Insulin-like Growth Factor-I Regulates the FAT/CD36 Expression in C2C12 Skeletal Muscle Cells (C2C12 골격근 세포에서 FAT/CD36 발현 조절에 있어 Insulin-like growth factor-I이 미치는 영향)

  • Fatty acid transporters are key mediators of skeletal muscle lipid metabolism. Several protein groups have been implicated in cellular long-chain fatty acid uptake or oxidation, including fatty acid transporter proteins (FATPs), the plasma membrane fatty acid-binding protein (FABPpm), and the fatty acid translocase (FAT/CD36). FAT/CD36 is highly expressed in skeletal muscle and known to be regulated by various factors such as exercise and hormones. Insulin-like growth factor-I (IGF-I) is a well-known regulator of skeletal muscle cells. However, it has not been studied whether there is any interaction between IGF-I and FAT/CD36 in skeletal muscle cells. In this study, the effects of IGF-I treatment on FAT/CD36 induction were examined. Differentiated C2C12 cells were treated with 20 ng/ml of IGF-I at different time points. Treatment of C2C12 cells with IGF-I resulted in increased FAT/CD36 mRNA and protein expression. After 24 and 48 hr of IGF-I treatment, FAT/CD36 mRNA increased 89% and 24% respectively. The increase of both proteins returned to the control level after 72 hr of IGF-I treatment, suggesting that the FAT/CD36 gene is regulated pretranslationally by IGF-I in skeletal muscle cells. These results suggest that IGF-I can regulate the expression of FAT/CD36 in skeletal muscle cells. In conclusion, IGF-I induces a rapid transcriptional modification of the FAT/CD36 gene in C2C12 skeletal muscle cells and has modulating effects on fatty acid uptake proteins as well as oxidative proteins.

Relationships between Speciation of Heavy Metals in Soil and Water Dropwort (Oenanthe javanica DC.) Cultivated near Industrial Complex (토양내 중금속 존재형태와 미나리중 함량과의 관계)

  • This experiment was conducted to investigate heavy metal speciation and bioavailability from soil to the edible parts of water dropwort near industrial complex. The soils and water dropwort were collected from the paddies cultivating water dropwort(10 sites), stream sediments(5 sites), and background soils(3 sites) near industrial complex. The total concentrations of Cd Cu, and Ni were higher than those of permissible level for soil contamination(Cd 3, Cu 100, Ni $50mg\;kg^{-1}$ in soil) suggested by Kloke(1979). Dominant chemical forms of Cd in paddies cultivating water dropwort and stream sediments were exchangeable form(49.1-56.3%), and those of Cu, Zn, and Ni were Fe and Mn oxide bound and residual forms. The mobility factor of heavy metals in paddies cultivating water dropwort and stream sediments was in the order Cd>Zn>Ni>Cu>Pb, specially, the mobility factor of Cd (62-72%) were relatively higher than that of other metals in soils. The total concentrations of Cd in soils showed significant positive correlation with the ratios of exchangeable and Fe and Mn oxide bound forms, while correlated negatively with residual form. Heavy metal contents in root parts were higher than those in top parts of water dropwort. The bioavailability of water dropwort varied considerably between the different parts and heavy metals. Cd, Cu and Ni contents in water dropwort were correlated with each fractions in soils. Specially, the exchangeable form of Cd and Ni in soils showed significant positive correlation with the those contents of water dropwort.

Deletion Timing of Cic Alleles during Hematopoiesis Determines the Degree of Peripheral CD4+ T Cell Activation and Proliferation

  • Capicua (CIC) is a transcriptional repressor that regulates several developmental processes. CIC deficiency results in lymphoproliferative autoimmunity accompanied by expansion of CD44hiCD62Llo effector/memory and follicular Th cell populations. Deletion of Cic alleles in hematopoietic stem cells (Vav1-Cre-mediated knockout of Cic) causes more severe autoimmunity than that caused by the knockout of Cic in CD4+CD8+ double positive thymocytes (Cd4-Cre-mediated knockout of Cic). In this study, we compared splenic CD4+ T cell activation and proliferation between whole immune cell-specific Cic-null (Cicf/f;Vav1-Cre) and T cell-specific Cic-null (Cicf/f;Cd4-Cre) mice. Hyperactivation and hyperproliferation of CD4+ T cells were more apparent in Cicf/f;Vav1-Cre mice than in Cicf/f;Cd4-Cre mice. Cicf/f;Vav1-Cre CD4+ T cells more rapidly proliferated and secreted larger amounts of IL-2 upon TCR stimulation than did Cicf/f;Cd4-Cre CD4+ T cells, while the TCR stimulation-induced activation of the TCR signaling cascade and calcium flux were comparable between them. Mixed wild-type and Cicf/f;Vav1-Cre bone marrow chimeras also exhibited more apparent hyperactivation and hyperproliferation of Cic-deficient CD4+ T cells than did mixed wild-type and Cicf/f;Cd4-Cre bone marrow chimeras. Taken together, our data demonstrate that CIC deficiency at the beginning of T cell development endows peripheral CD4+ T cells with enhanced T cell activation and proliferative capability.

