백혈구 공통 항원인 돼지 CD45는 PTPRC 유전자에 암호화 되어 있으며, CD45엑손의 선택적 스플라이싱에 따라 다른 T-세포에서 발현되는 티로신 인산분해효소이다. CD45는 기질인 TCR의 $CD3{\zeta}$ 사슬, Lck, Fyn, Zap-70 kinase의 인산화된 티로신에서 인산을 분해하여 T-세포 항원 수용체(TCR) 매개 신호전달을 조절한다. CD45의 조절이상은 많은 면역 질환과 관련이 있어서, CD45는 면역약물 개발에 표적이 되어왔다. TCR 신호전달의 조절효과를 가진 주요 구조적 특징을 특성화하기 위해, 사람의 알려진 CD45 구조를 템플릿으로 적용하여 돼지 CD45RO(가장 작은 CD45 isoform)의 단백질 구조와 예측된 돼지 CD45RO 모델구조에 $CD3{\zeta}$ 사슬의 ITAM(REEpYDV)를 도킹하여 CD45RO/ITAM 펩타이드 결합구조를 예측하였다. 돼지 CD45RO의 구조적 특징은 세포외영역의 구조견고성과 세포질 내 티로신 인산분해효소 도메인의 KNRY와 PTP signature 기능모티프(두 기능 모티프는 ITAM 펩타이드 결합부위의 좁은 입구로 역할)에 있었다. 주요 구조특성은 돼지 CD45RO-ITAM 펩타이드 결합구조 안정성과 결합친화력을 조절하면서 기질 선택성에 영향을 준다. 돼지 CD45RO의 구조적 특성은 T-세포에 특이적인 면역 조절제를 탐색하는 데에 적용될 것이다.
미란형 구강편평태선과 재발성 아프타성 구내염은 T림프구에 의해 매개되는 염증성 면역질환이다. T림프구에 영향을 주는 다양한 조절 인자들 중 CD28, CD45, CD152, CD154, CD279등의 mRNA가 미란형 구강편평태선 및 재발성 아프타성 구내염 환자들의 비자극성 전타액내에서 어떻게 발현되는지를 살펴보고, 이것의 진단학적 가치를 평가하고자 한다. 미란형 구강편평태선으로 진단받은 환자군 18명, 재발성 아프타성 구내염으로 진단받은 환자군 12명, 정상군 8명에게서 비자극성 전타액을 10분간 채득한 후, 상피세포를 분리하여 mRNA를 추출하였다. Real time PCR을 이용하여 각 군의 T림프구 조절 인자들의 mRNA 발현 양상을 비교분석하였다. 미란형 구강편평태선의 T림프구 조절인자들의 mRNA 발현 양상은 정상인과 비교 결과, CD45, CD279는 높게 측정되었고, CD154는 낮게 측정되었다. 재발성 아프타성 구내염 환자들의 T림프구 조절인자들의 mRNA 발현 양상은 정상인과 비교 결과, CD45, CD279는 높게 발현되었고, CD28, CD154는 낮게 발현되었다. 부가적으로 미란형 구강편평태선 환자군의 타액내 CD152의 발현 양상은 재발성 아프타성 구내염 환자군보다 높게 발현되었다. 미란형 구강편평태선과 재발성 아프타성 구내염 환자들의 비자극성 전타액내 T림프구 조절인자들의 mRNA 발현 양상은 각 질환의 진단에 기여할 수 있을 것으로 사료된다.
이 연구의 목적은 발치한 매복 과잉치에서 얻은 치수 유래 세포의 줄기세포 특성을 유세포 분석을 통해 알아보는 것이다. 15명의 환자로부터 채취한 정중 과잉치의 치수 세포를 계대 배양하였고, 3계대와 10계대 세포들을 유세포 분석을 이용하여 분석하였다. 간엽성 줄기세포 표지자 관찰에 사용된 항체 표지자(CD)는 양성 표지자로는 CD 73, CD 90, 그리고STRO-1 와 음성 표지자로는 CD 34, CD 45 이었다. 3계대에서 CD 73, CD 90은 각각 94.82%, 98.86%의 양성반응을, CD 34, CD 45는 각각 2.25%, 2.52%로 음성 반응을 보였으며, STRO-1은 20.93%를 나타냈다. 10계대에서는 CD 73, CD 90은 각각 96.62%, 98.61%의 발현을 보였지만, CD 34, CD 45는 각각 3.86%, 4.14%를 나타냈다. STRO-1은 35.62%로 발현되었다. 이상의 결과에서 과잉치 치수 유래 세포는 간엽성 줄기세포의 특성을 가지며, 3계대와 10계대 모두에서 간엽성 줄기세포의 특성을 유지하고 있다고 사료된다. 이에 빠른 성장 속도와 늦은 계대까지 유지되는 줄기세포능을 고려할 때, 치아 유래 줄기세포의 공여부로서 매복 과잉치의 충분한 활용 가능성을 확인하였다.
