Cadmium is considered one of the most toxic, non biodegradable heavy metal for the human and animals. The purpose of the present study was to investigate the changes in biochemical parameters of blood and accumulation of cadmium in various tissue caused by various levels of dietary cadmium chloride ($CdCl_2$) in broiler chicks. $CdCl_2$ was administered through drinking water to broiler chicks. In spectral analysis, $CdCl_2$ treatment caused a significant increase in Glutamate pyruvate transaminase (GPT), creatinine and uric acid levels in all treated groups. Intriguingly, the GPT, creatinine, and uric acid levels were significantly higher at 75 mg/kg as compared to the groups treated with high doses (100, 125 and 150 mg/kg) of $CdCl_2$. Atomic Absorption Spectrophotometer (AAS) was used for the determination of Cd accumulation in kidney, liver and Breast muscles. AAS analysis revealed that Cd accumulation is increased in breast muscles as compared to liver and kidney at higher doses of Cd than 75 mg/kg.
환경에 존재하는 중금속인 Cd은 생물에게 유입되며 먹이사슬을 통해 생물에게 축적된다. 본 연구에서는 카드뮴 노출에 따른 오염지표생물인 실지렁이(Tubifex tubifex)의 체내 농축 반응 및 스트레스 유전자 발현 패턴을 분석하기 위해, 퇴적물 내 카드뮴 노출 후 생존율 변화, 체내 농축 농도 측정, 그리고 스트레스 반응 유전자인 열 충격 단백질(HSP60 and HSP70)과 항산화효소인 GST의 발현 패턴을 관찰하였다, Cd 노출 퇴적물에 노출된 T. tubifex의 생존율은 노출 10일에 0.4 mg kg-1 Cd에서 93%, 1.87 mg kg-1 Cd에서 96%, 6.09 mg kg-1 Cd에서 93%로 관찰되었다. T. tubifex의 체내 Cd 농도는 퇴적물 내 Cd 농도에 비해 높게 나타났으며, 0.4 mg kg-1 Cd에서는 실험 10일에 18.4 mg kg-1 Cd, 1.87 mg kg-1 Cd에서 실험 1일에 13.06 mg kg-1 Cd, 6.09 mg kg-1 Cd에서 실험 10일에 79.11 mg kg-1 Cd로 가장 높게 나타났다. HSP60 유전자 발현은 6.09 mg kg-1 Cd 노출 퇴적물에서 시간 의존적으로 발현이 대조군에 비해 증가였으며, HSP70은 모든 농도에서 대조군에 비해 시간 의존적인 발현의 증가를 보였다. 또한, GST는 모든 농도에서 실험 1일에 대조군에 비해 발현이 증가한 후, 실험 10일까지 발현이 감소하였다. 이러한 연구결과는 퇴적물 내에 존재하는 Cd에 대한 실지렁이의 생태독성학적 및 분자유전학적 독성영향을 확인한 결과로, 환경 내 오염지표종으로 실지렁이를 이용한 환경독성평가의 기초자료로 활용될 수 있을 것이다.
This study was to investigate the cadmium removal effect of Korean green tea, black tea and oolong tea beverage on Cd administered rat, tissues and their excretions. Male Sprague-Dawley rats weighing 143±3.2g were divided into control and experimental groups. The control group were fed standard diet without cadmium . The experimental groups, which were fed standard diet containing 40 ppm Cd, were divided into 4 subgroups again , which were the groups given distilled water (CD group), 5% black tea (BT group), oolong tea (OT group ) and green tea (GT group), respectively. Five days before to sacrifice the rats, all 4 cadmium fed groups were supplied 1 ml of water with 600ppm Cd and control group were fed 1 ml of distilled water without Cd under the same dietary condition. After that, their excretion were collected separately for 3 days. In rat liver and kidney, accmulation of cadmium in 4 Cd administered groups were more than in control group and that of GT group was significantly less than CD group. In bone , also, accumulation of cadmium in 4 Cd administered groups was more than in control group and that of GT, OT,BT groups were much less than that of CD group. GT group was excreted more Cd in urine than Cd group. In feces, 3 tea feeding groups (BT, OT, GT group) were excreted Cd 1.7, 2.1, 2.4 times more than that of the CD group, respectively. We conclude that cadmium accumulations of GT feeding group in rat's liver, kidney and bone were much less than CD group , and the absorption and retention rate of GT group was significantly lower than CD group.
