Development of Parthenotes Produced by Various Treatments in Bovine

  • Development of effective activation protocols is of great importance for improving the success of cloning and subsequent transgenic. Three methods for oocyte activation, including 5μM ionomycin (5 min) alone, ionomycin+1.9 mM 6-dimetylaminopurine (DMAP, 3 hrs) and ionomycin+10㎍/㎖ cycloheximide(CHX, 3 hrs) were compared for their effects of pronuclei(PN) formation, development, developmental velocity and ploidy of parthenotes to IVF control in bovine. In group of ionomycin+DMAP, the oocytes having more 3 PN were significantly(P〈0.05) higher than in groups of ionomycin alone and of ionomycin+CHX (45.5% vs. 0 and 0%, respectively). Activation with the ionomycin alone, ionomycin+DMAP and ionomycin+CHX resulted in cleavage rates of 30, 85.5 and 57.9%, respectively. The blastocysts rate of parthenotes activated by ionomycin+DMAP treatment was significantly higher (12.3%, P〈0.05) than those of other treated groups. Chromosome analysis shows that ionomycin+DMAP treatment greatly increases the incidence of chromosomal abnormality of the parthenotes. When compared the developmental velocity at 24 hrs after insemination and activation, 27% eggs in IVF control and 55% in DMAP treatment out of total cleaved eggs developed to 2-cell stage, respectively. Developmental velocity of parthenotes activated by ionomycin +DMAP treatment was significantly (P〈0.05) faster than others. From the results, we may conclude that DMAP treatment to the oocytes accelerates developmental velocity resulting in both the higher incidence of chromosome abnormality and of PN formation suggesting that CHX combined with ionomycin is suitable DMAP for the purpose of successful nuclear transplantation.

Ellipsometry를 이용한 저 유전상수를 갖는 SiOCH박막의 광학특성 연구 (A Study of Optical Characteristics Correlated with Low Dielectric Constant of SiOCH Thin Films Through Ellipsometry)

  • We studied the optical characteristics correlated with low dielectric constants of low-k SiOCH thin films through ellipsometry. The low-k SiOCH thin films were prepared by CCP-PECVD method using BTMSM(Bis-trimethylsilylmethane) precursors deposited on p-Si wafer. The Si-O-CHx, Si-O-Si, Si-CHx, CHx and Si-H bonding groups were specified by FTIR spectroscopic spectra, and the groups coupled with the nano-porous structural organic/inorganic hybrid-type of SiOCH thin films which has extremely low dielectric constant close to 2.0. The structural groups includes highly dense pore as well as ions in SiOCH thin films affecting to complex refraction characteristics of single layer on the p-Si wafer. The structural complexity originate the complex refractive constants of the films, and resulted the elliptical polarization of the incident linearly polarized light source of Xe-light source in the range from 190 nm to 2100 nm. Phase difference and amplitude ratio between s wave and p wave propagating through SiOCH thin film was studied. After annealing, the amplitude of p wave was reduced more than s wave, and phase difference between p and s wave was also reduced.

Optimization of Post-Activation Systems to Improve the Embryonic Development in Porcine Parthenogenesis and Somatic Cell Nuclear Transfer

