Inhibitory effect of Ulmus davidiana Planch extracts on bone resorption mediated by processing of cathepsin K in cultured mouse osteoclasts

  • Objective: Ulmus davidiana Planch (Ulmaceae) has long been known to have anti-inflammnatory in the traditional Korean medicine. UD has been reported as a good enhancer for bone healing. Methods : In this experiment, we investigate the Inhibitory effects of UD on bone resorption using the bone cells culture. Different concentrations of crude extract of UD were added to mouse bone cells culture. The mitochondria activity of the bone cells after exposure was determined by colorimetric MIT assay. It was demonstrated that UD has potential effects on bone cells culture without any cytotoxicity. The most effective concentration of UD on bone cells were $100\;{\mu}g/ml$. Cathepsin K (Cat K) is the major cysteine protease expressed in osteoclasts and is thought to play a key role in matrix degradation during bone resorption. Results : When mouse long bone cells including osteoclasts and osteoblast were treated with the PI3-Kinase inhibitor, wortmannin (WT), WT prevented the osteoclast-mediated intracellular processing of Cat K. Similarly, treatment of osteoclasts-containing long bone cells with UD extracts prevented the intracellular maturation of Cat K, suggesting that UD may disrupt the intracellular trafficking of pro Cat K. This is similar to that of WT. Since secreted proenzymes have the potential to reenter the cell via mannose-6-phosphate (M6P) receptor, to prevent this possibility, we tested WT and UD in the absence or presence of M6P. Inhibition of Cat K processing by WT or UD was observed in a dose-dependent manner. Furthermore, the addition of M6P resulted in enhanced potency of WT and UD. Conclusion : UD dose-dependently inhibited in vitro bone resorption with a potency similar to that observed for inhibition of Cat K processing.

Spirometra decipiens (Cestoda: Diphyllobothriidae) Collected in A Heavily Infected Stray Cat from the Republic of Korea

  • Morphological and molecular characteristics of spirometrid tapeworms, Spirometra decipiens, were studied, which were recovered from a heavily infected stray cat road-killed in Eumseong-gun, Chungcheongbuk-do (Province), the Republic of Korea (=Korea). A total of 134 scolices and many broken immature and mature proglottids of Spirometra tapeworms were collected from the small intestine of the cat. Morphological observations were based on 116 specimens. The scolex was 22.8-32.6 mm (27.4 mm in average) in length and small spoon-shape with 2 distinct bothria. The uterus was coiled 3-4 times, the end of the uterus was ball-shaped, and the vaginal aperture shaped as a crescent moon was closer to the cirrus aperture than to the uterine aperture. PCR amplification and direct sequencing of the cox1 target fragment (377 bp in length and corresponding to positions 769-1,146 bp of the cox1 gene) were performed using total genomic DNA extracted from 134 specimens. The cox1 sequences (377 bp) of the specimens showed 99.0% similarity to the reference sequence of S. decipiens and 89.3% similarity to the reference sequence of S. erinaceieuropaei. In the present study, we report a stray cat heavily infected with S. decipiens identified by mitochondrial cox1 sequence analysis and morphological examinations of the adult worms.

The Effects of Donor Cell Type and Culture Medium on in vitro Development of Domestic Cat Embryos Reconstructed by Nuclear Transplantation

  • In this study we explored the possibility of performing nuclear transfer in the domestic cat and assessed the ability of different culture media to support in vitro development of reconstructed cat embryos. Donor somatic cells were derived from cultured cumulus cells or explants of oviduct tissue, and recipient cytoplasts from in vitro matured oocytes. A higher percentage of cleavage (84.6% and 86.5%) and development to the morula stage (35.9% and 44.2%) was found when reconstructed embryos receiving cumulus or oviduct cells were cultured in MK1 medium, compared with those cultured in CR1aa (58.7% and 72.5%, 13.8% and 13.6%, respectively). There was no significant difference between MK1 and CR1aa media with respect to the proportion developing to the blastocyst stage (15.4% and 17.3% vs 6.8% and 8.6%, respectively, p>0.05). There was no significant effect (p>0.05) of donor cell type (cumulus and oviduct cells) on the rates of fusion (65.0% and 52.5%), cleavage (84.6% and 86.5%), development to the morula (35.9% and 44.2%), and blastocyst (15.4% and 17.3%) stages when reconstructed embryos were cultured in MK1 medium. Similar results were found for the reconstructed embryos cultured in CR1aa medium. These results show that culture medium has a significant impact on the early development of reconstructed cat embryos, whereas donor cell type does not have a significant effect.

