Studies on the Efficient Embryo Transfer Methods using Inbred Embryos in Generation of Transgenic Mice (Inbred 마우스 수정란을 이용한 형질전환마우스 생산에 있어서의 효과적인 수정란 이식 방법에 관한 연구)

  • The objective of this study is to improve the efficiency of embryo transfer in generation of transgenic mice using inbred mouse (C57BL/6J) embryos. The embryos of C57BL/6J and BCF1 mice were superovulated by the standard protocol. One-cell stage of embryos were microinjected and the resulted one-or two-cell were transferred into one-or two-side oviducts of foster mother, respectively. When one-cell stage of embryos were transferred into one-side oviduct of 0.75 d.p.c. foster mother, the results were not ideal because of showing pregnancy ratios of 68.8$\pm$7.83% for C57BL/6J and 48.3$\pm$14.22% for BCF1, and development ratios of pups vs transferred embryos of 11.9$\pm$5.51% for C57BL/6J and 10.5$\pm$8.03% fur BCF1 . However, when two-cell stage of embryos were transferred into two-side oviducts of 0.5 d.p.c. foster mother, we got better results of 94.4$\pm$9.64% and 100$\pm$0% pregnancy ratio, and 22.1$\pm$0.4% and 21.8$\pm$0.38% development ratio for C57BL/6J and BCF1, respectively. Therefore, transferring two-cell stage of C57BL/6J embryos into two-side oviducts of 0.5 d.p.c. foster mother may be competitive to the result in BCF1 embryos. Even if there are a lot of other factors affecting these results, we conclude from these experiments that transfer of two-cell embryos into two-side oviducts of 0.5 d.p.c. foster mother is a more efficient and safe method than others in generating transgenic mice using inbred mice embryos.

Modifying Effect of Diallyl Sulfide on Colon Carcinogenesis in C57BL/6J-ApcMin/+ Mice

  • Diallyl sulfide (DAS), a flavoring compound derived from garlic, is considered to have cancer chemopreventive potential in experimental animals and humans. This study was designated to examine possible chemopreventive effects of DAS on colon carcinogenesis using genetically engineered transgenic $Apc^{Min/+}$ mice, a well-established animal model for familial adenomatous polyposis (FAP) and sporadic colorectal cancer. Male C57BL/6J-$Apc^{Min/+}$ mice were divided into three groups. Animals of group 1 were placed on the basal diet (AIN-76A) as non-treated controls. Animals of groups 2 and 3 were given DAS-containing diets (in doses of 100 and 300 ppm, respectively). All mice were sacrificed at the end of week 10 of the experiment. Histopathological investigation revealed that the incidence of colonic polyps was decreased dose-dependently by 19% (13/16) in group 2 and by 32% (13/20) in group 3 compared to the 100% incidence (10/10) in group 1. The multiplicity of colonic polyps per mouse was also slightly decreased by DAS treatment ($1.88{\pm}0.35$ in group 2 and $1.63{\pm}0.36$ in group 3) compared to $2.00{\pm}0.39$ in group 1. On the other hand, there were no significant differences in the numbers of total polyps per mouse in the small intestine between the groups. Taken together, we suggest that DAS may exert promising inhibitory effects on colon carcinogenesis in the transgenic $Apc^{Min/+}$ mice.

Questionable Reliability of Malondialdehyde to Measure Oxidative Stress in Sjögren's Syndrome: Preliminary Study

  • Purpose: To investigate the expression of malondialdehyde (MDA), lipid peroxidation marker for oxidative stress (OS), in autoimmune Sjögren's syndrome (SjS) by utilizing the SjS-prone C57BL/6.NOD-Aec1Aec2 (B6DC) mouse and the SjS patient plasma samples. Methods: The MDA concentrations in the lysates of the submandibular gland, liver, and serum samples from the SjS-prone B6DC mouse model were compared with those from the C57BL/6J as a control. A thiobarbituric acid reactive substance (TBARS) assay kit was used to measure MDA. Plasma samples from five SjS patients and five control subjects were also evaluated. Results: The MDA concentrations in experimental animals and controls were not significantly different. There were no significant differences between the plasma of SjS patients and of controls. Conclusions: The expression of MDA was investigated in the organs from the SjS-prone B6DC mouse for the first time and in the plasma samples of SjS patients. No significant differences were observed between SjS and control samples when MDA was the target molecule with the TBARS assay. MDA may not be a reliable marker to measure OS contrary to the published studies involving OS of SjS.