  • This study was conducted to establish the optimal chemical post-activation conditions in porcine embryonic development after parthenogenesis (PA) and somatic cell nuclear transfer (SCNT) using 4 different chemical compositions (cytochalasin B (CB), cyclohexamide (CHX), demecolcine (DC), 6-dimethylaminopurine (DMAP). Porcine embryos were produced by PA and SCNT and then, cultured for post-activation with CB ($7.5{\mu}g/mL$), CB ($7.5{\mu}g/mL$) + CHX ($10{\mu}g/mL$), CB ($7.5{\mu}g/mL$) +DC ($0.4{\mu}g/mL$), and CB ($7.5{\mu}g/mL$) + DMAP (2 mM). In PA embryonic development, cleavage rates have been significantly higher in CB group (94.7%) and CB+DMAP group (94.1%) than that of CB+CHX and CB+DC group (88.1 and 84.3%, respectively). There have been no significant differences in blastocyst formation rates among the four groups. In cell number of blastocyst was shown in CB group (42.3%) significantly higher than CB+CHX and CB+DC group (40.6 and 40.6%, respectively). In SCNT embryonic development, CB+DMAP group (89.7%) significant differences were found on embryo cleavage rates when compared with other three groups. Blastocyst formation rates in CB+DMAP group (26.9%) were significantly higher when compared with CB, CB+CHX, and CB+DC groups (25.5, 20.2, and 22.1%, respectively). In blastocyst cell number, CB+DMAP group (41.4%) was found higher significant difference compared with other three groups. Additionally, we have investigated survivin expression in early development stages of porcine SCNT embryos for more confirmation. Our results establish that CB group and CB+DMAP group for 4 h during post-activation improves pre-implantation improvement of PA and SCNT embryos.

Propidium monoazide와 real-time PCR을 이용한 살아있는 Enterococcus faecalis의 선택적인 검출 (SELECTIVE DETECTION OF VIABLE ENTEROCOCCUS FAECALIS USING PROPIDIUM MONOAZIDE IN COMBINATION WITH REAL-TIME PCR)

  • 세균의 검출에 있어서 polymerase chain reaction (PCR) 방법은 기존의 plate counting과 달리 빠르게 세균을 검출할 수 있다. 하지만 세균이 죽은 후에도 DNA는 장기간 존재할 수 있기 때문에, DNA에 기초한 분석은 살아있는 세균과 죽은 세균을 구분할 수 없다. 최근에 DNA extraction전에 propidium monoazide (PMA)를 처리하여 살아있는 세균만 선택적으로 검출하는 방법이 제시되었다. PMA는 손상된 세포막만 통과하여 죽은 세포의 DNA와 빛 노출 하에서 결합하여 PCR이 증폭되는 것을 막는다. Enterococcus faecalis는 근관치료의 실패에 있어서 중요한 원인이 되는 세균으로 제시되어 왔다. 그리고 chlorhexidine (CHX)은 E. faecalis의 제거에 있어서 효과적인 약제임이 밝혀졌다. 이번 실험의 목적은 세균 수의 측정에 있어서, PMA 처리와 real-time PCR 방법의 적용 가능성을 기존의 plate counting과 비교하여 알아보는 것이다. 또한 E. faecalis에 대한 2% CHX의 살균 효과를 PMA 처리 후 real-time PCR 방법을 사용하여 알아보는 것이다. 실험 방법으로 먼저 살아있는 세균과 죽은 세균을 다른 비율로 섞어서 PMA를 처리한 후 real-time PCR을 시행하여 PMA가 빛 노출 하에서 죽은 세균의 DNA와 결합하는 효과를 나타내는지 알아보았다. 다음으로 PMA 처리 후 realtime PCR 방법을 이용하여 살아있는 세균의 양을 측정한 것을 plate counting으로 얻은 CFU와 비교하였다. 마지막으로 2% CHX의 처리시간을 다르게 하였을 때 E. faecalis에 대한 살균 효과를 PMA 처리 후 real-time PCR 방법을 사용하여 알아보았다. 실험 결과로 살아있는 E. faecalis의 비율이 감소할수록 Ct value는 증가하였다. 그리고 PMA 처리 후 real-time PCR 방법을 이용하여 세균의 양을 측정한 것과 plate counting으로 얻은 CFU 사이에는 Optical density (OD) 값이 1.0일 때까지는 상관관계가 있었다. 하지만 OD 값이 1.5일 때는, PMA를 처리한 후 real-time PCR을 시행했을 때 측정된 살아있는 세균의 양이 감소하였음에 반해서 plate counting에 의한 CFU는 계속 증가하였다. 마지막으로 2% CHX을 오래 적용할수록 살아있는 E. faecalis의 상대적인 양이 감소하는 것을 PMA 처리와 real-time PCR 방법을 이용해 확인하였다.

