Association of the Porcine Cluster of Differentiation 4 Gene with T Lymphocyte Subpopulations and Its Expression in Immune Tissues

  • Cluster of differentiation 4 (CD4) is mainly expressed on $CD4^+$ T cells, which plays an important role in immune response. The aim of this study was to detect the association between polymorphisms of the CD4 gene and T lymphocyte subpopulations in pigs, and to investigate the effects of genetic variation on the CD4 gene expression level in immune tissues. Five missense mutations in the CD4 gene were identified using DNA pooling sequencing assays, and two main haplotypes (CCTCC and AGCTG) in strong linkage disequilibrium (with frequencies of 50.26% and 46.34%, respectively) were detected in the population of Large White pigs. Our results indicated that the five SNPs and the two haplotypes were significantly associated with the proportions of $CD4^-CD8^-$, $CD4^+CD8^+$, $CD4^+CD8^-$, $CD4^+$ and $CD4^+/CD8^+$ in peripheral blood (p<0.05). Gene expression analysis showed the mRNA level of the CD4 gene in thymus was significantly higher than that in lymph node and spleen (p<0.05). However, no significant difference was observed between animals with CCTCC/CCTCC genotype and animals with AGCTG/AGCTG genotype in the three immune tissues (p>0.05). These results indicate that the CD4 gene may influence T lymphocyte subpopulations and can be considered as a candidate gene affecting immunity in pigs.

신비탕 및 가미신비탕이 Allergy 천식 모델 흰쥐의 BALF내 면역세포 및 혈청 IgE에 미치는 영향에 관한 연구 (The Effects of Shinbi-tang and Gamishinbi-tang on Immune Cell and Serum OA-specific IgE in BALF in Rat Asthma Model)

  • Background : Asthma is a chronic inflammatory disorder under immunological influence. Shinbi-tang and Gamishinbi-tang are herbal decoctions used for treating asthma in traditional herbal medicine. Objective : To evaluate the effects of Shinbi-tang and Gamishinbi-tang on immune cell & serum OA-specific IgE in broncho-alveolar lavage fluid (BALF) in rat asthma model. Material and Methods : Rats were sensitized with ovalbumin (OA); at day 1 sensitized group and Shinbi-tang and Gamishinbi-tang groups were systemically immunized by subcutaneous injection of 1mg OA and 300mg of Al(OH)$_3$ in a total volume of 2ml. At the same time, 1 ml of 0.9% saline containing 6 x 10$^{9}$ B. pertussis bacilli was injected by i.p. 14 days after the systemic immunization, rats received local immunization by inhaling 0.9% saline aerosol containing 2%(wt/vol) OA. A day after local immunization, HAL fluid was collected from the rats. Rats were orally administered with each of Shinbi-tang and Gamishinbi-tang extract for 14 days from the day after local immunization. Lymphocyte, CD4+ T cell CD8+ T cell counts, CD4+/CD8+ ratio in BALF, change of serum OA-specific IgE level, CD4+ T cell CD8+ T cell percentages in the peripheral blood were measured and evaluated. Results : Shinbi-tang and Gamishinbi-tang showed an alleviating effect on asthmatic responses of rats. Shinbi-tang decreased total cell, lymphocyte, CD4+ T cell in BALF, serum OA-specific IgE level as compared with the control group. Gamishinbi-tang decreased total cell, lymphocyte, CD4+ T cell, and CD4+/CD8+ ratio in HALF as compared with the control group. CD4+/CD8+ ratio in HALF from Shinbi-tang group and serum OA-specific IgE level from Gamishinbi-tang group didn't show any significant variation from control group. CD8+ T cell in HALF, CD3+CD4+ T cell and CD3+CD8+ T cell percentages in peripheral blood showed no significant variation among groups. Conclusion : Shinbi-tang and Gamishinbi-tang alleviated asthmatic hypen-eactivity of the rat immune system through CD4+ T cell and serum IgE. Further the study of immune system modulating mechanism is expected.

