A STUDY OF THE EFFECT OF ZIZYPHUS FRUCTUS EXTRACTS ON MORPHOLOGY & CHEMOTAXIS OF GINGIVAL FIBROBLAST & PERIODONTAL LIGAMENT CELLS (치은섬유아세포와 치은인대세포의 형태와 화학주성에 미치는 대조추출물의 효과에 관한 연구)

  • The most important object of periodontal treatment is the perfect regeneration of destructed periodontal tissue. The healing of periodontal lesion is affected by several cells & factors, which result in formation of long juntional epithelium, root resorption, bony ankylosis or connective tissue attachment. And ideal healing is enhanced by epithilial exclusion or periodontal ligament cell activation. In this investigation, I studied the effect of Zizyphus Fructus extract which enhances biologic activity& collagen synthesis, on the chemotaxis & cell nature. The cells were obtained from interdental area & middle third area of the freshly extracted teeth for the orthodontic purpose. And they were fully incubated in${\alpha}-MEM$ solution containing $100{\mu]g/ml$ penicillin & $100{\mu]g/ml$ streptomycin followed by 6 generation incubation. The test cells were collected by trypsin-EDTA & centrifuge in the fully incubated cells, counted by Hernacyotmeter, incbated $5{\times}10^5/ml$ cells for 24 hours, re-incubated 24 hours in media containing natural extract and photographed. The cells were incubated for 4 hours in 48 well microchemotaxis chamber bisecting upper & lower chamber by 8ug/m pore polycarbonate membrane coating 5mg/ml gelatin solution. The migrated cells in microscope were counted, which meaned cell chemotaxis activity. The study had shown that the morphology of cell was spindle-shaped as the control group, and the subextract test groups were not significantly different. In gingival fibroblasts, the chemotaxis effect of PDGF was statistically significant compared to control group. The Zizyphus Fructus extract was more or less enhanced chemotaxis effect and in $1{\mu}g/ml$ concentration the chemotaxis effect was slightly elevated compared with $10{\mu}g/ml$ concentration. But, among the subextracts, it was not significantly defferent. In PDL cells, the chemotaxis effect of PDGF in statistically significant, and the zizyphus Fructus extract had shown the enhanced effect. The effect was slightly higher in $1{\mu}g/ml$ concentration than 10g/ml concentration,and no significance among the subextracts.

Temperature dependent 2,3-dihydroxybenzoic acid production in Acinetobacter sp. B-W (Acinetobacter sp. B-W의 온도 의존적 2,3-dihydroxybenzoic acid 생산)

  • A soil microorganism producing iron chelator (siderophore) under low iron stress (up to $2{\mu}M$ of iron) was identified as Acinetobacter sp. B-W by 16S rDNA sequence analysis, biochemical-, physiological tests and morphological analysis using electron microscope. Catechol nature of siderophore was detected by Arnow test. Although optimal cell growth was identified at $36^{\circ}C$ in iron-limited media, significant quantities of siderophore were produced only at $28^{\circ}C$. Biosynthesis of siderophore was strongly inhibited by growth at $36^{\circ}C$. Production of siderophore was completely inhibited by $10{\mu}M\;FeCl_3$. Iron chelator produced from Acinetobacter sp. B-W was purified from supernatant using butanol extraction, Sephadex LH-20 column chromatography and HPLC. Purified sideropore was identified as 2,3-dihydroxybenzoic acid by HPLC, TLC and IR analysis.

Influence of fin partitioning of a Rayeigh-Bénard cavity at low Rayleigh numbers

  • This computational study examines the augmentation of classic 2-D Rayleigh-$B{\acute{e}}nard$ convection by the addition of periodically-spaced transverse fins. The fins are attached to the heated base of the cavity and serve to partition the cavity into 'units' with different aspect ratios. The respective impacts upon heat transfer of the fin configuration parameters - including spacing, height, thickness and thermal conductivity - are systematically examined through numerical simulations for a range of laminar Rayleigh numbers (0 < Ra < $2{\times}10^5$) and reported in terms of an average Nusselt number. The selection of the low Rayleigh number regime is linked to likely scenarios within aerospace applications (e.g. avionics cooling) where the cavity length scale and/or gravitational acceleration is small. The net heat transfer augmentation is found to result from a combination of competing fin effects, most of which are hydrodynamic in nature. Heat transfer enhancement of up to $1.2{\times}$ that for a Rayleigh-$B{\acute{e}}nard$ cavity without fins was found to occur under favorable fin configurations. Such configurations are generally characterized by short, thin fins with half-spacings somewhat less than the convection cell diameter from classic Rayleigh-$B{\acute{e}}nard$ theory. In contrast, for unfavorable configurations, it is found that the introduction of fins can result in a significant reduction in the heat transfer performance.