  • Purpose: The periosteum is a well-known source of osteogenic precursor cells for tissue-engineered bone formation. However, cultured endothelial or endothelial-like cells derived from periosteum have not yet been investigated. This study focused on endothelial-like cell culture from the periosteum. Methods: Periosteal tissues were harvested from the mandible during surgical extraction of lower impacted third molars. The tissues were treated with 0.075% type I collagenase in phosphate-buffered saline (PBS) for 1 hr at $37^{\circ}C$ to release cellular fractions. The collagenase was inactivated with an equal volume of DMEM/10% fetal bovine serum (FBS) and the infranatant was centrifuged for 10 min at 2,400 rpm. The cellular pellet was filtered through a $100{\mu}m$ nylon cell strainer, and the filtered cells were centrifuged for 10 min at 2,400 rpm. The resuspended cells were plated into T25 flasks and cultured in endothelial cell basal medium (EBM)-2. Results: Among the hematopoietic markers, CD146 was more highly expressed than CD31 and CD34. The periosteal-derived cells also expressed CD90 and CD166, mesenchymal stem cell markers. Considering that the expression of CD146 was constant and that the expression of CD90 was lower at passage 5, respectively, the CD146 positive cells in passage 5 were isolated using the magnetic cell sorting (MACS) system. These CD146 sorted, periosteal-derived cells formed tube-like structures on Matrigel. The uptake of acetylated, low-density lipoprotein, labeled with 1,1'-dioctadecyl-3,3,3',3'-tetramethylindocarbocyanine perchlorate (DiI-Ac-LDL) was also examined in these cells. Conclusion: These results suggest that the CD146-sorted positive cells can be referred to as periosteal-derived CD146 positive endothelial-like cells. In particular, when a co-culture system with endothelial and osteoblastic cells in a three-dimensional scaffold is used, the use of periosteum as a single cell source would be strongly beneficial for bone tissue engineering.

  • Investigation was carried out to observe the dominant bacteria and the effect of metals(Cd & Zn) and its nitrate compound on growth of bacteria isolated from the three tributaries and lower reach of the Nakdong River. Mean CFU(log$_{10}$) level was highest in Kumho River(8.30 CFU), Nam River, Hwang River, and Mulgum followed. Staphylococcus xylosus, Staph. lentus, Pasteurella pneumotropica, Aeromonas hydrophilla were dominant species in each study site. Cadmium powder and Zinc powder showed strong effect to inhibit the growth of Micrococcus spp., Pasteurella pneumotropica, Aeromonas hydrophilla. But, nitrate compounds of Cd and Zn(Cd(NO$_3$)$_2$4$H_2O$, Zn(NO$_3$)$_2$6$H_2O$) did not clearly show the strong effect to inhibit the growth of dominants.s.

Light Scattering Amplification on Dye Sensitized Solar Cells Assembled by Hollyhock-shaped CdS-TiO2 Composites

  • To investigate the scattering layer effect of a $TiO_2$ multilayer in dye-sensitized solar cells (DSSCs), we designed a new DSSC system, assembled with a CdS-$TiO_2$ scattering layer electrode. A high-magnification SEM image exhibited hollyhock-like particles with a width of 1.5-2.0 ${\mu}m$ that were aggregated into 10-nm clumps in a hexagonal petal shape. The efficiency was higher in the DSSC assembled with a CdS-$TiO_2$ scattering layer than in the DSSC assembled with $TiO_2$-only layers, due to the decreased resistance in electrochemical impedance spectroscopy (EIS). The short-circuit current density ($J_{sc}$) was increased by approximately 7.26% and the open-circuit voltage ($V_{oc}$) by 2.44% over the 1.0 wt % CdS-$TiO_2$ composite scattering layer and the incident photon-to-current conversion efficiency (IPCE) in the maximum peak was also enhanced by about 5.0%, compared to the DSSC assembled without the CdS-$TiO_2$scattering layer.

