Effect of Timing of Nutrient Starvation during Transplant Production on the Growth of Runner Plants and Yield of Strawberry 'Seolhyang' (딸기 '설향' 육묘기 양분 공급 중단 시기가 자묘 생육 및 수량에 미치는 영향)

  • This study was conducted to investigate the effects of timing of nutrient starvation during transplant production on growth of runner plants and yield of strawberry 'Seolhyang' (Fragaria ${\times}$ ananassa). Nutrient solution supply at the level of EC (electrical conductivity) 0.8 $dS{\cdot}m^{-1}$ was terminated at interval about 10 days between July 25 and September 5. As a result, the growth of above-ground part was inhibited while root growth increased when the nutrient starvation treatment had been brought forward to July 25. It also reduced the T/R ratio significantly and chlorophyll content was tended to be lower than the other treatment. In addition, it significantly promoted the budding, flowering and harvest of first flower cluster. On the other hand, the period of harvest was delayed more than two weeks when the nutrients were continuously supplied after the middle of August. An accumulated marketable fruit yield per plant until the end of January and February was 169 and 266g, respectively in the treatment of nutrient starvation on July 25, which was 71 and 12% increase, respectively, as compared with those in the treatment of September 5. Therefore, the appropriate nutrient starvation in the late season of strawberry nursery period could be expected the increase in yield and income during the winter season by promoting the flower bud differentiation as reducing the endogenous nitrate level of the plantlet.

Influence of Incorporation Rates of Sulfur on Reduction of the Bicarbonate Injury in Hydroponic Cultivation of 'Ssanta' Strawberry (유황 처리수준이 '싼타' 딸기의 중탄산 피해 경감에 미치는 영향)

  • This research was conducted to determine the influence of incorporation levels of sulfur into a coir dust+pine bark medium (1:1, v/v) on the changes in the bicarbonate ($HCO_3{^-}$) concentrations and pH of soil solution, crop growth, and nutrient uptake of 'Ssanta' strawberry. In the preparing of the mixed medium, sulfur powder was added with the rate of 0 (control), 0.23, 0.45, 0.90, and $1.80g{\cdot}L^{-1}$ and Hoagland nutrient solution containing $240mg{\cdot}L^{-1}$ $HCO_3{^-}$ was supplied during the crop cultivation. The growth measurements and tissue analysis for the determination of nutrient contents were carried out 140 days after solution application and the soil solution analysis was performed every two weeks. As the level of sulfur was elevated, the overall growth of mother plants became better showing that the growth indexes except chlorophyll contents were the lowest in control treatment but the statistical differences were not found among the three treatments of $0.45g{\cdot}L^{-1}$ or higher sulfur. The higher the levels of sulfur incorporation, the higher the occurrence of runners and the growth of daughter plants. The length of the runners and the number of daughter plants occurred per mother plants were higher in the treatments of 0.90 and $1.80g{\cdot}L^{-1}$ than the three treatments of 0, 0.23, and $0.45g{\cdot}L^{-1}$, but the statistical differences were not observed between the 0.90 and $1.80g{\cdot}L^{-1}$ treatments. The rose of pH and $HCO_3{^-}$ concentrations in soil solution of root media continued all the cropping period, but those decreased slightly in the treatments of $0.90g{\cdot}L^{-1}$ or higher. The soil solution concentrations of $K^+$ and $PO_4{^3-}$ in the treatments of 0.90 and $1.80g{\cdot}L^{-1}$ was lower than those in other treatments and the statistical differences in the $Ca^{2+}$ and $Mg^{2+}$ concentrations were not observed among all treatments. The nutrient contents in tissue analyzed in this experiment were the lowest in the control treatments and those increased as incorporation rates of sulfur were elevated. Above results indicated that when 'Ssanta' strawberry is grown hydroponically and the root medium is coir dust and the pine bark (5:5, v/v) mix, the sulfur incorporation rate as pre-planting fertilizer has to be higher than $0.9g{\cdot}L^{-1}$ root medium to reduce the $HCO_3{^-}$ injury.

Effect of different light sources and ventilation on in vitro shoot growth and rooting of a rare and endangered species, Tsuru-rindo(Tripterospermum japonicum) (희귀 및 멸종위기 식물 덩굴용담의 기내생장에 미치는 광질 및 환기효과)

