The Effect of $Cd^{2+}$ on Stomatal Apertures of Epidermal Strips in Commelina communis L. ($Cd^{2+}$이 닭의장풀의 기공개폐에 미치는 영향)

  • The effect of $Cd^{2+}$ on stomatal apertures of epidermal strips and intact leaves in Commelina communis L. was investigated. Cadmium stimulated stomatal opening. The stomata, treated with 100 $\mu$M $Cd^{2+}$ opened to a degree of about 8.38 fm, but the stomata, treated with no cadmium opened to 3.74 ${\mu}{\textrm}{m}$. In order to show that there was no mechanical or osmotic impediment preventing the stomata in the epidermal strips, salicylic acid (SA) and abscisic acid (ABA) were used. The treatment of 100 $\mu$M SA and 1 $\mu$M ABA inhibited 14% and 28% of stomatal opening, respectively. Other heavy metals such as $Al^{3+} and Ag^{2+}$ were also used to investigate the effect of the stomatal apertures. The treatment of $Al^{3+} and Ag^{2+}$ also stimulated 19% and 41% of stomatal opening. To understand how cadmium open stomata, the effect of cadmium on the K+ influx into the epidermal strips was investigated. $Cd^{2+}$, SA, ABA inhibited 98%, 28%, 34% of K+ uptake respectively. 3-weeks old Commelina was transferred to and grown in Hoagland solution (0,5 mM $Cd^{2+}$, 10 mM $Cd^{2+}$) for 4 days and stomatal conductance were measured. The treatment of 5 mM $Cd^{2+}$ and 10 mM $Cd^{2+}$ showed about 51% and 70% inhibition of stomatal conductance, respectively Therefore, it could be concluded that stomata in epidermal strips and intact leaves behaved differently and cadmium- stimulated stomatal opening was due to the result of cadmium uptake into the epidermal strips instead of K+. Properties of CdS Thin Films Prepared by Chemical Bath Deposition as a Function of Thiourea/CdAc2 Ratio in Solution (CBD법으로 제작된 CdS 박막의 thiourea/CdAc2 농도비에 따른 특성)

  • In this paper CdS thin films, which were widely used window layer of the CdS/CdTe and the CdS/$CuInSe_2$ heterojunction solar cell, were grown by chemical bath deposition, which is a very attractive method for low-cost and large-area solar cells, and the structural, optical and electrical properties of the films was studied. As the thiourea/$CdAc_2$ mole ratio was increased, the deposition rate of CdS films prepared by CBD was increased due to increasing reaction velocity in solution and the optical bandgap was increased at higher thiourea/$CdAc_2$ mole ratio due to larger grain size and continuous microstructure. The minimum resistivity of the films was at thiourea/$CdAc_2$ mole ratio of 3.

  • CD4 and CD8 T cells are key players in the immune response against both pathogenic infections and cancer. CD4 T cells provide help to CD8 T cells via multiple mechanisms, including licensing dendritic cells (DCs), co-stimulation, and cytokine production. During acute infection and vaccination, CD4 T cell help is important for the development of CD8 T cell memory. However, during chronic viral infection and cancer, CD4 helper T cells are critical for the sustained effector CD8 T cell response, through a variety of mechanisms. In this review, we focus on T cell responses in conditions of chronic Ag stimulation, such as chronic viral infection and cancer. In particular, we address the significant role of CD4 T cell help in promoting effector CD8 T cell responses, emerging techniques that can be utilized to further our understanding of how these interactions may take place in the context of tertiary lymphoid structures, and how this key information can be harnessed for therapeutic utility against cancer.

