Development of Neuropathic Pain Behavior and Expression of CCL2/CCR2 and CX3CL1/CX3CR1 after Spinal Cord Hemisection

  • Purpose: The purpose of this study was to evaluate the development of pain behavior and the expression of CCL2/CCR2 and CX3CL1/CX3CR1 at above and below the level of hemisection of the spinal cord in a rat model. Methods: Spinal cords of adult female Sprague-Dawley rats (n= 16, 200~250 g, 6~8 weeks old) were hemisected at T13 on the right side to develop the spinal hemisection injury model. We compared behavioral responses of the hemisection and of a sham surgery group. Behavioral tests for motor function (by the BBB locomotor scale), and for pain response for mechanical and cold allodynia were assessed postoperatively (PO) for 21 days. Expression of mRNA for chemokines and their receptors (CCL2/CCR2 and CX3CL1/CX3CR1) below and above the level of the spinal cord dissection were examined by RT-PCR. Results: We observed gradual motor improvement and the development of mechanical and cold allodynia on the ipsilateral hindpaw after spinal hemisection injury. We also found upregulation of mRNA expression of CCL2/CCR2 both above and below the level of spinal cord dissection but CX3CL1/CX3CR1 mRNA expression. Conclusion: Upregulation of CCL2/CCR2 is associated with neuropathic pain after spinal hemisection injury. CCL2/CCR2 may play an important role in the development of neuropathic pain after SCI as well as of peripheral neuropathic pain. These findings may improve understanding of the pathophysiological mechanism of neuropathic pain after SCI.

  • Objectives The purpose of this study is to investigate the effect of SPDJTK(SoPungDoJeokTangKami) and concurrent administration of AJ(Atopy cream, Jawoongo)+SPDJTK on atopic dermatitis-like skin lesions by using in NC/Nga atopic dermatitis mouse induced by BMAC-induced mice. Methods Clinical skin score, hematology and Serum total IgE and IgG1 of NC/Nga atopic dermatitis mice were evaluated. Moreover, the cytokine level, total cell number, Immunohistochemical staining and Histological features of axillary lymph node(ALN), draining lymph node(DLN), peripheral blood mononuclear cells(PBMCs) and dorsal skin tissue were used in NC/Nga mice. Results Orally administrated SPDJTK with concurrent administration of SPDJTK and AJ decreased the clinical skin score, total cell number of WBC, eosinophils in blood, serum total IgE & IgG1, IL-5, IL-13, IFN-$\gamma$. Also, total cell number of ALN and dorsal skin tissue, absolute cell number of CD4+, CD8+, CD3+CD69+, CD3+CCR3+, CCR3+, CD4+CXCR5+ in ALN, absolute cell number of CD3+CCR3+, CCR3+ in DLN, granulocytes in PBMCs, activation cell number of CD3+CD69+, CCR3+, total cell number of CD3+ T cell in dorsal skin tissue were significantly decreased. Furthermore, thickness of epidermis, infiltrated inflammatory immune cell and mast cell in dermis, amount of Eotaxin2 mRNA, CCR3 mRNA in dorsal skin tissue, gene expression of IL-5, IL-13 mRNA in ALN, CD4+ Th cell in dorsal skin tissue and CCR3+ eosinophils in ALN were all significantly decreased. However, total number of DLN, absolute number of CD3e+ T cell and CD19+ B cell, absolute number of CD4+, number of Th cell in DLN and gene expression of foxp3 mRNA were significantly increased significantly. Conclusions Concurrent administration of SPDJTK and AJ on atopic dermatitis in NC/Nga atopic dermatitis mouse was very effective treatment for atopic dermatitis.

Measuring Efficiency of Korean Apartment Construction Firms using DEA (DEA 모형을 이용한 국내 아파트 건설기업(상장기업)의 효율성 분석)

  • The bankruptcy possibility of apartment construction firms has been incresed because of the environment of current apartment construction industry. This paper analyzes the efficiency of apartment construction firms using DEA model. We evaluate the CCR, BCC efficiency and RTS of 25 apartment construction firms. We also suggest the apartment construction firms which can be benchmarked based on analyzed information. The result shows that four enterprises whose values of CCR efficiency are 1, and ten enterprises whose values of BCC efficiency are 1. RTS indicates IRS of 21 firms and CRS of 4 firms.