  • Effects of light generated by LEDs on shoot growth and rooting of Tsuru-rindo(Tripterospermum japonicum) were evaluated. Apical shoots(one or two node with 3-4 leaves) were cultured on MS basal medium with 3% sucrose and maintained for four weeks under five different light qualities: fluorescent lamp(F), 100% red LED(R), 70% red LED+30% blue LED(R7B3), 50% red LED+50% blue(R5B5), or 100% blue LED(B). Rooting was promoted by both red light and fluorescent lamp, and the effect was further promoted under the ventilation. Red light enhanced shoot node elongation, whereas blue light appeared to suppress it. Growth of shoots and leaves were enhanced under the ventilation irrespective of the different light qualities. Under the ventilated condition, total fresh weight of plants was highest in R7B3 LED as 257.7 mg per plant. Dry matters, which are used for index of plant growth, were lowest under red light, whereas it was highest under blue light. The dry matter was inclined to getting higher by ascending the ratio of blue light and red light. Total chlorophyll content was highest in both R7B3 LED and R5B5 LED under ventilation as 29.5 and 31.2, respectively. Above results suggest that light quality optimization could be an important factor to foster in vitro growth of the species. Ventilation treatment appeared to be another important factor to induce normal shoot growth and rooting.

Identification of Maysin and Related Flavonid Analogues in Corn Silks (옥수수 수염에서 Maysin 및 유사물질의 동정)

  • This study was carried out to isolate and identify the maysin and related flavonoid analogues in corn silks. Silks were covered with silk bag to prevent pollination and were sampled at 3-5 days after silking. The silks were filled with 100% MeOH and stored at $0^{\circ}C$ until analysis. The MeOH extracts of corn silks were filtered and concentrated at 35-4$0^{\circ}C$. The ${CH}_2$${Cl}_2$ was added on the concentrated aqueous solution to remove the chlorophyll and lipids. The Cis open column (25mm$\times$54 cm) was washed and activated with serial treatment of 500$m\ell$ of 100% MeOH(twice)longrightarrow75% MeOH longrightarrow50% MeOHlongrightarrow30% MeOHlongrightarrow100% $H_2$O(2 times). The concentrated aqueous solution was applied to the $C_{18}$ column and washed with $H_2O$ several times to remove the sugars and water soluble pigments. Neochlorogenic acid, chlorogenic acid and 4-caffeoylquinic acid were eluted with 10% MeOH, and rhamosyl isoorientin was eluted with 30% MeOH, but maysin was eluted with 50% MeOH from the $C_18$ open column. Collected fractions were analyzed with HPLC by using revers-phase Ultras-phere $C_{18}$ column (4.6$\times$250mm, 5$\mu\textrm{m}$) and $H_2$O (10% MeOH containing 0.1% $H_3$${PO}_4$)/MeOH (100% MeOH containing 0.1% H$_3$PO$_4$) linear gradient from 20% to 90% MeOH for 35 minutes, a flow rate of 1 $m\ell$/min and detection at 340nm. The selected fractions were concentrated and applied to the silicic acid column. Maysin was eluted with 500$m\ell$ of 100% ethyl acetate from the silicic acid column for the first purification, and the purity of collected fractions was about 75%, but the purity from the second purification with the Cis column (1/2 $\times$ 43") was greater than 95%. FAB-MS spectral data was obtained with VG7O-VSEQ VG analytical fast atom bombardment mass (UK). $^1$H-NMR and $^{13}$ C-NMR data were obtained with Bruker DPX 400 MHz NMR spectrometers (German) in DMSO-d$_{6}$ at 400 and 100 MHz, respectively.vely.

Photosynthetic Characteristics and Primary Production by Phytoplankton with Different Water Quality of Influent in Open Waters of Constructed Wetlands for Water Treatment (수질정화용 인공습지 개방수역에서 유입수질에 따른 식물플랑크톤의 광합성특성 및 유기물생산력)

  • The photosynthetic characteristics and primary production by phytoplankton in open waters of two wetlands (the Banwol and the Donghwa wetland) of Sihwa Constructed Wetland with different water chemistry were investigated to provide the information for the wetland management considering the water treatment efficiency. During the study period (from March to October, 2005) the primary productivity in open waters ranged from 481 to 11,275 mgC $m^{-2}$ $day^{-1}$, which is very high compared with the eutrophic level of 600mgC $m^{-2}$ $day^{-1}$. From the analysis of the photosynthesis-irradiance (P-I) model parameters, the photosynthetic characteristics may be affected by different concentration and ratio of nutrient (N and P) between two wetlands. Assimilation number (AN) was higher in the Donghwa wetland (average AN: 8.5gC $gChl^{-1}$ $hr^{-1}$) with high P and low N/P ratio than the Banwol wetland (average AN: 5.8gC $gChl^{-1}$ $hr^{-1}$) with high N and high N/P ratio. This result indicates that AN may be concerned with phosphorus than nitrogen and low NIP ratio. Positive correlation (R=0.81) was observed between the initial slope and AN, implying that AN was high in case of phytoplankton having more active photosynthesis ability under low light. On the other hand, maximum photosynthesis (Pmax) was related positively with chlorophyll a concentration showing correlation coefficient of 0.47. In this study, considering the high primary production through phytoplankton photosynthesis in open waters of Sihwa Constructed Wetland, the produced organic matter by phytoplankton may affect the water quality within wetland and its efficiency of water treatment. Also, the photosynthetic characteristics may be affected by different nutrient enrichment (especially phosphorus) of wetlands. This study suggests that the production by phytoplankton and its characteristics in open water of constructed wetland for water treatment should be considered to improve the removal efficiency of organic matter.

