서열 분석)

  • The purpose of this work was to investigate the characterization and sequence of catechol 1,2-dioxygenase (Cl,2O) purified from Acinetobacter sp. KS-1 which was grown on benzoate as a sole carbon source. Cl,2O demonstrated its enzyme activity to catechol and 4-methylcatechol. The optimum temperature of Cl,2O was $35^{\circ}C$, and the optimal pH was in the range from pH 7.5 to 9.0. $Ag^{+}$, $Hg^{+}$, and $Cu^{2+}$ showed inhibitory effect on the activity of Cl,2O. Molecular weight of the enzyme was determined to approximately 36 kDa by SDS-PAGE and 7-terminal amino acid sequence of Cl,2O was analyzed as $^{1}MNYQQIDALVKQMNVDTAKG^{20}$and exhibited 95% sequence homology with that of Cl,2O from Acinetobacter radioresistens In addition, trypsin digestion and peptide mapping were performed for internal sequencing analysis. Molecular weights of three digested peptide fragments were analyzed as 966.3 Da, 1933.8 Da and 2081.7 Da by MALDI-TOF, which were matched with each internal sequences $^{1}SQSDFNLRR^{9}\, ^{1}HGNRPSHVHYFNSAPGYR^{18}\, ^{1}TIEGPLYVAGAPESVGFAR^{19}$) of. A. radioresistens. PCR product was amplified with the degenerated primers derived from N-terminal and each internal amino acid sequences.

Pressure Effects on the Ionic Dissociation of $[Co(en)]_3Cl_3\;and\;[Co(NH_3)_6Cl_3$ in Aqueous Solutions (수용액에서 $[Co(en)_3]Cl_3$$[Co(NH_3)_6]Cl_3$착물의 이온 해리에 미치는 압력의 영향)

  • The thermodynamic dissociation constants of $[Co(en)]_3Cl_3\;and\;[Co(NH_3)_6Cl_3$ in aqueous solutions at $25{\circ}$ and at the pressure from 1 bar to 2000 bars were measured by conductometric method. The dissociation constants were increased with pressure elevation because of volume decreasing by the formation of charged ions during dissociation reaction. That is, the $pK^T$ values of $[Co(en)]_3Cl_3$ were 2.16 for I bar, 2.08 for 500 bars, 2,08 for 1000 bars, 2.05 for 1500 bars and 2.03 for 2000 bars, respectively and those of $[Co(NH_3)_6Cl_3$ were 2.02 for 1 bar 1.96 for 500 bars, 1.90 for 1000 bars, 1. 88 for 1500 bars and 1. 87 for 2000 bars, respectively. Comparing and analyzing the values of Stokes' radii and $K^T$, the formation of ion pair compound was affected by not only the electrostatic interaction, but also the Interal Conjugate Base(ICB), which was increased by the elevation of the pressure.

Studies on acid protease produced from Aspergillus tubingensis I (Aspergillus tubingensis의 acid protease에 관한 연구 I)

  • The strain of Aspergillus, 6368A, producing acid protease showing high activity was isolated from soil, as a result of wide research about mold group. This strain was identified as a species of Aspergillus tubingensis by the investigation of morphological characteristics. The change of the enzyme production under the various media and culture condition was also studied. The optimum pH and stability of crude acid protease are 2.5, 2.0~4.5 and the optimum temeprature and thermal inactivation waas shown $50^{\circ}C,\;55^{\circ}C$, respectively. From the result of the study on the effects of metal ions, it was found that $MnCl_2,\;CoCl_2,\;CuCl_2,\;SrCl_2,\;and\;NiCl_2$ slightly increased the enzyme activity, on the other hand $ZnCl_2,\;CaCl_2,\;MgCl_2,\;SLS,\;and\;KMnO_4$ decreased it.