  • Perovskite type ceramic membranes which exhibit dual ion conduction (proton and oxygen ion conduction) can permeate water and can aid solving operational problems such as temperature gradient and carbon deposition associated with a working solid oxide fuel cell. From this point of view, it is crucial to reveal water transport mechanism and especially the nature of the surface sites that is necessary for water incorporation and evolution. $BaCe_{0.8}Y_{0.2}O_{3-{\alpha}}$ (BCY20) was used as a model proton and oxygen ion conducting membrane in this work. Four different catalytically modified membrane configurations were used for the investigations and water flux was measured as a function of temperature. In addition, CO was introduced to the permeate side in order to test the stability of membrane against water and $CO/CO_2$ and post operation analysis of used membranes were carried out. The results revealed that water incorporation occurs on any exposed electrolyte surface. However, the magnitude of water permeation changes depending on which membrane surface is catalytically modified. The platinum increases the water flux on the feed side whilst it decreases the flux on the permeate side. Water flux measurements suggest that platinum can block water permeation on the permeate side by reducing the access to the lattice oxygen in the surface layer.

Expression and characterization of the recombinant human galectin-3 (유전자 재조합 Human galectin-3의 발현과 성상)

  • Galectin-3 is known as an animal ${\beta}$-galactoside-binding lectin charicterized with S-type carbohydrate recognition domain. It plays a role in growth, adherence and movement of cells. It is, also, related to the cell transformation and metastasis of tumor cells. In this study, we have expressed and purified recombinant human galectin-3 (rHgalectin-3) using E coli system and asialofetuin affinity chromatography for the future development of monoclonal antibody to Hgalectin-3, which is suggested as the tumor marker for the gastric and thyroid gland cancers. Expressed protein was confirmed as the Hgalectin-3 by immunoblot with cross-reactive murine monoclonal antibody. Lectin activity and specificity of purified protein were, also, confirmed by the competitive inhibition with galectin-3 specific carbohydrate, lactose. Like physiological galectin-3, lectin activity of the molecule was not changed in nonreduced condition. Dimer formation, furthermore, was observed at high concentration of the protein even in the reduced condition, which is well known in physiological galectin-3. These results showed purified rHgalectin-3 has the same activity and molecular nature compared to the physiological galectin-3.

The Study on Degradation of Azo Dyes by Proteus sp. ST-1 (Proteus sp. ST-1에 의한 Azo계 색소의 분해에 관한 연구)

  • Direct Sky Blue-5B is an Azo dye known as general for staining of textile and leather, etc., and as materials which are difficult to be biodegraded in nature. The bacterium strain which could degrade direct Sky Blue-5B was isolated from activated sludge of dyeing factory and identified as Proteus sp. by experiment on morphological, cultural and biochemical characteristics, and so named Proteus sp. ST-1. The optimum condition of the strain for degradation of Sky Blue-5B were at about 35$^{\circ}C$ and PH 7~8. The strain had been capable of degradation with organic nitrogen effectively and had completely degraded 200mg/1 of the dye within 12hrs at 37$^{\circ}C$. The enzyme system related to degradation of Azo dye may be intracellular, and so degraded the dye after absorption into cell. The degradation products of Sky Blue-5B by Proton sp. 57-1 were analyzed by Gas Chromatography /Mass Spectrometry and Spectrophotomer, from this observation, it may be infered that the strain degraded the dye directly without any mediate.

The Effect of Different Type of Exercise on SOD, Neutrophils and T Lymphocytes (훈련방법의 차이가 SOD, Neutrophils 및 T세포에 미치는 영향)

  • Background: A physically active lifestyle and regular exercise training incurs many health benefits. One recently recognized benefit of regular moderate exercise is stress reduction and immune enhancement. Thus, a physical stress such as exercise may act at any number of points in the complex sequence of events collectively termed the immune response. Although exercise causes many propound changes in parameters of immune function, the nature and magnitude of such changes rely on several factors including the immune parameters of interest; type, intensity, and duration of exercise; fitness level or exercise history of the subject; environmental factors such as ambient temperature and humidity. Methods: This study was undertaken to investigate the effect of different type of exercise on superoxide dismutase (SOD), neutrophils, and T lymphocytes of Sprague-Dawley rats. Sprague-Dawley rats were randomly divided into three groups; a non-Trained group (NTG, n=6), a swim-Trained group (STG), and a treadmill-Trained group (TTG). The exercise regimen was designed in a treadmill (5 times/5 days/week) during 8-weeks for TTG, and swim training (5 times/5 days/week) during 8-weeks for STG, and the volume of exercise training was the same in both groups. Results: 8 weeks of regular swim and treadmill training significantly increased liver SOD concentration however, muscle SOD concentration was not statistically significant. In the level of neutrophils, TTG and STG showed significant difference, compared to NTG. TTG was the highest level of neutrophils. In the level of immune cell counts, there was significant difference among TTG, STG, and NTG both in the spleen and thymus. Conculsion: In conclusion, it can be stated that eight weeks swim and treadmill exercise training has beneficial effect in improving immune response and antioxidant defence capacity by augmenting immune cells and SOD activities of SD rats.

