운동요법, 운동.행동수정요법이 중년 비만여성의 비만도, 혈중지질 및 자아존중감에 미치는 효과 (The Effects of Exercise Therapy and Exercise-Behavior Modification Therapy on Obesity, Blood Lipids, and Self-esteem of the Obese Middle-aged Women)

  • To examine the effect of the exercise therapy, and exercise-behavior modifi- cation therapy on obesity, blood lipids and self-esteem of the obese middle-aged women. Method: A total of 35 middle-aged women (BMI: over 30) were selected for this research. Walking at a 50% intensity was administered 4 days a week for 12 weeks, while the behavior modification therapy performed for 60~90 minutes per week for 12 weeks. Result: Body weight and BMI has significantly reduced in the case of EG and E BG. The result of comparing body weight between groups showed significant difference between EG and CG, and E BG and CG whereas BMI showed significant difference between EG and CG only. TC, TG, LDL-C, %TC/HDL-C have shown significant decrease in EG and E-BG, while HDL-C displayed significant increase in EG and E BG. And HDL-C showed significant decrease in CG. As for comparison between groups, significant difference was noted in EG and CG, and E BG and CG at TC, HDL-C, LDL-C, and in EG and CG at %TC/HDL-C. Self-esteem displayed significant increase in EG and E BG; however, there was no significant different in CG. As for comparison between groups, there was significant difference noted in E BG and CG only. Conclusion: The results showed that the exercise therapy and the exercise-behavior modification therapy were effective in changing obesity, blood lipids and self-esteem of the obese middle-aged women.

Construction of Antibodies for Detection and Diagnosis of Cucumber green mottle mosaic virus from Watermelon Plants

  • We immunized BALB/c mice with purified Cucumber green mottle mosaic virus isolate HY1 (CGMMV-HY1). Through the selection of positive clones that were grown on the HAT medium, four sensitive monoclonal clones (CG99-01, CG99-02, CG99-03, and CG99-04) were selected from 500 Hypoxanthine-guanine phosphoribosyltransferase positive hybridoma cells. Four sensitive clones of CGMMV-HYI were determined as IgM type of the subclass of mouse immunoglobulins Ig group. The titer of monoclonal antiserum against CGMMVHY1 was estimated 1:12,800 by the indirect ELISA. Although monoclonal antibodies (MAbs) from CG99-01 and from CG99-04 cross-reacted with Zucchini green mottle mosaic virus and Kyuri green mottle mosaic virus, MAb from the cell line CG99-03 was highly specific to CGMMV. No MAbs cross-reacted with Cucumber mosaic virus-Fny. Only CG99-04 reacted with Pepper mild mottle virus weakly and CG99-02 reacted with both CGMMV and KGMMV. CGMMV was detected from the rind of watermelon fruit by DAS-ELISA of CGMMV-HY1, but not from the flesh of watermelon. Average seed transmission rate of CGMMV in watermelon was $24\%$ from symptomatic watermelon collected from 5 regions of Gyeongnam province. CGMMV was detected by DAS-ELISA with specific MAb of CGMMVHY1 periodically from root stock, during the sequential process for nursery seedling in Haman. Necrotic spots on cotyledons of root stock seedling progressed to reveal the typical symptomatology on the primary leaves of scion upon grafting. Here, we have established MAb based ELISA system, which could accurately detect CGMMV from watermelon seeds, nursery seedlings, transplants and field samples from greenhouse or open out door field as well.

테니스 선수의 어깨 통증과 플랫서브 동작의 견갑골 움직임 분석 (Analysis shoulder pain of tennis players and the movement of the scapula in flat serve)