Sensory Evaluation and Bioavailability of Red Ginseng Extract(Rg1, Rb1) by Complexation with ${\gamma}$-Cyclodextrin (${\gamma}$-cyclodextrin으로 포접한 홍삼추출물의 관능평가 및 Rg1, Rb1의 생체이용율)

  • In order to reduce the bitter taste and improve the bioavailability of red ginseng extract(RGE), inclusion complexes (RGE-CD) of the extract with ${\alpha}-,\;{\beta}-,\;{\gamma}$-cyclodextrin were prepared and studied for their sensory quality and bioavailability compared to RGE. By complexation, the bitter taste-reducing efficacies of ${\alpha}$-CD and ${\beta}$-CD were much lower than that of ${\gamma}$-CD. In comparative sensory analysis for the bitter taste, RGE-${\gamma}$-CD10, prepared using 10%(w/w) of ${\gamma}$-CD, showed a score of 1.93(decreased by about 78%) compared to RGE as the control. In addition, in sensory analysis for flavor, RGE-${\gamma}$-CD10showed a score of 5.60. Upon increasing the amount of ${\gamma}$-CD to 15%(w/w) and 20%(w/w), respectively, the bitter taste of RGE-${\gamma}$-CD was removed and the flavor of RGE disappeared(scores of 2.67 and 1.67, respectively). Therefore RGE-${\gamma}$-CD10 was chosen as an optimum. The same dosages of RGE and RGE-${\gamma}$-CD10 were orally administered to SD(Sprague-Dawley) rats on a saponin basis, and the plasma concentrations of ginsenoside Rg1 and Rb1 were measured over time to estimate the average AUC(area under the plasma concentration versus time curve) of the ginsenosides. After the oral administration, there were no significant differences in the AUC values of the RGE and RGE-${\gamma}$-CD 10 groups for ginsenoside Rg1. However, AUC values for ginsenoside Rb1 were $25.8{\mu}g{\cdot}hr/mL$ in the RGE group and $81.5{\mu}g{\cdot}hr/mL$ in the RGE-${\gamma}$-CD 10 group, respectively. Therefore, the bioavailability of ginsenoside Rb1 in the RGE-${\gamma}$-CD 10 group was significantly higher by up to 315% compared with that in the RGE group(p = 0.0029). These results show that the bitter taste of RGE can be simultaneously removed by the complexation of RGE and ${\gamma}$-CD(RGE-${\gamma}$-CD) along with increased bioavailability.

  • The standardized activated sludge for the biodegradation test of anion surfactants has been produced from the collected microorganisms in the soil and the wastewaters treatment plant. The activated sludge was kept under control of the pH, dissolved oxygen, microorganisms and inoculated the basal medium flasks with LAS and LAS mixed with heavy metals [Cd(II), Cu(II), Zn(II)]. Based of results, the inhibition effect(%) of heavy metals in LAS biodegradation were 1. All 1% when LAS 30mg/l-Cd(II), Cu(II) and Zn(II) 0.1mg/l, respectively 2. All 1${\sim}$10% when LAS 30mg/l-Cd(II), Cu(II) and Zn(II) 1mg/l, respectively 3. All 10${\sim}$40% when LAS 30mg/l-Cd(II), Cu(II) and Zn(II) 10mg/l, respectively 4. All 30${\sim}$65% when LAS 30mg/l-Cd(II), Cu(II) and Zn(II) 100mg/l, respectively And toxicity order of heavy metals to the microorganisms in LAS biodegradation were Cd>Cu>Zn in low concentration(0.1${\sim}$1mg/l)and Cd>Zn>Cu in high concentration(10${\sim}$100mg/l).

Soluble CD30: A Possible Serum Tumor Marker for Primary Effusion Lymphoma

  • Background: The serum level of soluble CD30 (sCD30) is known to be increased with several lymphomas and to correlate with prognosis. Primary effusion lymphoma (PEL) is a highly aggressive malignant lymphoma with poor prognosis, but the existence and significance of sCD30 in PEL have not yet been investigated in detail. Objectives: Since the membrane type of CD30 is frequently expressed on the surface of PEL cells, we compared the expression of the membrane type of CD30 and the production of sCD30 among PEL cell lines as well as other lymphomas. Methods: The expression of surface CD30 in various lymphoma cell lines was analyzed with flow cytometry ans sCD30 was quantified by ELISA. Results: Both surface and sCD30 were detected on PEL cell lines as well as on Hodgkin's lymphoma and adult T-cell leukemia/lymphoma cell lines. Surface CD30 and sCD30 levels of each cell lines correlated with each other. Conclusion: The serum level of sCD30 appear to be a useful biological tumor marker for the diagnosis and management of CD30-positive PEL.