Intestinal T cell lymphoma in a cat, Korea (고양이 장에서 발생한 T 세포 림프종)

  • An 11 year-old male Korean short-haired cat was presented to local animal hospital due to weight loss, vomiting, and intestinal hypomotility. After the cat was euthanized by poor clinical outcomes, necropsy was performed at Animal and Plant Quarantine Agency. At necropsy, the stomach was enlarged and had some nearly complete pellet food and the yellow mucous contents. The lumen of the middle and lower parts of the jejunum became narrow. Histopathologically, medium-sized lymphoid cells with hyperchromatic nuclei enclosed by scant cytoplasm were diffusely proliferated from mucosa to serosa of the small intestine. These findings were mainly observed in the jejunum and slightly in the duodenum and ileum. The monomorphous lymphocytes were 1 to 1.5 times larger than red blood cells and had few mitotic figures. Metastasis of the tumor cells to other organs was not observed. In the result of immunohistochemical analysis for identifying the origin of tumor cells, CD3 was expressed, but $CD79{\alpha}$ was not detected in the infiltrated cells. This case was diagnosed as T cell intestinal lymphoma in a Korean short-haired cat based on the clinical signs, gross findings, histopathology, and immunohistochemistry.

Exploring the Predictive Variables of Government Statistical Indicators on Retail sales Using Machine Learning: Focusing on Pharmacy (머신러닝을 이용한 정부통계지표가 소매업 매출액에 미치는 예측 변인 탐색: 약국을 중심으로)

  • This study aims to explore variables using machine learning and provide analysis techniques suitable for predicting pharmacy sales whether government statistical indicators built to create an industrial ecosystem based on data, network, and artificial intelligence affect pharmacy sales. Therefore, this study explored predictive variables and performance through machine learning techniques such as Random Forest, XGBoost, LightGBM, and CatBoost using analysis data from January 2016 to December 2021 for 28 government statistical indicators and pharmacies in the retail sector. As a result of the analysis, economic sentiment index, economic accompanying index circulation change, and consumer sentiment index, which are economic indicators, were found to be important variables affecting pharmacy sales. As a result of examining the indicators MAE, MSE, and RMSE for regression performance, random forests showed the best performance than XGBoost, LightGBM, and CatBoost. Therefore, this study presented variables and optimal machine learning techniques that affect pharmacy sales based on machine learning results, and proposed several implications and follow-up studies.

Denaturation and Inactivation of Antioxidative Enzymes due to Repeated Exposure to UV-B and Inhibitory Effect of RGP Lens (UV-B 반복노출에 따른 항산화효소의 변성 및 활성저하와 RGP렌즈의 차단효과)