Relationship of Fitness and Substance of Porphyrin Biosynthesis Pathway in Resistant Transgenic Rice to Protoporphyrinogen Oxidase (Protox) Inhibitor (Protoporphyrinogen oxidase (Protox) 저해제 저항성 형질전환 벼의 적응성과 Porphyrin 생합성 경로물질과 관련성)

  • The objectives of this study were to investigate fitness difference in growth and rice yield in herbicide-transgenic rice overexpressing Myxococcus xanthus and Arabidopsis thaliana protoporphyrinogen oxidase (Protox) genes and non-transgenic rice. We also aimed to determine whether these fitness differences are related to ALA synthesizing capacity, accumulation of terapyrroles, reactive oxygen species, lipid peroxidation, and antioxidative enzymes at different growth stages of rice. Plant height of the transgenic rice overexpressing M. xanthus (MX) and A. thaliana (AP37) Protox genes at 43, 50, and 65 days after transplanting (DAT) was significantly lower than that of WT. Number of tiller of PX as well as MX and AP37 at 50 and 65 DAT was significantly lower than that of WT. At harvest time, culm length and yield of MX, PX and AP37 and rice straw weight of MX and AP37 were significantly low compared with WT. The reduction of yield in MX, PX, and AP37 was caused by spikelets per panicle and 1000 grain weight, ripened grain, spikelets per panicle, 1000 grain weight, and ripened grain, respectively. On the other hand, 135 the reduction of yield in MX, PX, and AP37 was also observed in another yearly variation experiment. The reduction of rice growth in MX, PX, and AP37 was observed in seedling stage as well as growth duration in field. There were no differences in tetrapyrrole intermediate Proto IX, Mg-Proto IX and Mg-Proto IX monomethyl ester, reactive oxygen species ($H_2O_2$ and ${O_2}^-$), MDA, antioxidative enzymes (SOD, CAT, POX, APX, and GR) and chlorophyll between transgenic lines and wild type, indicating that accumulated tetrapyrrole intermediate and other parameters were not related to growth reduction in transgenic rice. However, ALA synthesizing capacity in MX, PX, and AP37 at one day after exposure to light and 52 DAT was significantly lower than that of WT. Further study is required to elucidate the mechanisms underlying the growth and yield difference between transgenic and WT lines.

Effect of Night and Daytime Temperatures on Growth and Yield of Paprika 'Fiesta' and 'Jubilee' (${\cdot}$야 온도가 착색단고추의 생육 및 수량에 미치는 영향)

  • This experiment was conducted to investigate the effect of night and daytime temperatures on growth and yield of paprika 'Fiesta' and 'Jubilee' under soil culture experiment in the vinyl houses during the 2003 and 2004 season. Total fruit yield was greater in 'Fiesta' than 'Jubilee' Marketable yield was not different between two cultivars, due to lower $\%$ marketable fruits in 'Fiesta'. mean Fruit weight was not different between two cultivars. Difference of yield between cultivars was due to fruit number, harves time and root condition. It was caused by cracked fruits to decrease $\%$ marketable fruits. Total yield was greater in nighttime temperature of $18^{\circ}C\;than\;15^{\circ}C$ and marketable yield was considerably greater because of $\%$ marketable fruits was higher. Mean fruit weight was slightly greater in nighttime temperature of $15^{\circ}C\;than\;18^{\circ}C$. Difference of fruit yield in treatments of nighttime temperature was due to fruit number and harvest time. There was not significant difference of yield between daytime temperature of $28^{\circ}C\;and\;31^{\circ}C$, but in $34^{\circ}C$, total and marketable yields were the least and mean fruit weight was the smallest because of decreased $CO_2$ concentration in the house, accerated vegetative growth, and the least chlorophyll content. There were no significant difference in photosynthetic rate, transpiration rate, maximal photochemical efficiency and antioxidant enzyme activities of all temperature treatments in this experiment. However it was clear that a little difference in error range of these results affects the source of crops in any case. It was not acknowledged that compensation effect by high temperature in daytime to the low temperature treatment in nighttime.