CD44 and CD133 as Cancer Stem Cell Markers for Gastric Cancer

  • Purpose: Currently, the two most influential gastric stem cell marker candidates are CD44 and CD133. The aim of this study was to make a comparison and determine the appropriate marker for use in gastric cancer stem cell research. Materials and Methods: We analyzed the expressions of CD44, CD133, and CD24 from the gastric cancer cell lines MKN45, MKN74, KATO-III, NCI-N87, SNU-1, SNU-216, SNU-601, SNU-638, and SNU-688 using flow cytometry. In addition, we measured the change in viability after applying 5 fluorouracil (5-FU) to the MKN45, MKN74, KATO-III, and NCI-N87 cell lines using a Cell Counting Kit 8. Results: CD133 expression was above moderate in the KATO-III, SNU-216, SNU-601 cell lines, whereas it was below 1% in the remaining cell lines. CD44 was expressed at levels above 5% in all gastric cancer cell lines. The effect of 5-FU on viability and CD133 or CD44 expression in the cell lines were not related. Conclusions: Expression of CD133 positive cells was insufficient in the gastric cancer cell lines. Therefore, of the cell lines tested, CD44 was the most appropriate tumor maker for research on gastric cancer stem cells.

The effects of zeatin on Cd2+ -induced physiological toxicities in Commelina communis L (Zeatin이 Cd2+에 의한 닭의장풀의 생리적 독성에 미치는 영향)

  • Three-week old Commelina communis was transferred and grown in Hoagland solution containing $100{\mu}M\;Cd^{2+},\;100{\mu}M\;Cd^{2+}+100{\mu}M\;kinetin,\;100{\mu}M\;Cd^{2+}+100{\mu}M\;zeatin\;and\;100{\mu}M\;Cd^{2+},\;200{\mu}M$ zeatin for 7 days, and then a number of physiological activities were investigated. In control, the length of the stem of plants was increased to 4.7cm, but in $Cd^{2+},\;Cd^{2+}+kinetin,\; Cd^{2+}+100{\mu}M\;zeatin\;and\;Cd^{2+}+200{\mu}M$ zeatin treatments, the growth of plants were increased to 1.5cm, 2.1cm, 3.9cm and 4.3 em, respectively. In the treatments of $Cd^{2+},\;Cd^{2+}+kinetin,\;Cd^{2+}+100{\mu}M\;zeatin\;and\; Cd^{2+}+200{\mu}M$ zeatin, total chlorophyll contents were reduced to $26\%,\;24\%,\;15\%\;and\;3\%$, respectively, on the contrast to the control. In chlorophyll fluorescence experiments, Fv/Fm ratios were also reduced to $44\%,\;21\%,\;17\%\;and\;5\%$ in the light intensity of $2100{\mu}Mmole\;E\;m^{-2}s^{-1}\;by\;Cd^{2+},\;Cd^{2+}+kinetin,\;Cd^{2+}+$100{\mu}M\;zeatin\;and\;Cd^{2+}+200{\mu}M$ zeatin treatments on the contrast to the control. Water stresses were increased to 2.6, 1.7 and 1.2 times by $Cd^{2+},\; Cd^{2+}+kinetin\;and\;Cd^{2+}+{\mu}M$ zeatin. On the other hand, combination of $Cd^{2+}+200{\mu}M$ zeatin reduced water stress to $0.12\%$. In $Cd^{2+}$ accumulation experiments $Cd^{2+}$transports were inhibited to $33\%\; 48\%\;and\;70\%\;by\;Cd^{2+}+kinetin,\;Cd^{2+}+100{\mu}M\;zeatin\;and\;Cd^{2+}+200{\mu}M$ zeatin. Therefore, it could be concluded that zeatin clearly reduced the toxicities of $Cd^{2+}$ by reducing the absorption of $Cd^{2+}$.