  • 본 연구는 테니스 1st 플랫서브 동작에서 어깨 통증 유무에 따른 견갑골의 움직임 차이를 확인하기 위해 테니스 선수 15명(남8, 여7)을 대상으로 3차원 영상분석시스템을 활용하여 견갑골의 앞쪽/뒤쪽 기울임, 아래쪽/위쪽 회전 및 안쪽/바깥쪽 회전에 대한 각속도와 모멘트를 산출하여 이를 집단 간 비교 분석을 하였다. 그 결과, 앞쪽/뒤쪽 기울임에 대한 최대 각속도는 실험군인 어깨통증집단(SPG)보다 대조군 집단(CG)이 빠른 경향을 보였으며, 팔로우 스로우 구간을 제외하고 모든 국면에서 안쪽/바깥쪽 회전 최대 각속도가 SPG가 CG보다 빠른 것으로 나타났다. 후기 코킹 구간에서의 앞쪽/뒤쪽 기울임 최대 모멘트가 SPG보다 CG가 더 크게 나타났고 통계적으로 유의한 차이를 보였다. 코킹 구간의 위쪽/아래쪽 회전 최대 모멘트 또한 SPG가 CG보다 작게 나타나면서 통계적으로 유의한 차이를 보였다. 안쪽/바깥쪽 회전 모멘트는 모든 국면에서 CG가 SPG보다 큰 것으로 나타났다.

운동선수들의 뇨 중 Human Chorionic Gonadotropin 분석 및 분석킷트 평가 (Assay of Human Chorionic Gonadotropin in Urine of Athletes and Evaluation of Assay Kit Performance)

  • Special attention has been paid to human chorionic gonadotropin (hCG) for athlete doping control because it stimulates the endogenous production of testosterone and epitestosterone without increasing the T/E ratio which is a doping indicator for the exogenous administration of testosterone. Even though the IOC banned the use of hCG, a detection method has not been decided upon since there are a variety of immunoassay kits available on the market. We evaluated three kits in terms of their performance characteristics. The assay value of the control sample varied depending on the kit, resulting in 198 mIU/ml for the MAIA kit, 172mIU/ml for the IRMA kit, and 143 mIU/ml for the MEIA kit. Considering the IOC inter-lab distribution of results(55-312 mIU/ml) using 27 different kits and the mean value (178$\pm$56 mIU/ml), all three kits are within the range of -15.8% - +5.6% of the mean value, which proves them useful for the hCG assay. The MEIA kit resulted in lower hCG values because it detects only intact hCG molecules, in contrast to the other two kits which detect intact hCG and -hCG together. However, it is suitable for screening purposes because of its advantage of being an automated system. When 123 urine samples of athletes were analyzed in 22 batches using this system, the variation of control values fell within $\pm$ 10% of the mean values, and an specimens tested negative with hCG values less than the detection limit of 2 mIU/ml.

손상된 말초신경의 재생에 미치는 전자침의 효과 (Effect of Electroacupuncture on the Regeneration of Injured Peripheral Nerve)

  • 손상된 말초신경이 정상상태로 회복되는데 침이 어떤 영향을 미치는지 확인하기 위하여 좌골신경을 인위적으로 손상시킨 후 침을 처리한 군 (AG)과 자연적으로 치유된 군 (CG)의 좌골신경내의 유발전위 증가율, 척수내 효소활성도의 변화 및 좌골신경의 미세구조를 비교.관찰하였다. 유발전위 증가율은 CG에서보다 AG에서 높았고, acid phosphatase 활성도는 1CG와 2AG에서 높게 나타났으나 시간이 경과함에 따라 정상상태로 회복되는 경향을 보였다. 손상된 좌골신경의 미세구조적 변화는 CG보다 AG에서 높은 회복율을 나타내었으며 단지 AG에서만 좌골신경 주위에 지방조직이 발달되어있는 것이 관찰되었다. 이상과 같이 손상된 말초신경이 정상상태로 회복되는 속도는 전자침을 처리한 군에서 높았으며, 특히 다른 치료기간보다 초기시기에서 침의 효과가 매우 높다는 것이 확인되었다.

