Evaluation of the Parameters of Soil Potassium Supplying Power for Predicting Yield Response, K2O Uptake and Optimum K2O Application Levels in Paddy Soils. -II.   • In order to predict the possible fertilizer requirement from the K supplying capacity of soil, the relative K activity ratio, Kas/Kai and Gapon coefficients, KG. were determined for the soil samples before flooding and at heading stage of rice in pot experiment. These parameters assumed as the K supplying capacity of soils were discussed through correlation with other factors such as grain yields or the amounts of $K_2O$ uptake by the rice plant. The results may be summarized as follows: 1. The KGo values in soils before flooding were 7.8, 6.6, and 7.1, whereas the Kas/Kai values were 1.37, 1.26 and 2.11, respectively, in clay, loam and sandy loam soils. 2. The significant yield responses to the application of potassium fertilizer were observed whenever the KG values in soils at heading stage become larger to the original KG values, regardless of any levels of fertilizer application. 3. The linear correlations between the exchangeable cation ratios [Kex./(Ca+Mg) ex.:me/100g] in soils and the potassium activity ratios ($[K^+]/\sqrt{[Ca^{{+}{+}}+Mg^{{+}{+}}]}$: mole/l) in equilibrium solutions were observed with different linear gradients according to the soil properties. 4. The Kas/Kai in the soils, estimated prior to the experiment, showed high correlations with the grain yields or the amounts of $K_2O$ uptake in the all treatments, while the Kas/Kai and the KGo in the soils at heading stage showed high correlations with the grain yields or the amounts of $K_2O$ uptake in only N 15 Kg/10a treatments. 5. The Kas/Kai and the KGo values determined in the soil at heading stage of rice showed high negative correlation each other and they could be used as soil factors for predicting potassium fertilizer requirement.

Representative Physical and Chemical Properties of Korean Soils by the Results from Detailed Soil Survey (우리나라 토양(土壤)의 대표적(代表的)인 물리화학적(物理化學的) 특성(特性) (정밀토양조사결과(精密土壤調査結果)를 중심(中心)으로))

  • These studies were aimed to find out the representative values of physico-chemical properties in Korean soils and to serve the basic informations to improve the soil properties for increasing the soil productivity. By the results of detailed soil survey, 14 physico-chemical properties (soil texture, moisture characteristics, organic matter content etc) were collected from 315 series of soils except Cheju soils, computerized the simple mean and the distributed area by weighted mean values with grouping of land use, drainage class and soil type. The results were as follows; 1. Simple mean values within the efficient soil depth in whole country soils were clay 20.0%, organic matter 2.03% and CEC 10.3me/100g. But weighted mean values with land area belonged to the same soil series were clay 18.0%, OM 1.85% and CEC 8.6me/100g. 2. Mean values of paddy soils weighted by area were clay 19.7%, OM 2.0% and CEC 9.1me/100g but those of upland soils were 17.9%, 1.8% and 8.1me/100g and forest soils were 16.7%, 1.8% and 8.6me/100g respectively. 3. Weighted mean values of moderately well soils with covered area were clay 18.9%, organic matter 1.7%, CEC 8.4me/100g but those of imperfectly drained soils were 21.3%, 2.2% and 9.5me/100g, and those of poorly drained soils were 15.1%, 1.8% and 9.9me/100g respectivcly. 4. Simple mean and area weighted mean values of clay content, pH, organic matter contents, cation exchange capacity and base saturation were calculated by the types of paddy and upland.

Determination of Application Rate of Composted Pig Manure for Wetland Rice (논토양에서 돈분톱밥퇴비 시용량 결정에 관한 연구)