Mechanical Properties and cytotoxicity of nitrided 630 Stainless by ion nitriding (플라즈마 이온질화에 따른 STS 630의 기계적 특성과 세포독성에 관한 융합적 연구)

  • In this study, we have examined the possibility to improve the material properties and biocompatibility of the 630 stainless steel for the application of medical instruments. The evaluation of mechanical properties and biocompatibility of the 630 stainless steel were studied by aging, nitriding, SEM measurement, Vicker's hardness, tensile strength and MTT cytotoxicity using IGF cells, respectively. The results showed that the tensile strength and Vicker's hardness of 630 stainless steel was increased with aging and ion nitriding time. The cytotoxicities of the 630 stainless decreased compared with the 420 stainless on MTT cytotoxicity using IGF cells. Ion nitriding of 630 stainless led to an enhanced application of medical instruments and biocompatibility performance.

GATA-3 is a Key Factor for Th1/Th2 Balance Regulation by Myristicin in a Murine Model of Asthma (Myristicin이 Ovalbumin으로 유도한 천식 생쥐모델에서 Th1/Th2 Balance를 조절하는 GATA-3에 미치는 효과)

  • Myristicin, l-allyl-3,4-methylenedioxy-5-methoxybenzene, was one of the major essential oils of nutmeg. However, its anti-allergic effect in the Th1/Th2 immune response was poorly understood. Recently, it was shown that T-bet and GATA-3 was master Th1 and Th2 regulatory transcription factors. In this study, we have attempted to determine whether myristicin regulates Th1/Th2 cytokine production, T-bet and GATA-3 gene expression in ovalbumin (OVA)-induced asthma model mice. Myristicin reduced levels of IL-4, Th2 cytokine production in OVA-sensitized and challenged mice. In the other side, it increased $IFN-{\gamma}$, Th1 cytokine production in myristicin administrated mice. We also examined to ascertain whether myristicin could influence eosinophil peroxidase (EPO) activity. After being sensitized and challenged with ovalbumin (OVA) showed typical asthmatic reactions. These reactions included an increase in the number of eosinophils in bronchoalveolar lavage fluid, an increase in inflammatory cell infiltration into the lung tissue around blood vessels and airways, and the development of airway hyper-responsiveness (AHR). The administration of myristicin before the last airway OVA challenge resulted in a significant inhibition of all asthmatic reactions. Accordingly, these findings provide new insight into the immunopharmacological role of myristicin in terms of its effects in a murine model of asthma.

Antimicrobial Activity of a Bacteriocin Produced by Enterococcus faecalis KT11 against Some Pathogens and Antibiotic-Resistant Bacteria

  • In this study, the antimicrobial activity of a bacteriocin produced by Enterococcus faecalis KT11, isolated from traditional Kargı Tulum cheese, was determined, and bacteriocin KT11 was partially characterized. The results showed that bacteriocin KT11 was antagonistically effective against various Gram-positive and Gram-negative test bacteria, including vancomycin- and/or methicillin-resistant bacteria. The activity of bacteriocin KT11 was completely abolished after treatment with proteolytic enzymes (proteinase K, ${\alpha}$-chymotrypsin, protease and trypsin), which demonstrates the proteinaceous nature of this bacteriocin. Additionally, bacteriocin KT11 remained stable at pH values ranging from 2 to 11 and after autoclaving at $121^{\circ}C$ for 30 min. In addition, the activity of bacteriocin KT11 was stable after treatment with several surfactants (EDTA, SDS, Triton X-100, Tween 80 and urea) and organic solvents (chloroform, propanol, methanol, ethyl alcohol, acetone, hexane and ethyl ether). Cell-free supernatant of E. faecalis KT11 was subjected to ammonium sulfate precipitation and then desalted by using a 3.5-kDa cut-off dialysis membrane. The bacteriocin activity was determined to be 711 AU/mL in the dialysate. After tricine-SDS-PAGE analysis, one peptide band, which had a molecular weight of ~3.5 kDa, exhibited antimicrobial activity. Because the bacteriocin KT11, isolated from E. faecalis KT11, exhibits a broad antimicrobial spectrum, heat stability and stability over a wide pH range, this bacteriocin can be used as a potential bio-preservative in foods. Additionally, bacteriocin KT11 alone or in combination with conventional antibiotics may provide a therapeutic option for the treatment of multidrug-resistant clinical pathogens after further in vivo studies.
