Overexpression of KiSS1 Induces the Proliferation of Hepatocarcinoma and Increases Metastatic Potential by Increasing Migratory Ability and Angiogenic Capacity

  • Liver cancer has a high prevalence, with majority of the cases presenting as hepatocellular carcinoma (HCC). The prognosis of metastatic HCC has hardly improved over the past decade, highlighting the necessity for liver cancer research. Studies have reported the ability of the KiSS1 gene to inhibit the growth or metastasis of liver cancer, but contradictory research results are also emerging. We, therefore, sought to investigate the effects of KiSS1 on growth and migration in human HCC cells. HepG2 human HCC cells were infected with lentivirus particles containing KiSS1. The overexpression of KiSS1 resulted in an increased proliferation rate of HCC cells. Quantitative polymerase chain reaction and immunoblotting revealed increased Akt activity, and downregulation of the G1/S phase cell cycle inhibitors. A significant increase in tumor spheroid formation with upregulation of β-catenin and CD133 was also observed. KiSS1 overexpression promoted the migratory, invasive ability, and metastatic capacity of the hepatocarcinoma cell line, and these effects were associated with changes in the expressions of epithelial mesenchymal transition (EMT)- related genes such as E-cadherin, N-cadherin, and slug. KiSS1 overexpression also resulted in dramatically increased tumor growth in the xenograft mouse model, and upregulation of proliferating cell nuclear antigen (PCNA) and Ki-67 in the HCC tumors. Furthermore, KiSS1 increased the angiogenic capacity by upregulation of the vascular endothelial growth factor A (VEGF-A) and CD31. Based on these observations, we infer that KiSS1 not only induces HCC proliferation, but also increases the metastatic potential by increasing the migratory ability and angiogenic capacity.

논 관개수중 영양물질의 함량변화와 천연공급량 (Changes of Nutrients Content and Natural Supplies by Irrigation Water during the Rice Cultivation)

  • 전라북도 진안군 마령면 평지리 일대 1.0ha의 논을 대상으로 벼농사 재배기간중 공급되는 관개수중에 함유된 영양물질과 오염물질의 함량 및 총 관개유량 측정을 통하여 논에 부하되는 영양물질과 중금속의 유입량을 시기별로 조사한 결과는 다음과 같다. 관개용수의 pH는 6.45${\sim}$8.34의 범위였으며, 전기전도도는 관개초기에만 $110{\mu}S/cm$ 수준을 유지하였으며 그밖에는 $90{\mu}S/cm$ 수준을 유지하였다. 전질소, 암모니아태질소 및 질산태질소의 함량은 각각 3.09${\sim}$12.46, 0.17${\sim}$1.34, 2.70~10.72mg/L 수준이었으며, 전인산의 함량은 0.09~0.22mg/L, orthophosphate는 검출되지 않았다. $Ca^{++}$는 0.89~5.12, $Mg^{++}$는 1.12~2.76, $Na^{+}$는 1.74~4.52, $Ka^+$는 1.34~4.18mg/L의 범위였으며 시기별로 큰 차이를 나타내지 않았다. 또한 $Cl^-$$SO_4\;^{2-}$는 각각 1.3~24.9, 3.5~20.3mg/L의 범위로 이앙초기에 높았다가 점차적으로 낮아지는 경향이었다. 중금속오염물질중 Cu와 Zn만이 각각 0.001~0.009, 0.007~0.091mg/L로 나타났을 뿐 Pb, Cd, Cr, Ni는 검출되지 않았다. 벼재배기간동안 1.0ha의 논에 $4,250m^3$의 관개수를 공급하였을 때 영양물질의 천연공급량은 전질소 29.68kg/1.0ha, 암모니아태질소 1.80kg/1.0ha, 질산태질소 24.57kg/1.0ha, 전인산 0.51kg/1.0ha으로 나타났다. 대표적인 양이온으로 $Ca^{++},\;Mg^{++},\;Na^+,\;K^+$는 각각 9.42, 7.09, 13.34, 9.08kg/1.0ha의 수준으로 공급되었으며 $Na^{+}>\;Ca^{++}>\;K^{+}>\;Mg^{++}$의 순서였다. 음이온으로 $Cl^-$$SO_4\;^{2-}$는 각각 40.27, 41.38kg/1.0ha으로 나타났다. 조사대상 6가지 중금속중 Cu와 Zn만이 관개수를 통하여 각각 0.17kg/1.0ha, 0.133kg/1.0ha 정도 논토양에 부하되었을 뿐 Pb, Cd, Cr, Ni은 검출되지 않았다.