Management Efficiency of Introduction Company of ERP System using DEA (DEA를 이용한 ERP시스템 도입기업의 경영효율성)

  • ERP is the general trend of industrial information system and many companies are introducing strategically to strengthen the competitive power. This paper analyzes the efficiency of introducing companies of ERP system using DEA model. We also suggest the introducing of ERP system which can be benchmarked based on analyzed information. We evaluate the CCR, BCC efficiency and RTS of 38 introducing companies of ERP system. The result shows that three enterprises whose values of CCR efficiency are 1, and seven enterprises whose values of BCC efficiency are 1. RTS indicates IRS of 22 companies. DRS of 2 companies and CRS of 4 companies.

Analysis of Three-Phase Multiple Access with Continual Contention Resolution (TPMA-CCR) for Wireless Multi-Hop Ad Hoc Networks

  • In this paper, a new medium access control (MAC) protocol entitled three-phase multiple access with continual contention resolution (TPMA-CCR) is proposed for wireless multi-hop ad hoc networks. This work is motivated by the previously known three-phase multiple access (TPMA) scheme of Hou and Tsai [2] which is the suitable MAC protocol for clustering multi-hop ad hoc networks owing to its beneficial attributes such as easy collision detectible, anonymous acknowledgment (ACK), and simple signaling format for the broadcast-natured networks. The new TPMA-CCR is designed to let all contending nodes participate in contentions for a medium access more aggressively than the original TPMA and with continual resolving procedures as well. Through the systematical performance analysis of the suggested protocol, it is also shown that the maximum throughput of the new protocol is not only superior to the original TPMA, but also improves on the conventional slotted carrier sense multiple access (CSMA) under certain circumstances. Thus, in terms of performance, TPMA-CCR can provide an attractive alternative to other contention-based MAC protocols for multi-hop ad hoc networks.

CCR Expression of Bronchoalveolar Lavage Fluid (BALF) Neutrophils and Chemotactic Activity of BALF

  • Asthma is an inflammatory airway disease and is characterized by the releases of inflammatory mediators including chemokines. They are mainly associated with the recruitment, activation and dysregulation of specific inflammatory cells, especially neutrophils in neutrophilic asthma. CC chemokines bind to CC chemokine receptors (CCRs) in the surface of their target cells. The aims of this study are to examine the CCR expression in neutrophils of bronchoalveolar lavage fluid (BALF) of asthmatic patients and to determine the alternation of migration and apoptosis of neutrophils by the BALF. We demonstrate that CCR3 strongly expresses in BALF neutrophils of asthmatic patients as compared to other CCRs and increases during apoptosis of the BALF neutrophils. The migration of asthmatic blood neutrophils increases in response to asthmatic BALF as compared to BALF of normal volunteer. In addition, asthmatic BALF includes the higher levels of IL-8 protein than normal BALF and it has no effect on apoptosis of asthmatic blood neutrophils. Taken together, our results indicate that CCR3 expression may be associated with unknown function of asthmatic BALF neutrophils and BALF may be involved in the recruitment of neutrophils into the airway, but not in the neutrophils apoptosis.

Efficiency Analysis of the Korean Listed Display Companies (국내 상장 디스플레이 기업의 효율성 분석)

  • Although the display industry plays an important role in the entire Korean economy, few empirical research has analyzed the efficiency of display companies. The purpose of this paper is to measure and analyze efficiency of korean listed display firms using DEA(Data Envelopment Analysis) models. We evaluate the CCR and BCC efficiency in DEA models and the return to scale of the Korean listed display companies. The benchmarking companies and efficiency value for the display companies with inefficiency are also provided to improve their efficiency. We analyzed the 44 listed companies consisted of 7 listed on KOSPI and 37 listed on KOSDAQ at the end of 2010. The analysis results show six companies whose values of CCR are 1, and fourteen firms whose values of BCC efficiency are 1. In additions, the six companies have the scalability efficiency. Eventually the efficiency analysis can provide the valuable information for inefficient companies to find benchmarking companies and to improve their efficiency.

Carbon Catabolite Repression (CCR) of Expression of the XylanaseA Gene of Bacillus stearothermophilus No.236