Objectives : This study was conducted to evaluate the acute toxicity and safety of Ssanghwa-tang (Shuanhetang in Chinese, Sou-wa-to in Japanese) in Crl : CD Sprague-Dawley (SD) rat though the current regulatory guideline. Methods : In this study, 10 rats of each sex were randomly assigned to two groups of 5 rats each and were administrated singly by gavage at dose levels of 0 and 2000 mg/kg/day of ssanghwa-tang water extract (SHT). After single administration of SHT, mortalities, clinical signs, body weight changes, gross findings were observed for the 15-day period. Results : Acute toxicity tests revealed that a single oral administration of SHT at dose levels of 2000 mg/kg did not affect clinical signs, body weight, and gross findings, evaluating the safety of SHT. The SHT treatment did not result in any toxicologically significant changes in mortality, clinical signs, body weight changes. Conclusions : These results showed that the single oral administration of SHT did not cause any toxic effect at the dose levels of 2000 mg/kg/day in rats. In conclusion, the median lethal dose (LD50) of SHT was considered to be over 2000 mg/kg/day body for both sexes.
초식성 요각류인 Tigriopus japonicus를 대상으로 사망률과 반수치사농도$(LC_{50})$ 및 호흡대사에 미치는 카드뮴의 급성 및 만성독성영향을 조사하였던바 다음과 같은 결과를 얻었다. 염분 32, 26 및 $18\%_{\circ}$에서 T. japonicus에 대한 카드뮴의 96시간 반수치사농도$(96hr.-LC_{50})$는 $20^{\circ}C$에서 각각 15.1, 13.7 및 9.9 ppm이었고, $10^{\circ}C$에서는 각각 21.6, 15.0 및 11.0ppm으로 나타나 수온의 감소와 염분의 증가에 따라 반수치사농도는 증가되었다. 96시간동안 카드뮴에 노출시킨 후 산소소비율은 $10^{\circ}C$에 비해 $20^{\circ}C$에서 높게 나타났으며, 카드뮴농도의 증가에 따라 보다 크게 감소되었다. 염분 $32\%_{\circ}$ 및 Cd농도 4ppm에서 T. japonicus의 생존률과 25일동안의 반수치수농도$(25days-LC_{50})$는 $10^{\circ}C$의 경우 각각 $5\%$와 0.05ppm이었고, $20^{\circ}C$에서는 $0\%$와 0.28ppm을 나타내었다.
Compared with organic fluorophores, semiconductor quantum dots (QDs) have the better properties such as photostability, narrow emission spectra coupled to tunable photoluminescent emissions and exceptional resistance to both photo bleaching and chemical degradation. In this work, CdSe/ZnS QDs nanobeads were prepared by the incorporation of CdSe/ZnS QDs with mesoporous silica to use as the optical probe for detecting toxic and bio- materials with high sensitivity, CdSe/ZnS core/shell QDs were synthesized from the precursors such as CdO and zinc stearate with the lower toxicity than pyrotic precursors. The QD-nanobeads were characterized by transmission electron microscopy, FL microscopy, UV-Vis and PL spectroscopy, respectively.
Contamination of heavy metals in water and sediments along Mangyeong Gang(river) has reached up to critical level The object of the study is to elucidate somed molluscs which inhabit along the river on respect to the modulatory role in reducing the pollution. For the purpose, molluscs which are common in numbers and biomass in the area were collected, and Cadmim(Cd) was subjected as a reference metal in the experiment. The corresponding species were; Cipango;aludina chinensis (muddy snail) and Scapharca subcrenata (seashell). As methods, sample species were kept in laboratory under the natural condition as possible. Soil were brought from the site of the collection, dried autoclaved and wetted with ordinary water. It wad utilized as media to maintain the collected species in vitro all the way of the experiment. CdCI$^{2}$ was mixe in the medium according to experimental design. On the result obtained in the study, it is summarized that molluscs which inhabit along Mangyeong Gang(river)consume heavy mital-containing matters, so far Cd is concerned in the study. The amout of Cd concentration in tissues of the benthic natured invertebrates were dose and time related, and MT-Cd was also similar trend. Thus, Cd may eventually combine with low molecular protein forming metalloprotein, then reduce the toxicity of the heavy metal.