  • A study was conducted for the determination of application rate of pig-dung composted with sawdust (referred as pig manure hereafter) for wetland rice. The study involved the field experiments with the cultivation of rice under different rates of application of pig manure in combination of different rates of chemical fertilizers, in a wetland rice soil. The field experiment involved following treatments : (I) Without fertilizer, (II) Standard rate of chemical fertilizers based on soil testing($98-73-71kg\;ha^{-1}$ as $N-P_2O_5-K_2O$), (III) $2.1Mg\;ha^{-1}$ of pig manure $>+80-37kg\;ha^{-1}$ of $N- K_2O$ as chemical fertilizer(Less $N-P_2O_5-K_2O$ contained in the compost), (IV) $4.2Mg\;ha^{-1}$ of pig manure+ $62-3kg\;ha^{-1}$ of $N-K_2O$as chemical fertilizer(Less $N-P_2O_5-K_2O$ contained in the compost), (V) $10Mg\;ha^{-1}$ of pig manure+ Treatment(II), (VI) $20Mg\;ha^{-1}$ of pig manure +Treatment(II). Number of tillers in treatment (I) were higher than other treatments in tillering and panicle formation stage. After heading stage, treatments (V) and (VI) have higher number of tillers, but treatment (III) and (IV) have fewer number of tillers during all growing stage. Uptake of NPK in rice plants was higher in treatment (VI), but the efficiency of N, P and K was higher in treatment (I), (III) and (IV). The yield of unhulled rice were in order of tretments (VI)>(V)>(IV)>(II)>(III), although the difference was not statistically significant. Inorganic nitrogen, available P and exchangable K contents in soil were highest at tillering stage in all treatments and became low from panicle formation to harvest stage. Available P in soil was increased by the application of pig manure upto 20 cm depth. Exchangeable cation contents in 40 to 60 cm soil depth was much higher in treatment (VI) than in other treatments. Treatment (V) and (VI) showed much higher losses of N. $P_2O_5 $ and $K_2O$ than other treatment. Though treatment (VI) tended yield higher than in other treatments, showed lodging and occurrence of leaf and neck blast in this treatment. Yield of unhulled rice in treatment (IV) was not significant statistically and reduced nutritional losses. It is conclude that treatment (IV) seems to be the most reasonable one for the application of pig manure in combination of chemical fertilizers.

Leaching and Distribution of Cation in Multi-layered Reclaimed Soil Column with Intermediate Macroporous Layer (대공극층위 형성 간척지 다층토주의 양이온 용탈 및 분포)

  • To investigate desalinization patterns of surface reclaimed saline-sodic soil (RSSS) with subsurface layer of macroporous medium, multi-layered soil columns were constructed. For the multi-layered soil columns, gypsum was treated at the rate of 5 cmolc $kg^{-1}$ in surface (top) while coal bottom ash (CBA) was placed into intermediate layer below the gypsum-treated surface soils followed by the reclaimed saline-sodic soil as bottom layer (BL). The lengths of top soil was 30 cm long while the lengths of the CBA were 20 and 30 cm long. The saturated hydraulic conductivities (Ksat) were $0.39{\times}10^{-4}$ and $0.31{\times}10^{-4}cm\;sec^{-1}$ for RSSS(30 cm)-CBA(20 cm)-BL(20 cm) and RSSS(30 cm)-CBA(20 cm)-RSSS(20 cm), respectively while the lowest $K_{sat}$. was $0.064{\times}10^{-4}cm\;sec^{-1}$ for RSSS(30 cm)-CBA(20 cm)+BL(20 cm). The time required to reach the lowest EC in eluent, 0.3 dS $m^{-1}$ from 33.9 dS $m^{-1}$ was shorter in multi-layered soil columns with GR-CBA than that of RS-SRS, representing that rate of desalinization was greater than 99%. Exchangeable Na decreased by 94.8~96.2 %, while exchangeable Ca increased by 98~129 %.

Patterns of Leaching and Distribution of Cations in Reclaimed Soil according to Gypsum Incorporation Rate (석고 혼합량에 따른 간척지토양의 양이온 용탈 및 분포)

  • Serious problems in reclaimed land for agriculture are high soil salinity and very poor vertical drainage. However, desalinization in these soils is very difficult. To identify the change of soil permeability by the gypsum incorporation in soils we observed elution patterns and salt distribution of the soil using soil columns packed with reclaimed saline soil with various rates of gypsum amendment. Saturated hydraulic conductivity ($K_{sat}$) of the top soil of reclaimed saline soils without gypsum incorporation was close 0 cm $hr^{-1}$ while $K_{sat}$ increased up to 0.3 cm $hr^{-1}$ with increasing amount of gypsum for 0.4% or more gypsum. Also $K_{sat}$ of the reclaimed saline soils for top soil was drastically increased to 1.0 cm $hr^{-1}$ or slightly greater after 141 hour's elution experiment. The cumulative amount of cation recovered in the effluent also increased in the order of gypsum incorporation rate 0.8% > 0.6% > 0.4 > 0.2%. Soil EC in soil columns decreased from initial 33.9 dS $m^{-1}$ to less than 0.4 dS $m^{-1}$ and exchangeable Ca 2+ increased by 32~140% according to gypsum incorporation rate.