수종의 근관세척액과 Listerine(R)의 항균성 비교 연구 (COMPARISON OF ANTIBACTERIAL EFFECT OF Listerine(R) WITH VARIOUS ROOT CANAL IRRIGANTS)

  • 본 연구는 여러 가지 근관세척액(NaOCl, CHX, EDTA)과 구강세정제로 사용되는 $Listerine^{(R)}$을 근관감염균주인 Porphyromonas gingivalis와 Enterococcus faecalis를 상대로 항균효과를 비교하고 $Listerine^{(R)}$이 근관세척액으로 사용가능한지를 확인하고자 시행하였다. 본 연구에서는 Porphyromonas gingivalis ATCC 3327과 Enterococcus faecalis ATCC 29212의 표준균주를 사용하였다. 실험을 위한 근관세척제로 0.1%, 0.2%, 1%, 2% Chlorhexidine(CHX)과 0.5%, 1%, 2.5%, 5.25% NaOCl, 0.5M EDTA(18.6% EDTA). 그리고 $Listerine^{(R)}$원액을 이용하였다. 항균효과의 비교는 액체배지상에서 균주의 혼탁도와 한천 확산법을 이용한 억제대 비교로 하였다. 모든 실험군은 대조군과 비교시 근관내 균주에 항균성을 나타냈다(p < 0.001). 모든 농도의 NaOCl, CHX, 그리고 EDTA는 실험균주에서 높은 항균성을 보였다. 모든 실험에서 $Listerine^{(R)}$은 다른 근관세척제에 비해 낮은 항균성을 보였다. 결론적으로, $Listerine^{(R)}$이 E. faecalis와 P. gingivalis에 대해 항균성을 보이나, 일반적으로 사용되는 근관세척 액에 비해서는 현저히 낮은 항균성을 나타냄으로 근관세척 액으로 사용은 적합하지 않음을 나타냈다.

Association of Anti-apoptotic Mechanism Due to House Dust Mite in Neutrophils with Protein Synthesis and Bad

  • House dust mite is an essential allergen in the pathogenesis of allergic diseases. Abnormal regulation of neutrophil apoptosis is an important pathogenic process in allergic diseases. In the present study, we investigated the effects of house dust mites on spontaneous apoptosis of neutrophils and its associated mechanisms. Extract of Dermatophagoides pteronissinus (DP) inhibited neutrophil apoptosis in a time-dependent manner. Cycloheximide (CHX), an inhibitor of translation, increased apoptosis of DP-treated neutrophils as well as control cells. The pro-apoptotic effect of CHX was blocked by DP in neutrophils. In addition, DP increased the phosphorylation of Bad in a time-dependent manner, indicating that it exerted an inhibitory effect on the function of Bad. These results suggest that DP has anti-apoptotic effects of neutrophils and may regulate protein synthesis and activation of Bad. Moreover, these findings may shed light on elucidation of allergy pathogenesis due to house dust mites.

치주질환치료를 위한 국소적용 서방출성 리오겔 (Injectable Sustained Release Gel as a Local Drug Delivery for Periodontal Diseases)

  • The purpose of this study was the development of sustained-release lyogel of chlorhexidine in the treatment of periodontal diseases. A sustained-release chlorhexidine lyogel (CHX-G) was formulated, based on Eudragit$^{(R)}$ (1~3%), polyvinyl pyrrolidone (PVP) (0~10%), triacetin (20~40%), hydroxy ethyl cellulose (HEC) (1%) and glycerin. In vitro studies were performed to determine the release rate of chlorhexidine from CHX-Gs using dialysis tube. Our results suggest that the release rate of chlorhexidine from lyogel could be controlled by changing the lyogel compositions.

