Transition over 35 Years in the Incidence Rates of Primary Central Nervous System Tumors in Shanghai, China and Histological Subtyping Based on a Single Center Experience Spanning 60 Years

  • Background: Only few epidemiological data on primary central nervous system (CNS) tumors in Shanghai have been reported. Methods: All cases of primary CNS tumors that were registered at Center for Disease Control and Prevention (CDC) were collected (1973-2007: urban Shanghai; 2003-2007: whole Shanghai city). Trends were analyzed using joinpoint analysis and rates were stratified by age, gender and region. Histological data were collected from both CDC and Huashan Hospital. Results: From 1973 to 2007, the five-year average incidence rate in urban Shanghai increased in both genders, especially in the elderly population. Joinpoint analysis showed the age-adjusted incidence rate for males increased first but then plateaued, whilst rates for females continued increasing over the 35 years. For the five-year status quo (2003-2007), rural had a higher age-adjusted incidence rate than urban populations, and females higher than males, especially those with advanced age. According to CDC (2003-2007) and Huashan Hospital (1951-2011), the two most common histological subtypes were neuroepithelial tumors (with male predominance) and meningiomas (with female predominance). Conclusions: In Shanghai, a steadily increased incidence rate of primary CNS tumors was observed in general, and in the elderly and female population in particular.

  • 이어진 블록계획 중에서 대칭 균형된 불완비 블록계획(Symmetrical Balanced Incomplete Block Design : SBIBD)을 이용하여 n-ary를 블록 완전이면교배(Complete Diallel Cross : CDC)계획을 설계하였다. 처리 수와 반복 수가 고정된 경우, 이렇게 설계된 계획이 균형된 불완비 블록계획을 이용해서 설계한 계획들 중에서 가장 효율이 높은 계획임을 보인다.

  • It is known that septin gene encodes the filament in Saccharomyces cerevisiae and it has importants roles in bud formation and cytokinesis. Four septin genes have been cloned in S. cerevisiae and it was found in Drosophila melanogaster and mouse. In this study, we cloned the septin gene in Schizosaccahromyces pombe by use of PCR technique. The septin gene in S. pombe has an 1,143 bp open reading frame and encodes a protein of 380 amino acids with a molecular weight of 42 kd. Comparison of the predicted amano acid sequences between the septin gene in S. pombe and CDC12 gene in S. cerevisiae reveals the 51.8% of simility.

  • Background: Therapeutic approaches using monoclonal antibodies (mAbs) against complement regulatory proteins (CRPs:i.e.,CD46,CD55 and CD59) have been reported for adjuvant cancer therapy. In this study, we generated a recombinant 1E8 single-chain anti-CD59 antibody (scFv-Fc) and tested anti-cancer effect.by using complement dependent cytotoxicity (CDC). Methods: We isolated mRNA from 1E8 hybridoma cells and amplified the variable regions of the heavy chain (VH) and light chain (VL) genes using reversetranscriptase polymerase chain reaction (RT-PCR). Using a linker, the amplified sequences for the heavy and light chains were each connected to the sequence for a single polypeptide chain that was designed to be expressed. The VL and VH fragments were cloned into the pOptiVEC-TOPO vector that contained the human CH2-CH3 fragment. Then, 293T cells were transfected with the 1E8 single-chain Fv-Fc (scFv-Fc) constructs. CD59 expression was evaluated in the prostate cancer cell lines using flow cytometry. The enhancement of CDC effect by mouse 1E8 and 1E8 scFv-Fc were evaluated using a cytotoxicity assay. Results: The scFv-Fc constructs were expressed by the transfected 293T cells and secreted into the culture medium. The immunoreactivity of the secreted scFv-Fc construct was similar to that of the mouse 1E8 for CCRF-CEM cells. The molecular masses of 1E8 scFv-Fc were about 120 kDa and 55 kDa under reducing and non-reducing conditions, respectively. The DNA sequence of 1E8 scFv-Fc was obtained and presented. CD59 was highly expressed by the prostate cancer cell line. The recombinant 1E8 scFv-Fc mAb revealed significantly enhanced CDC effect similar with mouse 1E8 for prostate cancer cells. Conclusion: A 1E8 scFv-Fc construct for adjuvant cancer therapy was developed.

