Ixora auricularis Chun & F. C. How ex W. C. Ko (Rubiaceae): A new record to Flora of Vietnam (Ixora auricularis Chun & F. C. How ex W. C. Ko: 베트남 미기록종)

  • Ixora auricularis (Rubiaceae), previously recognized as endemic to Yunnan (China), was reported in Vietnam. This plant differs from the closely related species Ixora coccinea in its auriculate shape of the base leaf, greater number of secondary veins, shorter stipules, calyx having more appendices, shorter corolla tube, and the oblanceolata shape of the corolla tip. Detailed information about its description as well as illustrations, color photographs and a species key of related species of Ixora in Vietnam are provided.

Excellent Strains Selection from Wild Cordyceps spp.(II) (야생동충하초로부터 우량균주선발(II))

  • Many Cordyceps spp. were collected for excellent strains selection at near Wanju-kun from May to October, 2000. They were identified. As the resulting, They were 2 genera and 4 species. 6 strains were got through 4 species culture. 6 strains are Cordyceps militaris f. albino, C, gracilaides, C. militaris and Isaria japoica. Among them, 4 species used for induction of artifitial fruiting body. 3 strains of them were selected for excellent strains. excellent strains are Cordyceps militaris and Isaria japoica.

Biochemical Characters of Polygalacturonase Produced by Botryosphaeria dothidea (사과 겹무늬썩음병균(Botryosphaeria dothidea)이 생산하는 Polygalacturonase의 생화학적 특성)

  • The polygalacturonase (PG) production in rotten apples by Botryosphaeria dothidea was purified by using gel filtration and ion exchange column chromatography, and the biochemical characters of PG were investigated. The purified PG appeared as a single band on sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) with approximate molecular weight of 49 kilodalton (kDa). The molecular weight was equal to the native molecular weight estimated by gel filtration. The Km and Vmax values of PG were 0.51 mg/ml and 90.9 $\mu$M/min/ml, respectively. Optimum pH was 4.0~5.0, and the PG activity was stable from pH 5.0~10.0. Optimum temperature of the enzyme activity was 4$0^{\circ}C$. The PG activity was relatively stable at 2$0^{\circ}C$, but it was reduced 45% at 4$0^{\circ}C$ and completely inactivated at 8$0^{\circ}C$. The PG activity was considerably inhibited by Cu2+, Zn2+, SDS and EDTA, whereas it was not effected by Ca2+, K+, Mg2+ or Na+ ions.

  • 1976년 7월부터 1979년 5월까지 3년간 진해만의 표층에서 격월로 식물성플랑크톤 군집 변화에 영향을 미치는 환경요인(수온, 염분도, pH, 투명도, 용존산소, Chlorophyll-a,c, NO$_{2}$-N 및 PO$_{4}$-P)에 대하여 조사했다. 9가지 환경요인이 식물성플랑크톤 군집 변화에 37.20% 영향을 미치고 있었으며 각각의 환경요인으로는 PO$_{4}$-P, 투명도, Chlorophyll-a, 수온, 염분도 등이 중요한 요인이었으며 용존산소, NO$_{2}$-N, pH 및 Chlorophyll-c 는 크게 영향을 미치지 않고 있었다.

The Metabolism of (2-$^{14}C$) Mevalonic Acid on Photoperiodic Induction in Grafted Solanum Andigena

  • The metabolism of sterol precurosor in leaves of Salanum andigena grafted between photoinduced and noninduced plant was investigated with the use of (2-$^{14}$ C) mevalonic acid. By the technique of the preparative gas-liquid chromatography, radioactive compounds of squalene, 4,4’-dimethylsterols and 4-demethylsterol were isolated and determined quantitatively. When labeled mevalonic acid n as applied to leaves radioactivity was extensively incorporated into non-saponifiable materials of lipid fraction and aqueous fraction (ethanol-water fraction). Radioactivity of 14C derived from (2-$^{14}$ C) mevalonic acid was transmissible from photoinduced plant to non-induced plant across the graft union, as tuberization hormone was, and incorporated into the sterols of the non-induced plant. Inhibitors of sterol biosynthesis, SK & F 7997 and nicotinic acid, are effective suppressors of tuber growing, if applied to leaves during photoinduction period. The experimental results suggest that certain substance containing isoprene unit, or sterol-like compound may participate in tuber growing.

Microwave-assisted extraction of paclitaxel from plant cell cultures (Microwave를 이용한 식물세포배양으로부터 paclitaxel 추출)

  • A simple and efficient microwave-assisted extraction procedure was developed and optimized for the extraction of paclitaxel from the plant cell cultures of Taxus chinensis. The biomass, immersed in a methanol-water mixture, was irradiated with microwaves in a closed-vessel system. The microwave-assisted extraction was compared with the existing conventional solvent extraction in terms of yield, extraction time, and solvent consumption. The use of microwave energy allows rapid recovery of paclitaxel from biomass and dramatically reduces extraction time and solvent usage compared to conventional solvent extraction. The paclitaxel was completely extracted from biomass by microwave-assisted extraction for 3 min at $50^{\circ}C$, for 6 min at $30^{\circ}C$ and $40^{\circ}C$, respectively.

  • We analyze relation between phytoplankton and marine environment based on data such as water temperature, phytoplankton, zooplankton, nutrient collected from the southern coast of Korea in the summer, 2004. The water temperature range of the study area was $20.5{\sim}31.5^{\circ}C$ and there was formed a water temperature frontal zone from $20.5^{\circ}C$ to $25.0^{\circ}C$ in Geojedo southern coast and Geomundo island. Especially, high density of nutrients were shown in the southern coast of Geojedo in which water temperature frontal zone was formed strongly, the concentration of chlorophyll-a which is appeared at the highest rate among the phytoplankton pigments was shown more than $0.4{\mu}g/L$ in the inside of frontal zone and zooplankton biomass was than $500mg/m^2$ in that area.