  • Recently, most studies concerning inorganic CdSe/ZnS quantum dot (QD)-polymer hybrid LEDs have been concentrated on the structure with multiple layers [1,2]. The QD LEDs used almost CdSe materials for color reproduction such as blue, green and red from the light source until current. However, since Cd is one of six substances banned by the Restriction on Hazardous Substances (RoHS) directive and classified into a hazardous substance for utilization and commercialization as well as for use in life, it was reported that the use of CdSe is not suitable to fabricate a photoelectronic device. In this work, we demonstrate a novel, simple and facile technique for the synthesis of ZnO-graphene quasi-core.shell quantum dots utilizing graphene nanodot in order to overcome Cd material including RoHS materials. Also, We investigate the optical and structural properties of the quantum dots using a number of techniques. In result, At the applied bias 10 V, the device produced bluish-white color of the maximum brightness 1118 cd/$m^2$ with CIE coordinates (0.31, 0.26) at the bias 10 V.

Morphological Change of Crosslinked ${\beta}$-Cyclodextrin after Recycling

  • The present study was carried out to examine the effect of crosslinked ${\beta}$-cyclodextrin(${\beta}$-CD) made by adipic acid on cholesterol removal rate and find the structural change after recycling on SEM observation. The size reduction and morphological changes were found during the recycling process and the profound changes were observed at 8th time reuse. After cholesterol removal in milk, the used crosslinked ${\beta}$-CD was washed for cholesterol dissociation and reused. In recycling study, the cholesterol removal rate at first trial was 92.5% in milk, which was mostly same as that using new crosslinked ${\beta}$-CD(92.4%). With repeated 10th reuse of crosslinked ${\beta}$-CD resulted in 81.4% of cholesterol removal in milk. Similar trend was found in cream and cholesterol removal was 91.5% at 1st trial and 83.4% at 10th trial. In both milk and cream samples, the removal rate at 1st reuse was not significantly different from that at 6th reuse(p>0.05). The present study indicated that crosslinked ${\beta}$-CD made by adipic acid resulted in the effective recycling efficiency, especially up to 6th reuse and morphological modifications were not distinguishable up to 8th reuse.

Effects of Kakamsodokum (KKSDU) on Atopic Dermatitis-like Skin Lesions in NC/Nga Mouse (아토피양(樣) 피부염 NC/Nga생쥐에서 가감소독음(加減消毒飮)의 투여가 피부염에 미치는 영향)

  • Objectives The purpose of this study is to investigate the effect of Kakamsodokum (KKSDU) on atopic dermatitis in an in-vitro experiment using an NC/Nga atopic dermatitis mouse, which has histological and clinical similarities to the condition in humans. Methods We evaluated $IL-1{\beta}$, IL-6, $TNF-{\alpha}$, $TGF-{\beta}$, IL-10 mRNA, CD4+/$IFN-{\gamma}+$, and CD4+CD25+foxp3+ in B and T cells of NC/Nga atopic dermatitis mouse by real-time PCR and intracellular staining in vitro. Results KKSDU medicines supressed $IL-1{\beta}$, IL-6, $TNF-{\alpha}$, $TGF-{\beta}$ mRNA and increased IL-10 mRNA in B cells. CD4+/$IFN-{\gamma}+$ and CD4+CD25+foxp3+ in T cells were increased by KKSDU. Conclusions KKSDU on atopic dermatitis might be very effective to the atopic dermatitis treatment.

Cadmium Accumulation and Elimination in the Tissues of the Manila Clam, Ruditapes philippinarum, after Sub-chronic Cadmium Exposure

  • Cadmium (Cd) accumulation and elimination were assessed in the tissues of the clam R. philippinarum at four experimental concentrations (control, 10, 20, 100, and $200\;{\mu}g/L$) over an exposure period of 2 weeks and an elimination period of 1 week. Cd accumulated in the digestive gland, gill, and residual clam tissues, and accumulation increased with time of exposure and concentration (100 and $200\;{\mu}g/L$). After 2 weeks of Cd exposure, the order of Cd accumulation in tissues was gill > digestive gland > residual tissues. An inverse relationship was observed between concentration factor (CF) and exposure level, but the CF showed an increase with exposure time. During the depuration period, Cd concentrations in the digestive gland, gill, and residual tissues decreased immediately on the cessation of exposure, except in individuals at the $200\;{\mu}g/L$ concentration. The Cd elimination rate from tissues decreased in the order of digestive gland > gill > residual tissues during the depuration period.