Genetic variance of Tuchomonns uaginclis isolates by Southern hybridization (Southern hybridization에 의한 질편모충의 유전학적 다양성)

  • In the present study, genomic DNAs were purified from Korean isolates (KT8, KT6, KT-Kim and KT-Lee) and foreign strains (CDC85, IR78 and NYH 286) of 1Trichomonas voslnalis, and hybridized with a probe based on the repetitive sequence cloned from T. uqfinolis to observe the genetic differences. By Southern hybridization, all isolates of T. uoSinoLis except the NYH386 strain had 11 bands. Therefore all isolates examined were distinguishable into 3 groups according to their banding patterns; i) KT8, KT6 and KT-Kim isolates had 11 identical bands such as 1 kb, 1.2 kb, 1.6 kb, 1.9 kb, 2.3 kb. 27 kb, 3.2 kb, 2.4 kb, 3.8 kb, 4.9 kb and 6.0 kb, ii) The metronidazole-resistant IR78 strain had the some bands as KT-Lee isolate at bands of 1 kb, 1.2 kb, 1.6 kb. 1.8 kb, 2.1 kb, 2.5 kb, 2.7 kb, 2.9 kb, 3.4 kb, 5.0 kb and 6.0 kb, Bands of CDC85, metronidazole-resistant strain, were similar to those of IR78 and KT-Lee, except that 3.2 kb replaced 2.9 kb. iii) NYH286 particularly had 12 bands and bun patterns were similar to IR78 with a few exceptions as follows; i) 6.2 kb in place of 6.0 kb, ii) 2.0 kb and 2.2 kb instead of 2.1 kb. Through the results obtained, genetic variance of T. uoginnlis isolates was demonstrated by Southern hybridization.

Implementation of Wideband SSM with Excellent Sideband and Carrier Suppression (우수한 측파대 및 반송파 억압을 갖는 광대역 단측파대 믹서의 구현)

  • In this paper, we present design method of wideband single-sideband mixer(SSM) with excellent sideband suppression and carrier suppression. We proposed SSM with 4-band split structure for uniform output flatness. From circuit simulation of this structure using Microwave OfficeTM, we got satisfactory results. We realized a SSM based on this results, then we experimented. The measured results show excellent agreements with the simulated results. We compared with two SSM's(target SSM(M Co. SM0218LC1CDC SSM) and our SSM) as well. The presented SSM is 3.35 times bigger, but sideband suppression is 3 dBc better and carrier suppression is 8 dBc better than those of comparative SSM, respectively.

The multifunctional RNA-binding protein hnRNPK is critical for the proliferation and differentiation of myoblasts

  • HnRNPK is a multifunctional protein that participates in chromatin remodeling, transcription, RNA splicing, mRNA stability and translation. Here, we uncovered the function of hnRNPK in regulating the proliferation and differentiation of myoblasts. hnRNPK was mutated in the C2C12 myoblast cell line using the CRISPR/Cas9 system. A decreased proliferation rate was observed in hnRNPK-mutated cells, suggesting an impaired proliferation phenotype. Furthermore, increased G2/M phase, decreased S phase and increased sub-G1 phase cells were detected in the hnRNPK-mutated cell lines. The expression analysis of key cell cycle regulators indicated mRNA of Cyclin A2 was significantly increased in the mutant myoblasts compared to the control cells, while Cyclin B1, Cdc25b and Cdc25c were decreased sharply. In addition to the myoblast proliferation defect, the mutant cells exhibited defect in myotube formation. The myotube formation marker, myosin heavy chain (MHC), was decreased sharply in hnRNPK-mutated cells compared to control myoblasts during differentiation. The deficiency in hnRNPK also resulted in the repression of Myog expression, a key myogenic regulator during differentiation. Together, our data demonstrate that hnRNPK is required for myoblast proliferation and differentiation and may be an essential regulator of myoblast function.

Measurement of Fast Neutron Spectrum and Flux in Central Thimble of TRIGA MARK-II Reactor

  • The measurements of the fast neutron flux and its spectrum have been carried out by the threshold detectors in the central thimble of TRIGA Mark-II reactor operating at 250 KW. The following reactions have been employed for these measurements, viz : Ni$^{58}$ (n, p) Co$^{58}$$Mg^{24}$ (n, p) Na$^{24}$$Al^{27}$ (n, $\alpha$) Na$^{24}$ . From the activation data the fast neutron spectrum were calculated by CDC-3600 computer making use of two semi-empirical methods. It has been verified that the validity of assumption of a fission spectrum in the central thimble exists only above 1 to 2 Mev energy level. With this spectrum, a fast neutron flux in the range of 1 $\times$ 10$^{12}$ n/$\textrm{cm}^2$-sec above the energy of 2.6 Mev was observed in the central thimble of TRIGA MARK-II reactor.
