Study on Anomalous Polarographic Behavior of Cd (II) in Tartrate Solution (타르타르酸염支持溶液에서의 Cd (II) 폴라로그람의 異常波에 關한 硏究)

  • The polarograms of Cd(II) in 0.15M tartrate solution were investigated in the range of pH values from 6.0 to 12.6 at 20$^{\circ}and 25$^{\circ}C. Up to pH value of 7.8, the limiting current and the halfwave potential were found to be constant, and when compared with the nitrate solution of the same $Cd^{++}$ concentration at the same ionic strength, the limiting current decreased by 28% and the halfwave potential shifted by -0.05 volt. The values of limiting current sharply decreased as the pH value exceeded 8.2 and the minimum values appeared at the pH values of 11.2~11.4. The values of halfwave potential gradually decreased as the pH value increased over 8.2 and the value of -0.78 volt was obtained at the pH value of 12.6. Possible mechanisms of electrode reaction were suggested and the anomalous behavior of reduction waves of Cd(II) in tartrate solution up to pH value of 9.4 was discussed.

  • The crystal structure of the title complex, Cd(SCN)₂{CH/sub 6/H/sub 10/(NH₂)₂}₂(1) has been analyzed by X-ray single crystallography. The complex 1 crystallizes in the monoclinic system P2₁/ space group with a=11.842(2), b=7.926(2), c=11.291(2) Å, β=106.73(3)°V=1014.8(4)Ų, Z=2, R₁=0.0518 and ωR₂=0.1315 for 1775 independent reflections. The central Cd(II) atom of this com-plex has a slightly distorted octahedral coordination geometry, with the 1,2-Diaminocyclohexane ligands functioning as an N,N'-bidentate and the thiocyanate ligands bonding through the sulfur atom in a trans arrangement.

Three Crystal Structures of Dehydrated $Cd^{2+}$ and $Rb^+$ Exchanged Zeolite A, $Cd_xRb_{12-2x}-A,$ x=4.0, 5.0 and 5.95

  • Three crystal structures of dehydrated Cd(II) and Rb(I) exchanged zeolite A, $Cd_{4.0}Rb_{4.0}-A (a = 12.204(3) {\AA}), Cd_{5.0}Rb_{2.0}-A (a = 12.202(1) {\AA}),$ and $Cd_{5.95}Rb_{0.1}-A (a = 12.250(2) {\AA}),$ have been determined by single-crystal X-ray diffraction techniques. Their structures were solved and refined in the cubic space group Pm3m at $21(1)^{\circ}C.$ All crystals were ion exchanged in flowing streams of mixed $Cd(NO_3)_2·4H_2O$ and $RbNO_3$ aqueous solution with total concentration of 0.05 M. All crystals were dehydrated at ca. $450^{\circ}C$ and $2×10^{-6}$ Torr for 2 days. In all of these structures, $Cd^{2+}$ ions are found on threefold axes, each nearly at the center of a 6-oxygen ring. The first three $Rb^+$ ions per unit cell preferentially associate with 8-oxygen rings, and additional $Rb^+$ ions, if present, are found on threefold axes in the large cavity. The final $R_1$ and $R_2$ values for the three structures are 0.087 and 0.079, 0.059 and 0.067, and 0.079 and 0.095, respectively.

The Protective Role of Gleditsiae fructus against Streptococcus pneumoniae (폐렴 구균에 대한 조협의 보호 역할 연구)

  • Natural products have been used to mitigate the effects of cancer and infectious diseases, as they feature diverse bioactivities, such as antioxidant, antibacterial, anti-inflammatory, and immunomodulatory effects. Here, we chose 10 natural products that are well-known as pulmonary enhancers and investigated their bactericidal effects on Streptococcus pneumoniae. In the disk diffusion assay, the growth of S. pneumoniae was significantly regulated by G. fructus treatment regardless of extraction method used. We first adopted spraying as a novel delivery method for G. fructus. Interestingly, mice exposed to G. fructus three times a day for 2 weeks were resistant to S. pneumoniae intranasal infection (shown both through body weight loss and survival rates compared to the control group). Moreover, we confirmed that exposure to G. fructus regulated the colonization of the bacteria despite the sustained inflammation in the lung after exposure to S. pneumoniae, indicating that migrated inflammatory immune cells may involve a host defense mechanism against pulmonary infectious diseases. While a similar number of granulocytes (CD11b+Ly6C+Ly6G+), neutrophils (CD11b+Ly6CintLy6G+), and monocytes (CD11b+Ly6CintLy6G-) were found between groups, a significantly increased number of alveolar macrophages (CD11b+CD11chiF4/80+) was detected in BAL fluids of mice pre-exposed to G. fructus at 5 days after S. pneumonia infection. Taken together, our data suggest that this usage of G. fructus can induce protective immunity against bacterial infection, indicating that facial spray may be helpful in enhancing the defense mechanism against pulmonary inflammation and in evaluating the efficacy of natural products as immune enhancers against respiratory diseases.