3-weeks old Commelina was transferred to and grown in Hoagland solution (Control, $100\muM$$Cd^{2+}$, $100\muM$$Cd^{2+}$+ $100\muM$ IAA, $100\muM$$Cd^{2+}$+ $100\muM$ IAA + 2 mM sucrose) for 3 weeks and then the effects of indole acetic acid (IAA) on the accumulation of $Cd^{2+}$ and growth of $Cd^{2+}$-treated Commelina were investigated. In the treatment of $Cd^{2+}$, $Cd^{2+}$ was uptaked to 1.74, and 51.36 ${\mu}g/g$ frwt. at the first week, but for three weeks, 0.51 and 34,53 ${\mu}g/g$ frwt, in leaf and stem respectively. When IAA was treated along with $Cd^{2+}$, $Cd^{2+}$ was uptaked to 0.18 and 8.63 ${\mu}g/g$ fiwt, at the first week, and for the incubation of 3 weeks, 0,51 and 45.0 ${\mu}g/g$ fiwt. in leaf and stem. In case of $Cd^{2+}$+IAA+sucrose, $Cd^{2+}$ was uptaked to 1.45 and 18.33 ${\mu}g/g$ frwt. at the first week, but for 3 weeks, 0,51 and 25.45 ${\mu}g/g$ fiwt. in leaf and stem. Likewise $Cd^{2+}$ uptake, the growth was also affected by $Cd^{2+}$ and IAA. During the incubation of 3 weeks, $Cd^{2+}$ reduced the stem growth about 8% in all weeks, but the treatment of IAA recovered the inhibition of stem growth caused by $Cd^{2+}$ to the degree of the control Therefore, it could be concluded that IAA altered the pattern of $Cd^{2+}$ uptake and the growth which were supposed to change $Cd^{2+}$ toxicity.

Purpose: Beyond the limited scope of non-specific polyclonal regulatory T cell (Treg)-based immunotherapy, which depends largely on serendipity, the present study explored a target Treg subset appropriate for the delivery of a novel epitope spreader Pep19 antigen as part of a sophisticated form of immunotherapy with defined antigen specificity that induces immune tolerance. Methods: Human polyclonal $CD4^+CD25^+CD127^{lo-}$ Tregs (127-Tregs) and $na\ddot{i}ve$$CD4^+CD25^+CD45RA^+$ Tregs (45RA-Tregs) were isolated and were stimulated with target peptide 19 (Pep19)-pulsed dendritic cells in a tolerogenic milieu followed by ex vivo expansion. Low-dose interleukin-2 (IL-2) and rapamycin were added to selectively exclude the outgrowth of contaminating effector T cells (Teffs). The following parameters were investigated in the expanded antigen-specific Tregs: the distinct expression of the immunosuppressive Treg marker Foxp3, epigenetic stability (demethylation in the Treg-specific demethylated region), the suppression of Teffs, expression of the homing receptors CD62L/CCR7, and CD95L-mediated apoptosis. The expanded Tregs were adoptively transferred into an $NOD/scid/IL-2R{\gamma}^{-/-}$ mouse model of collagen-induced arthritis. Results: Epitope-spreader Pep19 targeting by 45RA-Tregs led to an outstanding in vitro suppressive T cell fate characterized by robust ex vivo expansion, the salient expression of Foxp3, high epigenetic stability, enhanced T cell suppression, modest expression of CD62L/CCR7, and higher resistance to CD95L-mediated apoptosis. After adoptive transfer, the distinct fate of these T cells demonstrated a potent in vivo immunotherapeutic capability, as indicated by the complete elimination of footpad swelling, prolonged survival, minimal histopathological changes, and preferential localization of $CD4^+CD25^+$ Tregs at the articular joints in a mechanistic and orchestrated way. Conclusions: We propose human $na\ddot{i}ve$$CD4^+CD25^+CD45RA^+$ Tregs and the epitope spreader Pep19 as cellular and molecular targets for a novel antigen-specific Treg-based vaccination against collagen-induced arthritis.
Canine T-zone lymphoma (TZL) is a mature T-cell lymphoma in dogs. The diagnosis and sub-classification are impossible without biopsy or immunophenotyping by flow cytometry. An 11-year-old, spayed, female Golden Retriever presented with lymph node enlargement. Clinical examination was consistent with canine multicentric lymphoma. However, immunophenotyping revealed positive for CD3, CD4, CD5, CD8, CD21, TCRαβ, and MHCII but negative for CD34, CD45, CD79a, and TCRγδ. Histopathology revealed lymphocytes expanding to the cortex-preserving architecture and thinning of the nodal capsule, and CD3 positive but PAX-5 negative. Owing to the indolent nature of TZL, careful monitoring approach without clinical intervention was utilized.
