Organic Solvent and pH Induced Alteration of Product Specificity of CGTase

  • Cyclodextrin glucanotransferase [CGTase, E.C.2.4.1.19] is an extracellular enzyme, which catalyzes he formation of ${\alpha}$-, ${\beta}$-, ${\gamma}$- CDs from starch. Their proportions of formations depend on enzyme sources and reaction conditions. To understand what determines the product specificity of CGTases, we examined the alteration of product specificity of CGTase from Bacillus macerans by organic solvent sand pH. At acidic pH range less than pH 6 where the enzyme was unstable, the ratio of ${\alpha}$-/ ${\beta}$-CD production was increased 4 times more than that at neutral pH range. As we increased the concentration of 2-butanol, ${\alpha}$-/ ${\beta}$-CD ratio was proportionally increased but / ratio remained constant. The ${\alpha}$-/ ${\beta}$-CD ratio of products was increased in the reaction media which yielded low products.

Does Internet Increase the Market Efficiency? (인터넷이 시장의 효율성을 증가시키는가?)

  • The Internet is taking up every single life of us with a huge speed of growth. If the ideal market called 'perfect competition' in economics will ever come true in this digital world, the sellers won't be able to have monopolistic profits above the marginal cost any more, letting the resource allocation much more efficient. This paper attempts to test whether this theory is true in e-market as well through observing price differences between online and offline retailers of extremely homogeneous products, CDs. Since most results from previous research were supporting the inefficiency of e-market in price level, price adjustment, and especially price dispersion, this article designed the research methodology most carefully. The results of pervious works are partly due to the immaturity of the Internet market or due to the uniqueness of the American CD market, where oligopolistic market players are significantly dominant. The analysis of price data of 20 titles from 20 retailers for five weeks supports that online market is more efficient than offline market with statistical significance in all the three dimensions. We could conclude that the e-market is going much more efficient at least compared to the offline market and the more would it be unless the sellers resist and prevent comparison-shopping.

Complete genome sequence of Salmonella enterica strain K_SA184, multidrug resistance bacterium isolated from lamb (Ovis aries)

  • Salmonella enterica is a representative foodborne pathogen in the world. The S. enterica strain K_SA184 was isolated from the lamb (Ovis aries), which was collected from a local traditional market in South Korea. In this study, the S. enterica strain K_SA184 was sequenced using PacBio RS II and Illumina NextSeq 500 platforms. The final complete genome of the S. enterica strain K_SA184 consist of one circular chromosome (4,725,087 bp) with 52.3% of guanine + cytosine (G + C) content, 4,363 of coding sequence (CDS), 85 of tRNA, and 22 of rRNA genes. The S. enterica strain K_SA184 genome includes encoding virulence genes, such as Type III secretion systems and multidrug resistance related genes.

  • In eukaryotic cells, a key RNA processing step to generate mature mRNA is the coupled reaction for cleavage and polyadenylation (CPA) at the 3' end of individual transcripts. Many transcripts are alternatively polyadenylated (APA) to produce mRNAs with different 3' ends that may either alter protein coding sequence (CDS-APA) or create different lengths of 3'UTR (tandem-APA). As the CPA reaction is intimately associated with transcriptional termination, it has been widely assumed that APA is regulated cotranscriptionally. Isoforms terminated at different regions may have distinct RNA stability under different conditions, thus altering the ratio of APA isoforms. Such differential impacts on different isoforms have been considered as post-transcriptional APA, but strictly speaking, this can only be considered "apparent" APA, as the choice is not made during the CPA reaction. Interestingly, a recent study reveals sequential APA as a new mechanism for post-transcriptional APA. This minireview will focus on this new mechanism to provide insights into various documented regulatory paradigms.

