In vitro Effects of the Complex of Korean Citrus junos and Medicinal Herbs on the Cell Growth in CCD-986sk Human Fibroblast Cell Line

  • In order to investigate whether or not CCD-980sk cell line can be affected by Korean Citrus junos and medicinal herbs, we examined the MTT assay when we treated Korean Citrus junos and medicinal herbs in CCD-986sk human fibroblast cell line. The samples that added complex of grinded extracts of Korean Citrus junos and boiling-water extracts from Korean medicinal herbs were tested toy cell proliferation activity by means of a modification of the MTT assay. Among mixture of Citron 3 (Citron 3, less mellowed citron which was ripened for three months) and boiling-water extracts, the group Citron 3+Phellinus linteus showed significantly strong cell proliferation activity. And among mixture of Citron 4 (Citron 4, completely mellowed citron which was ripened for four months) and boiling-water extracts, the group Citron 4+Cordyceps militaris and Citron 4+Phellinus linteus showed significantly strong cell proliferation activity, respectively. These results suggest that complex of Korean Citrus junos and medicinal herbs could be an excellent candidate toy protection of human skin aging.

  • The purpose of this study was to investigate the effects of Cuscuta chinensis Lamark ethanol extract (CL) on wrinkle improvement. Cuscuta chinensis Lamark is known to contain dried saccharide, alkaloids, flavonoids, lignans and rein glycoside as major components of dried mature seeds of Cuscuta japonica Choisy. In this study, we evaluated the anti-wrinkle effects of CL and investigated bio-markers (e.g ; MMP-1, MMP-3, MMP-9, TIMP-1, type I procollagen) associated with skin wrinkle improvement. We tested the anti-wrinkle effect of CL using human fibroblast called CCD-986Sk cell. We observed an increase in MMPs, TIMP-1, and type 1 pro-collagen CL in CCD-986Sk cells irradiated with UVB at an intensity of $2mJ/cm^2$ for 60 seconds. As a result, CL decreased UVB-induced MMPs levels and mRNA expressions in CCD-986Sk cell. The levels and mRNA expressions of type I procollagen and TIMP-1 were increased by CL. These results suggest that CL has activities on improvement of skin wrinkle, which is induced by UVB radiation. Taken together, this study proposed the possibility of developing herbal medicine and functional herbal cosmetic materials with wrinkle-improving effects of Cuscuta chinensis Lamark.

Effects of ethanol extract of Nelumbo nucifera leaves on Anti-oxidation and Type I Procollagen Expression in CCD-986sk Cells (연잎 에탄올 추출물이 피부 주름에 미치는 영향)

  • The natural plants were screened for new cosmetic materials. Many parts of Nelumbo nucifera has been investigated for anti-wrinkling and anti-furrowing effects. The leaves. flowers and stamens of Nelumbo nucifera were extracted with ethanol or distilled water. The effects of cell viability and $H_2O_2-induced$ cytotoxicity were studied on CCD-986sk human fibroblast cell line by MTT assay. We measured various effects related to skin such as the synthesis of MMP-1 and type I procollagen. The ethanol extract of Nelumbo nucifera leaves (ENL) at concentrations of 30 ${\mu}g/ml$ showed maximum cell viability on CCD-986sk cells by 129.70%. and $H_2O_2-induced$ cytotoxicity was blocked by ENL (5. 10. 20. 30. and 40 ${\mu}g/ml$) concentration-dependently. The synthesis of MMP-1 and type I procollagen were positive in ENL. These results suggest that ENL has anti-oxidant and anti-furrowing effects.

Anti-aging effects of casuarictin on UVB-induced CCD-986sk cell (UVB유도성 CCD-986sk cell에서의 Casuarictin의 항노화 효과)

  • To develop a new functional agent for cosmetics, we investigated the anti-aging activities in fibroblasts of casuarictin. The anti-aging effect of casuarictin in CCD-986sk cell was as follows: it inhibited ROS expression increased by Ultraviolet B and suppressed pro-collagen expression. Also, casuarictin had inhibited Matrix metalloproteinase-1 expression. Therefore, the results suggested that casuarictin has considerable potential as a cosmetics ingredient with a anti-aging effect.