Heat-treatment Effects of CdS Thin Films Fabricated by Thermal Evaporation Method (진공증착법으로 제작한 CdS 박막의 열처리효과)

  • 열증착법에 의해서 온도 85$^{\circ}C$인 glass기판 위에 CdS 박막을 제작하였다. 두께가 200nm정도로 측정된 CdS박막은 공기 중에서 온도 25$0^{\circ}C$-55$0^{\circ}C$범위에서 각각 30분간 열처리 되었으며 이들 시료에 대하여 4-point probe, XRD, SEM, UV-Spectrophotometer 및 광발광 측정으로 전기적 광학적 특성을 조사하였다. 이들의 일련의 실험값은 열처리 온도 37$0^{\circ}C$ 근처에서 구조의 변화를 보여주었는데, 열처리 온도에 대한 비저항아니 XRD, SEM 의 측정은 cubic로부터 hexagonal구조로의 변환을 나타내었다. 특히 상온에서 측정한 광발광에서 green edge emission(GEE)피이크가 2.42eV를 나타내었는데 이 때의 발광 중심은 열처리할 때 생긴 S-vacancy에 보상된 산소로 이루어진 'CdO'의 악셉터준위에 기인하는 것으로 해석되며 그 이온화 에너지는 약 0.16eV이었다.

Differential Roles of Lung Dendritic Cell Subsets Against Respiratory Virus Infection

  • Respiratory viruses can induce acute respiratory disease. Clinical symptoms and manifestations are dependent on interactions between the virus and host immune system. Dendritic cells (DCs), along with alveolar macrophages, constitute the first line of sentinel cells in the innate immune response against respiratory viral infection. DCs play an essential role in regulating the immune response by bridging innate and adaptive immunity. In the steady state, lung DCs can be subdivided into $CD103^+$ conventional DCs (cDCs), $CD11b^+$ cDCs, and plasmacytoid DCs (pDCs). In the inflammatory state, like a respiratory viral infection, monocyte-derived DCs (moDCs) are recruited to the lung. In inflammatory lung, discrimination between moDCs and $CD11b^+$ DCs in the inflamed lung has been a critical challenge in understanding their role in the antiviral response. In particular, $CD103^+$ cDCs migrate from the intraepithelial base to the draining mediastinal lymph nodes to primarily induce the $CD8^+$ T cell response against the invading virus. Lymphoid $CD8{\alpha}^+$ cDCs, which have a developmental relationship with $CD103^+$ cDCs, also play an important role in viral antigen presentation. Moreover, pDCs have been reported to promote an antiviral response by inducing type I interferon production rather than adaptive immunity. However, the role of these cells in respiratory infections remains unclear. These different DC subsets have functional specialization against respiratory viral infection. Under certain viral infection, contextually controlling the balance of these specialized DC subsets is important for an effective immune response and maintenance of homeostasis.

Intracellular cAMP-modulated Gate in Hyperpolarization Activated Cation Channels

  • Hyperpolarization-activated nonselective cation channels (HCNs) play a pivotal role in producing rhythmic electrical activity in the heart and the nerve cells. In our previous experiments, voltage-dependent $Cd^{2+}$ access to one of the substituted cysteines in S6, T464C, supports the existence of an intracellular voltage-dependent activation gate. Direct binding of intracellular cAMP to HCN channels also modulates gating. Here we attempted to locate the cAMP-modulated structure that can modify the gating of HCN channels. SpHCN channels, a sea urchin homologue of the HCN family, became inactivated rapidly and intracellular cAMP removed this inactivation, resulting in about eight-fold increase of steady-state current level. T464C was probed with $Cd^{2+}$ applied to the intracellular side of the channel. We found that access of $Cd^{2+}$ to T464C was strongly gated by cAMP as well as voltage. Release of bound $Cd^{2+}$ by DMPS was also gated in a cAMP-dependent manner. Our results suggest the existence of an intracellular cAMP-modulated gate in the lower S6 region of spHCN channels.

