Optical absorption and emission studies have been carried out to understand the effect of deuterium on the solvent dependent photophysical properties of curcumin in deuterated solvents such as $CDCl_3,\;(CD_3)_2SO,\;(CD_3)_2CO,\;CD_3OD\;and\;CD_3CN$. Optical absorption spectral studies showed that there is no significant shift in absorption maxima compared to the non-deuterated solvent. The fluorescence maxima shows significant shift with polarity of solvent but not much affected by the deuteration. The fluorescence quantum yield of curcumin increased marginally in almost all the deuterated solvents, indicating reduction in the non-radiative pathways. The fluorescence decay was biexponential in all the solvents and the average fluorescence lifetime was not much affected with deuteration, but showed decrease with increasing solvent polarity. Based on these studies, it is concluded that intermolecular hydrogen transfer is only partially responsible for the excited state deactivation of curcumin.

There are several data in the literature indicating a great variety of pharmacological activities of Polypodium genus, which exhibit antiinflammatory and immunomodulatory activities. Since one of our main interests is to obtain natural immunoregulatory agents devoid of pharmacological adverse effects, we used flow cytometry analysis to highlight relative contributions of a water-soluble fraction of different concentrations of Polypodium rhizome extracts on lymphocyte subpopulations, NK and LAK activity. To measure their potential immunoregulatory activity a T cell proliferation assay in response to phytohemaglutinin (PHA) and mixed lymphocyte reactions were chosen. As a confirmatory bioassay we studied the effect of our extracts on CD45RO and CD95 antigen expressions. The results indicate that CD95 expression dramatically increases after peripheral blood lymphocyte activation and treatment with Polypodium leucotomus, cambricum and vulgare extracts, suggesting a powerful intrinsic pro-apoptotic effect.
$(1-x)BaTiO_3+(x)Cd_5(PO_4)_3Cl$ ceramics were prepared by the conventional ceramic technique, i.e., solid state reaction at high temperature. The concentration of $Cd_5(PO_4)_3C$ was varied from 0.01 to 0.15 mole fraction. In order to study the phase transitions of our ceramics, the Raman scattering spectra were measured as functions of concentration x and temperature. It was found that the soluble limit of $Cd_5(PO_4)_3Cl$ in $BaTiO_3$ was the x=0.05 composition and $BaTiO_3$ phase disappeared above x=0.10. A new phase identified as $Ba_4Ti_3P_2O_{15}$ was detected in all samples of our compositions. The Curie temperature shifts up to $130^{\circ}C$ as the concentration x increases from zero to 0.05 and shift down to $95^{\circ}C$ as further increases to 0.08. For the increase of the Curie temperature, it is suggested that it can result from the inhibition of displacement of $Ti^{4+}$ in the distorted octahedron due to well dispersed $Ba_4Ti_3P_2O_{15}$ and $Cd_5(PO_4)_3Cl$ phase.
We have synthesized new blue electroluminescent polyalkylfluorene-based copolymers [poly(F-co-Py)x:y, where x:y = 99:1 or 95:5 mole ratios] containing the hole-injecting pyrazole derivative [3,3'-(4,6-bis(octyloxy)-1,3-phenylene)bis(1,5-diphenyl-4,5-dihydro-1H-pyrazole] through Ni(0) mediated polymerization, and their electroluminescent properties were investigated. Electroluminescent (EL) devices were fabricated with ITO / PEDOT:PSS (110 nm) / copolymers or PF homopolymer (80 nm) / Ca (50 nm) / Al (200 nm) configuration. Each EL device constructed from the copolymer exhibited significantly enhanced brightness and efficiency compared with a device constructed from the PF homopolymer. The EL device constructed with poly(F-co-Py)99:1 exhibited the highest luminous efficiency and brightness (0.95 cd/A and $2,907\;cd/m^2$, respectively). The achieved luminous efficiency was an excellent result, providing almost a 4-fold improvement on the efficiency obtainable with the a PF homopolymer device. This enhanced efficiency of the copolymer devices results from their improved hole injection and more efficient charge carrier balance, which arises from the HOMO level (~5.83 eV) of the poly(F-co-Py)99:1 copolymer, which is higher than that of the PF homopolyme (~5.90 eV).