유통 콜라겐 제품 중 유해중금속 함량 모니터링 (Monitoring of hazardous heavy metals in circulated collagen products in Korea)

  • 국내 유통 중인 콜라겐 제품 120건을 대상으로 중금속 4종(납, 카드뮴, 비소, 수은) 함량에 대해 조사하였다. 수은은 금아말감화법을 이용한 수은분석기로 분석하였고 납, 카드뮴, 비소는 ICP-OES를 이용하여 분석하였다. 검사 결과 납의 평균 함량은 0.097±0.055 mg/kg이었고 건강기능식품, 기타가공품, 음료류, 과·채가공품에서 각각 평균 0.108±0.052mg/kg, 0.084±0.053 mg/kg, 0.131±0.047 mg/kg, 0.149 mg/kg 농도로 검출되었다. 카드뮴의 평균 함량은 0.026±0.011 mg/kg이었으며 검출된 제품은 모두 건강기능식품이었다. 비소의 평균 함량은 0.097±0.048 mg/kg이었고 건강기능식품, 기타가공품, 과·채가공품에서 각각 평균 0.091±0.055 mg/kg, 0.133 mg/kg, 0.086 mg/kg 농도로 검출되었다. 수은의 평균 함량은 0.0025±0.0016 mg/kg으로 건강기능식품, 기타가공품, 과·채가공품, 캔디류에서 각각 평균 0.0012 mg/kg, 0.0028±0.0018 mg/kg, 0.0013 mg/kg, 0.0031 mg/kg 농도로 검출되었다. 중금속 기준·규격이 있는 음료류(납 0.3 mg/kg, 카드뮴 0.1 mg/kg) 및 캔디류(납 0.2 mg/kg)는 모두 기준 이하로 검출되어 적합하였다. 기준·규격이 없는 제품도 국내외 식품의 중금속 기준과 보고된 식품 중금속 함량과 비교하였을 때 비교적 안전한 수준이라고 판단되었다.

Safety and Effectiveness of Passeo-18 Lux Drug-Coated Balloon Catheter in Infrainguinal Endovascular Revascularization in the Korean Population: A Multicenter Post-Market Surveillance Study

  • Objective: To evaluate the safety and clinical outcomes of the Passeo-18 Lux drug-coated balloon (DCB) in endovascular revascularization procedures under real-world conditions in a Korean population with atherosclerotic disease of the infrainguinal arteries, including below-the-knee (BTK) arteries. Materials and Methods: Eight institutions in the Republic of Korea participated in this prospective, multicenter, single-arm, post-market surveillance study. Two hundred patients with Rutherford class 2-5 peripheral arterial disease and infrainguinal lesions suitable for endovascular treatment were competitively enrolled. Data were collected at baseline, the time of intervention, discharge, and 1-, 6-, 12-, and 24-month follow-up visits. The primary safety endpoint was freedom from major adverse events (MAE) within 6 months (except when limiting the time frame for procedure- or device-related mortality to within 30 days), and the primary effectiveness endpoint was freedom from clinically driven target lesion revascularization (CD-TLR) within 12 months after the procedure. Results: A total of 197 patients with 332 target lesions were analyzed. Two-thirds of the patients had diabetes mellitus, and 41.6% had chronic limb-threatening ischemia. The median target lesion length was 100 mm (interquartile range: 56-133 mm). Of the target lesions, 35.2% were occlusions, and 14.8% were located in the BTK arteries. Rate of freedom from MAE was 97.9% at 6 months, and the rate of freedom from CD-TLR was 95.0% and 92.2% at 12 and 24 months, respectively. Subgroup analysis of 43 patients and 49 target lesions involving the BTK arteries showed rate of freedom from MAE of 92.8% at 6 months and rates of freedom from CD-TLR of 88.8% and 84.4% at 12 and 24 months, respectively. Conclusion: The results of the present study, including the BTK subgroup analysis, showed outcomes comparable to those of other DCB studies, confirming the safety and effectiveness of Passeo-18 Lux DCB in the Korean population.