  • 본 연구는 시뮬레이터 카메라 가상제어형 CG적용 영상제작시스템을 제공하는 것이 목적이다. 본 연구에서 제시하는 영상제작시스템은 촬영현장의 시뮬레이터 카메라와 CG적용 영상편집이 이루어지는 그래픽 툴의 가상카메라가 서로 동기화되도록 하고, 시뮬레이터 카메라의 위치, 움직임, 이동, 렌즈설정이 그래픽 툴에서 제공하는 카메라 제어 인터페이스를 통해 가상적으로 제어되도록 하여 그래픽 툴을 통해서 간편하고 용이하게 수행할 수 있는 시스템이다. 동서대학교 창작 애니메이션인 블레이즈 나이츠(Blaze Knights)를 사례로 하여, 시스템 적용전과 적용후의 진행율 및 작업양을 비교하여 연구결과의 우수성을 부각하였다. 연구결과에 따라 CG 아티스트의 작업효율이 증대되는 동시에 CG적용 영상의 장면 연출시 물리적 제약이 최소화되면서, 다양한 장면 연출의 가능 및 실사 촬영이후 영상 편집단계에서 요구될 수 있는 재촬영 분량도 최소화등 VFX업계에 기여도가 높을 것으로 기대한다.

Repeated Superovulation via PMSG/hCG Administration Induces 2-Cys Peroxiredoxins Expression and Overoxidation in the Reproductive Tracts of Female Mice

  • Superovulation induced by exogenous gonadotropin treatment (PMSG/hCG) increases the number of available oocytes in humans and animals. However, Superovulatory PMSG/hCG treatment is known to affect maternal environment, and these effects may result from PMSG/hCG treatment-induced oxidative stress. 2-Cys peroxiredoxins (2-Cys Prxs) act as antioxidant enzymes that protect cells from oxidative stress induced by various exogenous stimuli. Therefore, the objective of this study was to test the hypothesis that repeated PMSG/hCG treatment induces 2-Cys Prx expression and overoxidation in the reproductive tracts of female mice. Immunohistochemistry and western blotting analyses further demonstrated that, after PMSG/hCG treatment, the protein expression levels of 2-Cys Prxs increased most significantly in the ovaries, while that of Prx1 was most affected by PMSG/hCG stimulation in all tissues of the female reproductive tract. Repeated PMSG/hCG treatment eventually leads to 2-Cys Prxs overoxidation in all reproductive organs of female mice, and the abundance of the 2-Cys Prxs-$SO_{2/3}$ proteins reported here supports the hypothesis that repeated superovulation induces strong oxidative stress and damage to the female reproductive tract. Our data suggest that excessive oxidative stress caused by repeated PMSG/hCG stimulation increases 2-Cys Prxs expression and overoxidation in the female reproductive organs. Intracellular 2-Cys Prx therefore plays an important role in maintaining the reproductive organ environment of female mice upon exogenous gonadotropin treatment.

Structural and Biochemical Characterization of the Two Drosophila Low Molecular Weight-Protein Tyrosine Phosphatases DARP and Primo-1

  • The Drosophila genome contains four low molecular weight-protein tyrosine phosphatase (LMW-PTP) members: Primo-1, Primo-2, CG14297, and CG31469. The lack of intensive biochemical analysis has limited our understanding of these proteins. Primo-1 and CG31469 were previously classified as pseudophosphatases, but CG31469 was also suggested to be a putative protein arginine phosphatase. Herein, we present the crystal structures of CG31469 and Primo-1, which are the first Drosophila LMW-PTP structures. Structural analysis showed that the two proteins adopt the typical LMW-PTP fold and have a canonically arranged P-loop. Intriguingly, while Primo-1 is presumed to be a canonical LMW-PTP, CG31469 is unique as it contains a threonine residue at the fifth position of the P-loop motif instead of highly conserved isoleucine and a characteristically narrow active site pocket, which should facilitate the accommodation of phosphoarginine. Subsequent biochemical analysis revealed that Primo-1 and CG31469 are enzymatically active on phosphotyrosine and phosphoarginine, respectively, refuting their classification as pseudophosphatases. Collectively, we provide structural and biochemical data on two Drosophila proteins: Primo-1, the canonical LMW-PTP protein, and CG31469, the first investigated eukaryotic protein arginine phosphatase. We named CG31469 as DARP, which stands for Drosophila ARginine Phosphatase.