The CD4+CD25+ regulatory T cell (Treg) is a special kind of T cell subset. Studies have showed that Treg cells are involved in a number of physiological processes and pathologic conditions such as autoimmune diseases, transplantation tolerance and cancer. Tregs with unique capacity for immune inhibition can impair anti-tumour immunity and help tumor cells to escape from immune surveillance. The aim of our study was to investigate whether Tregs are involved in hepatocellular carcinoma (HCC). A BABL/C mouse with HCC in situ model was established to evaluate the Treg existence in carcinoma tissues and the changes of Tregs in spleen using flow cytometry and immunohistochemistry methods. Granzyme B expression in carcinoma tissues was analyzed by immunohistochemistry to investigate the tumor local immune status.The proportion of CD4+CD25+/CD4+ spleen lymphocytes of tumor bearing mice ($18.8%{\pm}1.26%$) was found to be significantly higher than that in normal mice ($9.99%{\pm}1.90%$) (P<0.01 ). Immunohistochemistry of spleen tissue also confirmed that there was an increase in Treg in tumor-bearing mice, while in carcinomas it showed Treg cells to be present in tumor infiltrating lymphocyte areas while Granzyme B was rarely observed. Anti-tumour immunity was suppressed, and this might be associated with the increase of Tregs. Our observations suggest that the CD4+CD25+Treg/CD4+ proportion in spleen lymphocytes can be a sensitive index to evaluate the change of Tregs in hepatocellular carcinoma mice and the Treg may be a promising therapeutic target for cancer.
CD44 is a particular adhesion molecule and facilitates both cell-cell and cell-matrix interactions. In particular, splice variants of CD44 are particularly overexpressed in a large number of malignancies and carcinomas. In this study, the $^{177}Lu$-labelled CD44 targeting antibody was prepared and bioevaluated in vitro and in vivo. Anti-CD44 was immunoconjugated with the equivalent molar ratio of cysteine-based DTPA-NCS and radioimmunoconjugated with $^{177}Lu$ at room temperature within 15 minutes. The stability was tested in human serum. An in vitro study was carried out in HT-29 human colon cancer cell lines. For the biodistribution study $^{177}Lu$-labelled anti-CD44 was injected in xenograft mice. Anti-CD44 was immunoconjugated with cysteine-based DTPA-NCS and purified by a centricon filter system having a molecular cut-off of 50 kDa. Radioimmunoconjugation with $^{177}Lu$ was reacted for 15 min at room temperature. The radiolabeling yield was >99%, and it was stable in human serum without any fragmentation or degradation. The radioimmunoconjugate showed a high binding affinity on HT-29 colon cancer cell surfaces. In a biodistribution study, the tumor-to-blood ratio of the radioimmunoconjugate was 43 : 1 at 1 day post injection (p.i) in human colon cancer bearing mice. The anti-CD44 monoclonal antibody for the targeting of colon cancer was effectively radioimmunoconjugated with $^{177}Lu$. The in vitro high immunoactivity of this radioimmunoconjugate was determined by a cell binding assay. In addition, the antibody's tumor targeting ability was demonstrated with very high uptake in tumors. This radioimmunoconjugate is applicable to therapy in human colon cancer with highly expressed CD44.
인간의 질병연구를 위한 소동물용 픽셀화 반도체 검출기 기반의 단일광자단층촬영(SPECT, single photon emission computed tomography)시스템 개발이 이루어지고 있다. 본 연구에서는 CdTe검출기 기반의 SPECT시스템의 고해상도 및 저선량 소동물 영상화 가능성을 알아보고자 NaI(Tl) 섬광결정 검출기로 구축된 SPECT 시스템과 비교 평가하였다. CdTe 검출기는 $44.8{\times}44.8$ mm의 크기이며 $0.35{\times}0.35{\times}5$ mm크기의 픽셀로 구성되어 있다. 검출기의 내인성 분해능은 0.35 mm 이며 이는 픽셀 크기와 동일하다. GATE 시뮬레이션 방법을 통하여 두 시스템간의 성능 평가를 수행하고 비교 분석하였다. 시스템의 공간 분해능과 민감도는 10 MBq의 $^{99m}Tc$ 점 선원을 사용하여 평가하였다. 복셀화된 MOBY (mouse whole-body) 팬텀을 사용하여 정량적 평가 및 흡수선량을 계산하였다. 점선원과 조준기 사이의 거리가 30 mm 일 때, NaI(Tl) 섬광결정 검출기 기반의 SPECT의 분해능은 1.54 mm, 민감도는 83 cps/MBq였으며, CdTe검출기 기반의 SPECT시스템의 분해능은 1.32 mm, 민감도는 116 cps/MBq로 더욱 향상된 공간 분해능과 민감도를 나타내었다. 두 시스템의 정량적 통계 분석은 CNR 계산을 통해 이루어졌으며, 주입 선량을 다양하게 설정하여 두 시스템에서의 CNR을 획득하였다. Mouse brain내 striatum의 주입선량이 160 Bq/voxel일 경우, CdTe검출기 기반의 SPECT에서 획득한 CNR은 2.30이었으며 섬광결정 검출기 SPECT에서 획득한 CNR은 1.85로 CdTe검출기 기반의 SPECT에서 더욱 큰 CNR을 지니고 있었다. 또한, CdTe기반의 SPECT를 사용할 경우 NaI(Tl) 섬광결정 검출기 기반의 SPECT 시스템을 사용하는 것보다 동일한 정량적 수치획득을 위한 소동물의 피폭선량을 감소시켜줄 수 있었다. 본 연구에서는 반도체 검출기 CdTe기반의 SPECT은 NaI(Tl) 섬광결정 검출기 SPECT 시스템보다 공간 분해능과 민감도 측면에서 높은 성능을 보였음을 증명하였다. 실제 시스템과의 검증 등의 추가 연구가 필요하지만, 본 연구 결과는 향후 피폭 선량을 줄이는 동시에 영상의 질을 높일 수 있는 소동물용 SPECT 시스템 구축에 응용될 수 있을 것이다.