Physicochemical Properties of the Synthetic Hectorite (합성 헥토라이트의 물리화학적 특성)

  • Hectorite was synthesized under hydrothermal conditions and its physicochemical properties have been investigated in terms of temperature, pH, and organic agent to observe the change of doll basal spacing. The IR, CEC, MB, swelling volume and specific surface area of the hectorite were measured for the characterization. The solid/liquid ratio of hectorite to distilled water before mixing with other materials was also determined for its use as a multi-functional material. The $d_{001}$ basal spacing decreased from $12.63\;\AA$ at room temperature to $10.19\;\AA$ at $650^{\circ}C$ in the heating tests. As the pH of hectorite slurry increased. the $d_{001}$ basal spacing decreased. reaching the lowest value of $13.33\;\AA$ at pH 7 and afterward, increased. All the fool basal spacings of the hectorite increased when it was intercalated with the following solvents: $12.86\;\AA$ in diethyl ether, $13.31\;\AA$ in acetonitrile. $13.59\;\AA$ in methanol, $14.05\;\AA$ in ethanol, $15.69\;\AA$ in acetone, and $17.42\;\AA$ in ethylene glycol. Our IR analysis results were in good agreement with those of other researchers. The CEC, MB, swelling volume and specific surface area of hectorite were determined to be 105 cmol/kg, 80 cmol/kg, $68\sim74ml/2g$ and $213m^{2}/g$, respectively. Also, the hectorite to distilled water ratio of 2 to 100 was found to be most favorable for mixing with other materials such as the solvents mentioned above.

Quantitative Real-Time PCR of Porcine Parvovirus as a Model Virus for Cleaning Validation of Chromatography during Manufacture of Plasma Derivatives (혈장분획제제 제조공정에서 크로마토그래피 세척 검증을 위한 모델바이러스로서의 Porcine Parvovirus 정량)

  • Chromatography has now been used successfully to provide the requisite purity for human plasma-derived biop-harmaceuticals such as coagulation factors and immunoglobulins. Recently, increasing attention has been focused on establishing efficient cleaning procedures to prevent potential contamination by microorganisms as well as carry-over contamination from batch to batch. The purpose of present study was to develop a cleaning validation system for the assurance of virus removal and/or inactivation during chromatography process. In order to establish an assay system for the validation of virus clearance during chromatography cleaning process, a quantitative real-time PCR method for porcine parvovirus(PPV) was developed, since PPV, a model virus for human parvovirus B19, has a high resistance to a range of physico-chemical treatment. Specific primers for amplification of PPV DNA was selected, and PPV DNA was quantified by use of SYBR Green I. The sensitivity of the assay was calculated to be 1.5 $TCID_{50}/ml$. The established real-time PCR assay was successfully applied to the validation of PPV removal and cleaning during SP-Sepharose cation chromatography for thrombin purification and Q-Sepharose anion chromatography for factor VIII purification. The comparative results obtained by real-time PCR assay and infectivity titrations suggested that the real-time PCR assay could be a useful method for chromatography cleaning validation and that it could have an additive effect on the interpretation and evaluation of virus clearance during the virus removal process.

Mössbauer Study of Tb2Bi1GaxFe5-xO12(x=0, 1) (Tb2Bi1GaxFe5-xO12(x=0, 1)의 뫼스바우어 분광연구)

  • $Tb_2Bi_1Ga_xFe_{5-x}O_{12}$(x=0, 1) fabricated by sol-gel and vacuum sealed annealing process. $Tb_2Bi_1Ga_xFe_{5-x}O_{12}$(x=0, 1) have been studied by x-ray diffraction(XRD), vibrating sample magnetometer, and $M\ddot{o}ssbauer$ spectroscopy. The crystal structures were found to be a cubic garnet structure with space group Ia3d. The determined lattice constants $a_0$ of x = 0, and 1 are $12.497\AA$, and $12.465\AA$, respectively. The distribution of gallium and iron in $Tb_2Bi_1Ga_xFe_{5-x}O_{12}$ is studied by Rietveld refinement. Based on Rietveld refinement results, the terbium and bismuth ions occupy the 24c site, iron ions occupy the 24d, l6a site, and nonmagmetic gallium ions occupy the 16a site. In order to verify the magnetic site occupancy of iron and gallium, we have taken $M\ddot{o}ssbauer$ spectra for $Tb_2Bi_1Ga_xFe_{5-x}O_{12}$(x=0, 1) at room temperature. From the results of $M\ddot{o}ssbauer$ spectra analysis, the absorption area ratios of Fe ions for $Tb_2Bi_1Fe_5O_{12}$ on 24d and 16a sites are 60.8 % and 39.2 %, respectively, and the absorption area ratios of Fe ions for $Tb_2Bi_1Fe_5O_{12}$ on 24d and 16a sites are 74.7 % and 25.3 %, respectively. It is noticeable that all of the nonmagnetic Ga atoms occupy the 16a site by vacuum annealing process.