The Effects of Indole Acetic Acid (IAA) on Cd$^{2+}$_induced Physiological Toxicities in Commelina communis L. (Cd$^{2+}$ 에 의한 닭의장풀의 식물 독성에 Indole acetic acid가 미치는 영향)

  • 3-weeks old Commelina was transferred to and grown in Hoagland solution (($\pm 100 \mu M \;Cd^{2+}, 100 \mu M \;Cd^{2+}+10 \mu M\;IAA,100 \mu M\; Cd^{2+}+100 \mu M \; IAA,100 \mu M \; Cd^{2+} +1 \mu M \; IAA)$) for three weeks and then a number of physiological activities was investigated. In the control the length of stem was increased to 7 cm after 3 weeks, but in the treatment of $Cd^{2+}$, 2.0 cm was grown. In cases of IAA with $Cd^{2+}$, the growth of the plants was increased to 3.7 cm, 5.0 cm and 3.3 cm in 100 $\mu$M, 10 $\mu$M and 1 mM IAA respectively, Cadmium stimulated stomatal opening. The stomata, treated with $Cd^{2+}$ opened to n degree of about 6.1 $\mu\textrm{m}$, but the stomata, treated with no cadmium opened to 4.5 $\mu\textrm{m}$. In the treatment of various concentation of IAA with cadmium the stomata opend to about 1 $\mu\textrm{m}$ more than that of cadmium alone. Cadmium reduced total chlorophyll content up to 13% for 3 weeks. In the treatment of $Cd^{2+}$ + IAA (10 $\mu$M and 100 $\mu$M) the clear change of total chlorophyll content was not observed, but in the addition of 1 mM IAA to $Cd^{2+}$ reduced the total chlorophyll content to about 9%. $Cd^{2+}$ reduced to 9% and 11% of Fv/Fm after two and three weeks respectively. In cases of IAA with $Cd^{2+}$, there were no clear changes of Fv/Fm. Cadmium reduced water potential to 67% after 3 weeks incubation, but in cases of IAA with $Cd^{2+}$, water potential was not clearly changed. Therefore, it could be concluded that the treatment of IAA showed clear alleviation of $Cd^{2+}$-induced several physiological toxicities.

The Study on Absorption of $Cd^{2+}$ and $Pb^{2+}$ by Persicaria thunbergii and Rumex crispus, and Their Phytochelatin (고마리와 소리쟁이에 의한 $Cd^{2+}$$Pb^{2+}$의 흡수 및 phytochelatin에 관한 연구)

  • When Persicaria thunbergii and Rumex crispus were treated with Cd($NO_3$)$_2$ and Pb($NO_3$)$_2$ of 5 or 10 mM for 5 days, the amount of bioaccumulation of $Pb^{2+}$ in the leaf of P. thunbergii was 2.87-8.08$\mu\textrm{g}$/g and that of $Cd^{2+}$ was 0.82-2.79$\mu\textrm{g}$/g. In the case of P. thunbergii, the concentration of $Pb^{2+}$ in the leaf was higher than that of $Cd^{2+}$. On the other hand, in R. crispus, the concentration of $Cd^{2+}$ and $Pb^{2+}$ were similar as follows ; 1.49$\mu\textrm{g}$/g in $Cd^{2+}$ 5mM, 2.90$\mu\textrm{g}$/g in Cd2+ 10mM, 1.83$\mu\textrm{g}$/g in $Pb^{2+}$ 5mM and 2.73$\mu\textrm{g}$/g in $Pb^{2+}$ 10mM. The remaining rate of heavy metals and the variation of pH in the cultured soil decreased as compared with control (100 % and pH 6.48) after 5 days as follows; to 77.l% and pH 6.39 in $Cd^{2+}$ 5mM, 90.2% and pH 5.79 in $Cd^{2+}$ 10 mM, 81.1% and pH 6.00 in $Pb^{2+}$ 5mM, and 85.7% and pH 5.80 in $Pb^{2+}$ 10 mM. The result of size exclusion chromatography, several phytochelatins were seperated from the extract of the leaf of both plants treated with heavy metals. The molecular mass of these phytochelatins were estimated as follows; in the case of P. thunbergii, about 4,300-8,600 da by $Cd^{2+}$ and about 3,200-9,700 da by $Pb^{2+}$, and in R. crispus, about 4,300 da by $Cd^{2+}$ and about 3,200-7,500 da by $Pb^{2+}$. In addition, $A_{254}$ of these phytochelatins were higher than $A_{280}$. [Phytochelatin, Persicaria thunbergii, Rumex crispus]