In vitro Effect of Ecklonia stolonifera Okamura Extract on the Cell Growth in CCD-986sk Human Fibroblast and Melanin Formation Inhibition in Clone M-3 Mouse Melanocyte Cell Line (청정해역 곰피추출물의 세포생리활성 연구)

  • In order to investigate whether or not CCD-986sk cell line can be affected by Korean Ecklonia stolonifera Okamura, we examined the MTT assay when we treated Korean Ecklonia stolonifera Okamura extract in CCD-986sk human fibroblast cell line. The sample were tested for cell proliferation activity by means of a modification of the MTT assay. Ecklonia stolonifera Okamura extract showed significantly strong cell proliferation activity at the range of from 6.25 mg $mL^{-1}$ to 1.56 mg $mL^{-1}$ compared with control group. And in order to search for inhibition agents of skin melanin formation, we tested for inhibition effect of melanin pigmentation of Korean Ecklonia stolonifera Okamura using Clone M-3 mouse melanocyte cell lines. when we treated the extracts of Ecklonia stolonifera Okamura to the mouse melanocyte cell lines, the sample showed a significantly little formation of melanin pigments compared with control group at the only range of 200 mg $mL^{-1}$. These results suggest that extract of Korean Ecklonia stolonifera Okamura may represents an excellent candidate for inhibition of melanin pigmentation and for protection of human skin aging at in vitro level.

Antioxidant and Antiwrinkle Effects of Persimmon Leaves extract (시엽(Persimmon Leaves) 에탄올 추출물의 항산화와 항주름 효과)

  • In this study, we investigated the antioxidant and anti-winkle activity in human fibroblast cell (CCD-986sk) of Persimmon Leaves (PL) as a cosmetic ingredient. As a result of investigating antioxidant activity through electron-donating ability and ABTS+ radical scavenging assay, the PL showed concentration-dependent antioxidant activity similar to ascorbic acid, a control group, at a concentration of 1,000 ㎍/ml. As a result of investigating the anti-wrinkle effect through elastase inhibition and collagenase inhibition assay, the PL showed concentration-dependent antioxidant activity similar to epigallocatechin gallate, a control group, at a concentration of 1,000 ㎍/ml. As a result of measuring the synthesis rate of pro-collagen type I and the inhibition rate of MMP-1 in UVB-induced CCD-986sk cells, the control group EGCG showed a 90.2% pro-collagen synthesis rate at 20 ㎍/ml and PL showed an 88.5% synthesis rate at 30 ㎍/ml. In addition, the inhibition rate of MMP-1 of 33.0% and 40.8% were confirmed in EGCG 20 ㎍/ml and PL 30 ㎍/ml, respectively. As a result of measuring the protein expression of pro-collagen type I and MMP-1 in the PL through western blot, it was confirmed that the protein expression of pro-collagen type I increased, and MMP-1 decreased when the PL was treated together compared to the UVB alone group. According to the above experimental results, it is expected to be used as a natural product material for cosmetics by confirming that the PL prevent photoaging caused by UVB stimulation and have antioxidant and anti-wrinkle effects.

α-Mangostin and Apigenin Induced Cell Cycle Arrest and Programmed Cell Death in SKOV-3 Ovarian Cancer Cells

  • Ovarian cancer is the fifth main cause of pre-senescent death in women. Although chemotherapy is generally an efficient treatment, its side effects and the occurrence of chemotherapeutic resistance have prompted the need for alternative treatments. In this study, ${\alpha}$-mangostin and apigenin were evaluated as possible anticancer alternatives to the chemotherapeutic drug doxorubicin, used herein as a positive control. The ovarian adenocarcinoma cell line SKOV-3 (ATCC No. HTB77) was used as model ovarian cancer cells, whereas the skin fibroblast line CCD-986Sk (ATCC No. CRL-1947) and lung fibroblast line WI-38 (ATCC No. CCL-75) were used as model untransformed cells. Apigenin and doxorubicin inhibited the growth of SKOV-3 cells in a dose- and time-dependent manner. After 72 hr exposure, doxorubicin was mostly toxic to SKOV-3 cells, whereas apigenin was toxic to SKOV-3 cells but not CCD-986Sk and WI-38 cells. ${\alpha}$-Mangostin was more toxic to SKOV-3 cells than to CCD-986Sk cells. A lower cell density, cell shrinkage, and more unattached (floating round) cells were observed in all treated SKOV-3 cells, but the greatest effects were observed with ${\alpha}$-mangostin. With regard to programmed cell death, apigenin caused early apoptosis within 24 hr, whereas ${\alpha}$-mangostin and doxorubicin caused late apoptosis and necrosis after 72 hr of exposure. Caspase-3 activity was significantly increased in ${\alpha}$-mangostin-treated SKOV-3 cells after 12 hr of exposure, whereas only caspase-9 activity was significantly increased in apigenin-treated SKOV-3 cells at 24 hr. Both ${\alpha}$-mangostin and apigenin arrested the cell cycle at the $G_2/M$ phase, but after 24 and 48 hr, respectively. Significant upregulation of BCL2 (apoptosis-associated gene) and COX2 (inflammation-associated gene) transcripts was observed in apigenin- and ${\alpha}$-mangostin-treated SKOV-3 cells, respectively. ${\alpha}$-Mangostin and apigenin are therefore alternative options for SKOV-3 cell inhibition, with apigenin causing rapid early apoptosis related to the intrinsic apoptotic pathway, and ${\alpha}$-mangostin likely being involved with inflammation.