  • I. Objectives Intracanal medicament with antibacterial action is required to maximize the disinfection of the root canal system. The purpose of this study was to evaluate the efficacy of 2% chlorhexidine digluconate(CHX) and controlled release device(CRD) containing 20% CHX and chitosan coating, compared to calcium hydroxide as intracanal medicament against E. faecalis. II. Materials and Methods One hundred and twenty intact freshly extracted bovine incisors were used and were 0.5% NaOCl. Middle 1/3 portion of roots were sliced into 4mm thick section and cementum was removed using diamond burs and external diameter was approximally 6mm.(omitted)

Comparison of Neurotoxicity Induced by Some Glutathione Depletors in Mouse Cortical Cell Cultures

  • We examined the neurotoxic effects of 3 glutathione (GSH) depletors, buthionine sulfoximine (BSO), diethyl maleate (DEM) and phorone, under the presence of trolox, cycloheximide (CHX), pyrrolidine dithiocarbamate (PDTC) or MK-801 in primary mouse cortical cell cultures. All three depletors induced neuronal death in dose and exposure time dependent manner, and decreased total cellular GSH contents. The patterns of the neuronal death and the GSH decrements were dependent on the individual agents. DEM $(200\;{\mu}M)$ induced rapid and irreversible decrement of the GSH. BSO (1 mM) also decreased the GSH irreversibly but the rate of decrement was more progressive than that of DEM. Phorone (1 mM) reduced the GSH content to 40% by 4 hr exposure, that is comparable to the decrement of BSO, but the GSH recovered and reached over the control value by 36 hr exposure. BSO showed a minimal neurotoxicity $(0{\sim}10%)$ at the end of 24 hr exposure, but marked neuronal cell death at the end of 48 hr exposure. The BSO (1 mM)-induced neurotoxicity was markedly inhibited by trolox or CHX and partially attenuated by MK-801. DEM induced dose-dependent cytotoxicity at the end of 24 hr exposure. Over the doses of $400\;{\mu}M,$ glial toxicity also appeared. DEM $(200\;{\mu}M)-induced$ neurotoxicity was markedly inhibited by trolox or PDTC. Phorone (1 mM) induced moderate neurotoxicity (40%) at the end of 48 hr exposure. Only CHX showed significant inhibitory effect on the phorone-induced neurotoxicity. These results suggest that the GSH depletors induce neuronal injury via different mechanisms and that GSH depletors should be carefully employed in the researches of neuronal oxidative injuries.

구강소독제의 적용 시간 및 기계적 세척 여부에 따른 타액 내 세균 수의 감소 효과 (Salivary Bacterial Counts on Application Time of Oral Antiseptic Agents and Mechanical Irrigation)

  • Purpose: The purpose of this study is to establish the acceptable intraoral application time of antiseptic agents and evaluate the effect of mechanical irrigation. Methods: A total of 80 subjects were selected for this study. Saliva secreted at the resting state was taken. The subjects were divided into 8 experimental groups, and kept 10% povidone-iodine (PVI) or 0.2% chlorhexidine gluconate (CHX) for 20 or 40 seconds in their oral cavity with/without irrigation of the oral cavity with sterilized normal saline, respectively. Then, the saliva was taken and diluted with phosphate buffered saline and then plated onto 5% sheep blood agar plates, which were incubated. Colony forming unit (CFU) was measured for the salivary bacterial counts. Results: After application of PVI and CHX, all the experimental groups showed statistically significant decrease in CFU (P<0.01). Group 2 (PVI, 40 s) showed more significant reduction rate in CFU than group 4 (CHX, 40 s; P<0.01). Group 6 (PVI, 40 s, irrigated) showed more significant reduction rate than group 2 (PVI, 40 s; P<0.01). Group 2 (PVI, 40 s) showed more significant reduction rate than group 1 (PVI, 20 s; P<0.01). Conclusion: Application of PVI for 40 seconds and mechanical irrigation with sterilized normal saline showed the best result among the 8 groups in terms of the reduction rate of salivary bacterial counts.