Effects of salinity and standard toxic metals on fertilization and embryo development rates were investigated in the sea urchin, Strongylocentrotus nudus. Spawning was induced by injecting 1ml of 0.5 M KCl into the coelomic cavity. The fertilization and embryo development rates were below 20% when salinity was 25 psu or lower, but were above 90% when salinity was between 30 and 35 psu. The fertilization and embryo development rates in the control condition (not including Cu and Cd) were greater than 90%, but decreased with a high negative correlation (r) of 0.89 and 0.91 with the increasing of Cu and Cd concentrations, respectively. These results suggest that salinity concentrations for successful fertilization and normal embryogenesis of S. nudus are between 30 and 35 psu, and the biological assays of fertilization and embryo development rates using S. nudus are useful methods for the ecological toxicity test of marine pollution elements.
본 연구는 카드뮴으로 유발된 생쥐 간중독에 대한 키토산올리고당의 효과를 알아보기 위하여 시도되었다. Mouse를 대상으로 카드뮴 (5.0 mg/kg; i.p.) 단독투여군(group Cd), 카드뮴과 키토산올리고당 (0.5% solution) 동시투여군 (group Cd+Chi) 그리고 키토산올리고당 1주일 전처치군 (group Chi7+Cd)으로 구분한 후 간손상 억제효과를 알아보기 위해 혈청중 효소활성도와 glutathione peroxidase (GSH-Px)의 활성도 그리고 glutathione reductase (GR)의 활성도를 비교측정하였다. 또한 간조직의 형태학적 손상을 확인하기 위해 조직학적 관찰을 실시하였다. 혈청 효소활성결과, AST, ALT 그리고 LDH의 활성도는 키토산올리고당을 처치한 Cd+Chi군과 Chi7+Cd군이 Cd군보다 감소되었다. 그러나 TP와 Alb의 활성도는 키토산올리고당을 처치한 Cd+Chi군과 Chi7+Cd군이 Cd군보다 증가되었다. GSH-Px활성은 Cd군에 비해 Cd+Chi군과 Chi7+Cd군이 현저하게 증가되었다. GR활성은 Cd군에 비해 Cd+Chi군과 Chi7+Cd군이 현저하게 감소하였다. 광학현미경적 관찰 결과, Cd군은 카드뮴 독성에 의한 간세포의 괴사가 관찰되었다. Cd+Chi군과 Chi7+Cd군은 정상군과 유사한 소견을 보였다. 이상과 같은 결과로, 키토산올리고당이 생쥐 간조직에 미치는 카드뮴의 독성을 감소시킴을 알 수 있었다.
Objectives : The aim of this study was to confirm whether or not coral has a preventive effect on development of atopic dermatitis induced by house mite(dermatophagoides pteronyssinus) in NC/Nga mice. Methods : This study was undertaken by using a reliable Atopic dermatitis mouse model demonstrating similar immune response. Lobophytum crassum was administered orally to NC/Nga mouse for 3 weeks. In order to verify the effectiveness of Lobophytum crassum in atopic dermatitis treatment, its role in immune factors were observed in NC/Nga mice. Results : ALT, AST, BUN and creatine levels were all within in the normal ranges in MC-1 200 and 400 (mg/kg) treated groups, indicating no induced toxicity. MC-1 200 and 400 (mg/kg) groups decreased of atopic dermatitis skin manifestation in NC/Nga mouse of MC-1 200 and 400 (mg/kg) groups compared to that of the control group and decreased the ratio of WBC and lymphocyte in blood. Also, MC-1 200 and 400 (mg/kg) groups significant decreased the ratio of CD4+, CD8+, CD11b+/Gr1+, B220/CD23 and CD4/CD25 immune cell ratio in ALN. Finally MC-1 200 and 400 (mg/kg) groups significantly increased the ratio of CD4+, CD8+, B220/CD23 and CD4/CD25 immune cells in DLN. Conclusions : Theses results suggested that Lobophytum crassum has suppressive effects on aberrant and overactive immunological activities in dermatophagoides pteronyssinus-induced dermatitis mice of NC/Nga.