  • New electroluminescent materials base on anthracene chromophore, [9.10-bis(${\alpha}$-naph -thylethenyl) anthracene (${\alpha}$-BNA)] were newly synthesized. The anthracene derivatives with bulky substituent possessed high melting point and they gave stable amorphous films through vacuum - sublimation methods. Three types of electroluminescent devices were fabricated with double layer and triple layer structure : ITO/TPD/emission layer/MgAg, ITO/emission layer/ OXD-7 and ITO/ TPD/ emission layer/OXD-7/MgAg, respectively. In three types of devices with the emissive layer of ${\alpha}$-BNA, efficient orange electroluminescence was observed. In the triple layer device whit a emitting layer of 20 nm thickness , maximum luminance was about 10000 cd/ $m^2$ at an applied voltage of 10v and maximum external quantum efficiency was 1.0%.

유해오염물질에 급성 노출된 단각류 Monocorophium acherusicum의 지연 사망률에 관한 연구 (Delayed Mortality of Benthic Amphipods Monocorophium acherusicum Exposed to Various Pollutants in Seawater(Cd, Cu, Hg, TBT, Ammonia and Phenanthrene))

  • 다양한 유해오염물질에 급성 노출된 단각류 Monocorphium acherusicum의 노출 기간 이후에 발생하는 지연 사망(latent mortality)이 반수치사농도(LC5O)산출에 어떤 영향을 미치는 지를 규명하기 위한 일련의 실험이 수행되었다. 본 연구에서는 실험생물을 카드뮴, 구리, 수은과 같은 중금속, tributyltin(TBT), 암모니아 그리고 방향성탄화수소인 phenanthrene에 각각 96시간 동안 노출시킨 후 깨끗한 해수에 옮겨 다시 6일 동안 배양하면서 사망률을 조사하였다. 실험결과 구리, TBT, 암모니아, phenanthrene과 같은 물질에 노출된 M. acherusicum의 사망률은 노출이 끝난 이후에도 계속적으로 증가하는 지연 사망이 관찰되었으며, 이에 따라 기존의 방법_으로 산출된 96-h LC50보다 지연 사망을 고려한 새로운 LC50이 크게 낮아지는 경향이 관찰되었다. 지연사망률을 고려하지 않은 기존의 독성시험 결과는 지연 사망의 영향을 반영하지 못하므로 실제 현장에서 발생할 수 있는 오염물질의 영향을 과소평가 할 가능성이 있다. 따라서 지연 사망률에 대한 고려는 실제 현장 개체군에 대한 유해오염물질의 영향을 보다 정화하게 예측하는 데에 활용될 수 있을 것이다.

Preparation of Novel Fused Ring Spiro[benzotetraphene-fluorene] Derivatives and Application for Deep-Blue Host Materials

  • A series of novel fused-ring spiro compounds, spiro[benzo[ij]tetraphene-7,9'-fluorene] (SBTF) derivatives containing an end-capping aryl substituent at both the C3 and C10-positions hasbeen designed and synthesized via multi-step Suzuki coupling reactions. 3-(1-Naphthyl)-10-phenylSBTF (1N-PSBTF), 3-(2-naphthyl)-10-phenylSBTF (2N-PSBTF) and 3-[4-(1-naphthyl)phenyl]-10-phenylSBTF (NP-PSBTF) showed improved glass transition temperatures ($T_g$) with good thermal stability. Their photophysical, electrochemical, and electroluminescent properties were investigated and were used to construct blue organic light emission diodes (OLEDs). The typical OLED devices showed excellent performance; the NP-PSBTF-based device exhibited highly efficient deep blue-light emission with a maximum efficiency of 5.27 cd/A (EQE, 4.63%) with CIE (x = 0.133, y = 0.144). According to these characteristics, these deep-blue light emitting materials have sufficient potential for fluorescent OLED applications.