Chemical characteristics of Rainwater in Suwon (수원지역 강우의 화학적 특성)

  • To evaluate the acidity and chemical characteristics of rainwater in Korea, its pH and ion concentrations were investigated in Suwon from April to December, 2006. In addition, to estimate the contribution of ions on its acidity, ion composition and neutralization effect of major cations were investigated. Ion balance and electrical conductivity balance between measured and estimated values showed a high correlation. The mean pH and EC in rainwater collected during the investigation periods were 4.7 and $17.6{\mu}S\;cm^{-1}$, respectively. The monthly variation in EC showed a clear seasonal pattern, which had the lowest value of $9.1{\mu}S\;cm^{-1}$ in July and increased remarkably in November. $Na^+$ was the most abundant cation and followed by $NH_4{^+}>Ca^{2+}>H^+>Mg^{2+}>K^+$. Among them, $Na^+$ and $NH_4{^+}$ accounted for more than 65% of the total cations. In case of anions, the relative abundance was $SO_4{^{2-}}>NO_3{^-}>Cl^-$. About 67% of the total anions in rainwater was $SO_4{^{2-}}$, which showed $119.0{\mu}eq\;L^{-1}$ as mean value during the monitoring periods. Furthermore, 94% of the soluble sulfate in rainwater was identified as nss-$SO_4{^{2-}}$(non-sea salt sulfate). We also found that $NH_4{^+}$ and $Ca^{2+}$ contributed greatly in neutralizing the rain acidity, especially in dry season.

Effects of Production Performance, Immunity and Egg Quality by Raising on Exercise Yard in Laying Hens (산란계 운동장 사육이 생산성, 면역성 및 계란의 품질에 미치는 영향)

  • The present study examined effects of production performance, immune activity and egg quality by raising on exercise yard in laying Hens, the results of which could be used as baseline data to enhance animal welfare and the safety of livestock products. A total of 90 Hy-line-Brown laying hens of 13 weeks old were used in the experiment for 38 weeks. The cage group (Cage group) was raised in a cage, where an area of $0.084m^2$ was assigned to two hens, while hens in the exercise yard $1.1m^2$ group ($1.1m^2$ group) was assigned to a combination of a chicken house ($0.11m^2$) and a exercise yard ($1.1m^2$) per a hen. Hens in the exercise yard $2.2m^2$ group ($2.2m^2$ group) was assigned to a combination of a chicken house ($0.11m^2$) and a exercise yard ($2.2m^2$) per a hen. Treatment was replicated 3 times with ten birds per replication. Ten birds were arranged according to randomized block design. While initial egg production rate was significantly higher in the Cage group, $1.1m^2$ group exhibited a slightly higher rate in the mid- and late-stage of the experiment, although the difference was not statistically significant. Exercise yard treatment groups exhibited a higher feed intake rate than the Cage group up until the hens were 39 weeks old (P<0.05), but the difference dissipated from that age on. The age at first egg in the exercise yard treatment groups was 16 days later than that for the Cage group (P<0.05), although differences in the quality of the eggs were not observed. The results of immune activity test showed that IgA in the exercise yard treatment groups was significantly higher than that in the Cage group (P<0.05). IgG, IgM, and corticosterone were also higher in the exercise yard treatment groups. The soil in exercise yard increased of organic matter and decreased of cation exchange capacity (CEC) in raised hens. In sum, raising hens in a exercise yard raise style decreased the rate of initial egg production, which was followed by a slight increase during the latter part of the experiment. The exercise yard raise hens' immune activity was heightened.