  • Previous work has identified that only the catabolite responsive element A (creA; previously called cre-2) out of two potential cre sequences (cre-1: nucleotide +160 to +173 and cre-2: +173 to +186), recognized within the coding region of the xylanaseA gene (xynA) of Bacillus stearothermophilus No.236, was actually, was actually involved in the carbon catabolite repression(CCR) of xynA expression in B. subtilis. However, the level of CCR of xynA expression in the original B.stearothermophilus No.236 strain (70-fold repression). Therefore, to search for an additional cre element in the promoter region, the upstream region of the xynA gene was subcloned by chromosome walking, and as a result, another potential cre element (nucleotide -124∼-137; designated creB) was recognized in this region. The cre-like sequence revealed a high homology to the cre consensus sequence. The xylanase activity of B. subtilis MW15 bearing pWPBR14 (containing creA and creB) cultured in a medium containing xylose as the sole carbon source was about 7.7 times higher than that observed for the same culture containing glucose. B. subtilis MW15 bearing pWPBR23 (containing only creA) produced an activity about 2.4 times higher. This pattern of CCR was confirmed using derivatives of xynA::aprA fusion plasmids. Furthermore, a measurement of the amounts of the xynA transcript showed a similar pattern as that for the production of xylanase. In addition, the synthesis of xylanase in B. subtilis QB7115 [a catabolite control protein A (ccpA) mutant strain] carrying pWPBR14 was almost completely relieved from glucose repression. Together, these results lead to a conclusion that the CCR of the expression of the xynA gene is mediated by CcpA binding at creA and creB sites in B. subtilis.

A Study on the Mechanism of Urinary and Biliary Excretion of Chloramphenicol in the Dog (개에 있어서 Chloramphenicol의 뇨(尿) 및 담즙중(膽汁中) 배설기전(排泄機轉)에 관(關)한 연구(硏究))

  • A study on the mechanism of biliary and urinary excretion of chloramphenicol has been performed in the dog. 1) Chloramphenicol administered intravenously to dogs with ligated renal pedicle, readily appeared in bile greater than in plasma. 6.9% of a 50mg /kg i. v. dose of chloramphenicol were excreted into bile within 100 minutes. During the same periods of above experiment, the bile/plasma concentration ratios(B/P ratios) were 46 to 87. 2) Chloramphenicol injected into the vein of dog was rapidly excreted into urine. 18% of the administered dose were excreted into urine within 70 minutes. In the same periods of this experiment, Ccm/Ccr ratios were greater than 1.0 in most cases. 3) In experiment of simultaneous measurement of biliary and urinary excretion of chloramphenicol, Ccm/Ccr ratios were less than 1.0 and B/P ratios were 50 to 52. 4) In experiment measured simultaneously biliary and urinary excretion both Ccm/Ccr and $C^Hcm$(hepatic clearance) were significantly declined by probenecid, but not affected by 2,4-DNP and aminophylline although 2,4-DNP increased only bile flow and aminophylline both bile and urine volume. 5) Ccm/Ccr and $C^Hcm$ were increased in proportion to increment of plasma concentration ranging from 3.3 to 30 mg% of chloramphenicol. But when plasma concentration were increased to 70mg %, Ccm/Ccr were not increased and $C_Hcm$ were reduced about 30% in comparison with values obtajned at 30mg% of chloramphenicol. 6) Free/Bound(free to bouid from) ratios ranging from 1.0 to 90.0mg% of chloramphenicol were 76.2+3.72% $(mean{\pm}S.E.)$ Above results suggest that chloramphenicol is excreted into bile by a process of active trasport, that excretion of chloramphenicol into urine was made up with dual process, reabsorption and secretion, and that renal secretion was attained by active trasport process although renal reabsorption process could not understand.

The Role of the Peripheral Chemokine, CCL3, in Hyperalgesia following Peripheral Nerve Injury in the Rat (신경손상에 의해 유발된 과민통반응에서 말초 케모카인 CCL3의 역할)

  • Background: Upregulation of one type of the pro-inflammatory chemokine (CCL2) and its receptor (CCR2) following peripheral nerve injury contributes to the induction of neuropathic pain. Here, we examined whether another type of chemokine (CCL3) is involved in neuropathic pain. Methods: We measured changes in mechanical and thermal sensitivity in the hind paws of naïve rats or rats with an L5 spinal nerve ligation (SNL) after intra-plantar injection of CCL3 or met-RANTES, an antagonist of the CCL3 receptor, CCR1. We also measured CCL3 levels in the sciatic nerve and the hind paw skin as well as CCR1 expression in dorsal root ganglion (DRG) cells from the lumbar spinal segments. Results: Intra-plantar injection of CCL3 into the hind paw of naive rats mimicked L5 SNL-produced hyperalgesia. Intra-plantar injection of met-RANTES into the hind paw of rats with L5 SNL attenuated hyperalgesia. L5 SNL increased CCL3 levels in the sciatic nerve and the hind paw skin on the affected side. The number of CCR1-positive DRG cells in the lumbar segments was not changed following L5 SNL. Conclusions: Partial peripheral nerve injury increases local CCL3 levels along the degenerating axons during Wallerian degeneration. This CCL3 binds to its receptor, CCR1, located on adjacent uninjured afferents, presumably nociceptors, to induce hyperalgesia in the neuropathic pain state.