NMR Spectroscopic Analysis on the Chiral Recognition of Noradrenaline by β-Cyclodextrin ( β-CD) and Carboxymethyl- β-cyclodextrin (CM- β-CD)

  • ${\beta}$-CD and CM- ${\beta}$-CD as chiral NMR shift agents were used to resolve the enantiomers of noradrenaline (NA). The stoichiometry of each complex formed between the CDs and the enantiomers of NA was found to be 1 : 1 through the continuous variation plots. The binding constants (K) of the complexes were determined from $^1H$ NMR titration curves. This result indicated that both ${\beta}$-CD and CM- ${\beta}$-CD formed the complexes with the S(+)-NA more preferentially than its R(-)-enantiomer. The K values for the complexes with ${\beta}$-CD ($K_{S(+)}$ = 537 $M^{-1}$ and $K_{R(-)}$ = 516 $M^{-1}$ was larger than those with CM- ${\beta}$-CD ($K_{S(+)}$ = 435 $M^{-1}$ and $K_{R(-)}$ = 313 $M^{-1}$), however, enantioselectivity (${\alpha}$) of S(+)- and R(-)-NA to CM- ${\beta}$-CD ( ${\alpha}$ = 1.38) was larger than that to ${\beta}$-CD ( ${\alpha}$ = 1.04), indicating that CM- ${\beta}$-CD was the better chiral NMR solvating agents for the recognition of the enantiomers of NA. Two dimensional rotating frame nuclear Overhauser enhancement spectroscopy (ROESY) experiments were also performed to explain the binding properties in terms of spatial fitting of the NA molecule into the macrocyclic cavities.

Changes of Microbial Community Depending on Different Dissolved Oxygen in Biological Nitrogen Removal Process (생물학적 질소제거 공정에서 용존산소변화에 따른 미생물의 군집변화)

  • PCR-DGGE method was applied to analyze changes of microbial community in simultaneous nitrification and denitrification (SND) bioreactor with various DO concentrations. In the analysis of eubacterial community, band profiles of DGGE were similar with 2 or 1 mg/L DO concentrations in the reactor. Experimental results led to 16 different bacteria being identified, including 5 dominant strains(3 strains of Uncultured Bacterium, 1 strains of Bacillus, 1 strains of Uncultured Bacteroidetes). DGGE results at 0.5 mg/L DO concentration led to 12 strains being identified, including 7 dominant strains(5 strains of Uncultured Bacterium, 2 strains of Zoogloea sp.). DGGE results at 0.1 mg/L DO concentration led to 11 strains being identified, including 3 dominant strains(1 strains of Uncultured Bacterium, 2 strains of Zoogloea sp.). In DGGE band profiles of $\beta$-AOB($\beta$-Ammonia Oxidizing Bacteria), only one band was observed. This band had 97% similarity with Nitrosomonas sp. done DNB Y20. This band was clearly observed at the 2, 1 and 0.5 mg/L DO concentrations, while the brightness of the band at 0.1 mg/L DO concentration was mostly dimmed. In DGGE band profiles of denitrification process, 5 bands(3 strains of Uncultured organism containing nirS, 2 strains of Uncultured organism containing nirK) were observed. Among those bands, the brightness of one band was gradually increased at the lower DO concentrations. This band has 86% identity with Uncultured organism clone eS1 cd1 nirS gene, partial cds. Based on this result, it could be concluded that Uncultured organism clone eS1 cd1 nirS gene, partial cds is a predominant microorganism in the denitrification process.

  • Cells of Deinococcus puniceus $DY1^T$ are Gram-positive, coccus-shaped, and crimson color-pigmented. Strain $DY1^T$ was isolated from soil irradiated with 5 kGy gamma radiation and showed resistance to UVC and gamma radiation. In this study, we report the complete genome sequence of a bacterium Deinococcus puniceus $DY1^T$ is consist of circular chromosome comprised of 2,971,983 bp, with the G + C content of 62.5%. The complete genome sequence was obtained using the PacBio RS II platform, it included 2,617 coding sequences (CDs), 2,762 genes, and 88 pseudogene.

