Heat Inducible Expression of the CDC70 Gene Under the Control of Heat Shock Element in Saccharomyces Cerevisiae

  • In order to express the CDC70 gene of Saccharomyces cerevisiae by heat shock, we have designed heat inducibe hybrid promoters using the Drosophila melanogaster heat shock elements (HSEs). A 220 bp-long upstream fragment of the D. melanogaster hsp70 gene comprised of four HSEs was placed upstream of the putative proximal TATA box of the CDC70 gene. Hybrid promoters containing different fusion joints were tested for their ability to drive the CDC70 gene expression by heat shock. The results showed that the HSEs of D. melanogaster conferred the heat-induced CDC70 gene expression, but the heat inducibility was much lower than that in D. melanogaster.

Genes involved in mating processes of saccharomyces cerevisiae (효모의 접합과정에 관여하는 유전자의 연구)

  • In order to elucidate and characterize the signal transduction pathway(s) whereby yeast cells respond to mating pheromone, we have isolated mutants which are able to conjugate in the absence of the alpha-factor receptor. Sixty-one suppressors of a ste2-deletion mutation which also confer a ts conditional "start" arrest phenotypw have been subjected to genetic analysis. The mutants could be assigned to three complementation groups designated CDC70, CDC72 and CDC73, which are unlinked to each other as well as to the previously identified start genes. Quantitation of mating ability of the cdc70, cdc72 and cdc73 mutations in a ste2-deletion background gives levels ranging from 0.1% to 0.3% of wild type, depending on the allele and the gene. The results indicate that the signals from mating pheromone might be mediated by the CDC70, CDC72 and CDC73 products. products.

  • The gene CDC70 encoding the${\alpha}-subunit$ of G protein has been known to be a component involved in mating pheromone signalling in the yeast, Saccharomyces cerevisiae. To isolate mutations of the genes involved in the signal transduction, Saccharomyces cerevisiae the strain bearing the cdc70-5 mutation was mutagenized to be forced to recover the ability of colony-formation at restrictive temperature, which means the new mutation can suppress the temperature sensitivity of the cdc70-5 phenotypes. Among these suppressors, $sir^-$ and $mat{\alpha}2^{-}$ mutations are excluded because of no relationship to signal transducer. And the selected suppressors were analyzed for the linkage relationships by the tetrad analysis. Out of fifteen suppressors isolated, twelve were classified into four linkage groups, designated as sga1, sga2, sga3, sga4 by the tetrad analysis. The other three genes were determined for the linkage.

  • The signal transduction pathway leading to the activation of the p70s6k plays an important role in the progression of cells from G0/G1 to S phase of the cell cycle but remains incompletely characterized. We investigated the role of the Rho family G protein Rac1 in H2O2-mediated p70s6k activation. Transient expression of a dominant negative mutants of the small GTP-binding proteins Rac1 (Rac1N17) and Cdc42(Cdc42N17) showed reduced levels of slower migration on Western blots of one-dimensional SDS-PAGE in p70s6k and ERK1/2 by PDFG stimulation.   • We have investigated the action of sodium nitrite on the growth and morphologic changes of T uaginolis and on the treatment of subcutaneous abscess by trichomonad in mice. Sodium nitrite inhibited the growth of metronidazole-sensitive KT9 isolate and metronidazole-resistant CDC85 strain of T vcsinalis as concentration of 6 mM and 10 mM respectively Intraperitoneal injection of sodium nitrite (70 ㎍, 100 ㎍, 130 ㎍/g body weight) did not reduce the size of abscess produced by subcutaneous inoculation of T uasinnlis in mice. T uosinnlis, treated with sodium nitrite at concentration giving about 50% inhibition of growth, showed fissures, many vacuoles and electron-translucent zone in the cytoplasm by transmission electron microscopy. In the case of CDC85 treated with 9 mM sodium nitrite, hydrogenosomal matrical change , destruction of hydrogenosomal membrane, autophagic vacuoles, disappearance of Golgi complex and polysome were notably observed . With above results, it is assumed that sodium nitrite inhibits the growth of metronidazole-sensitive and - resistant strains of T. ucsinalis and induces the morphological changes of 7 uusinalis although it does not affect in reducing of abscess size by vagiginalis in mice.

  • With increasing environmental demands in surface treatment of steel sheets, the passivation layers containing hexavalent chromium $(Cr^{+6})$ are being replaced by non-chromium or trivalent chromium compounds. After review on the various types of inorganic compounds, the zirconates was chosen as the candidate for alternative to sodium dichromate in the aspect of its barrier properties with excellent adhesion to organics. The ammonium zirconium carbonate (AZC) and sodium hexafluorozirconate (SFZ) could be reach $70-80\%$ level of CDC (cathodic dichromate) treatment by their single applications. But high porosity in the AZC layer and poor electrical conductivity of SFZ solution limit the single application of zirconate. Mixed composition of zirconates to compensate their inferiorities or incorporation of organic compounds to seal the porosity seems to be inevitable to match up the target level of Cr-free passivation of tinplate.

Study on Rector Dynamic Response by Cross Correlation Method (상호상관함수법에 의한 원자로 동특성에 관한 연구)

  • The reactor noise analysis was performed by the cross correlation method using two detectors to determine the dynamic stability and the system parameter of TRIGA Mark-II reactor under critical and subcritical conditions. The a values turned out to be 46.67 and 70.04 respectively at zero power and full power under critical condition, while 79.47 and 97.59 respectively at the safety rod dropping and the regulating rod dropping under subcritical condition. Prompt neutron life time of TRIGA Mark-II reactor measured 107 and 160 $\mu$sec, and shut down margin was -10.03$\times$10-4 at the safety rod dropping and -29-43$\times$10-4 at the regulating rod dropping CDC 3100/MSOS digital computer, HITACHI 505 Analog computer, and preamplifier, bandpass filter, FM modulator or demodulator designed for this specific purpose, were employed for the present investigation.