  • Purpose: The present study was conducted to reveal the correlation of structural denaturation and decrease of enzyme activity when the antioxidative enzymes, superoxide dismutase (SOD) and catalase (CAT) were repeatedly exposed to UV-B, and further investigate whether the denaturation and inactivation of those enzymes can be effectively blocked by using UV-inhibitory RGP lens. Methods: Each enzyme solution was prepared from the standardized SOD and CAT, and repeatedly exposed to UV-B of 312 nm for 30 minutes, 1 hour and 2 hours a day over 1, 2, 3, 4 and 5 days. Structural denaturation of SOD and CAT induced by repeat UV-B irradiation was confirmed by the electrophoretic analysis, and their enzyme activity was determined by the colorimetric assay using the proper assay kit. At that time, the change in structure and activity of the antioxidant enzymes directly exposed to UV-B was compared to the case that UV-B was blocked by UV-inhibitory RGP lens. Results: SOD exposed repeatedly to UV-B showed the polymerization pattern in the electrophoretic analysis when it repeatedly exposed for 30 min a day, however, the change of its activity was less than 10%. On the other hand, CAT repeatedly exposed to UV-B reduced size and density of the electrophoretic band which indicated a structure denaturation, and its activity was significantly decreased. In the case that the repeat exposure time was longer, CAT activity was completely lost even though some enzyme band occurred in the electrphoretic analysis. In addition, the degeneration of CAT due to UV-B irradiation was inhibited to some extent by using RGP lens with a UV-B blocking of 63.7%, however, it was not completely inhibited. Conclusions: From these results, it was revealed that the structural denaturation of antioxidative enzymes was not perfectly correlated with the reduction in enzyme activity according to the type of enzyme. It is recommended to minimize the exposure time to UV when wearing contact lens, or wear the contact lenses having UV blocking rate of the FDA Class I blocker or the sunglasses having equivalent UV-blocking rate for reducing the damage of antioxidative enzymes induced by UV.

A Quantitative Ultrastructural Study on the Effects of Ischemia and Reperfusion on the Rat and Cat Hearts (허혈 및 재관류가 흰쥐 및 고양이 심장에 미치는 영향에 관한 형태계측학적 연구)

  • To understand the structural changes of the myocardial myocytes and endothelial cells in ischemic and reperfused heart, and to elucidate their roles in those conditions, the authors observed cat and rat myocardium ultrastructurally and evaluated them with morphometric techniques. In cat, mild ischemia and moderate degree reperfusion injury was induced by ligation of the anterior interventricular branch of left coronary artery and reperfusion. In rat, severe ischemia and irreversible reperfusion iniury was made using in vitro Langendorff techniques. In normal cat myocytes, the volume densities of cytoplasm, myofibrils, mitochondria, sarcoplasmic reticulum and T tubules were $0.11{\pm}0.013,\;0.51{\pm}0.096,\;0.25{\pm}0.082,\;0.09{\pm}0.008,\;0.02{\pm}0.010$ (Mean${\pm}$S.D.) respectively, and the myofibril/mitochondria ratio was $2.33{\pm}1.379$. The numerical density and average volume of mitochondria were $0.76{\pm}0.210/{\mu}m^3$ and $0.33{\pm}0.057{\mu}m^3$ respectively. In normal cat endothelial cells, the volume densities of cytoplasm, cytoplasmic vesicles, tubular systems (including endoplasmic reticulum and Golgi apparatus) and mitochondria were $0.43{\pm}0.023,\;0.28{\pm}0.007,\;0.22{\pm}0.021,\;0.03{\pm}0.014$ respectively. The mean thickness of endothelial cells was $230{\pm}45.2{\mu}m$. The numerical density and average volume of cytoplasmic vesicles were $508{\pm}55.0/{\mu}m^3,\;578{\pm}104.8nm^3$ respectively. In cat myocytes which received mild ischemic injury, the volume densities of organelles were not changed significantly in ischemic and reperfusion states. In reperfusion group myocytes, the numerical density of mitochondria was decreased significantly and the average volume was increased significantly. In endothelial cells, the volume density of tubular system in ischemic group and the average volume of cytoplasmic vesicles in reperfusion group were increased significantly. In rat myocytes which received severe ischemic injury, the volume density and average volume of mitochondria were increased significantly, and the volume density of sarcoplasmic reticulum and numerical density of mitochondria were decreased significantly in both ischemic and reperfusion groups. In ischemic and reperfused endothelial cells, the volume density and numerical density of cytoplasmic vesicles, the volume density of cytoplasm were decreased significantly. The volume densities of tubular system were increased significantly in both ischemic and reperfused groups. The volume density of mitochondria in ischemic group and the average volume of cytoplasmic vesicles in reperfusion group showed significant increase. The authors, based on the above observations, conclude that the mitochondria of myocytes and the cytoplasmic vesicles of endothelia are the first group of targets in ischemic and reperfusion injury and in this respect, the degree of ischemic insult is not significant. The role of myocyte mitochondria in reperfusion injury may be insignificant, but endothelial cells may contribute actively to reperfusion injury.