Effects of Testosterone on White Adipose and Liver Tissues in Male Castrated C57BL/6J Mice

  • Obesity is defined as increased mass of adipose tissue, conferring a higher risk of cardiovascular and metabolic disorders such as diabetes, hyperlipidemia, and coronary heart disease. To get a better understanding of the role of a male sex hormone testosterone on obesity, we thus measured the effects of testosterone on white adipose tissue (WAT) mass, adipocyte histology and hepatic lipid accumulation in male castrated (CAST) C57BL/6J mice. Compared to male CAST control mice, testosterone-treated mice had the decreased WAT mass and the increased the number of adipocytes. Especially, histological data showed that the adipocyte size was reduced in a dose-dependent manner and was most effective at dose 150 $\mu$g per mouse for testosterone. In addition, the administration of testosterone resulted in the inhibition of hepatic lipid accumulation compared with control mice. Our results suggest that testosterone regulates adipocytes development and hepatic lipid metabolism, resulting in the prevention of obesity in male CAST mice.

Induction of Squamous Cell Carcinomas and Adenocarcinomas in Mouse Lung by Intratracheal Instillation of Benzo(a)pyrene and Urethan (Benzo(a)pyrene 및 Urethan의 마우스 기관내(氣管內) 주입(注入)에 의한 편평상피암(扁平上皮癌)과 선암(腺癌)의 발생(發生))

  • 성숙한 A/J, $C_{57}BL/6N$, DBA/2 및 NIH-GP계(系) 마우스에 benzo(a) pyrene과 charcoal powder를, $C_{57}/6N$ 및 NIH-GP계(系) 마우스에 Urethan을 기관내(氣管內) 주입(注入)하여 폐장(肺臟)의 종양발생(腫瘍發生)과 조직변화(組織變化)를 관찰(觀察)한 바 다음과 같은 결과(結果)를 얻었다. Benzo(a)pyrene을 주입(注入)한 군(群)에서는 편평상피암(扁平上皮癌) 및 선암(腺癌), 그리고 편평상피암(扁平上皮癌)과 선암(腺癌)의 혼합 발생예(例)를 볼 수 있었다. 이와 같은 병변(病變)은 A/J 및 $C_{57}BL/6N$계(系) 마우스에서 발생률(發生率)이 높았으며 A/J계(系) 마우스에서 편평상피암(扁平上皮癌)의 발생(發生) 및 분화(分化)가 현저하였다. 한편 DBA/2 및 NIH-GP계(系) 마우스에서는 종양(腫瘍) 발생률(發生率)이 극히 낮았다. Benzo(a)pyrene의 주입시(注入時) 동일 계통의 마우스에서도 대량을 주입(注入)할 때 선암(腺癌)보다 편평상피암(扁平上皮癌)의 발생(發生)이 많았으며 A/J계(系) 마우스에서는 편평상피암(扁平上皮癌) 단독(單獨) 발생예(發生例)가 다수 관찰(觀察)되었고 $C_{57}BL/6N$계(系) 마우스에서는 선암(腺癌) 및 선암(腺癌)과 편평상피암(扁平上皮癌)의 혼합 형태(形態)가 주(主)로 관찰(觀察)되었다. 그리고 기관지상피(氣管支上皮)의 편평상피화생(扁平上皮化生) 및 편평상피암(扁平上皮癌)의 발생시(發生時) alcian blue-PAS 양성반응세포가 관여함을 알 수 있었고 선종양조직(腺腫樣組織) 발생예(發生例)에서도 편평상피화생(扁平上皮化生) 및 편평상피암(扁平上皮癌)으로 분화(分化)되는 경우도 관찰(觀察)되었다. Urethan 주입군(注入群)은 극히 낮은 종양발생률(腫瘍發生率)을 나타내어 발암물질(發癌物質)의 종류(種類), 용량(用量) 및 마우스의 계통(系統)에 따라 종양(腫瘍)의 발생률(發生率) 및 형태(形態)의 차이가 인정되었다.

Anti-obesity Effect of Jeoreongchajeonja-tang in a High-fat Diet-induced Obesity Mouse model (고지방식이로 유도된 비만 생쥐모델에서 저령차전자탕의 항비만 효과)