The Experimental Study on the Suppression Effect of Asthma and Immune Response Improvement of Platycodi Radix Herbal-acupuncture (길경약침(桔梗藥鍼)의 천식억제(喘息抑制) 및 면역조절효과(免疫調節效果)에 대(對)한 실험적(實驗的) 연구(硏究))

  • Objectives : The aim of this study was to investigate the effect of Asthma-depression and Immunoregulation with PR-HAS(Herbal-acupuncture with Platycodi Radix infusion solution) injection at Joksamni(St36) on ovalbumin-induced asthma in mice. Methods : C57BL/6 mice were sensitized and challenged with OVA(ovalbumin) for 12 weeks(once a week). The experimental group(OVA-PR-HA) wase treated with concentrations(1%) of PR-HAS at Joksamni(St36) for the later 8 weeks(3times/week). The second experimental group(OVA-Needle prick) was treated with Needle-Prick at Joksamni(St36) for the later 8 weeks(3times/week). Results : 1. The weight and total cells in the mice lung treated with PR-HA decreased significantly compared with that of control group. 2. Total leukocytes and eosinophils in BALF of the mice group treated with PR-HA decreased remarkably compared with those of control group. 3. The sticking of collagen on histological analysis of lung sections, the mice group treated with PR-HA decreased significantly compared with those of control group. 4. The concentrations of IL-4, IL-5, IgE in BALF, and IL-4, IL-5, IL-13 in serum of the mice group treated with PR-HA decreased significantly compared with those of control group. 5. The number of $Gr-1^+/CD11b^+\;and\;CD11b^+$ cells in the lungs of the mice group treated with PR-HA decreased significantly compared with that of control group. 6. The numbers of $CCR3^+\;cells,\;CD4^+\;cells\;and\;CD8^+\;cells$ in the lungs, and $CD3e^+/CD69^+$ in the lungs of the mice group treated with PR-HA decreased significantly compared with those of control group. 7. The mRNA expression of ${\beta}-actin,\;TNF-{\alpha}$, IL-4, IL-5,IL-13 in the mice group treated with PR-HA with RT-PCR decreased significantly compared with those of control group. Conclusion : The concentrations of IL-4, IL-5, IgE in BALF, and IL-4, IL-5, IL-13 in serum of the mice group treated with PR-HA decreased significantly compared with those of control group. The number of $Gr-1^+/CD11b^+\;and\;CD11b^+$ cells in the lungs of the mice group treated with PR-HA decreased significantly compared with that of control group. The numbers of $CCR3^+\;cells,\;CD4^+\;cells\;and\;CD8^+\;cells$ in the lungs, and $CD3e^+/CD69^+$ in the lungs of the mice group treated with PR-HA decreased significantly compared with those of control group. Doping Effect of CdO on the Oxidation of Carbon Monoxide over CdO-${\alpha}-Fe_2O_3$System (CdO-${\alpha}-Fe_2O_3$촉매상에서 일산화탄소의 산화반응에 대한 CdO의 첨가 효과)

  • The oxidation reaction of CO on the catalysts 4 mol%, 8 mol%, and 12 mol% Cd-doped ${\alpha}-Fe_2O_3$ is individually investigated. Regardless of Cd doping level, over-all reaction order for the oxidation of CO is 1.5; the first order with respect to CO and the one-half order with respect to $O_2$. Over the temperature range of 350∼$460^{\circ}C$, the activation energy for CO oxidation is 10.10∼11.30Kcal/mol. From the agreement between the kinetic data and conductivity measurements, the reaction mechanism is suggested. Especially from the effect of Cd doping, the fact that catalytic activity of ${\alpha}-Fe_2O_3$ is due to the excitation of electrons which are traped on oxygen vacancy is found, and the adsorption sites for reactant molecules are found.

A Study on the Preparation of CdS Doped $SiO_2$ Glass Coating Films by Sol-Gel Method (졸-겔법에 의한 CdS 분산 $SiO_2$ Glass 코팅막의 제조에 관한 연구)

  • CdS doped SiO2 glass coating films which are good candidates for the nonlinear optical materials were prepared by the Sol-Gel method. TEOS, C2H5OH, H2O and HCl were used as starting materials to obtain SiO2 matrix solutions. Then Cd(NO3)2.2H2O and CS(NH2)2 were dissolved into the SiO2 matrix solutions. Coating was performed several times in order to increase the thickness of coated film by the dip-coating method. Then heat treatments were carried out to control the size of CdS microcrystals doped in SiO2 glass matrix with respect to temperatures and times. CdS-doped SiO2 transparent coating films were successfully obtained. CdS crystals were changed from cubic to hexagonal type about $600^{\circ}C$.