The Study of Hepatic Antioxidative Enzyme Activity and Eletrophoresis in Mice After Treatment with Paraquat and/(or) Ginseng Saponins (Paraquat를 투여한 생쥐 간에서 홍삼 사포닌의 항상화 효소 활성과 전기영동에 관한 연구)

  • This study examined effects of the active ingredients from ginseng on paraquat(PQ) toxitity. Mice were given PQ(25mg/kg, ip) and then they were given total saponins (TS; 5mg/kg, orally), protopanaxadiol (PD; 5mg/kg, orally) and protopanaxatriol(PT; 5mg/kg, orally) per day for periods of 1,3 & 7 days. We measured the activities of superoxide dismutase (SOD), electrophoretic isozyme band, catalase (CAT) were compared in the liver of mouse that dose with PQ and/or TS, PD and PT. The activities of SOD, CAT were generally higher in PQ+PD group than others groups. Especially the activity of SOD was the highest in PQ+PD group than others groups. SOD isozyme separated into three bands by electrophoresis. One band was located to near the anode side and two bands were cathode side. As the results of treated with KCN, we were confiremed that the Cu, Zn-SOD was located to near the anode side but the Mn-SOD were cathode side. Our results suggested that an antioxidant effect of ginseng saponins elevated a protection ability to an oxidative damage by direct action of SOD, CAT and reinforced the synthetic ability of endogenous antioxidant material in living organism. Particularly, PD was a effective antioxidant compared with others.

Photochemical Damage and Responses of Antioxidant Enzymes in Rice Leaves Induced to Light-Chilling (Light-chilling에 의해 유도된 벼 잎에서의 광합성 변화와 항산화 효소의 반응)

  • We investigated photooxidation and responses of antioxidant enzymes involved in scavenging reactive oxygen species (ROS) after light-chilling ($4^{\circ}C$) for 2 days and post chilling ($25^{\circ}C$) in rice leaves. Chilling leaves indicated a 50% reduction in photosynthetic efficiency ($F_v/F_m$ ratio) and a 48% increase of $H_2O_2$, respectively, compared to the control group. In comparison with the control, activities of superoxide dismutase (SOD) and glutathione reductase (GR) increased at light-chilling and post-chilling. CuZn-SOD and Mn-SOD among SOD forms were detected in rice leaves, while Fe-SOD was not found. The increase of SOD and GR activity may serve as a basis for defense against chilling injury as it dismutase superoxide generated by light-chilling. Catalase (CAT) activity decreased during light-chilling, while activity of APX showed remarkable increase during light-chilling in rice leaves. Among CAT isoforms analyzed by 10% native PAGE, activities of isoform -2 and -3 were inhibited during light-chilling. From the elevated APX activity and decreased CAT activity, we suggest that these two enzymes show mutual supplementary relationships, indicating different tendency during light-chilling.

Cat Behavior Pattern Analysis and Disease Prediction System of Home CCTV Images using AI (AI를 이용한 홈CCTV 영상의 반려묘 행동 패턴 분석 및 질병 예측 시스템 연구)

  • Cats have strong wildness so they have a characteristic of hiding diseases well. The disease may have already worsened when the guardian finds out that the cat has a disease. It will be of great help in treating the cat's disease if the owner can recognize the cat's polydipsia, polyuria, and frequent urination more quickly. In this paper, 1) Efficient version of DeepLabCut for pose estimation, 2) YOLO v4 for object detection, 3) LSTM is used for behavior prediction, and 4) BoT-SORT is used for object tracking running on an artificial intelligence device. Using artificial intelligence technology, it predicts the cat's next, polyuria and frequency of urination through the analysis of the cat's behavior pattern from the home CCTV video and the weight sensor of the water bowl. And, through analysis of cat behavior patterns, we propose an application that reports disease prediction and abnormal behavior to the guardian and delivers it to the guardian's mobile and the server system.