Solubility and Physicochemical Stability of Ondansetron Hydrochloride in Various Vehicles (용제 중 염산온단세트론의 용해성 및 안정성)

  • The solubility and stability of ondansetron hydrochloride (OS) in various vehicles were determined. The effect of cyclodextrins (CD) on the solubility of OS in water was determined by equilibrium solubility method. The solubility of OS at $32^{\circ}C$ increased in the rank order of isopropyl myristate (IPM) < propylene glycol laurate (PGL) ${\ll}$ propylene glycol monolaurate < propylene glycol monocaprylate (PGMC) < poly(ethylene glycol) 400 < diethylene glycol mono ethyl ether (DGME) < ethanol < poly(ethylene glycol) 300 < water (36.1 mg/ml) ${\ll}$ propylene glycol (PG) (283 mg/ml). The addition of PG or DGME to non-aqueous vehicles such as IPM, PGL and PGMC markedly increased the solubility of OS. The addition of CDs in water increased the solubility. Apparent stability constant for the CD complexation with OS was calculated to be $25.5\;M^{-1}$ for $2-hydroxypropyl-{\beta}-CD\;(2HP{\beta}CD)$. Twenty mM ${\beta}-CD$, 69.4 mM sulfobutyl ether ${\beta}-CD$ and 115.4 mM $2HP{\beta}CD$ increased the aqueous solubilty of OS 1.27, 2.18 and 1.85 times, respectively. Molecular Cloning and Enzymatic Characterization of Cyclomaltodextrinase from Hyperthermophilic Archaeon Thermococcus sp. CL1

  • Genome organization near cyclomaltodextrinases (CDases) was analyzed and compared for four different hyperthermophilic archaea: Thermococcus, Pyrococcus, Staphylothermus, and Thermofilum. A gene (CL1_0884) encoding a putative CDase from Thermococcus sp. CL1 (tccd) was cloned and expressed in Escherichia coli. TcCD was confirmed to be highly thermostable, with optimal activity at $85^{\circ}C$. The melting temperature of TcCD was determined to be $93^{\circ}C$ by both differential scanning calorimetry and differential scanning fluorimetry. A size-exclusion chromatography experiment showed that TcCD exists as a monomer. TcCD preferentially hydrolyzed ${\alpha}$-cyclodextrin (${\alpha}$-CD), and at the initial stage catalyzed a ring-opening reaction by cleaving one ${\alpha}$-1,4-glycosidic linkage of the CD ring to produce the corresponding single maltooligosaccharide. Furthermore, TcCD could hydrolyze branched CDs (G1-${\alpha}$-CD, G1-${\beta}$-CD, and G2-${\beta}$-CD) to yield significant amounts (45%, 40%, and 46%) of isomaltooligosaccharides (panose and $6^2$-${\alpha}$-maltosylmaltose) in addition to glucose and maltose. This enzyme is one of the most thermostable maltogenic amylases reported, and might be of potential value in the production of isomaltooligosaccharides in the food industry.

Identification of DEA Determinant Input-Output Variables : an Illustration for Evaluating the Efficiency of Government-Sponsored R&D Projects (DEA 효율성을 결정하는 입력-출력변수 식별 : 정부지원 R&D 과제 효율성 평가를 위한 실례)

  • In this study, determinant input-output variables are identified for calculating Data Envelopment Analysis (DEA) efficiency scores relating to evaluating the efficiency of government-sponsored research and development (R&D) projects. In particular, this study proposes a systematic framework of design and analysis of experiments, called "all possible DEAs", for pinpointing DEA determinant input-output variables. In addition to correlation analyses, two modified measures of time series analysis are developed in order to check the similarities between a DEA complete data structure (CDS) versus the rest of incomplete data structures (IDSs). In this empirical analysis, a few DEA determinant input-output variables are found to be associated with a typical public R&D performance evaluation logic model, especially oriented to a mid- and long-term performance perspective. Among four variables, only two determinants are identified : "R&D manpower" ($x_2$) and "Sales revenue" ($y_1$). However, it should be pointed out that the input variable "R&D funds" ($x_1$) is insignificant for calculating DEA efficiency score even if it is a critical input for measuring efficiency of a government-sonsored R&D project from a practical point of view a priori. In this context, if practitioners' top priority is to see the efficiency between "R&D funds" ($x_1$) and "Sales revenue" ($y_1$), the DEA efficiency score cannot properly meet their expectations. Therefore, meticulous attention is required when using the DEA application for public R&D performance evaluation, considering that discrepancies can occur between practitioners' expectations and DEA efficiency scores.