  • Objective This study investigated the effects of Jeoreongchajeonja-tang in a high-fat diet-induced obesity mouse model. Methods The study examined 9-week-old male mice (C57bl/6J) divided into four groups: the normal(C57bl/6J-Nr), control (high-fat diet only; HFD-CTL), positive-control (high-fat diet with Garcinia cambogia), and experimental (high-fat diet with Jeoreongchajeonja-tang; HFD-JCT) groups. After 7 weeks, the body weight, food efficiency ratio, organ weight, and visceral fat weight of the mice were measured. Blood serum tests, mRNA, liver histopathology, and epididymis adipocytes were also examined. Results Compared with the Control(HFD-CTL) group, the Experimental(HFD-JCT) group given Jeoreongchajeonja-tang showed significant reductions in absolute body weight and food efficiency ratio. The serum alanine aminotransferase, total-cholesterol, triglyceride, low-density lipoprotein (LDL)-cholesterol, high-density lipoprotein (HDL)-cholesterol, insulin-like growth factor-1, and leptin levels were significantly lower in the experimental group than in the control group. The serum adiponectin levels were significantly higher in the experimental group than in the control group. Compared with the control group, the experimental group showed significant reductions in absolute abdominal subcutaneous fat, epididymal adipose tissue, kidney adipose tissue, intestine adipose tissue, and liver, kidney and spleen adipose tissue weights. The C/EBP-${\beta}$, leptin, and SREBP1c/ADD1 mRNA expression were significantly lower in the experimental group than in the control group, while the UCP-2 and adiponectin mRNA expression were significantly higher. Compared with the control group, the experimental group showed a significant reduction in the absolute adipocyte area in the liver and epididymal adipose tissue. Conclusion Jeoreongchajeonja-tang has an anti-obesity effect. Additional clinical studies are expected.

Immune Tolerance in Murine Islet Transplantation Across HY Disparity (HY 항원 불일치 췌도 이식에 의한 면역 관용의 유도)

  • Background: Minor histocompatibility HY antigen, as a transplantation antigen, has been known to cause graft rejection in MHC (major histocompatibility complex) matched donor-recipient. The aim of our study is to investigate the role of male antigen (HY) disparity on MHC matched pancreatic islet transplantation and to examine the mechanism of the immune reaction. Methods: Pancreatic islets were isolated and purified by collagen digestion followed by Ficoll gradient. The isolated islets of male C57BL6/J were transplanted underneath the kidney capsule of syngeneic female mice rendered diabetic with streptozotocine. Blood glucose was monitored for the rejection of engrafted islets. After certain period of time, tail to flank skin transplantation was performed either on mouse transplanted with HY mismatched islets or on sham treated mouse. The rejection was monitored by scoring gross pathology of the engrafted skin. Results: HY mismatched islets survived more than 300 days in 14 out of 15 mice. The acceptance of second party graft (male B6 islets) and the rejection of third party graft (male BALB/c islets) in these mice suggested the tolerance to islets with HY disparity. B6 Skin with HY disparity was rejected on day $25{\pm}7$. However, HY mismatched skin transplanted on the mice tolerated to HY mismatched islets survived more than 240 days. Tetramer staining in these mice indicated the CTL recognizing MHC Db/Uty was not deleted or anergized. Conclusion: The islet transplantation across HY disparity induced tolerance to HY antigen in C57BL6 mouse, which in turn induced tolerance to HY mismatched skin, which otherwise would be rejected within 25 days. The MHC tetramer staining suggested the underlying mechanisms would not be clonal deletion or anergy.

A Vinegar-processed Ginseng Radix (Ginsam) Ameliorates Hyperglycemia and Dyslipidemia in C57BL/KsJ db/db Mice

  • Having idea to develop more effective anti-diabetic agent from ginseng root, we comprehensively assessed the anti-diabetic activity and mechanisms of ginsam in C57BL/KsJ db/db mice. The db/db mice were divided into 4 groups; diabetic control (DC), ginsam at a dose of 300 or 500 mg/kg (GS300 or GS500) and metformin at a dose of 300 mg/kg (MT300). Ginsam was orally administered for 8 weeks. GS500 reduced the blood glucose concentration and significantly decreased an insulin resistance index. In addition, GS500 reduced the plasma non-esterified fatty acid, triglyceride, and increased high density lipoprotein-cholesterol as well as decreased the hepatic cholesterol and triglyceride. More interestingly, ginsam increased the plasma adiponectin level by 17% compared to diabetic control group. Microarray, quantitative-polymerase chain reaction and enzyme activity results showed that gene and protein expressions associated with glycolysis, gluconeogenesis, and fatty acid oxidation were changed to the way of reducing hepatic glucose production, insulin resistance and enhancing fatty acid $\beta$-oxidation. Ginsam also increased the phosphorylation of AMP-activated protein kinase and glucose transporter expressions in the liver and skeletal muscle, respectively. These changes in gene expression were considered to be the mechanism by which the ginsam exerted the anti-diabetic and anti-dyslipidemic activities in C57BL/KsJ db/db mice.
