Cortex Mori Inhibits the CGG-specific IgE-Dependent Histamine Release

  • Cortex Mori, the root bark of mulbery tree has been used as an antiphlogistic, diuretic, and expectorant in herbal medicine. The purpose of this study is to evaluate chicken gamma globulin (CGG)-specific IgE-induced morphologic and functional changes in rat peritoneal mast cells (RPMC), and to determine whether Cortex Mori could inhibit the CGG-specific IgE-depeildent mast cell degranulation and histamine release from RPMC. Results are 1) the degranuration and histamine release from RPMC were not induced within 1 hour after addition of Cortex Mori alone, 2) the CGG and CGG-specific IgE-Induced degranulation from RPMC was observed within 10 minutes, 3) the histamine release from RPMC sensitised with CGG-specific IgE was induced by tile addition of CGG, 4) CGG-specific IgE-dependent degranulation rate in RPMC pretreated with Cortex Mori was significantly Inhibited, compared to that of control group without Cortex Mori pretreatment, and 5) the CGG-specific IgE-dependent histamine release from RPMC was significantly inhibited by pretreatment with Cortex Mori. These data suggest that Cortex Mori contains some substances with capabilities to inhibit CGG-specific IgE-dependent degranulation and histamine release from RPMC.

  • Recently the velocity stack inversion is having many attentions as an useful way to perform various seismic data processing. In order to be used in various seismic data processing, the inversion method used should have properties such as robustness to noise and parsimony of the velocity stack result. The IRLS (Iteratively Reweighted Least-Squares) method that minimizes ${L_1}-norm$ is the one used mostly. This paper introduce another method, CGG (Conjugate Guided Gradient) method, which can be used to achieve the same goal as the IRLS method does. The CGG method is a modified CG (Conjugate Gradient) method that minimizes ${L_1}-norm$. This paper explains the CGG method and compares the result of it with the one of IRSL methods. Testing on synthetic and real data demonstrates that CGG method can be used as an inversion method f3r minimizing various residual/model norms like IRLS methods.

The Effect of Cheonmagudeng-um gagam(CGG) on Spontaneous Hypertensive Rat(SHR) (천마구등음가감(天麻鉤藤飮加減)이 고혈압 유관인자 및 SHR 병태모델에 미치는 영향)

  • Objectives : This study was examined to investigate the effects of Cheonmagudeng-um gagam (CGG) extract on spontaneous hypertension. Methods : For the study of CGG, we divided rats into three groups. The normal group was Wister Kyoto rats (WKY). The control group was spontaneously hypertensive rats (SHR). The treatment group was SHR which were administered CGG extract (SHR-CGG). SHR-CGG were orally administered CGG extract that was diluted in distilled water at the various concentrations for 4 weeks (234.5 mg/kg) and SHR were orally administered the same dosage of plain distilled water as SHR-CGG. Then we measured anti-oxygen effects, ACE inhibitory activity, weight of heart and kidney, blood pressure, heart rate, plasma aldosterone, electrolyte, creatinine, uric acid, BUN, and observed the cortex of the cardiac muscle, kidney, and adrenal gland. Results : CGG increased DPPH scavenging activity and SOD similar activity depending on the concentration. CGG significantly decreased ROS, TNF-${\alpha}$, IL-6, IL-$1{\beta}$, heart weight, blood pressure, heart rate, aldosterone, and BUN in SHR. CGG increased ACE inhibition activity depending on the concentration. CGG inhibited the heart, kidney and adrenal gland tissue injury that is caused by hypertension. Conclusions : These results suggest that CGG is effective in treatment and prevention of hypertension.

Determination of Critical Generator Group Using Accelerating Power and Synchronizing Power Coefficient in the Transient Energy Function Method

  • This paper proposes an algorithm for determining critical generator lists using accelerating power and synchronizing power coefficient (SPC), and critical generator group (CGG) from CGG candidates, which is a combination of critical generators. The accurate determination of CGG provides a more accurate energy margin while providing system operator with information of possible unstable generator group. Classical transient energy function (TEF) method selects the critical generators with big corrected kinetic energy of each generator at the moment of fault removal. However, the generator with small acceleration after fault, that is, the generator with small corrected kinetic energy, is also likely to belong to CGG if the generator has small synchronizing power. The proposed algorithm has been verified to be effective compared with the classical TEF method. We utilized the power system of Korean Electric Power Corporation(KEPCO) as a test system.