Vascular Plants Distributed in Baekdudaegan Mountains (Gitdaebaegibong~Mt. Cheonghwasan) (백두대간(깃대배기봉~청화산)에 분포하는 관속식물상)

  • The purpose of this study is to provide the baseline data for conservation and management of Korean forest ecosystem by surveying and analyzing the vascular plants distributed from Gitdaebaegibong to Cheonghwasan in Baekdudaegan Mountains. The results are as follows. The numbers of vascular plants in the whole survey section were summarized as 771 taxa including 103 families, 379 genera, 623 species, 4 subspecies, 121 varieties and 23 forms. There were 377 taxa in A-section, 395 taxa in B-section, 278 taxa in C-section, 325 taxa in D-section, 534 taxa in E-section, and 406 taxa in F-section. The rare plants were 32 taxa including Megaleranthis saniculifolia, Rodgersia podophylla, Iris ensata var. spontanea, and Gastrodia elata. In IUCN Red List categories, there were 1 taxon of CR, EN, and DD each, 11 taxa of the VU, and 18 taxa of the LC. The Korean endemic plants were 26 taxa including Asarum versicolor, Clematis fusca var. coreana, Vicia chosenensis, Stewartia pseudocamellia, Carex okamotoi, and Luzula sudetica var. nipponica. The specific plants by floristic region were 143 taxa including 3 taxa of grade V, 12 taxa of grade IV, 41 taxa of grade III, 42 taxa of grade II, and 45 taxa of grade I. The naturalized plants were 41 taxa including Rumex crispus, Ailanthus altissima, Erechtites hieracifolia, Erigeron annuus, and Poa pratensis. The invasive alien plants were 4 taxa including Rumex acetocella, Sicyos angulatus, Ambrosia artemisiifolia, and Aster pilosus. The plants adaptable to climate change were 43 taxa including 14 taxa of endemic plants, 2 taxa of southern plants, and 27 taxa of northern plants.

Growth Acceleration and Acclimatization of In Vitro Plantlets derived from Apical Meristem of Sweet Potato (고구마의 경정조직 유래 기내 소식물체의 생장촉진과 순화)

  • The single node cuttings of sweet potato (cv. Mokpo #29) plantlets maintained in vitro were cultured with (MF+) or without membrane filter (MF-) under photomixotrophic (PM), hetrotrophic (HT) and autotrophic (AT) conditions. Shoot length was the greatest (11.9cm) in 3$0^{\circ}C$ (HT) treatment and it was the shortest (3.4 cm) in $25^{\circ}C$ (PM) treatment. Nodal explants cultured in 3$0^{\circ}C$ treatment looked more vigorous than those of $25^{\circ}C$ in appearance, and node number was the greatest (10.5 per plantlet) among the treatments. But plantlets grew in 3$0^{\circ}C$ (HT) treatment were observed all overgrown. The size in leaf area was about 2 times greater and shoot length was about 2 times shorter in PM than in HT condition. Percent dry matter of shoots was 5.9% (HT) and 7.4% (PM) in $25^{\circ}C$ treatment and 6.1% (HT) and 7.4% (PM) in 3$0^{\circ}C$ treatment. Plantlets cultured in the MF+ treatments were less succulent than those cultured in the MF- treatment. Vitrified plantlets were examinated 14.8% (both $25^{\circ}C$ and 3$0^{\circ}C$) in PM condition and 22.2% ($25^{\circ}C$) and 31.5% (3$0^{\circ}C$) in HT condition. Sucrose was necessary for the survival of in vitro plantlets. In the sucrose-free medium, explants cultured in the MF- had turned yellow and were dead after 30 days of culture. But explants cultured in the MF+ were alive and produced plantlets with shoot and root (AT). On the other hand, the survival of explants on the MS basal medium (sucrose-free and hormone-free) depended entirely upon the MF attachment.

Cytological Analysis of Microspores during Temperature Pretreatment in Anther Culture of Capcicum annuum L. (고추의 약배양 시 온도 전처리에 따른 소포자의 세포학적 변화 분석)

  • Inoculated anthers of Capsicum annuum L. were subjected to 4 and 32$^{\circ}C$ pretreatment and their influence on the microspore viability, early cytological changes and the induction frequency of microspore embryo was investigated. Viability of freshly isolated microspores was between 62 and 64%. During temperature pretreatment, microspore viability showed a rapid decrease and this tendency enhanced with the 32$^{\circ}C$ pretreatment. Irrespective of temperature pretreatment, microspore viability declined to nearly zero after nine days. Before temperature pretreatment, most of the microspores in anthers were at late uninucleate stage. Several types of multinuclear microspores appeared from the 2 day after culture onwards, together with many degenerated and non-induced microspores. The 32$^{\circ}C$ pretreatment gave higher proportions of embryogenic microspore than other treatment. However, the temperature pretreatment had no clear effect on the frequencies of symmetrical binucleate rnicrospore. The multinucleate grains might originate either by symmetrical or asymmetrical division. After 2 days of pretreatment at 25 and 32$^{\circ}C$ , degenerated microspore increased above 50%. In contrast, during 4$^{\circ}C$ treatment, nucleus of most microspores remained intact for 14 days. The 32$^{\circ}C$ pretreatment produced more embryos than 4$^{\circ}C$ treatment. The most effective period of 32$^{\circ}C$ pretreatment was 4 days. In contrast, effective period of 4$^{\circ}C$ pretreatment was 2 days and longer time had deleterious effect on induction of microspore embryo.