MicroRNA-203 As a Stemness Inhibitor of Glioblastoma Stem Cells

  • Glioblastoma stem cells (GBM-SCs) are believed to be a subpopulation within all glioblastoma (GBM) cells that are in large part responsible for tumor growth and the high grade of therapeutic resistance that is so characteristic of GBM. MicroRNAs (miR) have been implicated in regulating the expression of oncogenes and tumor suppressor genes in cancer stem cells, including GBM-SCs, and they are a potential target for cancer therapy. In the current study, miR-203 expression was reduced in $CD133^+$ GBM-SCs derived from six human GBM biopsies. MicroRNA-203 transfected GBM-SCs had reduced capacity for self-renewal in the cell sphere assay and increased expression of glial and neuronal differentiation markers. In addition, a reduced proliferation rate and an increased rate of apoptosis were observed. Therefore, miR-203 has the potential to reduce features of stemness, specifically in GBM-SCs, and is a logical target for GBM gene therapy.

Gamma radiation shielding properties of poly (methyl methacrylate) / Bi2O3 composites

  • This work investigated the gamma-ray shielding performance, and the physical and mechanical properties of poly (methyl methacrylate) (PMMA) composites embedded with 0-44.0 wt% bismuth trioxide (Bi2O3) fabricated by the fast ultraviolet (UV) curing method. The results showed that the addition of Bi2O3 had significantly improved the gamma shielding ability of PMMA composites. Mass attenuation coefficient and half-value layer were examined using five gamma sources (Cs-137, Ba-133, Cd-109, Co-57, and Co-60). The high loading of Bi2O3 in the PMMA samples improved the micro-hardness to nearly seven times that of the pure PMMA. With these enhancements, it was demonstrated that PMMA/Bi2O3 composites are promising gamma shielding materials. Furthermore, the fast UV curing exerts its great potential in significantly shortening the production cycle of shielding material to enable rapid manufacturing.

Combination Therapy of Lactobacillus plantarum Supernatant and 5-Fluouracil Increases Chemosensitivity in Colorectal Cancer Cells

  • Colorectal cancer (CRC) is the third most common cancer in the world. Although 5-fluorouracil (5-FU) is the representative chemotherapy drug for colorectal cancer, it has therapeutic limits due to its chemoresistant characteristics. Colorectal cancer cells can develop into cancer stem cells (CSCs) with self-renewal potential, thereby causing malignant tumors. The human gastrointestinal tract contains a complex gut microbiota that is essential for the host's homeostasis. Recently, many studies have reported correlations between gut flora and the onset, progression, and treatment of CRC. The present study confirms that the most representative symbiotic bacteria in humans, Lactobacillus plantarum (LP) supernatant (SN), selectively inhibit the characteristics of 5-FU-resistant colorectal cancer cells (HT-29 and HCT-116). LP SN inhibited the expression of the specific markers CD44, 133, 166, and ALDH1 of CSCs. The combination therapy of LP SN and 5-FU inhibited the survival of CRCs and led to cell death by inducing caspase-3 activity. The combination therapy of LP SN and 5-FU induced an anticancer mechanism by inactivating the Wnt/β-catenin signaling of chemoresistant CRC cells, and reducing the formation and size of colonospheres. In conclusion, our results show that LP SN can enhance the therapeutic effect of 5-FU for colon cancer, and reduce colorectal cancer stem-like cells by reversing the development of resistance to anticancer drugs. This implies that probiotic substances may be useful therapeutic alternatives as biotherapeutics for chemoresistant CRC.