MOLECULAR BIOLOGIC ANALYSIS OF FMR-1 GENE TRINUCLEOTIDE REPEATS IN AUTISTIC PATIENTS (자폐장애 환자에서 FMR-1 유전 삼염기 반복의 분자생물학적 분석)

  • Objectives:There has been a rapid expansion of studies aimed at elucidating the genetic basis of autistic disorder, especially it’ relationship to fragile-X syndrome. The detection of fragile X chromosome(Xq27.3) by cytogenetic analysis has revealed many difficulties in testing. Therefore, to explore the relationship between autistic disorder and fragile X syndrome, this study administered molecular biologic methods which examined an unstable CGG repeat within the fragile X mental retardation-1(FMR-1) gene. Methods:Ninety nine autistic children and eight normal control children were tested. The number of CGG repeats within FMR-1 gene was measured after amplification by PCR, and cytogenetic analysis was also carried out to detect fragile site Xq27.3. Southern blot hybridization, using StB12.3 and/or Pfxa3 probe, was done for the patients showing expansion of more than 50 CGG repeats (premutation). Results:All but two autistic patients had no expansion in CGG repeats by PCR and there was no significant statistical difference in number of CGG repeat in comparison with normal control. Two autistic patients, considered as premutation by PCR analysis, had no full mutation or premutation by Southern blot hybridization. All autistic children tested did not have any abnormal karyotype or fragile site Xq27.3. Conclusions:These results suggest that autistic patients may not have abnormality in FMR-1 gene or abnormal expansion in CGG repeat. In conclusion, fragile X syndrome may not be antecedent of autistic disorder.

Molecular diagnosis of fragile X syndrome in a female child (여아 환자에서의 취약 X 증후군의 분자유전학적 진단)

  • Purpose : Fragile X syndrome (FXS) is the most common heritable cause of cognitive impairment. FXS is caused by hyperexpansion and hypermethylation of a polymorphic CGG trinucleotide repeat in the 5' untranslated region of the fragile X mental retadation-1(FMR1) gene. Combination of Southern blotting and simple polymerase chain reaction(PCR) amplification of the FMR1 repeat region is commonly used for diagnosis in females. To give a definite diagnosis in a female child suspected of having FXS, we carried out the molecular diagnostic test for FXS using the recently developed Abbott Molecular Fragile X PCR Kit. Methods : The PCR amplification of the FMR1 repeat region was performed using the Abbott Mdecular Fragile X PCR Kit. The amplified products were analyzed by size-separate analysis on 1.5% agarose gels and by DNA fragment analysis using Gene scan. Results : Agarose gel and Gene scan analyses of PCR products of the FMR1 repeat region showed that the patient had two heterozygous alleles with a normal 30 repeats and full mutation of >200 repeats whereas her mother had two heterozygous alleles with the normal 30 repeats and premutation of 108 repeats, suggesting that the premutation of 108 repeats in her mother may have led to the full mutation of >200 repeats in the patient. Conclusion : We diagnosed FXS in a female patient using a simplified molecular diagnostic test. This commercially available diagnostic test for FXS, based on PCR, may be a suitable alternative or complement method to Southern blot analysis and PCR analysis and/or methylation specific(MS)-PCR analysis for the molecular diagnosis of FXS in both males and females.

Design Strategy for the Implementation of Cooperative Group Games in Motion Based Arcade Game System

  • Cooperative Group Games (CGG) emphasize participation, challenges and fun by cooperation among group members rather than competition. CGGs have been proven to be an efficient education method that teaches the value of cultivating cooperation skills, discipline, and sense of public order for tackling problems together through various types of interactions. When integrated with computer game technology, the general classic CGG can be reborn with new educational and entertaining aspects. To combine the joy of physical movement of group games and the richness of computer game contents, a motion based arcade CGG has been developed in this study, based on the original ideas and structures of classic off-line CGGs. While implementing the classic game concepts in arcade environment, various design attributes have been considered and applied, which were supposed to promote cooperative game play. Overall, the process of the implementation and test results of our four CGGs suggest several design strategies for effective arcade CGGs.