미란형 구강편평태선과 재발성 아프타성 구내염 환자들의 비자극성 전타액내 T림프구 조절인자들의 발현 양상 (Expression Pattern of T Lymphocyte Regulatory Factors in Unstimulated Whole Saliva of Erosive Oral Lichen Planus and Recurrent Aphthous Stomatitis Patients)

  • 미란형 구강편평태선과 재발성 아프타성 구내염은 T림프구에 의해 매개되는 염증성 면역질환이다. T림프구에 영향을 주는 다양한 조절 인자들 중 CD28, CD45, CD152, CD154, CD279등의 mRNA가 미란형 구강편평태선 및 재발성 아프타성 구내염 환자들의 비자극성 전타액내에서 어떻게 발현되는지를 살펴보고, 이것의 진단학적 가치를 평가하고자 한다. 미란형 구강편평태선으로 진단받은 환자군 18명, 재발성 아프타성 구내염으로 진단받은 환자군 12명, 정상군 8명에게서 비자극성 전타액을 10분간 채득한 후, 상피세포를 분리하여 mRNA를 추출하였다. Real time PCR을 이용하여 각 군의 T림프구 조절 인자들의 mRNA 발현 양상을 비교분석하였다. 미란형 구강편평태선의 T림프구 조절인자들의 mRNA 발현 양상은 정상인과 비교 결과, CD45, CD279는 높게 측정되었고, CD154는 낮게 측정되었다. 재발성 아프타성 구내염 환자들의 T림프구 조절인자들의 mRNA 발현 양상은 정상인과 비교 결과, CD45, CD279는 높게 발현되었고, CD28, CD154는 낮게 발현되었다. 부가적으로 미란형 구강편평태선 환자군의 타액내 CD152의 발현 양상은 재발성 아프타성 구내염 환자군보다 높게 발현되었다. 미란형 구강편평태선과 재발성 아프타성 구내염 환자들의 비자극성 전타액내 T림프구 조절인자들의 mRNA 발현 양상은 각 질환의 진단에 기여할 수 있을 것으로 사료된다.

삼출건비탕(蔘朮健脾湯)이 흰쥐의 면역반응(免疫反應)에 미치는 영향(影響) (Effects of Samchulkunbitang on the Immunosuppression Induced by Methotrexate in Rats)

  • Introduction The effects of Samchulkunbitang on the immunosuppression induced by methotrexate in rats were investigated in this experument. The multiple parameters of immunity assessed in each rats includes the rate of body weight loss, weight changes in thymus, spleen and axillary lymphnode. The number of lymphocytes and CD4+ T cell count in the blood, thymus, spleen and axillary lymphnode were also measured. Methodology Male Sprague-Dawley rats were chosen as an experiment object and were divided into 4 groups by a random selection. Each group consisted of 6 rats. The normal group didn't receive any treatment. The control group was administered with methotrexate for 5 days. The sample Ⅰ group was administered with Samchulkunbitang for 5 days. The sample Ⅱ group was administered with both Samchulkunbitang and methotrexate for 5 days. The dosage of medication was 2cc/day, 1cc given at 10 AM and another 1cc given at 5 PM. Results The rate of body weight loss was significantly increased in the Sample Ⅰ and Sample Ⅱ group. The weight of thymus and spleen were significantly increased in Sample Ⅰ and Sample Ⅱ groups While the weight of axillary lymphnode did not show much increase. No significant differences were measured among the experimental groups. Blood lymphocyte count, blood CD4+ T cell count, spleen lymphocyte count, axillary lymphnode lymphocyte and CD4+ T cell count were significantly increased in Sample Ⅰ and Sample Ⅱ groups while spleen CD4+ T cell count did not show much increase. No significant differences were measured among the experimental groups. Conclusion As one can witness from the above results, administration of Samchulkunbitang played potent role in increasing immune system among the rats treated with methotrexate which induces immunosuppression. Overall increase of lymphocyte count and CD4+ T cell count in the sample groups with Samchulkunbitang effectively proves its ability to boost the immune system.

보비탕(補脾湯)이 methotrexate로 유발(誘發)된 흰쥐의 면역기능저하(免疫機能低下)에 미치는 영향(影響) (Effect of Bobitang water extracts on immunosuppression induced by Methotrexate in SD rats)