Effects of FBS(Fetal Bovine Serum) and pFF(Porcine Follicular Fluid) on In Vitro Maturation and Development of Porcine Parthenogenetic and Nuclear Transfer Embryos

  • In this study, in vitro maturation system using fetal bovine serum (FBS) or porcine follicular fluid (pFF) was investigated to produce comparable oocytes to those derived from in vivo. Control group of oocytes was cultured in TCM 199 supplemented with 0.1% polyvinyl alcohol (PVA). Other three groups of oocytes were cultured in TCM 199 supplemented with 10% FBS, 10% pFF or 5% FBS + 5% pFF, respectively. After 44 h maturation, oocytes with the first polar body were activated with two electric pulses (DC) of 1.2 kv/cm for 30 ${\mu}sec$. Also, matured oocytes of four groups were reconstructed and fused. Reconstructed embryos were cultured in PZM-3 under 5% $CO_2$ in air at $38.5^{\circ}C$ for 6 days. The oocytes matured in the medium supplemented with FBS or/and pFF showed significantly higher maturation rates (64.0 vs. 73.9 to 85.2%). In PA embryos, cleavage rates (89.7 vs. 77.1 to 86.6%) and blastocysts rates (30.0 vs. 16.2 to 26.2%) were significantly higher in pFF group (p<0.05). In NT embryos, there was no difference among treatments in cleavage rate, but the blastocyst rates (28.5 vs. 15.5 to 24.6%) were significantly higher in pFF group (p<0.05). The apoptosis rate was significantly higher (p<0.05) in the control than other groups (10.8 vs. 4.9 to 8.2% for PA, 3.1 vs. 0.5 to 1.3% for NT). In order to select the comparable oocyte to in vivo oocytes, each group of oocytes was stained with Brilliant cresyl blue (BCB) after 42h maturation. The matured oocytes were separated according to color of cytoplasm; stained group (BCB+) and unstained group (BCB-). The oocytes matured in the presence of FBS or/and pFF showed significantly higher staining rates (70.3 to 72.7 vs. 35.1%) (p<0.05). To verify the fact that the supplementation of FBS or/and pFF can increase the maturation rates, cdc2 kinase activity, the catalytic subunit of MPF, was determined. The cdc2 kinase activity of the oocytes matured in the medium supplemented with FBS or/and pFF was significantly higher than control group (6.7 to 9.3 vs. 3.8). In conclusion, the supplementation of FBS or/and pFF can support in vitro maturation rate of porcine oocytes through the increment of cdc2 kinase activity level in the cytoplasm.

세포능식촉진인자(MPF)의 특이적 억제제 생산 토양균주의 검색 및 억제제의 분리와 특성 규명에 관한 연구

  • 본 연구에서는 토양균이 생성하는 2차 대사산물로부터 세포증식 촉진인자 (Maturation Promoting Factor. MPF)인 cdc2/cdk2-cyclin의 복합체를 특이적으로 억제하는 물질을 분리하고 그 물질의 생리활성을 조사하였다. 토양으로부터 순수분리된 300여개의 토양균에서 생성된 배양액을 취하여 MPF의 특이적인 기질인 합성 peptide (CSH103 :HATPPKKKRK)를 사용하여 인산화 활성을 측정하였다. 그중 MPF 활성 억제능이 90% 이상인 19개의 균주를 1차적으로 선정한 후 각각에 대하여 열/pH에 대한 안정성, 각종 용매에 대한 추출성 등 이화학적 성질을 규명하였으며 이중 균주 LPL 931로부터 MPF 활성 억제제를 분리하고자 하였다. 예비실험의 결과로부터 토양균 LPL 931을 대량배양하여 열처리하고, 비이온성 수지인Amberlite XAD-2에 결합시키고 70% acetone으로 용출시켰다. 이 추출물로부터 ethylacetate와 n-butanol을 사용하여 MPF 억제 활성물질을 추출하였다. 이 추출액을 실리카겔 관 크로마토그래피, 분취 TLC, 분취 HPL를 하여 MPF 활성 억제 분획을 분리하였다.

Cost Effectiveness of Interventions to Promote Screening for Colorectal Cancer: A Randomized Trial

  • Objectives: Screening for colorectal cancer is considered cost effective, but is underutilized in the U.S. Information on the efficiency of "tailored interventions" to promote colorectal cancer screening in primary care settings is limited. The paper reports the results of a cost effectiveness analysis that compared a survey-only control group to a Centers for Disease Control (CDC) web-based intervention (screen for life) and to a tailored interactive computer-based intervention. Methods: A randomized controlled trial of people 50 and over, was conducted to test the interventions. The sample was 1224 partcipants 50-70 years of age, recruited from Kelsey-Seybold Clinic, a large multi-specialty clinic in Houston, Texas. Screening status was obtained by medical chart review after a 12-month follow-up period. An "intention to treat" analysis and micro costing from the patient and provider perspectives were used to estimate the costs and effects. Analysis of statistical uncertainty was conducted using nonparametric bootstrapping. Results: The estimated cost of implementing the web-based intervention was $40 per person and the cost of the tailored intervention was $45 per person. The additional cost per person screened for the web-based intervention compared to no intervention was $2602 and the tailored intervention was no more effective than the web-based strategy. Conclusions: The tailored intervention was less cost-effective than the web-based intervention for colorectal cancer screening promotion. The web-based intervention was less cost-effective than previous studies of in-reach colorectal cancer screening promotion. Researchers need to continue developing and evaluating the effectiveness and costeffectiveness of interventions to increase colorectal cancer screening.