Physiological Activities of Cudrania tricuspidata Extracts (Part I) (꾸지뽕나무 추출물의 생리 활성(제1보))

  • Cudrania tricuspidata has been used for a long time as a traditional herb medicine in Korea nad China. This paper has shown the experimental results about the physiological activities of water-, ethanol-, ethyl acetate-soluble fractions from ethanolic extracts of leaves, stems and roots of Cudrania tricuspidata. The effects of these fractions on the growth of various cells have exhibited that the ethyl acetate fractions from leaves, stems and roots inhibited significantly the growths of macrophage(RAW 264.7 cell), melanoma cell(B16-F10 cell), fibroblast cell(CCD-986sk cell), and lung carcinoma cell(A549 cell). The water and ethanol fractions of leaves and ethanol fraction of stems demonstrated better antioxidant activities scavenging radicals than other fractions when compared with the concentrations of different fractions for scavenging free radical DPPH (di(phenyl)-(2,4,6-trinitrophenyl) iminoazanium).

Enhancement of the Cosmeceutical Activity by Nano-encapsulation of Thiamine Di-lauryl Sulfate (TDS) with antimicrobial efficacy (항균 효능이 있는 비타민 B1 유도체(Thiamine Dilauryl Sulfate:TDS)의 나노입자화를 통한 기능성 향장 활성 증진)

  • This study was to improve cosmetical activity of thiamine di-lauryl sulfate (TDS) by encapsulation of nanoparticle with lecithin. Results showed that most of the nanoparticles containing the TDS were well formed in round shape with below 150 ~ 200 nm diameter as well as they were fairly stable in various pH ranges by measuring zeta potentials. The nanoparticles of TDS resulted in 85% cell viability of human normal fibroblast cells (CCD-986sk) when added at the highest concentration (1.0 mg/mL). The nanoparticles of Acer mono sap showed highest free radical scavengering effect as 88.1% in adding sample (1.0 mg/mL), compared to TDS solution of non-encapsulation (81.6%). The nanoparticles of TDS reduced the expression of MMP-1 on UV-irradiated CCD-986sk cells down to as 41.4%. The TDS solution and nanoparticles showed significant anti-microbial activities agaionst the salmonella typhimurium and listeria monocytogenes at 5 and 6 days as compared with control. Anti-microbial activities of TDS nanoparticles were similar to positive control. These results indicated that TDS nanoparticles may be a source for functional cosmetic agents capable of improving cosmetical activity such as antioxidant, whitening, and anti-wrinkling effects and can be further developed as natural preservative in cosmetics.

Skin Whitening and Skin Immune Activities of Different Parts of Acer mono and Acer okamotoanum (고로쇠 및 우산고로쇠나무의 부위별 미백 및 피부면역 활성)

  • UV-protection skin whitening and immune activities several parts of Acer mono and Acer okamotoanum were investigated. The bark of both Acer mono and Acer okamotoanum had higher yields than other parts as 2.67% and 2.45%. The cytotoxicity of the extracts were lower than 21.64% against human skin cell(CCD-986sk) line in adding 1.0 mg/mL of the highest concentration. The bark extracts of Acer mono greatly reduced the expression of MMP-1 on UV-irradiated CCD-986sk cells down to as 30%. At 1.0 mg/mL of bark extration of Acer mono, $PGE_2$ expression was also significantly decreased. Generally, the bark extracts of Acer mono and Acer okamotoanum had higher activity than other parts, but, interestingly, wood extract of Acer okamotoanum showed strong inhibition effect on melanin production by Clone-M3 cells as 79.25%. From these results, we could conclude that the bark extract from Acer mono and Acer okamotoanum had skin-whitening activity as well immune enhancement activity.