Background: The leukocyte common antigen (CD45) is a transmembrane-type protein tyrosine phosphatase that has five isoforms. Methods: We generated seven murine mAbs against human CD45 by injecting cells from different origins, such as human thymocytes, PBMCs, and leukemic cell lines. By using various immunological methods including flow cytometry, immunohistochemistry, and immunoprecipitation, we evaluated the reactivity of those mAbs to CD45 of thymus as well as tonsil lysates. Furthermore, we transiently transfected COS-7 cells with each of gene constructs that express five human CD45 isoforms respectively, and examined the specificities of the mAbs against the transfected isoforms. Results: In case of thymocytes, lymphocytes, and monocytes, all the seven mAbs demonstrated positive reactivities whereas none was reactive to erythrocytes and platelets. The majority of immune cells in formalin-fixed paraffin-embedded thymus and tonsil tissues displayed strong membranous immunoreactivity, and the main antigen was detected near 220 kDa in all cases. Among the mAbs, four mAbs (AP4, DN11, SHL-1, and P6) recognized a region commonly present in all the five isoforms. One mAb, YG27, recognized four isoforms (ABC, AB, BC, and O). Two mAbs, P1 and P14, recognized the isoforms that contain exon A encoded regions (ABC and AB). Conclusion: In this study, we confirmed that AP4, DN11, SHL-1, YG27 and P6, are mAbs reactive with the CD45 antigen whereas P1 and P14 are reactive with the CD45RA antigen.
가용성 전분으로부터 CGTase 효소에 의한 Cyclodextrin(CD) 동족체의 생산 실험을 dead-end형 막모듈을 설치한 막-효소 반응기에서 수행하였다. 회분 반응기에서의 CD 동족체 생산시 생산물 저해작용 및 생산된 CD의 다른 환원당으로의 분해에 의해 반응시간에 따른 전분의 CD 동족체로의 총 전환율이 최대 45%를 나타내었다가 급격히 감소하였다. 반면 분획분자량 10,000 달톤의 분리막이 설치된 막-효소 반응기에서의 CD 동족체 생산시 CD 동족체를 생산 즉시 반응기로부터 막을 통해 연속 분리할 수 있어 총 전환율이 35%로 일정하게 유지되었다. 10% 전분용액 및 2 atm 조작압력하에서 막-효소 반응기를 24시간 운전하였을 때 CD 동족체의 누적 생산량은 약 3.7 kg/m$^2$이었다.
In this paper, CdS/Cd(Cu)S thin films and diodes were manufactured via a chemical bath deposition (CBD) process, and the effects of $NH_4Cl$ and TEA(triethylamine) on the properties of the films were examined. The addition of $NH_4Cl$ significantly increased the thickness of the CdS and Cd(Cu)S films, however, the addition of TEA decreased the thickness in both cases slightly. The addition of $NH_4Cl$ along with TEA increased the film thickness more effectively compared to the addition of only $NH_4Cl$. The thickness of the CdS film prepared from an aqueous solution of 0.007 M $CdSO_4$, 1.3 M $NH_4OH$, 0.03 M $SC(NH_2)_2$, 0.0001 M TEA and 0.03 M $NH_4Cl$ was 310 nm. Dark resistivity of the CdS film was $1.2{\times}10^3\;{\Omega}cm$ and the photo resistivity with $500\;W/cm^2$ irradiation of white light was $20{\Omega}cm$. The Cd(Cu)S/CdS thin film diodes prepared by CBD showed good rectifying characteristics.
To investigate the cadmium (Cd) toxicity on the testis, male rats were treated with 1, 2, 4 and 8 mg/kg of Cd by IP. According to histochemical studies, Cd-treated testis tissue showed death of spermatozoa, death of Sertoli cells, death of all the spermatogenic cells, and finally disappearance of basal lamina of seminiferous tubules with increasing doses, and showed decreased ground substances and Leydig cells, increased inflammatory cells and fibroblasts, and fibroblasts, and finally disappearance of ground substances and all the cells except fibroblasts within interstitial tissues with increasing doses. According to biochemical studies, two kinds of proteins, 25 and 45 kDa, were dramatically disappeared from the total protein of rat testis treated with Cd comparing to normal testis. The result of electrophoresis of total protein suggests that actin (45 kDa), presumed on its mmolecular weight and amount, in the testis-cells is the primary target of Cd poisoning. Although its exact mechanism is not clear, the disappearance of two proteins when testis is exposed to Cd should give some clues to understnad the mechanism of necrosis of testis tissue crumbling by heavy metal pollutant such as Cd.