  • Objectives : In order to investigate the effect of Bobitang on SD rats with deteriorated immunity caused by methotrexate. Methods : The test sample were dosed once a day for 14 days by gastric gavage at a dosage 1,000, 500 and $250mg/kg/10m{\ell}$ from 2 days after last MTX-dosing, and the changes on body weight and gains, spleen weight and total blood leukocyte numbers, total lymphocyte numbers, the percentage of B-cell, T-cell, CD3+CD4+ T-cell, CD3+CD8+ T-cell and CD4+/CD8+ T-cell ratios in the blood and spleen were observed. Results : The changes on body weight gains, the spleen weight, the total blood leukocyte numbers, the total lymphocyte numbers in the blood and spleen, the ratio of T-cell in the blood and spleen, the ratio of CD3+CD4+ T-cell in the blood and spleen were increased significantly in BBT Extracts groups as compared with control group. The ratio of B-cell in the blood and spleen was not increased significantly in BBT Extracts groups as compared with control group. The percentage of CD3+CD8+ T-cell in the blood and spleen was decreased significantly in BBT Extracts groups as compared with control group. The ratio of CD4+/CD8+ T-cell in blood and spleen was increased significantly in BBT Extracts group as compared with control group. Conclusions : According to the above results, Bobi-Tang has an effect of increasing immune responses on SD rats with deteriorated immunity caused by methotrexate.

한우와 젖소 신생 송아지의 말초혈액 림프구아군 특성비교 (Evaluation of character on lymphocyte subpopulations from peripheral blood in Hanwoo and Holstein neonatal calves)

  • The present study was undertaken to establish reference values for the composition blood lymphocyte populations and compare forty three Hanwoo neonatal calves (KC) with twenty one Holstein calves (HC) by blood cell count and immunophynotying. The percentages of CD2+, CD4+, CD8+, CD26+, ACT2+, MHC class, MHC class II and WC1+ T cells, B cells were determined by flow cytometry. The number of lymphocyte and monocyte in HC were higher than those of KC. However, the number of neutrophils was higher in HC than KC. The proportions of CD2+, CD4+, CD8+, MHC class, and WC1+ lymphocytes remained relatively stable during the study period, while there was a moderate increase in the relative percentage of CD26+, ACT2+, MHC class II and B cell from birth to approximately 3 weeks of age. Marked differences in the relative proportions of the lymphocyte subpopulations were noted between the individual calves. The present study shows that the T-cell subpopulations are present in peripheral blood of KC at levels comparable with HC, while the MHC class II and B cell population of KC increases significantly with age. The absolute number of WBC in KC was due to the decrease of absolute number of neutrophil rather than the increase of lymphocyte. The results indicated that KC have significantly higher number of neutrophils, and proportion of MHC class II and B cell than HC.

Polymorphism, Expression of Natural Resistance-associated Macrophage Protein 1 Encoding Gene (NRAMP1) and Its Association with Immune Traits in Pigs

  • Natural resistance-associated macrophage protein 1 encoding gene (NRAMP1) plays an important role in immune response against intracellular pathogens. To evaluate the effects of NRAMP1 gene on immune capacity in pigs, tissue expression of NRAMP1 mRNA was observed by real time quantitative polymerase chain reaction (PCR), and the results revealed NRAMP1 expressed widely in nine tissues. One single nucleotide polymorphism (SNP) (ENSSSCG00000025058: g.130 C>T) in exon1 and one SNP (ENSSSCG00000025058: g.657 A>G) in intron1 region of porcine NRAMP1 gene were demonstrated by DNA sequencing and PCR-RFLP analysis. A further analysis of SNP genotypes associated with immune traits including contain of white blood cell (WBC), granulocyte, lymphocyte, monocyte (MO), rate of cytotoxin in monocyte (MC) and $CD4^-CD8^+$ T lymphocyte subpopulations in blood was carried out in four pig populations including Large White and three Chinese indigenous breeds (Wannan Black, Huai pig and Wei pig). The results showed that the SNP (ENSSSCG00000025058: g.130 C>T) was significantly associated with level of WBC % (p = 0.031), MO% (p = 0.024), MC% (p = 0.013) and $CD4^-CD8^+$ T lymphocyte (p = 0.023). The other SNP (ENSSSCG00000025058: g.657 A>G) was significantly associated with the level of MO% (p = 0.012), MC% (p = 0.019) and $CD4^-CD8^+$ T lymphocyte (p = 0.037). These results indicate that the NRAMP1 gene can be regarded as a molecular marker for genetic selection of disease susceptibility in pig breeding.