경남지역 논 토양의 중금속 관리를 위한 기초 자료를 제공하고자 260 지점을 대상으로 2011년에 Cd, Cu, Ni, Pb, Zn 및 As 전함량과 0.1N HCl 가용성 Cr 함량을 분석하였다. 논토양의 중금속 평균 함량은 Cd 0.143 (범위 0.003-0.537) mg $kg^{-1}$, Cr 0.322 (0.002-0.986) mg $kg^{-1}$, Cu 25.41 (6.03-76.19) mg $kg^{-1}$, Ni 16.36 (2.63-32.94) mg $kg^{-1}$, Pb 18.67 (4.16-87.02) mg $kg^{-1}$, Zn 71.76 (22.99-153.58) mg $kg^{-1}$, As 3.516 (0.002-19.481) mg $kg^{-1}$ 였다. 논토양에서 Cd 함량은 산록경사지에서 가장 높은 반면 Ni과 As 함량은 홍적대지, Zn 함량은 해안평탄지에서 가장 높았다. 또한, Cd, Cu 및 Zn 함량은 미사질식양토에서 미사질양토, 사양토 및 양토 보다 높은 경향이었다.
Extract of Centella asiatica(ECA), which is poorly water-soluble extract from the Centella asiatica is known to express excellent wound healing properties. $ECA-{\beta}-cyclodextrin$$(asiaticoside-{\beta}-cyclodextrin\;and\;genin-{\beta}-cyc1odextrin)$ solid dispersion system, which was prepared by freezedrying method, was formulated as gels containing poloxamer 407 and propylene glycol, and evaluated with respect to their viscosity, stability, skin permeation and drug amount in the skin of hairless mouse. The average molar ratio $asiaticoside-{\beta}-CD$ and $genin-{\beta}-CD$ was 1:1.7 and 1:22, respectively. When the molar ratio of genin and ${\beta}-CD$ was 1:5, madecassic acid made 100% solid dispersion system and asiatic acid about 65%. In dissolution study, >99% of asiaticoside from $asiaticoside-{\beta}-CD$ was dissolved in 5 minutes, and >99% madecassic acid and >64% asiatic acid from $genin-{\beta}-CD$. The apparent viscosity of poloxamer 407 gels with $ECA-{\beta}-CD$ solid dispersion system increased in proportion to poloxamer 407 and propylene glycol concentration. In the accelerated stability test, all $ECA-{\beta}-CD$ poloxamer 407 gels showed that asiaticoside was most stable and madecassic acid stable and asiatic acid similar to stability of gel with free ECA. The permeation amount of asiaticoside in poloxamer gels through hairless mouse skin decreased as the concentration of poloxamer 407 increased. When propylene glycol was added at the level of 10%, the permeation amount of asiaticoside at poloxamer gels through hairless mouse skin increased but from 15% it decreased. The permeation of asiaticoside into the skin of hairless mouse was estimated to be about $0.10\;{\mu}g/cm^2$.
감국의 유산균 발효를 통한 감국 배양물(CILL)의 비만억제 식품의 소재로서 가능성을 알아보기 위해 고지방식이로 유도된 C57BL/6 mice에서의 체중, body fat mass, T cell 등을 측정하였다. 체중 변화에서 CILL 군 (25.15±2.44 g)은 4주부터 체중 감소를 보였으며, 1주차부터 8주차까지 낮은 증체량을 유지했다(1.00±0.53 g). 8주 체중(30.38±4.17 g)은 HFD 군(60% high fat diet, 34.99±2.09 g) 대비 13.15%의 체중 감소를 보였다. Fat mass는 10.3022±2.8813 g로 감소했으며, 간 절대 중량에서 HFD 군에 비해 감소하였다. CD4+ T cell 4.84±1.33%, CD8+ T cell 7.02±2.26%, CD4+CD8+ T cell 1.46±0.81%로 HFD 군에 비해 높게 측정되었다. 결과적으로 CILL은 비만억제 효과가 있으며, CILL내의 비만억제 물질을 선별한다면 효과적인 비만억제 식품 소재로서의 가능성이 있을 것으로 사료된다.