Estimation of POC Export Fluxes Using 234Th/238U Disequilibria in the Amundsen Sea, Antarctica; Preliminary Result (남극 아문젠해에서 234Th/238U 비평형법을 사용한 유광대에서 심층으로의 입자상 유기탄소 침강플럭스 추정; 예비결과)

  • In order to understand the carbon cycle in the Amundsen Sea of the Southern Ocean, the export fluxes of particulate organic carbon from the euphotic zone to deep water estimated using ${\psi}$/${\psi}$ disequilibrium method. Seawaters in 14 water columns were collected during February and March 2012, and analyzed for total and dissolved ${\psi}$, and particulate organic carbon. Total ${\psi}$ activities in the water column showed deficiency and excess relative to those of ${\psi}$ depending on the water depth. Deficiency of total ${\psi}$ in the euphotic zone showed mirror images both with chlorophyll-a and fluorescence, and was consistent with the loss of nitrate, which indicated the effect of biological activity. In addition, deficiency of total ${\psi}$ from deep water was associated with the increase of total dissolvable Fe/Mn concentration. Excess total ${\psi}$ activity presented below the euphotic zone might be related to particulate ${\psi}$ concentrated in this water depth. Mean export flux of ${\psi}$ estimated using the steady state model was $867{\pm}246dpmm^{-2}day^{-1}$. Mean export flux of particulate organic carbon, which were estimated by the product of total ${\psi}$ flux and ratio of POC/${\psi}$ ($7.08{\pm}4.27{\mu}molCdpm^{-1}$) in the sinking particles, was $5.9{\pm}3.9mmolCm^{-2}day^{-1}$. These fluxes were similar levels to those in the Weddell Sea during February and March 2008. Export ratios (ThE) relative to the primary production in the euphotic zone were in the range of 3-54% (av. 28%).

Effects of Seed Soaking of Kinetin on Alleviating Copper Toxicity during Germination in Rice (Kinetin 침종처리가 벼 발아중 구리 독성 경감에 미치는 영향)

  • The study was carried out to determine the proper concentration of plant growth regulator, kinetin on alleviating copper toxicity for two rice cultivars of seed germination. The results were as followings : Soaking treatment of kinetin 10$^{-3}$ M increased the germination rate of both cultivars, Ilpumbyeo and Hyangmibyeo 1 by 92% and 88% as compared with copper treatment (60ppm). But the soaking treatment effect of plant growth regulator, kinetin was not recognized at the kinetin 10$^{-4}$ M and 10$^{-5}$ M. Chlorophyll content of both rices was higher than that of Hyangmibyeo 1. Copper content of Ilpumbyeo was higher in leaf than in seed part. At the 3 days after treatment of copper 60ppm, both cultivars of treatment of kinetin 1O-3M showed the somewhat thin bands at the 35 and 40kDa compared with others. A new protein band pattern was only appeared to kinetin 1O-3M at approximately 54.4kDa(M. W) at the 7 days after treatment of copper 60ppm in llpumbyeo cultivar, SOD activity of copper 60ppm treatment increased in 3DAT, but there were no significant differences in 5 and 7DAT of two cultivars. Free proline contents of copper 60ppm treatment in llpumbyeo were remarkably increased about 4.996$\mu$M. In particular, free proline content of kinetin l0$^{-3}$ M in Ilpumbyeo was 5.008$\mu$M in 3DAT. In case of Hwangmibyeo 1, free proline content of copper 60ppm was 5.825$\mu$M compared with an untreated control showing 2.34l$\mu$M. The effects of kinetin treatment were recognized to promote the root growth and germination rate under copper toxicity(60ppm) condition in both cultivars.

Effect of Root Zone Temperature on the Growth and the Leaf Mineral Contents of Apple(Malus domestica Borkh) Trees (근권(根圈) 온도환경(溫度環境)이 사과나무의 생육(生育) 및 엽중(葉中) 무기성분함량(無機成分含量)에 미치는 영향)

  • This study was conducted to investigate the influence of root zone temperature on the growth of shoot and root and the mineral contents in leaf of 'Fuji/M26' apple tree. Shoot growth and enlargement of trunk girth increased linearly with increasing root zone temperature. Fresh and dry weight of root reached maximum at $35^{\circ}C$. Water content of root increased with rising root zone temperature. The chlorophyll content of leaves showed insignificant difference with root zone temperature. Leaf water potential was high at $35^{\circ}C$ at 15 day after treatment but 60 day after treatment this was decreased. The nitrogen content of the leaves was not different by root zone temperature whereas the phosphorus content of the leaves was increased at $30^{\circ}C$ in 1993 and at $25^{\circ}C$ in 1994. The potassium content of the leaves reached a maximum at $30^{\circ}C$ in 1993 and $25^{\circ}C$ in 1994. In 1994 the calcium content of the leaves was increased with rising root zone temperature and with lengthening duration of treatment but no such differences were found in 1993. The magnesium content of the leaves was highest at $25^{\circ}C$ in 1993 and at $20^{\circ}C$ in 1994. The nitrogen and potassium content of the roots were increased linearly with root zone temperature in 1993 and 1994 and the magnesium and phosphorus content of the roots were high at $35^{\circ}C$ in 1994 but no such differences were found in the calcium content of the roots.