녹용 에탄올 분획이 생쥐의 T-Lymphocyte에 미치는 영향 (Effect of Ethyl Alcohol Fraction of Cervus nippon on Mouse T-Lymphocyte)

  • In this study, the effect of 70% ethyl alcohol fraction of Cervus nippon(CN-E) on mouse T-lymphocyte was investigated in vivo. The administration of CN-E(100 mg/kg) enhanced the proliferation of thymocytes, the population of $CD4^+CD8^-$ single-positive cells and the production of $interferon-{\gamma}$ in thymocytes and splenocytes. The administration of CN-E did not induce DNA fragmentation and reduce mitochondrial transmembrane potential in thymocytes. These results indicate that the CN-E contams a stimulative component on the proliferation of thymocytes, the population of $T_H$ cells and the production of $interferon-{\gamma}$ in T-lymphocytes.

고강도운동 지속시간이 rat의 림프구 세포사에 미치는 영향 (Effects of the Duration of Highly Intensive Exercise on Lymphocyte Cell Death in Rats)

  • 고강도운동의 지속시간이 백혈구 조성과 T-림프구 활성 보조인자로서의 $CD4^+$$CD8^+$수준의 변화 그리고 림프수의 세포사에 미치는 영향을 조사하기 위해 쥐실험을 하였다. 고강도 운동을 매일 20, 60, 그리고 120분 동안 8주간 실시하였다. 혈액내의 총 백혈구 수는 20분간 운동을 했을 때 상승하였고 이것은 다시 120분 까지 대조군의 수준 이하로 감소하였다. 림프구의 수준변화 패턴 역시 운동시간의 영향을 받았으며 그 변화 정도는 총 백혈구의 그것과 유사하였다. 고강도운동을 실시한 쥐의 혈액 내 $CD4^+$$CD8^+$의 수준은 운동시간이 120분간 지속될 때까지 변화가 없었기 때문에 T-림프구의 활성에는 영향을 주지 않는 것으로 보인다. 거의 모든 초기단계 및 후기 단계의 림프구의 세포자멸사는 운동시간에 영향을 받지 않았으나 120분간 운동한 그룹에서 후기단계의 림프구 자멸사 수준이 증가되는 것으로 보아 이때 세포노화의 촉진이 일어났으리라 사료된다. 운동시간이 길어질수록 림프구의 괴사 수준이 증가되는 것을 확인 하였고 이에 따라 고강도운동에 의한 림프구 손상과 면역력 저하의 가능성이 예상된다. 본 연구에서 장시간 동안의 고강도 운동은 림프구의 염증관련 기능과 세포 수에 있어서의 손상 등을 일으켜 면역력의 저하를 초래할 수 있다고 보며 따라서 적어도 본 연구조건에 한해서 20분 이상의 고강도운동은 건강유지 차원에서 바람직하지 않다고 판단된다.

Gamichunggumgangwha-tang (CG, Jiaweiqingjinjianghuo-tang) and Gamiyukmigiwhang-tang (YM, Jiaweiliuweidihuang-tang) are herbal tonics for asthma from traditional herbal medicine. Objective : To evaluate the effect of CG and YM on immune cell & serum OA-specific IgE in broncho-alveolar lavage fluid (BALF) in a rat asthma model. Materials and Methods : Rats were sensitized with OA; at day I sensitized group and CG and YM groups were systemically immunized by subcutaneous injection of 1mg OA and 300mg of Al(OH)3 in a total volume of 2ml. At the same time, 1 ml of 0.9% saline containing $6{\times}10^9$ B. pertussis bacilli was injected by Lp. 14 days after the systemic immunization, rats received local immunization by inhaling 0.9% saline aerosol containing 2% (wt/vol) OA. A day after local immunization, BALF was collected from the rats. Rats were orally administered with each of CG and YM extract for 14 days since the day after local immunization. Lymphocyte, CD4+ T cell and CD8+ T cell counts, CD4+/CD8+ ratio in BALF, change of serum OA-specific IgE level, CD4+ T cell and CD8+ T cell percentages in the peripheral blood were measured and evaluated. Results : CG and YM showed an alleviating effect on asthmatic responses of rats. CG decreased total cell, lymphocyte, CD4+ T cells in BALF, and serum OA-specific IgE level as compared with the control group. YM decreased lymphocytes as compared with the control group. CD4+/CD8+ ratio in BALF from the CG and YM groups and serum OA-specific IgE level from the YM group didn't show any significant variation from the control group. Conclusion : CG alleviated asthmatic hyperreactivity of the immune system through CD4+ T cells and serum IgE. Further the study of this immune system modulating mechanism is expected.