The Document of Museum of Chosen General Government and its systemic management of document (일제하 총독부 박물관 문서와 관리체계)

  • The Museum of Chosen General Government(MCGG) was a supreme organ to take charge of business affairs of historical remains Japanese imperial rule. The MCGG was established in Kyongbok Palace in 1915. The MCGG was changed the reorganization of the Chosen General Government(CGG) setups, the MCGG was to maintain the cultural assets, to excavate the ruins and to put on display the remains. However, the Japanese colonist took advantage of the MCGG for political purposes. They didn't use the MCGG to promote the research of Korea culture. Therefore, the MCGG was an organization to belong to the Department of Education of the CGG. In this reason, the MCGG produced the amount of public document to business affairs. Now, This document left in the Museum of Korea. We have seen the document to study the cultural policies and the cultural assets of the CGG. This document includes the abundant information for the historical remains and ruins at that time. Accordingly, this document will help to survey the archaeological research and historical research. In addition, this document will help to manage the cultural assets. What then is the advantage of this document? The first is to see the cultural policies of the CGG through this document. The Japanese colonist took advantage of Korea history, which was low-grade culture, to justify rule of the colony. Therefore, they needed collect Korean assets to verity their theory. The second is to see the administration system of the MCGG. Indeed, this document includes information of organization of the MCGG, the policies and the process of the MCGG. In substance, we can see the systemic proceedings of the MCGG. The third is to provide historical materials to the historian. This document has the persons to plan the colonial culture policy of the MCGG, and events to rule the Korea culture. Moreover, the document of the MCGG would help to inquire into the truthfulness of history and to get the national identity.

Fragile-X Mental Retardation: Molecular Diagnosis in Argentine Patients

  • Fragile-X-syndrome (FXS) is the most common type of inherited cognitive impairment. The underlying molecular alteration consists of a CGG-repeat amplification within the FMR-1 gene. The phenotype is only apparent once a threshold in the number of repeats has been exceeded (full mutation). The aim of this study was to characterize the FMR-1 CGG-repeat status in Argentine patients exhibiting mental retardation. A total of 330 blood samples from patients were analyzed by PCR and Southern blot analysis. Initially, DNA from 78 affected individuals were studied by PCR. Since this method is unable to detect high molecular weight alleles, however, we undertook a second approach using the Southern blotting technique to analyze the CGG repeat number and methylation status. Southern blot analysis showed an altered pattern in 14 out of 240 (6%) unrelated patients, with half of them presenting a mosaic pattern. Eight out of 17 families (47%) showed a (suggest deleting highlight). The characteristic FXS pattern was identified in 8/17 families (47%), and in 4 of these families 25% of the individuals presented with a mosaic model. The expansion from pre-mutation to full mutation was shown to occur both at the pre and post zygotic levels. The detection of FXS mutations has allowed us to offer more informed genetic counseling, prenatal diagnosis and reliable patient follow-up.

Modified Graphene Oxide-Based Adsorbents Toward Hybrid Membranes for Organic Dye Removal Application

  • In this study, the channels-contained hybrid membranes have been fabricated through the incorporation of glass fibers and GO sheets (GO/glass fibers, GG), or a mixture of chitosan/GO (CS/GO/glass fibers, CGG), as hybrid membranes using in organic dye removal. The material properties are well investigated the terms in the morphological, chemical, crystal, and thermal characterizations for verifying interactions in their formed structure. These hybrid membranes have been fitted well in pseudo-second order and Langmuir models that are associated with chemical adsorption and a monolayer approach, respectively. The highest adsorption ability of methylene blue and methyl orange reached 59.40 mg/g and 229.07 mg/g (GG); and 287.47 mg/g and 252.91 mg/g (CGG), which is more enhanced than that of previous GO-based other adsorbents. Moreover, the dye separation on these membranes could be favorable to superb sealing and trapping dye molecules from water instead of only the dye connection occurring on their surface, regarding the physically sieving effect. The membranes can also be reused within two and three adsorbing-desorbing cycles on the GG and CGG ones, respectively. These membranes can become future adsorbents to be applied for wastewater treatment due to their structural features.
